In order to detect the way of absorption and metaboism of the urea it is sprayed on the surface of the leaves of sunflower. The sunflowers used in this study are grown in different conditions such that the one in nittogen aboundant and the other in nitrogen deficient soil, respectively. The urea-N, ammonia-N, amide-N, and 80% alcohol soluble-N in the leaves were quantitatively determined. All of the nitrogenous components measured are generally tended to increased with rising the concentration of urea except only amide-N at 24 hours after sprayed, and these were highly significances. It seemed that hydrolizing of urea into ammonia and carbon dixide and the assimilation of ammonia into other organic nitrogenous constituents were rapid in the young leaves than in the mature. It is interest that the amide content, in the young leaves and nitrogen defieient one were enhanced with the increasing concentration of urea, although in the mature leaves it did not show any change in the urea treatment. It is presumed that the assimilation rate of ammonia and the urease activity were lower in the matture leaves than in the young and nitrogen deficient leaves. No significance at 5% level showed all of the nitrogenous components except total nitrogen between nitrogen abundant and deficent leaves. Urea content was a high peak at first 12 hours, ammonia at 48 hours, and amide and alcohol soluble nitrogen at 96 hours, whence decrease4d the content of these constituents gradually. The total nitrogen content is not incrased obviously by only one time of urea spray in this study. When the concentration of urea was relatively high there appeared the wilting spots on t도 edge of leaves. As a whole, it seemed that sprayed urea was rapidly absorbed and taken part in nitrogen metabolism within relatively short period.
The temporal and spatial analyses of total nitrogen (TN) fractionation were conducted in order to understand 1) total nitrogen components in streams and 2) their patterns in rainy and dry seasons. The result showed that the concentration of nitrogen components in stream water was lower in non-urban area and getting higher in urban area. Dissolved total nitrogen (DTN) was 95~97.7% of total nitrogen in streams, and the proportion of dissolved organic nitrogen (DON) and ammonia nitrogen ($NH_3-N$) was higher with increasing urban area. The concentration of total nitrogen and nitrate nitrogen ($NO_3-N$) were highest in winter among four seasons. The result was showed that concentration of $NH_3-N$ was same variation as concentrations of TN and $NO_3-N$ in urban-rural complex and urban areas, except rural areas. During rainy season, concentrations of particulate organic nitrogen (PON) and $NH_3-N$ increased in rural areas and decreased in both urban-rural complex and urban areas. Correlation between total nitrogen components and land uses was positively correlated with site > paddy, and negatively correlated with forest. The variation of total nitrogen concentration was determined by $NO_3-N$ in non-urban areas, by $NO_3-N$ and $NH_3-N$ in urban-rural complex and by $NH_3-N$ in the urban areas.
This study estimated the effect of hydraulic residence time(HRT), influent total ammonia nitrogen(TAN) concentration, temperature and pH in the packed-bed bioreactor using immobilized nitrifiers. Removal rate of ammonia nitrogen was increased with decreasing HRT and the optimum HRT was 0.2 hour when influent TAN was $2g/m^3$. At this point, removal rate was $226.1\;g/m^3{\cdot}day$ and removal efficiency was 88.8%. Removal rate of ammonia nitrogen was Increased with increasing TAN concentration. Removal rate and efficiency of ammonia nitrogen were kept constant at $20{\sim}35^{\circ}C$ and pH $8{\sim}9$ value.
This study investigated growth and hematological changes in parrotfish Oplegnathus fasciatus (~200 g/fish) reared under different total ammonia nitrogen (TAN) concentrations (0, 4, or 8 mg/L) for 6 weeks. Survival rates of parrotfish in all experimental groups did not significantly differ, as they were all ~100%. Although specific growth rate (SGR), weight gain, and daily feed intake in the high TAN concentration group (TAN8) were significantly lower than in the other two groups, there was no significant difference between the TAN4 group and the control group, (TAN0), indicating that parrotfish have a strong resistance to ammonia toxicity. As for temporal changes of the major blood components, cortisol increased as a result of stress caused by the high ammonia concentration in the TAN8 group. For this reason, the concentrations of energy sources such as glucose and total cholesterol were reduced. However, there was little difference among all experimental groups in concentrations of liver function glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT), and nutrient factors, such as total protein and albumin.
Nitrifier consortium immobilized in Ca and Ba-alginate beads were packed into two bioreactors and the performances of bioreactors were evaluated for the removal of ammonia nitrogen from synthetic aquaculture water. The total ammonia nitrogen (TAN) concentration of the influent was continually kept about 2g TAN/㎥. At the HRT of 0.6hr, ammonia nitrogen removal rate of two bioreactors were about 52.6 and 51.0g TAN/$\textrm{m}^3$/day, respectively. At the respect of ammonia nitrogen removal, two bioreactor showed the similar abilities. The second trial with nitrifier consortium immobilized in Ca-alginate bead was carried out to evaluate the ammonia nitrogen removal rate for 35 days. The highest ammonia nitrogen removal rate was 82g TAN/$\textrm{m}^3$ when HRT was about 0.3hr.
This study was conducted to determine the changes of nitrogen compounds in lamina and midrib during the curing process with the different curing methods, such as priming and stalk-cut curing. After KB 108 burley tobacco was cultivated by the different fetilization levels such as standard and 20% higher, only the tips and leaf were harvested. Though the contents of total alkaloid and total nitrogen were similar in lamina, midrib showed a very low contents of those components by the different curing method and fertilization levels. The content of nitrate-nitrogen in lamina increased during a middle of curing process, and then these compound was decreased during an end of curing process by stalk-cut curing method. Nitrate-nitrogen contents in the lamina by the priming curing showed a high level caused by rapid drying process during an end of curing process. That component in midrib was 5-6 times higher than that of lamina. The contents of ammonia-nitrogen in the lamina and midrib were increased during a curing process. Though those amount in tips showed a similar by a different fetilization and curing method, midrib of stalk-cut curing showed a slow increasing during a curing process.
Two in vivo experiments were conducted to evaluate the effect of novel urease inhibitor hydroquinone (HQ) on ammonia release rate from urea hydrolysis, nitrogen balance, nutrient digestibility and efficiency of microbial protein synthesis. In Exp. 1, twelve crossbred cannulated lambs were randomly assigned within initial body weight block to one of four HQ treatments, which included 0 (control), 30, 60 or 80 mg HQ/kg DM intake. Ammonia concentration and pH of ruminal fluid were immediately measured at 0, 2, 4, 6 and 8 h after feeding. Increasing the dose of HQ tended (p<0.15) to linearly decrease NH3 formation. The ammonia peak concentration (2 h post-feeding) in animals receiving HQ was approximately one-half of that in animals not receiving HQ (p<0.01), and a relatively sustained ammonia release could be obtained at the dose of 30 or 60 mg HQ/kg DM. In Exp. 2, sixteen intact crossbred lambs (weight $40{\pm}0.8kg$) were used in a $2{\times}2$ factorial design experiment. The four rations consisting of soybean meal-based (SBM) or urea-based (Urea) nitrogen source with or without HQ (S1, S0, U1 and U0) were fed in digestion and N balance trials. Apparent digestibility of major nutrients except that of ADF was not affected by either nitrogen source or addition of HQ. Regardless of nitrogen source, supplementation of HQ significantly improved ADF digestibility (p<0.05). The various ration had no effects on N metabolism in the presence of HQ. There was significant difference between total purine derivatives (PD), estimated efficiency of microbial N synthesis (p<0.05) and urea-N excretion (p<0.01) in the urine for the SBM ration and for the Urea ration. However, HQ had little influence on efficiency of microbial N synthesis as proportion of daily intake of total tract digestible OM (p>0.05). No interactions between main nitrogen source and HQ were measured throughout the trial. Results of this study suggest that addition of HQ to ration may improve ADF digestion with having no negative effect on N metabolism and microbial protein production.
Objective: An experiment was conducted to investigate the effects of a specific mixture of essential oils (MEO), containing thyme, clove and cinnamon EO, on rumen microbial fermentation, nutrient apparent digestibility and blood metabolites in fistulated sheep. Methods: Six sheep fitted with ruminal fistulas were used in a repeated measurement design with two 24-d periods to investigate the effect of adding MEO at 0 (control), 0.8, and 1.6 mL/d on apparent nutrient digestibility, rumen fermentation characteristics, rumen microbial population and blood chemical metabolites. Animals were fed with a 50:50 alfalfa hay:concentrate diet. Results: Ruminal pH, total volatile fatty acids (VFA) concentration, molar proportion of individual VFA, acetate: propionate ratio and methane production were not affected with MEO. Relative to the control, Small peptides plus amino acid nitrogen and large peptides nitrogen concentration in rumen fluid were not affected with MEO supplementation; while, rumen fluid ammonia nitrogen concentration at 0 and 6 h after morning feeding in sheep fed with 1.6 mL/d of MEO was lower (p<0.05) compared to the control and 0.8 mL/d of MEO. At 0 h after morning feeding, ammonia nitrogen concentration was higher (p<0.05) in sheep fed 0.8 mL/d of MEO relative to 1.6 mL/d and control diet. Ruminal protozoa and hyper ammonia producing (HAP) bacteria counts were not affected by addition of MEO in the diet. Relative to the control, no changes were observed in the red and white blood cells, hemoglobin, hematocrit, glucose, beta-hydroxybutyric acid, cholesterol, total protein, albumin, blood urea nitrogen and aspartate aminotransferase and alanine aminotransferase concentration. Apparent total tract digestibility of dry matter, crude proten, organic matter, and neutral detergent fiber were not influenced by MEO supplementation. Conclusion:The results of the present study suggested that supplementation of MEO may have limited effects on apparent nutrient digestibility, ruminal fermentation and protozoa and HAP bacteria count, blood cells and metabolites.
Budgets for water, nitrogen, and chemical oxygen demand (COD) were determined in two 0.012 ha earthy-bottom ponds stocked with Israeli strain common carp at an initial stocking density of $20\;fish/m^3$. Total ammonia nitrogen (TAN) concentrations increased continuously but later decreased in pond A as a consequence of high nitrification. COD concentrations increased during the experimental period due to the accumulation of feed input. Nutrient budgets showed that feed represented $94-95\%$ of nitrogen input and about 99% of organic matter input. Fish harvest accounted for $40\%$ of nitrogen and organic matter input. Seepage and water exchange removed $15-17\%$ of nitrogen input but only $1-2\%$ of organic matter. Draining of the ponds removed $20-26\%$ of input nitrogen, mostly in inorganic forms, but removed only minus organic matter. Fish and water column respiration accounted for $39\%$ of organic matter input, and benthic respiration accounted for $7-12\%$ of organic matter input. No significant change of nitrogen and organic matter in both pond bottoms were found during the three-month growth period. The unrecovered input nitrogen, about $6.3-13\%$, was lost through denitrification and ammonia volatilization. On a dry matter basis, fish growth removed $31\%$ of total feed input and left $69\%$ as metabolic wastes.
Nitrifier consortium entrapped in Ca-alginate bead were packed into aerated packed bed bioreactor and non aerated packed bed bioreactor and the performances of two bioreactors were evaluated for the removal of ammonia nitrogen from synthetic aquaculture water. Total ammonia nitrogen(TAN) removal rate was decrease in aerated packed bed bioreactor below 0.3hr of hydraulic residence time(HRT), but increased in non aerated packed bed bioreactor until 0.5hr of HRT. At HRT of 0.05hr, TAN removal rate of non aerated packed bed bioreactor was about 335g TAN/㎥/day and the optimum ratio of packing height and inside diameter of reactor (H/D) was 4. The performance of two bioreactors indicated that non aerated packed bed bioreactor was better than aerated packed bed bioreactor in ammonia removal from synthetic aquaculture water.
