산불 진화용 소화약제의 산림생태환경 영향 평가 - 식물 및 토양생물독성에 대하여 - (An Forest Ecological Environment Impact Assessment of Forest Fire Suppression Chemicals - To Plants & Soil Organism -)

  • 전 세계적으로 산불이 빈번히 발생되고 대형화산불이 끊임없이 발생되고 있다. 따라서 산불진화를 보다 효과적으로 신속히 하기위해 산불진화용 소화약제가 사용되고 있다. 산불용 소화약제는 일반적으로 물리적 형태에 따라 분말, 액상, 포(泡), 겔(gel)로 구분되어 사용되고 있다. 우리나라에서는 직접적인 산불진화용 소화약제인 강화액과 포소화약제를 사용하고 있어 본 연구에서는 이러한 소화약제들의 식물독성(종자발아율, 소나무 용기묘 치사율) 및 토양생물 독성(지렁이 급성독성) 등 산림생태에 대한 독성평가를 실시하였다. 그 결과, 강화액 및 포소화약제 모두 영향을 미치는 것으로 나타났으며 특히, 강화액의 경우 모든 독성실험방법에 대해 실험개체 모두 100%의 치사율을 나타내어 독성이 가장 강한 것으로 조사되었다. 따라서 향후, 산불진화용 소화약제에 대한 산림생태환경독성 영향평가 방법과 기준제시가 필요할 것으로 판단된다.

물벼룩(Daphnia magna)에 대한 중금속의 급성 및 만성 독성 (Acute and Chronic Toxicity of heavy Metals to Daphnia magna)

  • The toxicity values heavy metals were evaluated by immobilization and chronic reproduction impairment tests, using Daphnia magna. Acute tests were evaluated by the inhibition of their mobilization after 24hrs without food addition. The tests of reproductive impairment were investigated for 21 days by food addition and exchange or water. The effect of each concentration was assessed by Probit analysis and t-test. The results obtained from this study were as follows : 1) The change of pH and DO was not significant in the acute tests, while, in the reproductive tests, pH was increased by 0.3~1.4 and DO also increased. 2) The $E_iC_{50}$ values of immobilization to Daphnia magna in artificial fresh water were $0.030mg/{\ell}(Cu),\;0.054mg/{\ell}(Cd),\;0.12mg/{\ell};(Cr),\;0.74mg/{\ell}(Pb),\;3.4mg/ {\ell}(As)$ and the $NOE_iC$ values were $0.010mg/{\ell}(Cu),\;0.018mg/{\ell}(Cd),\;0.010mg/{\ell}(Cr),\;0.10mg/{\ell}(Pb),\;and\;$1.8mg/{\ell}(As)$. 3) The $E_rC_{50}$ values of reproductive impairment to Daphnia magna were $13.8\mu\textrm{g}/{\ell}(Cu),\;2.9\mu\textrm{g}/{\ell}(Cd),\;15.5\mu\textrm{g}/{\ell}(Cr),\;61.7\mu\textrm{g}/{\ell}(Pb),\;759\mu\textrm{g}/{\ell}(As)$, and $NOE_rC$ values were $0.95\mu\textrm{g}/{\ell}(Cu),\;$0.54\mu\textrm{g}/{\ell}(Cd),\;1.2\mu\textrm{g}/{\ell}(Cd),\;$7.4\mu\textrm{g}/{\ell}(Pb),\;110mu\textrm{g}/{\ell}(As)$. The results of tests using OECD artificial culture water were more sensitive than natural water for culturing. The presented data show that an artificial culture water is suitable in the experiment of bioassay for assessing the toxicity of marterials.

  • Compare to the toxicity tests using experimental animals, the GLP application and compliance in toxicity studies using cell culture systems may be less straightforward elucidated in the two documents published by the OECD Working Croup on GLP 'The Application of the GLP Principles to Short Term Studies (1999)' and 'The Application of the Principles of GLP to in vitro Studies (2004)' The object of this presentation is to show how to interpret the GLP principles and to apply with actual performances in a well known toxicity test using cell culture, chromosome aberration study. The presentation will cover test substance, test system (cell line), study environment management, documentation, quality assurance, and study protocol and report.

  • The ecotoxicity assessment of nanomaterials (NMs) in the environment is actively conducted throughout the world because of the concerns about their potential risk from usage and release into the environment, as well as their unique physiochemical properties. Ecotoxicity tests for NMs have been conducted using various species and methods; however, in spite of these efforts, the characteristics and toxicity of NMs have not been defined. The fish embryo toxicity test (FET) has been conducted extensively to evaluate the toxicity of NMs as an alternative to a whole-body test in fish. In this study, we collected and analyzed the trends of nanotoxicity on the early development of freshwater fish. The model nanomaterials are carbon NMs ($C_{60},\;C_{70},\;C_{60}$(OH)n and carbon nanotube). Their adverse effects were extensively investigated based on the properties of NMs, test species, and diverse exposure conditions.

A NEWLY DEVELOPED CONTINUOUS TOXICITY TEST SYSTEM USING A LUMINOUSLY MODIFIED TERRESTRIAL BACTERIUM

  • Freshwater borne bacteria transformed with luxAB-containing plasmid were optimized for the toxicity tests of various organic carbons and heavy metals. The EC$\sub$50/ values obtained from tests using the most sensitive bacterium to toxicants, YH9-RC, revealed to be much less than those from the Microtox$\^$/. In addition, some physiological characteristics of this bacterium under the toxic stress conditions such as potential bioluminescence, specific growth rate, and intracellular ATP contents, reproducibly and reliably correlated to the toxicity of the chemicals exposed. The higher concentrations of COD in wastewater samples, the lower EC$\sub$50/ values, therefore the developed toxicity test was found to be easily applicable to the toxicity test for wastewater samples and effluents. The conditions for constructing 384-multiwell plate containing freeze-dried bacterium were also optimized through the addition of 0.16 M trehalose before freeze-drying. Consequently, the advanced test system featuring a continuous measurement of the toxicity, an automated real-time monitoring of its results, and an alerting function was designed and constructed in combination with the microbiological, mechanical, and electronic compartment.

Viable Alternatives to in vivo Tests for Evaluating the Toxicity of Engineered Carbon Nanotubes

  • Carbon nanotubes (CNTs) stand at the frontier of nanotechnology and are destined to stimulate the next industrial revolution. Rapid increase in their production and use in the technology industry have led to concerns over the effects of CNT on human health and the environment. The prominent use of CNTs in biomedical applications also increases the possibility of human exposure, while properties such as their high aspect ratio (fiber-like shape) and large surface area raise safety concerns for human health if exposure does occur. It is crucial to develop viable alternatives to in vivo tests in order to evaluate the toxicity of engineered CNTs and develop validated experimental models capable of identifying CNTs' toxic effects and predicting their level of toxicity in the human respiratory system. Human lung epithelial cells serve as a barrier at the interface between the surrounding air and lung tissues in response to exogenous particles such as air-pollutants, including CNTs. Monolayer culture of the key individual cell types has provided abundant fundamental information on the response of these cells to external perturbations. However, such systems are limited by the absence of cell-cell interactions and their dynamic nature, which are both present in vivo. In this review, we suggested two viable alternatives to in vivo tests to evaluate the health risk of human exposure to CNTs.

알루미늄 가공 공장 배출 방류수의 독성 원인물질 탐색 (Application of Toxicity Identification Evaluation Procedures for Toxic Effluents from the Aluminum Rolling Industry)

  • Objectives: The objective of this study is to identify toxicants causing acute toxicity in effluents from the aluminum rolling industry that violate the discharge limits in Korea. Methods: Whole effluent toxicity tests (WET) were conducted on effluent discharged from the aluminum rolling industry following the US EPA WET test methods. We collected effluent samples three times and evaluated acute toxicity by using Daphnia magna. We employed toxicity identification evaluation (TIE) procedures to identify toxicants causing toxicity in the effluent. Results: No specific chemical groups were identified in the seven different manipulations applied to the of wastewater effluent samples showing 1.3 toxic units (TU) according to the TIE phase I procedures. Water quality parameters for water hardness, electric conductivity and heavy metals (Mn) were 4,322 mg/l as $CaCO_3$, 11.39 mS/cm, and $5,551{\mu}g/l$, respectively. Considering water hardness and reference toxicity, high concentrations of Mn can be disqualified from the causative toxicants. Consequently, high ionic concentrations of $Na^+$(1,648 mg/l), $Ca^{2+}$(1,048 mg/l), $Mg^{2+}$(1,428 mg/l) and $SO_4{^{2-}}$(7,472 mg/l) were identified to be causative toxicants. Water hardness and electric conductivity exceed the $EC_{50}$ value obtained by biological toxicity tests using Daphnia magna. Conclusion: According to TIE procedures, high salt concentration is determined to be a major toxicant in the effluent of agro-industrial wastewater treatment plants receiving wastewater from the aluminum rolling industry.

Heterorhabditis megidis Gwangju Strain (Nematoda: Heterorhabditidae)으로부터 분리한 Photorhabdus temperata의 어류 및 쥐 독성평가 (Toxicity Assessment of Photorhabdus temperata Isolated from Heterorhabditis megidis Gwangju Strain (Nematoda: Heterorhabditidae) in Fish and Rat)

  • Photorhabdus is a bacterial symbiont of entomopathogenic nematodes of the genus Heterorhabditis in the family Heterorhabditidae. Photorhabdus is known to have nematicidal activity in addition to insecticidal activity. P. temperata isolated from Korean indigenous H. megidis Gwangju strain also produced high control efficacy against root-knot nematode Meloidogyne incognita and root-lesion nematode Pratylenchus penetrans. P. temperata has drawn interest as a potential bionematicide for the control of root-knot nematodes thereby. For the registration as an organic agricultural material, the toxicity of P. temperata was assessed by the acute toxicity test in carp (Cyprinus carpio) and acute oral and dermal toxicity tests in Sprague-Dawley rat (Rattus norvegicus) in compliance with the guidelines of the Rural Development Administration (RDA). In the acute toxicity test in fish, neither lethality nor abnormal responses of carp were observed. Body length and weight of carp and changes in DO concentrations and pH values were not significantly different between the treated group and the untreated control. In the acute oral and dermal toxicity tests, clinical signs, abnormal behavior, mortality, and pathological findings were not observed in all the experimental rats. The weight increment of all rats was normal. Acute toxicity results of P. temperata in fish and rats belonged to categories III, IV, and IV of RDA, respectively. Toxicity results of the present study indicated that P. temperata could be a safe and promising bionematicide against root-knot nematodes and root lesion nematode.

  • Toxicity tests in our laboratory are conducted usually with mass-reared organisms. They are under the same environmentel conditions throughout seasons and are supplied at specific age. A total of 38 species of aquatic organisms are being reared. We have attempted to establish pruified strains or to select clones of various parthenogenic organisms. The merits or demerits of our culturing test organisms are discussed. The differences in the susceptibility among clones or strains of test organism are also discussed. For a single species test, algae, daphnia, fish are often used. However, we usually use early stages, but occasionally, adults fish are used for reproduction tests. As an another important aspect, the toxicity through food chains has been studied. In this study, we select a pair of species belonging different trophic levels. The differences between single species tests and multispecies tests will be discussed. Even a single species test intends to assess the effects of chemicals on ecosystem levels, however, this idea is not applicable to ecosystems. Single species tests with standard organisms and multispecies tests are contradictory in concept. One type of multispecles tests is indoor microcosms being composed of severel species artificially assembled, and another is composed of natural components (both indoor and outdoor). We have used three types of outdoor mesocosms using ponds and three types of artificial streams. The mesocosms is useful to not only to analyze the floral or faun61 changes but also to study the fate or behaviour of chemicals in naturd environments. Lastly, usefulness of the field observation or experiments or semi-field experiments will be discussed. This will enhance the exploitation of early warning systems utilizing indicator organisms or animal behaviour.aviour.

수은, 납 및 구리에 대한 붕어(Carassius auratus)와 돌고기(Pungtungia herzi) 자어의 급성독성 (Acute Toxicity of Carassius auratus and Pungtungia herzi Lavra on Mercury, Lead and Copper Exposure)

  • Larva of Carassius auratus and Pungtungia herzi were exposed to mercury ($HgCl_2$), lead ($PbCl_2$) and copper ($CuSO_4$) to determine acute toxicity. The toxicity tests were conducted triplicate and the $LC_{50}$ values (24, 48, 72 and 96 hours) were determined for two species. Data obtained from the toxicity tests were evaluated using the probit analysis. Although sensitivities of two species to mercury were relatively similar, C. auratus was more susceptible than P. herzi to the exposure of lead but P. herzi was more sensitive than C. auratus on copper. The sensitivity on three metals tested may be ranked in the following order from highest to lowest toxicity on larval stage of these fish: mercury > copper > lead. It is suggested that acute toxic test at the larval stage of C. auratus and P. herzi, indigenous species in Asia area, is an important part of the ecotoxicological assessment of some heavy metals.
