Production Conditions and Properties of Glucose Isomerase from Streptomyces griseolus (Streptomyces griseolus기원의 포도당 이성화효소의 생성 조건과 성질)

  • Cultural characteristics of Strptomyces griseolus isolated from the soil were investigated. This strain was disclosed to utilize D-xylose, and D-glactose in preference order as a carbon source with the formation of glucose isomerase. The addition of sweet potato starch also proved effective promoting the total enzyme activity measured at 29% higher than the control. Corn cob, one of waste agricultural resources, was hydrolyzed in 2~3% $H_2SO_4$ solution at $100^{\circ}C$, 3~5 hours to produce a xylose syrup which gave rise to the recovery of 19.9% in a batch system and 28.2% in a repeated system. By the addition of both 2% of xylose syrup(Be'28) prepared by and us 65% of corn steep liquor (total nitrogen 1.2%), enzyme induction was maximized. The enzyme activity was stimulated by the xylose and the cell growth by the C.S.L. Also, remarkable increase of enzyme activity was noticed by the addition of protein acid hydrolysate 86.2% higher than the control. $QO_2$ of the biomass cultured in 30L capacity jarfermentor recorded low oxygen requirement of 251.2 1/hr. Maximum activity of glucose isomerase was observed noted at the 9th hour after inoculation which is 2 hours faster than the stationery was observed noted at the 9th hour after inoculation which is 2 hours faster than the stationery phase of the biomass growth. Glucose isomerase from the strain was activated by adding the $Co^{++}\;and\;Mg^{++}$ with optimum temperature of $73^{\circ}C$ and pH of 7.2. Conversion ratio of 60% glucose to frutose was 42.5% after 70 hours reaction.

디젤로 오염된 토양의 효과적인 Bioventing

  • In this work, cost effective venting is considered by comparing flow rates of 5$m\ell$/min, 10$m\ell$/min, and 20$m\ell$/min. Studies were performed on a soil artificially contaminated with diesel oil (the initial TPH(Total Petroleum Hydrocarbon) concentration of 7098mg/kg), and nutrient condition was C:N:P rate of 100:10:1. The soil has a sandy texture with pH of 6.8, 2.16 ~2.38% organic matter, a total porosity of 47~52% and field capacity 16.2~ 17.2%. The column experiments was made of glass column of 60cm length and 10cm I.D. at controlled temperature of 2$0^{\circ}C$($\pm$2.5$^{\circ}C$). The efficiency of continuous flow rate of 5, 10 and 20$m\ell$/min resulted in separately 61.3%, 58.1%, and 55% reduction of initial TPH concentration(7098mg/kg). Hydrocarbon utilizing microbial count and dehydrogenase activity in air flow of 5$m\ell$/min were higher than those of the others. The first order degradation rate of n-alkanes ranging from C10 to C28 was higher than that of pristane and phytane as isoprenoids. The $C_{17}$/pristane and $C_{18}$phytane ratios for monitoring the degree of biodegradation were useful only during the early stages of oil degradation. Degradation contributed from about 89% to 93% of TPH removal. Volatilization loss of diesel oil in contaminated soil was about 7% to 11%, which was significantly small compared to degradation.n.

Conversion of Shrimp Shell by Using Serratia sp. TKU017 Fermentation for the Production of Enzymes and Antioxidants

  • A chitinase (CHT) and a protease (PRO) were purified from the culture supernatant of Serratia sp. TKU017, with shrimp shell as the sole carbon/nitrogen source. The molecular masses of CHT and PRO determined by SDS-PAGE were approximately 65 kDa and 53 kDa, respectively. CHT was inhibited by $Mn^{2+}$ and $Cu^{2+}$, and PRO was inhibited by most tested divalent metals and EDTA. The optimum pH, optimum temperature, pH stability, and thermal stability of CHT and PRO were pH 5, $50^{\circ}C$, pH 5-7, and <$50^{\circ}C$, and pH 9, $40^{\circ}C$, pH 5-11, and <$40^{\circ}C$, respectively. PRO retained 95% of its protease activity in the presence of 0.5 mM SDS. The result demonstrates that PRO is an SDS-resistant protease and probably has a rigid structure. The $4^{th}$-day supernatant showed the strongest antioxidant activity (70%, DPPH scavenging ability) and the highest total phenolic content ($196{\pm}6.2\;{\mu}g$ of gallic acid equiv./ml). Significant associations between the antioxidant potency and the total phenolic content, as well as between the antioxidant potency and free amino groups, were found for the supernatant. With this method, we have shown that shrimp shell wastes can be utilized and it is effective in the production of enzymes and antioxidants, facilitating its potential use in industrial applications and functional foods.

Comparison of Tiletamine-zolazepam, Tiletamine-zolazepam-xylazine and Ketamine-xylazine anesthesia in dogs (개에서 Tiletamine-zolazepam, Tiletamine-zolazepam-xylazine 및 Ketamine-xylazine의 마취효과)

  • This study was conducted to compare the anesthetic effects of intravenous tiletamine-zolazepam(TZ, 7mg/kg TZ), tiletamine-zolazepam-xylazine(TZX, 7mg/kg TZ and 1.1mg/kg X) and ketamine-xylazine(KX, 10mg/kg K and 1.1mg/kg X). Fifteen mixed-breed healthy dogs($3.5{\pm}1.0kg$) were randomly assigned to the three treatment groups(TZ, TZX, KX) with 5 dogs in each group. The mean surgical anesthesia time was $25.6{\pm}4.2$, $62.6{\pm}6.2$ and $21.0{\pm}3.7$ min in TZ-, TZX- and KX-anesthetized dogs, respectively. The duration of the loss of response to toe-web needle prick and to visceral pain was significantly increased in the TZX group with $40.0{\pm}15.8$ min and $44.0{\pm}5.5$ min, respectively(p<0.01). Heart rate decreased significantly below baseline in TZX and KX groups(p<0.05, p<0.01) whereas it increased above baseline in TZ group. Respiratory rate remained unchanged or increased above baseline in TZ group, but decreased significantly from 10 to 30 min in TZX(p<0.01, p<0.05) and at 10 min in KX group(p<0.05). Body temperature decreased significantly below baseline in all three groups(p<0.01, p<0.05). Hematologic(PCV, RBC, WBC) and serum chemistry values(GOT, GPT, BUN, creatinine, total protein, glucose) were monitored before anesthesia, after recovery from anesthesia and 1, 3 and 7 days postanesthesia. All hematologic values remained generally within normal ranges, and GOT, GPT, BUN, creatinine and total protein values were within normal ranges during the period. Glucose values for TZX and KX groups increased greatly after recovery from anesthesia. We conclude that tiletamine-zolazepam-xylazine provides effective surgical anesthesia in dogs and in many cases may be preferable to conventional ketamine-xylazine regimen.

The Effect of Angelicae gigantis radix according to Heat-process on Anti-Oxidant and Anti-Thrombotic (초법에 따른 당귀의 항산화 및 항혈전 효과)

  • Objectives: Arachidonic acid is control the thromboxane A2 (TXA2) and prostacycline (PGI2) synthesis, TXA2 increase lead to thrombus produced by induces platelet aggregation and vasoconstriction. Angelicae gigantis radix (RAR) is mainly used blood deficiency and stagnation. In previous studies, RAR has been reported that a vasodilating and blood clotting delay effects. In this study, investigate that anti-oxidant and anti-thrombotic effects of RAR by heat-process.Methods: The heated angelicae gigantis radix sample were made by 140, 180, and 220 ℃ and 4, 6, 9 and 12 min using water or 30% ethanol. The anti-oxidant effects were measured by total polyphenol, total flavonoid, DPPH and ABTS radical scavening activation. Anti-thrombotic effect conducted in samples that are determined to be effective through the anti-oxidant experiment such as angelicae gigantis radix roasted 180℃, and 220℃ and angelicae gigantis radix roasted with 30% ethanol 180℃, and 220℃.Results: Anti-oxidant parameters were efficacious in high temperature roasted AR. Also AR and EAR increased a inhibitory activity of FXa compared with RAR. The blood coagulation time of administration groups were significantly increased compare with control group. The TXB2 was significantly decreased in AR and EAR.Conclusions : We confirmed that whether AR and EAR administration has anti-oxidant and anti-thrombotic effect or not. As the results, AR and EAR were improved anti-oxidant effects and blood biochemistry compare with control group. This study provides scientific evidence that AR and EAR are have an anti-oxidant effect and anti-thrombotic effect, it expected that there is no difference between the two.

The anti-atopic effect of natural carbonated hot spring water on DNCB-induced NC/Nga mice under different temperature (DNCB로 아토피가 유도된 NC/Nga생쥐에서 천연 탄산 온천수의 온도별 항아토피 효과)

  • Objectives : The aim of this study is to investigate the effect of natural carbonated hot spring water (NCHW) on DNCB-induced NC/Nga mice, an atopic dermatitis (AD) model. Methods :At first, NC/Nga mice were prepared and induced to have atopic lesion on their back skin by dinitrochlorobenzene (DNCB). They swam for 20 minutes everyday in tanks where the one is filled with $28^{\circ}C$ NCHW, the other is with $25^{\circ}C$ carbonated hot spring water and another is with artificial carbonated hot spring water (ACHW). After 3 weeks, We assessed the skin clinical score and macroscopic appearance, total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and histological changes. Results : There are meaningful results of improving atopic lesion-state by relieving the count of total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and recovering skin clinical score in the group with $28^{\circ}C$ NCHW in comparison with the other groups. Conclusions : The NCHW may have potential as an effective treatment for atopic dermatitis.

Evaluation of Trophic State of a Small-scale Pond (Wonheung) in Ecological Park (소규모 생태연못(원흥이 방죽)의 부영양화 평가)

  • Many small-scale ponds that serve as ecological habitat, recreation and irrigation are faced to eutrophication problem, which causes aesthetic nuisance and ultimately loss of their functions. Thus accurate evaluation of the trophic state of these ponds is essential to provide rational information to the stakeholders so that they can develop effective management actions. In this study, the trophic state of a small pond (Wonheung) that experiencing water quality degradation due to vicinity land development was assessed using various Trophic State Indexes (TSIs) and statistical analysis including Principal Components Analysis (PCA) based on the field monitoring data obtained from May to December, 2007. The results showed that the pond is under eutrophic state with average total nitrogen (T-N) and total phosphorus (T-P) concentrations of $708.1{\mu}g/L$ and $59.3{\mu}g/L$, respectively. The factor loading plot obtained from PCA showed distinct two influencing factors, PC 1 and PC 2. PC 1 was grouped by T-P, Chlorophyll a (Chl-a), suspended solids (SS), TN/TP ratio, and transparency that all strongly related to the eutrophication state, while PC 2 by temperature, conductivity, dissolved oxygen (DO) and turbidity that explains the seasonal water quality variations. The limiting factor was identified as light rather than phosphorus by both T-N/T-P ratio and TSI indexes analysis. The results and methodology adopted in this study can be used for water quality assessment for other small ponds and lakes.

Changes in Color Value and Chemical Components of Hoelen by Various Drying Methods (건조방법에 따른 복령의 색도 및 화학성분의 변화)

  • In this study, the quality changes of Hoelen was investigated, which were color differences, browning degrees, total phenolic compounds and electron donating abilities to optimize drying condition using hot air and far infrared drying by changing conditions such as temperatures, air velocities and radiation distances. In color values, ${\Delta}b\;value$ decreased with increasing temperatures, while ${\Delta}L$, ${\Delta}a$ and ${\Delta}E\;value$ increased. The browning degrees showed the similar tendency with the color values. Total phenolic components of Hoelen decreased with increasing drying temperature and air velocity, and decreasing radiation distance. On the other hand, there was no significant difference in the changes of electron donating abilities (EDA) of Hoelen among drying methods and conditions. As a result, the hot air drying was more effective method than the far infrared was.

Assessment of Seasonal Variation in Water Quality in Daedong Lake (대동호의 시기별 및 계절별 수질변화 평가)

  • BACKGROUND: Most lakes have increased concerns about water pollution due to the inflow of non-point sources caused by human activities. Therefore, the lake water quality survey was conducted in order to propose effective plans for water quality management by analyzing the characteristics of lakes and the change of water quality. METHODS AND RESULTS: In order to investigate the physicochemical water quality in Daedong lake, water quality analysis was undertaken from July 2018 to June 2019. Water temperature was ranged from 7.8 to 34.3℃ and pH varied from 6.9 to 10.2. The concentration of Dissolved oxygen, Suspended solid, Biochemical Oxygen Demand (BOD), and Chemical Oxygen Demand (COD) were 5.6 ~ 17.2 mg/L, 2.4 ~ 35.3 mg/L, and 4.5 ~ 15.1 mg/L, and 0.9 ~ 2.8 mg/L, respectively. The Total Nitrogen (T-N) concentration ranged from 0.974 ~ 2.126 mg/L, and Total Phosphorus (T-P) concentration ranged from 0.014 ~ 0.057 mg/L. The Chlorophyll-a (Chl-a) ranged from 2.7 ~ 37.9 mg/㎥. Through Carlson TSIm assessment using T-P and Chl-a results, evaluating trophic state, Daedong lake was evaluated as mesotrophic. CONCLUSION: Water pollution management plan needs such as nutrient removal technology and nonpoint source management for prevention of eutrophication in Daedong lake.

Determination of Dairy Cow Food Intake using Simulated Annealing (시뮬레이티드 어닐링을 이용한 젖소의 급이량 산정)

  • The daily food intake for dairy cows has to be effectively controlled to breed a sound group of cows as well as to enhance the productivity of the cows. But, feed stuffs are fed in the common bulk for a group of cows in most cases despite that the individual food intake has to be varied. To obtain the feed for each cow, both the nutrient requirements and the nutrient composition of fred have to be provided in advance, which are based on the status of cows such as weigh marginal weight amount of milk, fat concentration in milk, growth and milking stages, and rough feed ratio, etc. Then, the mixed ration fur diet would be computed by the nutrient requirements constraints. However, when TMR (Total Mixed Ration) is conventionally supplied for a group of cows, it is almost impossible to get an optimal feed mixed ration meeting the nutrient requirements of each individual cow since the constraints are usually conflicting and over-constrained although they are linear. Hence, addressed in this paper is a simulated annealing (SA) technique to find the food intake for dairy cows, considering the characteristics of individual or grouped cows. Appropriate parameters fur the successful working of SA are determined through preliminary experiments. The parameters include initial temperature, epoch length. cooling scheduling, and stopping criteria. In addition, a neighborhood solution generation method for the effective improvement of solutions is presented. Experimental results show that the final solution for the mixture of feed fits the rough feed ratio and some other nutrient requirements such as rough fiber, acid detergent fiber, and neutral detergent fiber, with 100 percent, while fulfilling net energy for lactating, metabolic energy, total digestible nutrients, crude protein, and undegraded intake protein within average five percent.
