  • The genus of Bacteroides has been isolated from vertebrate animal feces. Bacteroides sp. KGMB 02408 was isolated from fecal samples obtained from a healthy Korean. The wholegenome sequence of Bacteroides sp. KGMB 02408 was analyzed using the PacBio Sequel platform. The genome comprises a 5,771,427 bp chromosome with a G + C content of 39.50%, 5,005 total genes, 18 rRNA genes, and 74 tRNA genes. Furthermore, we found that strain KGMB 02408 had some genes for oxidoreductases and menaquinone biosynthesis in its genome based on the result of genome analysis.

  • We investigated occurrence and molecular phylogeny of the toxic epiphytic dinoflagellate Ostreopsis at seven sampling sites in the coastal waters off Jeju Island of Korea in April, 2017. During the sampling period, surface water temperature ranged from 15.7 to $18.3^{\circ}C$ and salinity was relatively constant, ranging from 33.4 to 34.9. Of a total of 13 macroalgal species collected from all sampling sites, Ostreopsis cells were observed from 8 macroalgal species and the highest cell abundance ($157.5cells\;g^{-1}$) was recorded on the red alga Grateloupia filicina at St. 6. LSU rDNA D8/D10 sequences of all Korean Ostreopsis strains isolated from the 4 sampling sites were 100% identical. Molecular phylogentic analyses (BI and ML) inferred from LSU rDNA alignment showed that the Korean Ostreopsis strains placed into the previously described the Ostreopsis sp. 1 clade, which contained strains isolated from the temperate coastal waters of Japan. The Korean Ostreopsis sp. 1 strain grew in a wide range of temperature ($10-30^{\circ}C$) and salinity (25-30), with its maximum growth rate of $0.49d^{-1}$ at $25^{\circ}C$ and salinity of 30, indicating that they can be tolerated in temperate areas.

  • The genus of Olsenella has been isolated from vertebrate animal mouth, rumen, and feces. Olsenella sp. KGMB 04489 was isolated from fecal samples obtained from a healthy Korean. The whole-genome sequence of Olsenella sp. KGMB 04489 was analyzed using the PacBio Sequel platform. The genome comprises a 2,108,034 bp chromosome with a G + C content of 65.50%, 1,838 total genes, 13 rRNA genes, and 52 tRNA genes. Also, we found that strain KGMB 04489 had some genes for hydrolysis enzymes, and antibiotic biosynthesis and resistance in its genome based on the result of genome analysis.

The Preventive Effect of Bacillus polyfermenticus KJS-2 and Bacillus mojavensis KJS-3 on Triton WR-1339-induced Hyperlipidemia (Triton WR-1339로 유도된 고지혈증에 대한 Bacillus polyfermenticus KJS-2와 Bacillus mojavensis KJS-3의 예방효과)

  • The purpose of this study was to evaluate the possibility that administration of Bacillus polyfermenticus KJS-2 (BP2), Bacillus mojavensis KJS-3 (Moja3), and their mixtures could control serum lipid levels. We observed changes in the blood cell level, metabolic function evaluation, and blood lipid levels after two weeks of oral administration of these microbial strains to hyperlipidemia-induced rats. Measurements of major cell changes in the white blood cells (WBC) indicated no significant effects due to the administration of the microbial strains. Platelet (PLT) levels decreased by 18.4% in the Triton WR-1339-treated group (NCON) and recovered to the control (CON) group levels in the positive control (PCON) group and the microbial strain-administered groups (p<0.05). No functional changes were observed in red blood cells (RBC) by Triton WR-1339-induced hyperlipidemia. The blood AST, ALT, BUN, and creatinine levels did not indicated effects on liver and kidney function, and all rats administered the microbial mixture recovered. The blood lipid levels in the microbe-treated groups indicated reduced levels of triglyceride (TG) and total cholesterol (TC), and increased levels of serum HDL cholesterol. The HMG-CoA inhibition rate of 7-O-succinyl macrolactin A (SMA) produced by BP2 showed similar activity at a concentration of 1,000 times lower than that achieved with atorvastatin. The administration of the microbial strains to the Triton WR-1339-induced rat model of hyperlipidemia resulted in reduced weight gain without affecting the food and water intake. Thus, blood circulation can be improved by controlling serum lipid levels by the combined administration of the BP2 and Moja3 microbial strains.

  • The impact protection system consists of an arrangement of circular sheet pile cofferdams-denoted dolphin structuredeeply embedded in the seabed, filled with crushed rock and closed at the top with a robust concrete cap. Centrifuge model tests were performed to investigation the behaviors of dolphins in this study. Total 7 quasi-model tests and 11 dynamic model tests were performed. The main experimental results can be summarized as follows. Firstly, The experimental force-displacement results for quasi-static tests show a limited influence on the initial stiffness of the structure from the change in fill density and the related change in the stiffness of the fill. And by comparing the dissipation at the same dolphin displacement it was found that the denser fill increase the dissipation by 16% for the 20m dolphin and by 23% for the 30m dolphin. The larger sensitivity for the large dolphin is explained by a larger contribution to the dissipation from strain in the fill. In low level impacts the dynamic force-response is up to 26~58% larger than the quasi-static and the dissipation response is showed larger in small displacement. Hence, it is concluded conservative to use the quasi-static response characteristics in the approximation of the response, and it is further concluded that the dolphin resistance to low level impacts is demonstrated to be equivalent and even superior to the high level impacts.

Numerical Study on Seismic Performance Evaluation of Circular Reinforced Concrete Piers Confined by Steel Plate (강판으로 보강된 원형철근콘크리트교각의 내진성능 평가에 관한 해석적 연구)

  • This study quantitatively evaluated the performance improvement of a circular reinforced concrete pier under dynamic load with strengthening using a steel plate. Various three-dimensional elements were applied using the finite element program ABAQUS. The analytical parameters included the ratios of the steel cover length to the pier's total height and the ratios of the steel cover thickness to the pier diameter for inelastic-nonlinear analysis. The lower part of the pier had fixed boundary conditions, and lateral repetitive loads were applied at the top of the pier. The pier was investigated to evaluate the dynamic performance based on the load-displacement curve, stress-strain curve, ductility, energy absorption capability, and energy ratio. The yield and ultimate loads of piers with steel covers increased by 3.76 times, and the energy absorption capability increased by 4 times due to the confinement effects caused by the steel plate. A plastic hinge part of the column with a steel plate improved the ductility, and the thicker the steel plate was, the greater the energy absorption capacity. This study shows that the reinforced pier should be improved in terms of the seismic performance.

Wrinkle Improvement, Whitening Effect of Boseong Camellia sinensis Vaccum Distilled Extract and Antioxidant, Antibacterial Efficacy of its Hydrothermal Extract (보성녹차 감압증류추출물의 주름개선, 미백 효능과 열수추출물의 항산화, 항균효과)

  • In this study, Camellia sinensis Leaf cultivated in Boseong was extracted by vacuum distillation and hydrothermal extraction methods, and we performed in vitro experiments such as antioxidant, anti-wrinkle, whitening, and antibacterial activity test and HPLC analysis. Wrinkle improvement (elastase inhibition assay) and whitening effect (mushroom tyrosinase inhibition assay) of Boseong vacuum distilled extract have excellent results and IC50 values of vacuum distillation extract was measured at a lower concentration than that of the hydrothermal extract. The antioxidant activity of the Boseong hydrothermal extract was excellently measured in DPPH radical and ABTS+ radical scavenging activity, SOD-like activity, and total polyphenol content. In the antibacterial experiment, the Boseong hydrothermal extract formed a clear zone for the Staphylococcus aureus and Cutibacterium acnes strain. As a result of HPLC analysis, EGCG and caffeine in the hydrothermal extract were 40.29 mg/g, 9.94 mg/g respectively, and caffeine in the vacuum distilled extract was 3.40 mg/g. Therefore, we examined that Boseong Camellia sinensis vacuum distilled extract has excellent anti-wrinkle and whitening effect, and its hydrothermal extract has good antioxidant and antibacterial efficacy.

Distribution of Toxin Genes and Antimicrobial Resistance of Vibrio parahaemolyticus Isolated from Seafood in Gwangju (광주지역 유통·판매 수산물에서 분리된 장염비브리오의 독소유전자 분포 및 항생제 내성 조사)

  • The purpose of this study is to investigate the distribution of toxin genes and antimicrobial resistance of Vibrio parahaemolyticus isolated from seafood in Gwangju. A total of 335 seafood, including 163 shellfish, 97 fish, and 36 mollusk, were tested in this study. As a result, V. parahaemolyticus was detected in 123 (36.7%) of 335 seafood. The tdh gene was not detected in all strains, while the trh gene was detected in 3 strains (2.4%). According to antimicrobial susceptibility test, 116 strains (94.3%) represent resistance to ampicillin, and 1 strain (0.8%) represents resistance to trimethoprim/sulfametoxazole. However, all strains were sensitive to 9 antimicrobial agents, including amikacin, chloramphenicol, tetracycline, and more. Therefore, the risk of V. parahaemolyticus isolated from seafood in Gwangju is considered low, but continuous monitoring of V. parahaemolyticus in seafood is required.

Antimicrobial resistance studies in staphylococci and streptococci isolated from cows with mastitis in Argentina

  • Background: Staphylococcus aureus and Streptococcus agalactiae are the main cause of clinical mastitis in dairy cattle in Argentina, whereas coagulase-negative staphylococci (CNS) and environmental streptococci are the main cause of subclinical mastitis. Bacteria isolated from infected animals show increasing antimicrobial resistance. Objectives: This study aims to determine the antimicrobial resistance of staphylococci and streptococci isolated from milk with mastitis, and to genotypically characterize the methicillin-resistant (MR) staphylococci. Methods: Isolation was performed on blood agar and identification was based on biochemical reactions. Antimicrobial susceptibility was according to the Clinical and Laboratory Standards Institute guidelines. The antimicrobial resistance genes, SCCmec type and spa type were detected by the polymerase chain reaction method. Results: We isolated a total of 185 staphylococci and 28 streptococci from 148 milk samples. Among the staphylococcal isolates, 154 were identified as CNS and 31 as S. aureus. Among the 154 CNS, 24.6% (n = 38) were resistant to penicillin, 14.9% (n = 23) to erythromycin, 17.5% (n = 27) to clindamycin, 6.5% (n = 10) to cefoxitin and oxacillin. Among the S. aureus isolates, 16.1% (n = 5) were resistant to penicillin, 3.2% (n = 1) to cefoxitin and oxacillin (MRSA). Six MR isolates (5 CNS and 1 MRSA) were positive to the mecA gene, and presented the SCCmec IVa. The MRSA strain presented the sequence type 83 and the spa type 002. Among the 28 streptococcal isolates, 14.3% (n = 4) were resistant to penicillin, 10.7% (n = 3) to erythromycin and 14.3% (n = 4) to clindamycin. Conclusions: The present findings of this study indicate a development of antimicrobial resistance in main bacteria isolated from cows with mastitis in Argentina.

Evaluation of Strength and Deformability of a Friction Material Based on True Triaxial Compression Tests (진삼축압축시험을 통한 마찰재료의 강도 및 변형 특성 평가)

  • Knowledge of the failure behavior of friction materials considering their intermediate principal stress is related to an understanding of situations where these materials might be used: for example, the stability of deep-seated boreholes and fault slip analysis. This study designed equipment for physically implementing true triaxial compression and used it to assess specimens of plaster, a friction material. The material's mechanical behaviors are discussed based on the results. The applicability of the 3D failure criteria are also reviewed. The tested specimens were molded cuboids of width, length, and height 52, 52, and 104 mm, respectively. A total of 24 true triaxial compression tests were performed under various combinations of 𝜎3 and 𝜎2 conditions. Conventional uniaxial and triaxial compression tests were employed to estimate the mechanical properties of the plaster for use as parameters for 3D failure criteria. Examining the stress-strain relations of the plaster materials showed that a large difference between the intermediate principal stress and the minimum principal stress indicated strong brittle behavior. The mechanical behavior of the plaster used here reflects the change of intermediate principal stress. Nonlinear multiple regression analysis on the test data in the principal space showed that the modified Wiebols-Cook failure criterion and the modified Lade failure criterion were the most suitable 3D failure criteria for the tested plaster.
