The Change of Major Cellular Fatty Acids Composition and Morphology of Escherichia coli Affected by Toxic Substances

  • This study was performed to compare the growth rates, cellular fatty acid compositions and morphology by using electron microscope of Escherichia coli (E. coli) grown in various conditions including different concentrations of phenol, CdCl₂ and HgCl₂. Ninety eight E. coli strains were isolated from Naktong river and human feces. The content of unsaturated fatty acids, especially 16:1ω7c and 18:1ωc increased as the concentration of phenol and CdCl₂ increased. The content of unsaturated fatty acid increased up to 50 ppb of HgCl₂, but decreased at 75 ppb of HgCl₂. There were more unsaturated fatty acids than saturated fatty acid in the presence of toxic substances. However, the ration was reversed when the affected E. coli was transferred to toxic substance free fresh trypticase soy broth medium. Also, by using transmission electron microscope these cells were observed to various morphological deformation by heavy metals and their deposition on the surface. From these results, we suggested that the changes of major fatty acids composition and morphology of E. coli may be considered to indicate contaminated levels of heavy metals or organic solvents. The information presented here may be useful in predicting effects of heavy-metal and organic solvent contamination in streams and provides a basis for further studies of metal or organic solvent effects on microbial communities.

Bioassays On marine Organisms: Acute Toxicity Test Of mercury, Cadmium And Copper To Arkshell, Anadara broughtonii, From Jin-Dong Bay, And To Oyster, Crassostrea gigas, From Kwang-Do Bay, South Coast Of Korea

  • Short-term acute toxicity of mercury, cadmium and copper to arkshell, Anadara broughtonii, and to oyster, Crassostrea gigas, was determinedby static bioassays from 20 May to 27 June in 1977. During the observations of the opening rate of the shell mercury was the most sensitive toxicant of the three toxic substances to the test animals and caused them to close their shellvalves together after being exposed to a mercury solution for an hour during the test. Opening rate to cadmium and copper increased gradually at the higher concentration. the 96hr-LC50 values for the test animals are 4.84mg/l for mercury and 1.86mg/l for cadmium, while the 72hr-LC50 value for copper is 0.31mg/l. the death rate of oysters for cadmium showed lower than that of the mercury and copper test solutions. The 96hr-LC50 values of mercury, copper and cadmium were 1.1mg/l, 2.54mg/l and 19.5mg/l, respectively. For oysters mercury was the most toxic substance, and cadmium was the least toxic one. the medium lethal time (LT 50) value decreased gradually at higher concentration of heavy metals. The LT 50 of 2mg/l was found within 96 hours ofr copper, 104 hours for mercury and 121 hours for cadmium. The lethal threshold concentrations for 7 days were found to be about 1mg/l for mercury and copper, and 2mg/l for cadmium.

  • Drug intoxication in children and adolescents is not uncommon in Korea. But the tendency of intoxication is changing with some factors, such as national surveillance system, Naderism and increasing concern among physicians. But the death rate of intoxication among adolescents is increasing in spite of decreasing total death rate of intoxication among children and adolescents. Therefore the physician must be concerned about the basic management of intoxication and figure out the common toxic substance among children and adolescents.

  • 여천지역은 화학물질 누출사고 등 안전 사고 발생의 잠재적 위험이 높으나 (김영성 등, 1999), 복잡한 해안선의 여수 반도 해안지역에 산을 등지고 있어 해륙풍에 의한 일 변화와 지형 조건에 따른 국지 변화등 복잡한 형태의 바람장이 예상되는 지역이다. 이러한 지역에서의 확산현상은 환경영향평가에서 흔히 이용되고 있는 균일한 바람장 분포를 가정한 대기화산 예측과는 다른 결과를 보이며 (오현선, 김영성, 1999), 따라서 단순한 가우시안 플룸 모델을 이용한 확산 예측은 적용범위가 매우 제한적일 수밖에 없다.(중략)

  • There is considerable interest in how microbiological processes can affect the behaviour of metal contaminants in natural and engineered environments and their potential for bioremediation. The extent to which microorganisms can affect metal contaminants is dependent on the identity and chemical form of the metal and the physical and chemical nature of the contaminated site or substance. In general terms, microbial processes which solubilize metals increase their bioavailability and potential toxicity, whereas those that immobilize them reduce bioavailability. The balance between mobilization and immobilization varies depending on the metal, the organisms, their environment and physico-chemical conditions.

  • Streptomyces속에서 분이한 물질의 생물학적 활성을 검색하던 중 어유에 대한 독성물질을 강하게 생성하는 균 1수를 선별하여 그 생화학적 성질을 검토하였다. 1. 분이된 물질의 독성은, 예로서 Pseudorasbora parva T. et. S. (체중 0.~0.5g, 신장 3.0~4.0cm)에 대한 살어역은 5ug/ml에서 30분, 25ug/ml에서 50분이었으며 Carassius carassius L. (체중 2.0g, 유장 5.0cm)온 50ug/m1용액에서 70분간 생존할 수 있었다. 2. 본 물질은 산성 및 유기용매에서는 비교적 안정하여 pH 4.0에서 15시간까지 처이하였을 경우, 또 CHCI$_3$, acetone 용액에서 약 5일간까지 처이하였을 때에도 거의 독성이 감소되지 않았다. alkali성 및 열에 대하여서는 불안정한 물질로써 pH 8.0에서 5분간 처이하였을 때 물질이 상당히 실활되었으며 6$0^{\circ}C$以上의 온도에서는 비교적 격히 실활되었다. 3. 25ug/m1 수용액에서 Pseudorabora parva에 대하여 약 20분간 이상 처이로서는 회생이 불가능함을 알았다. 이상과 같은 성질을 가진 본 독성물질은 선별된 균주의 배양여액을 pH 3.0으로 하강하여 NaCl로서 포화시킨 후 CHCl$_3$에 전용하여 농축하르로써 비교적 간단한 방법으로 분리할 수 있었다.

오약순기산(烏藥順氣散) 및 중성어혈(中性瘀血) 약침(藥鍼)이 흰쥐 좌골신경 압좌 손상 후 통증 감소와 신경 재생에 미치는 영향 (Effects of Ohyaksungi-san(Wuyaoshungi-san)and Jungsongouhyul Pharmacopuncture on Pain Reduction and Nerve Regeneration after Crush Injury in Rat Sciatic Nerve)

  • Objectives : This study was designed to evaluate the effects of Ohyaksungi-san(Wuyaoshungi-san) and Jungsongouhyul pharmacopuncture on pain reduction and nerve regeneration after crush injury in rat sciatic nerve. Methods : Animal model was produced through crush injury of right sciatic nerve and they were divided into four groups; Group I: no treatment control group; Group II: experimental group treated with Ohyaksungi-san(Wuyaoshungi-san); Group III: experimental group treated with Jungsongouhyul pharmacopuncture; Group IV: experimental group treated with Ohyaksungi-san(Wuyaoshungi-san) and Jungsongouhyul pharmacopuncture. For the assessment of pain, this study was observed the paw withdrawal latency(PWL) and immunoreactivity on the substance-P. For the assessment of nerve regeneration, the sciatic functional index(SFI) and immunoreactivity on the BDNF were measured. Results : 1. In the assessment of pain, the PWL of experimental groups was significantly higher than control group and group IV was significantly higher than other groups at the all days. 2. In immunohistochemical response of substance-P, as time passes, the immunoreactivity of all groups were decreased gradully. Especially, group IV had the lowest immunoreactivity. 3. In the assessment of SFI, the SFI of experimental groups were significantly higher than control group. 4. In immunohistochemical response of BDNF, the BDNF immunoreactivity of all groups was significantly higher than control group and especially, group IV had the highest immunoreactivity at the 14 days after injury. 5. H & E stain was used on the liver and kidney to investigate toxic effect of Jungsongouhyul pharmacopuncture and Ohyaksungi-san(Wuyaoshungi-san) on on 21 days after injury. However there were no any toxic effects both control group and experimental groups. Conclusions : On the basis of these results, we propose that Ohyaksungi-san(Wuyaoshungi-san) and Jungsongouhyul pharmacopuncture were related to pain reduction and motor nerve recovery, also decreased substance-P expression and increased BDNF expression after crush injury of sciatic nerve, especially these two treatments could be more effective when they were combined simultaneously.

Estimation of Environmental Distribution for Benzoyl peroxide Using EQC Model

  • Benzoyl peroxide is a high production volume chemical, which was produced about 1,375 tons/year in Korea as of 2001 survey. Most of them are used as initiators in polymerization, catalysts in the plastics industry, bleaching agents for flour and medication for acne vulgaris. The substance is one of the sever chemicals of which human and environmental risks are being assessed by National Institute of Environmental Research under the frame of OECD SIDS Program. It has a melting point of 104-106 $^{\circ}C$ and has solubility of 9.1 mg/1 in water at 25 $^{\circ}C$. The substance was readily biodegradable (83 % after 21days) and had toxic effects to aquatic organisms. The range of 72 hr-EbC50 (biomass) for algae was 0.07-0.44 mg/1 and 48 hr-EC50 for daphnia was 0.07-2.91 mg/1. The LC50 of acute toxicity to fish was 0.24-2.0 mg/1. Although the toxic effects of benzoyl peroxide to aquatic organisms were investigated, environmental monitoring data were not studied. In this study, distribution of the chemical among multimedia environment was estimated using EQC model based on the physical-chemical properties to evaluate the risk of benzoyl peroxide in environment. In level I, II calculation the chemical was distributed to soil (68.3 %) and water (28.7 %). In level III calculation it was primarily distributed to soil (99.9 %) and overall residence time of 3.4 years was estimated. Benzoyl peroxide could be persistent in environment.

지능형 실시간 대기확산 시스템에 관한 연구 (A Study on Smart Real-time Atmospheric Dispersion System)

  • 대부분의 산업시설이 유해물질을 다루고 있기 때문에 산업시설의 사고발생은 막대한 물적, 인적 피해를 발생시키므로 안전관리, 중대형 사고 예방 및 위험 예측 등이 중요시 되고 있다. 특히, 유해물질 누출사고는 누출물질, 저장시설, 대기 상태에 따라 유독가스의 확산 속도와 범위가 달라지므로 이를 모사하는 대기확산 시스템이 이용되어왔다. 모사에 기반하는 대기확산 시스템은 산업시설 구조물 설계 단계에서 주로 사용되며 누출 사고 시 즉각적인 처리와 대응이 어렵다. 실시간 대기정보 데이터를 이용한 연구 및 사례는 존재하지만 시스템의 성능저하 및 전문적 지식의 결여로 신속한 처리 및 대응이 미흡하였다. 본 논문은 에너지 플랜트 환경에서 특수성과 효율성을 강화하여 유해화학물질 누출로 인한 대기확산범위를 즉각적으로 산출하고 누출 시점 및 지점을 지능형 알고리즘으로 결정함으로써 사고예방형 실시간 지능형 대기확산 시스템을 구현한다.

침몰선박 위해도 평가항목 및 평가지수 개선에 관한 연구 (A Study on the Improvement of Risk Assessment Items and Index for Sunken Ship)

  • 본 연구에서는 침몰선박 위해도 평가항목 및 평가지수를 개정하고 신규항목을 도출하기 위해 전문가를 대상으로 AHP 기법의 설문을 실시하였다. 설문조사 결과, 사고원인 및 조류영향 등 두개의 신규항목을 도출하였으며 하나의 그룹으로 평가되었던 독성액체물질과 연료유적재량, 폭발성가스는 각각 평가하는 것이 적합한 것으로 나타났다. 이에 따라 기존 일곱 개 평가항목을 열한 개 항목으로 조정하여 평가항목별 지수를 분석한 결과, 독성액체물질, 유출가능성, 폭발성가스, 연료유적재량, 해역환경민감도, 해상교통환경, 사고원인, 조류, 여유수심, 선박종류, 선박규모 순으로 평가지수가 나타났다. 특히, 신규 도출된 평가항목 지수를 기존 위해도 평가항목과 비교해 보았을 때 해역환경민감도와 유출가능성 항목은 기존 평가지수 보다 높게 나타났고, 여유수심 항목의 평가지수는 더 낮게 나타났다.
