Evaluating the Micronucleus Induction Potential for the Genotoxicity Assay Using the Human Skin Model, KeraSkinTM (인공피부모델 KeraSkinTM을 이용한 유전독성 평가)

  • Micronucleus test is genotoxicity assay for detection of micronuclei in the cytoplasm of interphase cells. The reduction and replacement of in vivo toxicity testing on animals require the development of in vitro models to predict the genotoxicity or other tests for cosmetic products. In this study, we evaluated a genotoxicity assay for topically applied chemicals using a three-dimensional human reconstructed skin model, KeraSkin$^{TM}$. Two genotoxins, mitomycin C (MMC) and methyl methanesulfonate (MMS), induced significant dose-related increases in cytotoxicity and micronuclei induction in the skin model. In contrast, two non-genotoxins, 4-nitrophenol (4-NP) and trichloroethylene (TCE), induced cytotoxicity but not micronucleus formation. In conclusion, micronucleus test using human skin model may be useful for predicting in vitro genotoxic potentials of cosmetic products.

Antioxidant and Anti-Inflammatory Effects of Kamisipjeondaebotang in RAW 264.7 Cells (가미십전대보탕의 RAW 264.7 세포에서 항산화 및 항염증 효과)

  • As Kamisipjeondaebotang (KSD) extract is an herbal ingredient, safety is very important due to possible cell poisoning or heavy metal toxicity to organs when administered to humans or animals. Accordingly, this study examined the antioxidant and anti-inflammatory effects of KSD extract to confirm its medicinal safety by using RAW 264.7 cells after heavy metal screening, functional index test of the liver and kidney, and cell survival rate test. Heavy metals were not found in KSD extracts or were less than standard amounts. Liver function indices such as aspartate aminotransferase and alanine aminotransferase revealed low values and kidney function indices such as creatinine and blood urea nitrogen were not significantly different from the normal group. This proved the safety to the human. RAW 264.7 cells showed no poisoning compared to the control group in terms of survival rate. Regarding the antioxidant effect of KSD extract, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and 2,2'-azino-bis(3-ethylbenzothiazo-line-6-sulphonic acid) radical scavenging activity increased at concentrations over $10{\mu}g/mL$. The anti-inflammatory effect of KSD extract significantly decreased based on the amount of nitric oxide at concentrations of 10 and $100{\mu}g/mL$ compared to the control group. Expression of interleukin (IL)-$1{\beta}$ and IL-6 decreased in a concentration-dependent manner. There was no significant difference in tumor necrosis factor-${\alpha}$ level. Based on the results, KSD can be regarded as a safe antioxidant with anti-inflammatory effects for fracture treatment.

  • $\beta$-glucan is a fiber-type complex sugar (polysaccharide) derived from the cell wall of baker's yeast, oat and barley fiber, and many medicinal mushrooms, such as maitake. The primary uses of $\beta$-glucan are to enhance the immune system, to lower blood cholesterol levels and to treat tumor. $\beta$-glucan has no systemic toxicity in mice, therefore it needed clinical trail to prove efficacy and safety for human. The subjects total 56 healty volunteers were divided into two groups including taken $\beta$-glucan tablet group and placebo group. Subjects were taken two tablets per oral for 4 weeks. They had agreed to take part in this experiment, and didn't take any other clinical trail products. After 4 weeks blood of subjects were checked. The check list are TNF-$\alpha$, INF-$\gamma$, IL-2, IL-4, total WBC, differential WBC, RBC, hemoglobin, platelet, MCV, MCH, MCHC, HCT, Na, K, Ca, Cl, AST, ALT, ALP, $\gamma$-GTP, total protein, triglyceride, total cholesterol, total bilirubin, albumin, uric acid, creatinine, BUN, pH, protein, glucose, ketone body, blood, bilirubin. We evaluated efficacy by cytokines that compare before and after taking. Collected data were analyzed as two sample t-test, chi-square test and ANOVA using SAS V.9.1.This study results are that in TNF-$\alpha$ of $1^{st}$ efficacy measurement item, all of two groups figure were increased significantly compare to before figure. In IL4 of $2^{nd}$ efficacy measurement item, experimental group figure were decreased significantly but placebo group figure were increased. The conclusions show that based on the above results, $\beta$-glucan has favorable effect to enhance immune system, especially IL4 results showed that it has effect to improve the allergic immune system.

Studies on a Toxin/Antitoxin System in Streptococcus iniae (어류병원균 Streptococcus iniae의 toxin/antitoxin system에 대한 연구)

  • Streptococcus iniae is a typical fish pathogen causing streptococcosis and it can also cause zoonotic infectious diseases. We studied S. iniae FP5228 isolated from infected olive flounder in Wando, Korea. In a study to find virulence factors in FP5228, we found that the number of live bacteria decreased dramatically in culture medium containing S. iniae FP5228 for more than 24 hr. This phenomenon was hypothesized to be related to Toxin ${\zeta}$ and Antitoxin ${\varepsilon}$ genes, components of the Toxin/ Antitoxin (TA) system on the 14 kb plasmid of FP5228. We used a protein overexpression system to identify it. The pBP1140 vector system was constructed to regulate the expression of Toxin ${\zeta}$ and Antitoxin ${\varepsilon}$ by IPTG and Arabinose. E. coli/pBP1140 strain grew slowly in early growth under toxin expression condition, and it was confirmed by microscopic observation that the strain became longer. S. iniae CK287, lacking a 14 kb plasmid of S. iniae FP5228 strain, was constructed. CK287 bacterial cells did not show rapid killing during culture, and the ability to produce biofilm was also decreased, and toxicity was weakened in cytotoxicity test and fish test. These results suggest that the TA system is involved in physiological regulation and expression of virulence factors in S. iniae FP5228.

  • CO I gene sequence analysis was applied to earthworms that had been used as test animals in toxicity test in Institute of Kyeongbook Agrochemicals and earthworms used as vermicomposting agents in the farm of Youngdong province to identify their species names. In terms of molecular species, the former was identified as Eisenia fetida and the latter was Eisenia andrei. Cocoons produced from Eisenia fetida was more than those from Eisenia andrei. And No. of adults developed from eggs of Eisenia fetida was more or less higher than those developed from eggs of Eisenia andrei. These results were contradictory to previous reports on two Eisenia spp.. When Eisenia fetida was crossed with Eisenia andrei, hybridized eggs were produced and adults were developed from those eggs, but cocoons and adults were much less than those from non-crossed Eisenia fetida or Eisenia andrei. This indicated that two Eisenia spp. were not distinctly different biological species because there was no complete 'reproductive isolation' between Eisenia fetida and Eisenia andrei. However, this also meant that Eisenia fetida and Eisenia andrei had already been on the tract of speciation.

A Study of Environmental Conditions of Survival Rate and Relative Growth Rate in Female Gametophyte of Undaria pinnatifida for Toxicity Assessment (생태독성평가를 위한 미역(Undaria pinnatifida) 암배우체 생존율 및 상대성장률의 환경조건 연구)

  • The ecotoxicity test method using Undaria pinnatifida spore is challenging to use throughout the year. Since U. pinnatifida female gametophytes can be cultured in the laboratory, they can be used for ecotoxicity testing at any time. Changes in female gametophyte survival rate and relative growth rate in U. pinnatifida exposed to various environmental conditions were analyzed. The female gametophyte of U. pinnatifida was exposed to salinity (5~40 psu), temperature (5~30℃), pH (4~10), and light intensity (0~120 μmol photon m-2 s-1). Based on the highest average value, the survival rate of female gametophyte was highest at a temperature of 20℃, salinity 27.5 psu, pH 8, and light intensity 30 μmol photon m-2 s-1. And the relative growth rate was highest at a temperature of 15℃, salinity 35 psu, pH 9, and light intensity of 60 μmol photon m-2 s-1. As a result of this study, the method using the optimal conditions for the survival rate and relative growth rate is expected to be a practical test method that can complement the current method.

  • We developed insect-resistant GM rice(Bt transgenic rice) by inserting the mCry1Ac1 a modified gene from the soil bacterium, Bacillus thuringiensis. The Bt transgenic rice expressing the Bttoxin mCry1Ac1 was tested for the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. M. anguillicaudatus and C. carpio fed 100% ground rice in suspension, using either Bt rice or non-GM counterpart rice(Nakdong). The Bt rice used for the test were confirmed to have the mCry1Ac1 gene expression by the immuno-strip and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between Bt rice and non-GM counterpart rice. The 96hr-LC50 values showed no difference between Bt rice(>1,000mg/L) and non-GM rice(>1,000mg/L). We concluded that there was no significant difference in toxicity for non-target organisms(M. anguillicaudatus and C. carpio) between Bt rice and non-GM counterparts.

Analysis of Applicability of Rapid Hardening Composite Mat to Railway Sites (초속경 복합매트의 철도현장 적용성 분석)

  • The Rapid Hardening Composite Mat (RHCM) is a product that improves the initial strength development speed of conventional Geosynthetic Cementitious Composite Mats (GCCM). It offers the advantage of quickly securing sufficient strength in railway slopes with insufficient formation level, and provides benefits such as preventing slope erosion and inhibiting vegetation growth. In this study, an analysis of the practical applicability of RHCM in railway settings was conducted through experimentation. The on-site applicability was assessed by categorizing it into fire resistance, durability, and stability, and conducting combustibility test, ground contact pressure test, and daily displacement analyses. In the case of South Korea, where a significant portion of the territory is composed of forested areas, the prevention of slope fires is imperative. To analyze the fire resistance of RHCM, combustibility tests were conducted as an essential measure. Durability was assessed through ground contact pressure tests to analyze the deformation and potential damage of RHCM caused by the inevitable use of small to medium-sized equipment on the construction surface. Furthermore, daily displacement analysis was conducted to evaluate the structural stability by comparing and analyzing the displacement and behavior occurring during the application of RHCM with railway slope maintenance criteria. As a result of the experiments, the RHCM was analyzed to meet the criteria for heat release rate and gas toxicity. Furthermore, the ground contact pressure was observed to be consistently above 50 kPa during the curing period of 4 to 24 hours under all conditions. Additionally, the daily displacement analyzed through field site experiments ranged from -1.7 mm to 1.01 mm, confirming compliance with the criteria.

Evaluation of applicability of xanthan gum as eco-friendly additive for EPB shield TBM soil conditioning (친환경 첨가제로서 잔탄검의 토압식 쉴드 TBM 쏘일 컨디셔닝 적용성 평가)

  • The Earth Pressure Balance (EPB) shield Tunnel Boring Machine (TBM) is widely used for underground tunnel construction for its advantages, such as eliminating the need for additional facilities compared to the slurry shield TBM, which requires Slurry Treatment Plant (STP). During EPB shield TBM excavation, a soil conditioning technique is employed to enhance the physical properties of the excavated soil by injecting additives, thus broadening the range of applicable ground conditions to EPB shield TBMs. This study explored the use of xanthan gum, a type of biopolymer, as an alternative to the commonly used polymer additive. Biopolymers, derived from biological sources, are fully biodegradable. In contrast to traditional polymers such as polyacrylic acid, which contain environmentally harmful components, xanthan gum is gaining attention as an eco-friendly material due to its minimal toxicity and environmental impact. Test conditions with similar workability were established through slump tests, and the rheological characteristics were assessed using a laboratory pressurized vane shear test apparatus. The experiments demonstrated that, despite exhibiting similar workability, the peak strength in the flow curve decreased with increasing the content of xanthan gum. Consequently, a correlation between the xanthan gum content and peak strength was established. Replacing the traditional polymers with xanthan gum could enable stable EPB shield TBM operation by reducing equipment load, in addition to offering environmental benefits.

Cytotoxicity and Genotoxicity of Newly Developed Calcium Phosphate-based Root Canal Sealers (신개발 인산칼슘계 근관 봉함재의 세포독성 및 유전독성에 관한 연구)

  • The purpose of this study was to compare the cytotoxicity by MTT test and genotoxicity by Ames test of new calcium phosphate-based root canal sealers (CAPSEAL I, CAPSEAL II) with commercially available resin-based sealers (AH 26, AH Plus) , zinc oxide eugenol-based sealers (Tubliseal EWT, Pulp Canal Sealer EWT), calcium hydroxide-based sealer (Sealapex), and tricalcium phosphate based sealers (Sankin Apatite Root Canal Sealer I, II, III). According to this study, the results were as follows : 1. The extracts of freshly mixed group showed higher toxicity than those of 24 h set group in MTT assay (p<0.001). 2. CAPSEAL I and CAPSEAL II were less cytotoxic than AH 26, AH Plus, Tubliseal EWT, Pulp Canal Sealer EWT Sealapex and SARCS II in freshly mixed group (p<0.01). 3. AH 26 in freshly mixed group showed mutagenicity to TA98 and TA100 with and without S9 mix and AH Plus extracts also were mutagenic to TA100 with and without S9 mix. 4. Tubliseal EWT, Pulp Canal Sealer EWT and Sealapex in freshly mixed group were mutagenic to TA100 with S9 mix. 5. Among those of 24 h set groups the extracts of SARCS II were mutagenic to TA98 with and without S9 mix and AH 26 showed mutagenic effects to TA98 with S9 mix. 6. No mutagenic effect of CAPSEAL I and CAPSEAL II was detected. 7. There is no statistically significant difference between CAPSEAL I and CAPSEAL II at MTT assay and Ames test in both freshly mixed group and 24 h set group.
