용수재이용을 위한 하수처리 유출수의 UV 소독 후 광회복 조사 (Photoreactivation Study of Wastewater Treatment Effluent Disinfected by UV-disinfection for Water Reuse)

  • Photoreactivation of microorganism following UV-disinfection is one of the research topics of interest in assessing the UV-disinfection performance. Apparent photoreactivation was examined under fluorescent lamp and solar radiation as well as in darkness. Total coliform, fecal coliform, and Escherichia coli were used as indicator microorganisms, and their concentration was monitored with time after UV-disinfection. Under the darkness, their initial concentration of 10∼30 MPN/100 mL increased to the level of 100 MPN/100 mL after 24 hours, which implied that part of damaged microorganisms by UV-disinfection might be repairable with time. Under the fluorescent lamp, photoreactivation was more apparent that their concentration increased up to 1,000 MPN/100 mL which might significantly impair the water uses specially in reuse of reclaimed wastewater. However, their concentration further decreased down to below 2 MPN/100 mL under the solar radiation primarily due to additional disinfection by solar radiation rather than photoreactivation. Samples not disinfected by UV-disinfection also demonstrated substantial decrease of their concentration under solar radiation from about 5,000 MPN/100 mL to less than 30 MPN/100 mL in 24 hours. But direct reuse of effluent without disinfection is not recommended because natural decay by solar radiation may take time and be affected by climatic conditions. The result suggests that photoreactivation of pathogenic microorganisms may not be concerned in agricultural reuse of reclaimed wastewater because solar radiation may provide further disinfection after UV-disinfection.

Feasibility of Reclaimed Wastewater and Waste Nutrient Solution for Crop Production in Korea

  • BACKGROUND: Development of water recycle technologies is important for human health and sustainable agriculture. However, few studies have been conducted to examine the purification methods or the water quality of reclaimed wastewater in Korea. METHODS AND RESULTS: In this study, the different wastewaters including reclaimed wastewater and waste nutrient solution (NS) were evaluated. The changes of water quality in reclaimed wastewater and NS were determined using ultraviolet (UV) treatment and sand filtration with charcoal. Our results showed that one of the most critical limitations of reusing wastewater was the presence of harmful pathogens that possibly cause human health risks. CONCLUSION(s): This study suggests that the application of UV treatment or combined with sand filtration on reclaimed wastewater and waste NS effectively removes the total coliform bacteria below the harmful or acceptable level. For future studies, a long-term field monitoring after applying reclaimed wastewater or NS is needed.

  • Effluent of wastewater treatment plant is to be disinfected to protect drinking water sources. DOF (Dissolved Ozone Flotation) was developed to meet this purpose. DOF was developed by combining DAF system with ozone. DAF system has good floating power with numerous microbubbles, and ozone has strong oxidation capability. And DOF system has good floating power and strong oxidation capability simultaneously. When DOF was applied to secondary wastewater effluent, color of 11CU in raw water which was secondary effluent was reduced to 1CU by the DOF system. Removal rate of other water quality parameters treated by DOF were also higher than that by DAF, which were proved the strength of oxidation capability of ozone. When ozone concentration of 3.3mg/l were applied in DOF system, general aerobic bacteria were reduced to 5CFU/ml from TNTC (Too many Numbers To Count). With the same ozone concentration, total coliform were not detected at all. These figures are under the numbers of drinking water regulation. These microbes were the target parameters of DOF. It was proved that DOF was very effective in disinfection of wastewater treatment plant effluent as well as in removal of color, turbidity, and T-P.

경작기 농경배수에 의한 오염물질의 배출 (Discharge of the Pollutants from Rice Paddies during the Period of Cultivation)

  • In this study, discharge of the pollutants from the rice paddies during cultivation and its pattern were investigated. The pH in the returned and rainfall water from ten different paddies was between 6.9 and 7.4, which is not associated with the cultivation time and farming style of individual paddy. TN and TP concentrations were highest in the beginning of first top dressing (fertilization), which indicates that proper top dressing and returned water management is crucial to reduce their discharge loads. In particular, TN concentration was more or less constant until tillering stage. This is thought to be due to the runoff of soil-adsorbed nitrogen and compost. Average SS concentration in the returned and rainfall water was 28 mg/L, and organics was predominantly soluble form, and highest COD 25 mg/L during transplanting, lowest 11.3 mg/L and average 11.3 mg/L. Density of TC (total coliform bacteria) was from 1000/100 mL to 114,000/100 mL and FC (fecal coliform) was 5 to 16% of TC density.

  • The current study demonstrates the ability of the marine cyanobacterium Oscillatoria willei BDU130511 to disinfect raw sewage. Within a holding time of 3 h under laboratory conditions, the organism drastically reduced in the total bacterial and coliform counts at various pH levels, in both unbuffered and buffered sewage, thereby suggesting a potential role for cyanobacteria in wastewater treatment.

충무김밥의 영양적 및 미생물적 품질 평가 (Nutritional and Microbiol Quality Assessment of Chungmukimbab)

  • 이 연구는 통영지방에서 전통적으로 만들어지고 있는 충무김밥의 영양적 및 미생물학적 품질평가를 시도하였다. 일반김밥 중에서는 열량, Ca, Fe, 비타민$B_l$ 및 비타민$B_2$등이 한국 성인 남성의 권장량에 비하여 낮은 것으로 나타났다. 따라서 영양권장량을 맞추기 위해서는 부식과 과일 및 음료 등을 함께 섭취하여야 할 것으로 사료되었다. 시장에서 구입한 일밥김밥과 충무김밥의 총 호기성균 및 대장균군 수는 일반 김밥에서 $5.50{\pm}0.38 log_{10}CFU/g,\;2.10{\pm}0.47 log_{10}$ MPN/100g, 충무김밥에서 $5.61{\pm}0.42 log_{10}CFU/g,\;1.75{\pm}0.34log_{10}$ MPN/100 g로 각각 나타났다. 충무김밥의 원재료에 대한 총호기성 균수는 김밥, 주꾸미무침, 무김치에서 $3{\sim}4 log_{10}CFU/g$, 어묵에서 $4{\sim}5log_{10}$ CFU/g를, 대장균군은 김밥, 주꾸미무침, 무김치에서 $1{\sim}2log_{10}CFU/g$, 어묵에서 $2{\sim}3log_{10}CFU/g$으로 분석되었다. 충무김밥의 재료별 대장균과 포도상 구균의 검출율은 김밥에서 각각 10.0%,주꾸미무침에서 15.0, 10.0%, 무김치에서는 0, 10.0%가 각각 검출되었다. 일반김밥과 충무김밥을 $15^{\circ}C$에 저장하여 하룻밤 방치한 후에 측정한 균수는 일반김밥과 충무김밥에서 생균수가 1.04 및 $0.60 log_{10}CFU/g$, 대장균군이 0.97 및 $0.72 log_{10}MPN/100g$이 각각 증가하였다. 충무김밥 재료의 24시간 후에 총호기성 균수의 증가는 $0.83{\sim}1.33 log_{10}CFU/g$증가되는 것으로 조사되었다.

전해수 처리에 의한 미역의 대장균군 억제효과 (Effect of Electrolyzed Water for Reducing Coliform Bacteria on Undaria pinnatifida)

  • 생미역에 고농도 장시간(50%, 75%, 90%/30분), 고농도 단시간(50%, 75%/1분, 2분), 저농도 장시간(15%, 30%/30분), 저농도 단시간(15%/5분, 10분)의 전해수를 처리하고 증류수로 세척한 뒤 미역의 전체와 표면의 생균수, 대장균군, 곰팡이수, 색도, 관능평가를 실시하였다. 고농도 단시간의 경우 미생물의 생육을 효과적으로 억제하였지만 관능적으로 낮은 점수를 받았고 고농도 단시간의 경우 관능적으로는 다른 처리구에 비해 비교적 우수하지만 대장균군이 검출되어 효과적이지 못한 것으로 나타났다. 저농도 장시간의 경우 대장균군의 생육을 억제시켰으나 장시간 침지에 의해 전해수취가 약간 남에 따라 보다 짧은 시간인 5분과 10분으로 전해수 침지 횟수를 달리하여 실험하였다. 15% 전해수 5분 1회 처리구의 경우 증류수 처리구에 비해 관능적으로는 선호도가 높았지만 미생물의 생육을 효과적으로 억제하지 못하였다. 결론적으로 15% 농도의 전해수를 10분간 1회 처리하는 것이 증류수 처리구와 비교 시 총균수, 대장균군수 및 곰팡이수를 약 1-2 log cycle 정도 억제함으로써 검출되지 않아 미생물 오염을 억제시키는데 효과적이며, 관능평가에서도 미역 원물 자체의 비린맛 및 비린향을 감소시킬 수 있어 최적의 조건임을 확인할 수 있었다. 이러한 연구결과로 미역을 생산단계에서 화학적 처리인 전해수 세척을 이용해 대장균군의 생육을 억제해 보다 위생적이고 안전한 제품을 생산할 수 있을 것으로 사료된다.

감마선을 이용한 녹즙의 위생화 (Sanitizing Effect of $\gamma$-Irradiation on Fresh Vegetable-extract Juices)

  • The sanitizing effect of irradiation on the fresh vegetable extract juices was investigated. Total bacteria, coliform bacteria and total ascorbic acid were determined during the storage periods at 4oC. Chlorophyll, carotenoid, tannin, electron donating ability and peroxidase activity were determined immediately after irradiation. Results showed that the viable cells were detected below the level of 105 CFU/ml during 12 days with doses of 3 and 5 kGy. Total ascorbic acid and tannin contents increased immediately after irradiation. However, irradiation didn't affect chlorophyll and car otenoid contents, electron donating ability, and peroxidase activity. It was considered that irradiation was effective in sanitizing fresh vegetable extract juices.

A Study of Frozen Desert Containing Canola and Soybean Oils as a Replacement for Milk Fat

  • This study was done to determine how added vegetable oils affect the physical, chemical and microbial properties of experimental frozen desserts. There were no differences in the percentages of total fat and total solids in the seven frozen desserts. Freezing points did not differ among treatments. There were significant differences in viscosity among the frozen desserts containing up to 80% of vegetable oils. Oil substitution at 30% or higher significantly decreased viscosity when compared to the milk fat control. The hardness of frozen desserts decreased significantly with increasing addition of oils. Oil substitution at 20% or higher significantly decreased hardness. Substitution of canola and soybean oils for milk fat increased melting rate. Initiation of fluid release in the control was slower than in frozen products with added oils. The SPC values and coliform counts of all frozen products were not significantly changed. Cholesterol content decreased significantly in the products as the vegetable oil content was raised. Frozen desserts containing 10% or 60% of vegetable oils in the total fat contained 91.8%, 73.5% or 32.5%, respectively, of the cholesterol in the control.

급식시설의 손 세척을 위한 70% 알콜 소독제 사용 및 세척방법의 적용효과 분석 (Effectiveness of 70% Alcohol Solution and Hand Washing Methods on Removing Transient Skin Bacteria in Foodservice Operation)

  • Hand washing is an important component of hygiene program for food handlers. Hands can be a source of direct or indirect contamination of foods with pathogenic microorganisms. In this study, the effectiveness of hand washing methods and the use of 70% alcohol solution against transient skin bacteria was tested in an university foodservice facility. 70% alcohol solution is sprayed for 5 seconds automatically when hands are placed in the dispenser. Samples were taken using swab technique in meat cutting area, vegetable trimming area, and vegetable cutting area: before and after washing hands according to planned methods, and after being sprayed with 70% alcohol solution after washing hands. The bacteriological analysis of total plate counts, coliform, fecal coliform of food handlers' hands was done. Statistical data analysis was completed with Mann-Whitney U test and Kruskal-Wallis model using the SPSS program. The levels of initial contamination of workers' hand were significantly different by the work areas($x^2$=9.156, p<0.01). Workers in the vegetable trimming area had more heavily soiled hands than in the other work areas. Mean of TPC counts and coliform was 8.97×$10^5$ CFU/12.4$cm^2$, 2.93×$10^2$ MPN/12.4$cm^2$ respectively, but fecal coliform was not detected. Transient bacteria were removed from hands after washing and using 70% alcohol solution but were not removed completely. Mean reduction percentage in TPC varied among work areas and ranged from 93.19% to 94.99%, and in coliforms from 97.31% to 100%. A significant difference in TPC was found between before and after hand disinfection (Z=-2.714, p<.01) and between standardized hand washing procedures and un-standardized hand washing procedures(z=-2.301, p<.01). Subjects using the hand sanitizer showed a great elimination of TPC(99.45% reduction), but this effect was valid only after following proper washing procedures. Based on the results, the most effective hand washing method was recommended as the combination of the standardized hand washing procedure with warm-water and use of the 70% alcohol solution. The results can be used to develop hand hygiene programs and training strategies for enhancing hand hygiene practices for food handlers in foodservice operations.
