Effect on the Inoculation of Bacillus on the Growth of Chinese Cabbage and Sesame and on Microbial Flora in Soils (Bacillus subtilis 접종이 배추 및 참깨의 생장(生長)과 토양(土壤) 미생물상(微生物相)에 미치는 영향(影響))

  • An antagonistic bacteria was isolated from rhiaosphere of pepper and corn and identified as Bacillus (B.) subtilis. These B. subtilis B-5 was transformed and marked with the plasmid pCPP4 which possess neomycine resistan. gene. The marked stranins showed growth inhibition to Rhizoctonia (R.) solani, Fusarium (F.) solani, and F. oxysporum in vitro, and were used in studying growth promoting effects on sesame and cabbage. All the identified strains utilize glucose, sucrose, fructose, lactose, mannitol and sorbitol as carbon source, but not rhamnose, and the marked strains also showed characteristics similar to wild-type strains. Germination rate of chinese cabbage and sesame seeds was increased by about 10% or more in the plot to which these strains were inoculated and the effect was higher in soil than in petri dish. The early growth promoting effects of these strains appeared higher, as compared with control plot, in the plots to which B. subtilis B-5 and pathogenic fungi was inoculated together. When the marked strains, B. subtilis B-5NEOr, were inoculated in the rhizosphere of chinese cabbage and sesame with $1.1{\times}10^8CFU/g$ dry soil, the number of inoculated strain was decreased slowly to the level of $10^5{\sim}10^6CFU/g$ dry soil after 4 weeks and the number of Pseudomonas spp. maintanied the level of $10^5CFU/g$ dry soil throught total period, but the number of fungi was decreased rapidly from the early level of $10^8CFU/g$ dry soil to $10^3CFU/g$ dry soil after 4 weeks.

Phytotoxicity of Endophytic Fungi and Characterization of a Phytotoxin Isolated from Gliocladium catenulatum from Pinus densiflora (식물내생곰팡이의 제초활성 검정 및 소나무에서 분리한 Gliocladium catenulatum이 생산하는 제초활성 물질의 특성 규명)

  • This study was conducted to discover new phytotoxins which may be used as lead molecules for the development of new herbicides. A total of 187 endophytic fungi were isolated from 11 plant species, which were collected from 8 locations in Korea. Their herbicidal activities were screened in vivo by herbicidal and duckweed bioassays after they were cultured in potato dextrose broth and rice solid media. Both fermentation broth and solid culture extract of Gliocladium catenulatum F0006 isolated from red pine (Pinus densiflora) showed 70% herbicidal activity only against cocklebur (Xanthium strumarium) out of the 10 weeds tested. Solid culture extract of F0034 isolated from arrowroot (Pueraria thunbergiana) exhibited 20 to 100% herbicidal activities against all of the weeds. Especially, shattercane (Sorghum bicolor), barnyardgrass (Echinochloa crus-galli), large crabgrass (Digitaria sanguinalis), and fall pauicum (Panicum dichtomiflorum) were sensitive to the solid culture extract of F0034. In addition, solid culture extract of F0043 isolated from red pine displayed 20% to 70% herbicidal activities only against 5 grass species, but not against 5 broad-leaf plant species. On the other hand, as the results of duckweed assay, 8 fermentation broths showed 100% growth inhibitory activity at concentrations less than 5.0% of culture supernatants and 12 solid cultures had a potent inhibitory activity against duckweed growth. A toxic metabolite was purified from the solid cultures of G. catenulatum F0006 by repeated column chromatography and bioassay. It caused a phytotoxic syndrome only on cocklebur out of the 10 weeds tested; it completely killed cocklebur seedlings at $500\;{\mu}g/ml$ and showed 85% herbicidal activity against cocklebur at $100\;{\mu}g/ml$. The molecular weight of the toxic metabolite is 238 daltons and its structure determination is underway.

An Investigation of the Hazards Associated with Cucumber and Hot Pepper Cultivation Areas to Establish a Good Agricultural Practices (GAP) Model (오이와 고추생산 환경에서의 GAP 모델 개발을 위한 위해요소 조사)

  • To analyze the hazards associated with cucumber and hot pepper cultivation areas, a total of 72 samples were obtained and tested to detect the presence of biological (sanitary indicative, pathogenic bacteria and fungi) and chemical hazards (heavy metals and pesticide residues). The levels of sanitary indicative bacteria (aerobic plate counts and coliforms) and fungi were ND-7.2 and ND-4.8 log CFU/(g, mL, hand, or $100cm^2$) in cucumber cultivation areas, and ND-6.8 and 0.4-5.3 log CFU/(g, mL, hand, or $100cm^2$) in hot pepper cultivation areas. More specifically, the soil of hot pepper cultivation areas was contaminated with coliforms at a maximum level of 5.6 log CFU/g. Staphylococcus aureus was detected only in glove samples at a level of 1.4 log CFU/$100cm^2$ and Bacillus cereus was detected in the majority of samples at a level of ND-4.8 log CFU/(g, mL, hand, or $100cm^2$). Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella spp. were not detected. Heavy metal (Zn, Cu, Ni, Pb, and Hg) chemical hazards were detected at levels lower than the regulation limit. Residual insecticides were not detected in cucumbers; however, hexaconazole was detected at a level of 0.016 mg/kg (maximum residue limit: 0.3 mg/kg) in hot peppers.

  • Effects of the indigenous Vesicular-arbuscular mycurrhizal fungi(VAMF) on early growth response of greenhouse grown crops were experimented. This study was done to evaluate the benefit of indigenous VAMF inoculation on the early growth and the subsequent growth after transplanting of some crops such as cucumber, tomato, hot pepper, eggplant, and melon. Leaf area, shoot dry weight, and plant length of mycorrhizal greenhouse crops showed the tendency of significant or no significant increase over control plants receiving no inoculation. The levels of VA mycorrhizal colonization were increased with plant growth, and infection rates of horticultural crop except hot pepper around one week after transplanting were decreased, while that of 8 weeks after emergence of mycorrhizal seedlings were increased again and infected by around 50% at harvesting time. In spore densities in the rhizosphere soil of craps experimented, the number of spore ranged from $72.7{\pm}26.3$ to $100{\pm}10.3g^1$ on dried soil basis and high density showed in both cucumber and tomato. Total nitrogen contents in shoots were lower in the mycorrhizal plants than non-mycorrhizal one, whereas P uptake in mycorrhizal hot pepper and tomato were highly ramarkable. The K contents in the shoots of mycorrhizal cucumber and eggplent were highly enhanced. Inoculation of the indigenous VAMF enhanced shoot Ca and Mg in both tomoto and melon. The contents of Fe, Zn, Mn and Cu in shoots of mycorrhizal crops were higher than non-mycorrhizal plants and vice versa in case of eggplent. Inoculation of the indigenous VAMF to horticultural crops were effective for alleviation of transplanting shock, and pretransplanting infection improved subsequent growth by reducing the time required for establishment of a functional mycorrhizal symbiosis following transplanting.

Analysis of Terpenoids as Volatile Compound Released During the Drying Process of Cryptomeria japonica (삼나무 건조 중 발생하는 휘발성 유기화합물 Terpenoids의 분석)

  • The aim of this study was to investigate the terpenoids of Total Volatile Organic Compounds (VOCs) released during drying of Cryptomeria japonica using the thermal extractor (TE). Considering the drying process of C. japonica, temperatures of TE were set at $27^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$, $100^{\circ}C$, and $120^{\circ}C$, respectively. As the result, the emission factors of VOCs and terpenoids were increased as temperature increased. The amount of terpenoids included in VOCs emission factors were 87.5%, 81.6%, 83.6%, 90.1%, and 97.3% depending on above temperatures, respectively. Especially at$100^{\circ}C$ and $120^{\circ}C$, the amount of terpenoids were measured more than 90%. ${\delta}$-cadinene was the highest yield at each temperature and 32 types of terpenoids were collected. Emitted terpenoids were classified into the sesquiterpene group which consists of 15 carbon sources. These 32 sesquiterpenes were used for determining the useful bioactivity such as antifungal activity by the agar dilution. As the result, they showed the antifungal activity against Trichophyton rubrum, Trichophyton mentagrophytes, Microsporum gypseum. The 5,000 ppm concentration of terpenoids showed a strong activity with 100% against the 3 fungi. At the 1,000 ppm concentration of terpenoids, the antifungal activities against three fungi were 95.2%, 98.7%, and 97.3%, and their activities were a little inhibited at 100 ppm concentration.

The Effects of Mortierella alpina Fungi and Extracted Oil (Arachidonic Acid Rich) on Growth and Learning Ability in Dam and Pups of Rat (흰쥐의 Mortierella alpina 균사체와 추출유의 섭취에 의한 생육 효과와 학습능력 비교)

  • Mortierella alpina, a common soil fungus, is the most efficient organism for production of production acid presently known. Since arachidonic acid are important in human brain and retina development, it was undertaken the growing effect containing diet as a food ingredient. Arachidonic acid rich oil derived from Mortierella alpina, was subjected to a program of studies to establish for use in diet supplement. This study was compared the growth and learning effect of fungal oil rich in arachidonic acid by incorporated into diets ad libitum. Sprague-Dawley rats received experimental diets 5 groups (standard AIN 93 based control with beef tallow, extract oil 8%, and 4%, and Mortierella alpina in diet 10% and 20%) over all experiment duration (pre-mating, mating, gestation, lactation, and after weaning 4 weeks). Pups born during this period consumed same diets after wean for 4 weeks. There was no statistical significance of diet effects in reproductive performance and fertility from birth to weaning. But the groups of Mortierella alpine diet were lower of weight gain and diet intake after weaning. The serum lipids were significantly different with diet groups, higher TG in LO (oil 4%) group of dams, and higher total cholesterol in LF (M. alpina 10%) of pups, although serum albumin content was not significantly different in diet group. The spent-time and memory effect within 4 weeks of T-Morris water maze pass test in dam and 7-week- age pups did not differ in diet groups. On the count of backing error in weaning period of pups was lower in HO(extracted oil 8%) group. In the group of 10% and 20% Mortierella alpina diet, DNA content was lower in brain with lower body weight, but liver DNA relative to body weight was higher than control. Further correlation analyses would be needed DNA and arachidonic acid intakes, with Mortierella alpina diet digestion rate.

  • Quercus acutissima ectomycorrhizae were classified as apical type, linear type, clavate type, diffuse type, pyramidal type, coralloid type, and nodular type. The surface texture of the fungal mantle at the initial stage of mycorrhizal formation was velvety. The surface texture of Pisolithus tinctorius(Pt) mycorrhizae at 30 September was well-developed felty mantle, Yellowish white. Except Pt all mycorrhizae formed by ectomycorrhizal fungi used to experiment were white with felty mantle. Mycorrhizae at the initial stage of mycorrhizae formation were creamy or creamy brown and swelled with thin mantle. Transverse and longitudinal sections showed radially-elongated cortical cell layers and epidermal cell with Hartig net. The transversal wideth of radially-elongated cortical and epidermal cells in the mycorrhizae with thick mantle on 30 September did not different with the mycorrhizae with thin mantle on the initial stage. Pt #250 formed coralloid mycorrizae but Pt KJ-1 did not although they are same species. On the mean length of linear type ectomycorrhizae of Pt KJ-1(2.21mm) was 1.5 times longer than that of Pt #250(1.32mm). The total dry weight of seedlings inoculated with Pt KJ-1, Pt #250, Lycoperdon pedicellatum, Scleroderma verrucosum were significantly heavier than those of suillus granulatus, Laccaria laccata.

Herbicidal and Antifungal Activities of the aqueous extracts of Several Naturalized Plants (수종의 귀화식물 수용성추출물의 제초 및 항균 활성 탐색)

  • The study researched germination of the plants and growth of experimented bacteria according to concentration of water extract in order to provide basic data for developing natural agricultural resources by using naturalized plants including Solidago altissima, Amaranthus retroflexus and Sida spinosa. As concentration of water extract increased, most of test plants showed a decrease in relative germinability. Sida spinosa(r=-0.540, p<0.01), Physalis wrightii(r=-0.693, p<0.01), Amaranthus retroflexu(r=-0.724, p<0.01), Solidago altissima(r=-0.728, p<0.01) and Eclipta prostrata(r=-0.779, p<0.01) showed tendency of decrease in relative germinative power in order, respectively. For average germination period, as concentration of the processed group increased, the time for germination increased (r = 0.769, p<0.01) and according to donor plants and test plants, there was a little difference. Also, as concentration of water extract of donor plant, length of above-aerial part(r=-0.587, p<0.01), length of underground part(r=-0.741, p<0.01), fresh weight(r=-0.574, p<0.01) and generation of root hair decreased. An then, for growth of test fungi according to concentration of water extract of donor plants, growths of Botrytis cinerea(r=-0.266, p<0.05), Diaporthe citri(r=-0.323 p<0.01), Colletotrichum gloeosporioides(r=-0.512, p<0.01), Pythiumultimum(r=-0.581, p<0.01) and Rhizoctonia solani(r=-0.806, p<0.01) were repressed in order, respectively. For total amount of content of phenol with herbicidal and Antifungal activities, S. altissima $17.3{\pm}0.5mg/g$, A. retroflexus $13.1{\pm}0.3mg/g$, P. wrightii $12.0{\pm}0.4mg/g$, S. spinosa $9.5{\pm}0.1mg/g$ and E. prostrata L. $4.1{\pm}0.1mg/g$ showed in order, respectively. As these results are summarized, donor plants which were naturalized, have competitive advantage because they release phenolic compounds with allelopathic effect and affect on germination, growth and fungi growth on underground flora compared to native plants and they have eligibility for natural herbicide and germicide.

Comparison of Ethylene Oxide (E.O.) and Irradiation Treatment on the Sterilization of Spices (향신료(香辛料)의 Ethylene Oxide 처리(處理)와 감마선(線) 조사(照射)와의 살균효과 비교)

  • E.O. and gamma irradiation treatment on the sterilization of ground samples of 5 different types of spices(red and black pepper, onion, garlic and ginger) were investigated. Populations of mesophilic bacteria, mesophilic spores, acid tolerant bacteria and fungi in various samples were $10^4-10^6/g,\;10^3-10^5/g,\;10^3-10^5/g\;and\;10^3-10^4/g$, respectively. Coliforms and osmophilic molds were found only in red and black pepper as $10^3-10^4/g$. A radiation dose of 5 to 7 kGy proved sufficient to redure the viable cell count of the total bacteria and fungi to the level of $10^3/g$ and they were sterilized completely by radiation dose of 10 kGy or more. Coliforms, mesophilic spores and acid tolerant bacteria were sterilized at 5,7 and 10 kGy, respectively. In the mean time $D_{10}$ values of each spices ranged from 1.38 to 2.88 kGy. Comparison of E.O. and gamma irradiation treatment showed that E.O. treatment was less effective than radiation in controlling microbial contamination in spices.

Cultural Characteristics of Ectomycorrhizal Mushrooms

  • Ectomycorrhizal (ECM) mushrooms play a major role in plant growth promotion through symbiotic association with roots of forest trees. They also provide an economically important food resource to us and therefore they have been studied for their artificial cultivation for decades in Korea. We have secured bio-resources of ECM mushrooms from Korean forests and performed their physiological studies. To investigate the cultural characteristics, the fungi were cultured under different conditions (medium, temperature, pH of the medium, inorganic nitrogen source). More than 90% of total 160 strains grew on three solid media (potato dextrose agar, PDA; sabouraud dextrose agar, SDA; modified Melin-Norkrans medium, MMN). The rate of mycelial growth on malt extract agar (MEA) was lower than those of three media (PDA, SDA, MMN). None of the Tricholomataceae strains grew on MEA. Many strains of ECM mushrooms were able to grow at the temperature range of $15{\sim}25^{\circ}C$ on PDA, while they showed poor growth at $10^{\circ}C$ or $30^{\circ}C$. In particular, the growth rates of both Gomphaceae and Tricholomataceae were significantly lower at $10^{\circ}C$ than at $30^{\circ}C$. The optimal pH of many strains was pH 5.0 when they cultured in potato dextrose broth (PDB). Fifty-seven percent of tested strains grew well on medium containing ammonium source than nitrate source. Many strains of Tricholomataceae showed a notable growth on ammonium medium than nitrate medium. Twenty-three percent of strains preferred nitrate source than ammonium source for their mycelial growth. The production and activity of two enzymes (cellulase and laccase) by ECM fungi were also assayed on the enzyme screening media containing CMC or ABTS. Each strains exhibited different levels of enzymatic activities as well as enzyme production. The number of laccase-producing strains was less than that of cellulase-producing strains. We found that 77% of tested strains produced both cellulase and laccase, whereas 2% of strains did not produce any enzymes. The morphological characteristics of mycelial colony were also examined on four different solid media. Yellow was a dominant color in mycelial colony and followed by white and brown on all culture media. ECM mushrooms formed mycelial colonies with a single or multiple colors within a culture medium depending on the strains and culture media. The most common shape of mycelial colony was a circular form on all media tested. Other families except for Amanitaceae formed an irregular colony on MMN than PDA. All strains of Tricholomataceae did not form a filamentous colony on all media. The pigmentation of culture media by mycelial colonies was observed in more than 50% of strains tested on both PDA and SDA. The degree of pigmentation on PDA or SDA was higher than MMN and brown color was dominant than yellow color. The production of exudates from mycelial colony was higher on PDA than MMN. Brown exudates were mainly produced by many strains on PDA or SDA, whereas transparent exudates were mainly produced by strains on MMN. We observed the mycelial colonies with a single or multiple textures in just one culture plate. Wrinkled or uneven colony surfaces were remarkably observed in many strains on PDA or SDA, while an even colony surface was observed in many strains on MMN. Sixty percent of Tricholomaceae strains formed wrinkled surface on PDA. However, they did not form any wrinkle on MMN plate. Cottony texture was observed in mycelia colonies of many strains. Velvety texture was often observed in the mycelial colonies on SDA than PDA and accounted for 60% of Suillaceae strains on SDA.
