Screening of New Bioactive Materials from Microbial Extracts of Soil Microorganism (I) Antimicrobial Activity from 200 Sampled Using Microdilution Assay

  • The microdilution assay recommended by NCCLS (National Committee for Clinical Laboratory Standards) is one of the standardized methods of antibiotic susceptibility test. This method has been widely used clinically to obtain MIC values of antibiotics on pathogenic microorganisms. It is more convenient, rapid and simple to test many samples than other test methods such as agar diffusion assay and broth macrodilution assay. The screening of antimicrobial agents from microbial extracts is too laborious in its process. Therefore, a number of screening methods having more simple procedure have been developed. In our laboratory, we applied microdilution assay for screening the antimicrobial agents. This assay showed dose-response results and was more sensitive than disc diffusion assay in our system. We tested 200 samples of microbial extracts originated from 100 microbial strains and selected several samples as potential candidates. In this report, we show that the microdilution assay is more convenient method in screeing of antibiotic susceptibility than those previously reported.

The Plant Cellular Systems for Plant Virus Movement

  • Plasmodesmata (PDs) are specialized intercellular channels that facilitate the exchange of various molecules, including sugars, ribonucleoprotein complexes, transcription factors, and mRNA. Their diameters, estimated to be 2.5 nm in the neck region, are too small to transfer viruses or viral genomes. Tobacco mosaic virus and Potexviruses are the most extensively studied viruses. In viruses, the movement protein (MP) is responsible for the PD gating that allows the intercellular movement of viral genomes. Various host factors interact with MP to regulate complicated mechanisms related to PD gating. Virus replication and assembly occur in viral replication complex (VRC) with membrane association, especially in the endoplasmic reticulum. VRC have a highly organized structure and are highly regulated by interactions among the various host factors, proteins encoded by the viral genome, and the viral genome. Virus trafficking requires host machineries, such as the cytoskeleton and the secretory systems. MP facilitates the virus replication and movement process. Despite the current level of understanding of virus movement, there are still many unknown and complex interactions between virus replication and virus movement. While numerous studies have been conducted to understand plant viruses with regards to cell-to-cell movement and replication, there are still many knowledge gaps. To study these interactions, adequate research tools must be used such as molecular, and biochemical techniques. Without such tools, virologists will not be able to gain an accurate or detailed understanding of the virus infection process.

Analysis of the Surface Characterisitics of Microstickies by Contact Angle Measurement (접촉각 측정을 통한 스틱키의 표면화학적 특성 분석)

  • Increase in the utilization rate of recycled paper has significantly increased the problem associated with stickies. Despite the effort to eliminate contaminants from recycled furnishes, stickies are still the most serious obstacle in using recycled paper. The amount of micro stickies that are too small to be eliminated by screening, tends to increase significantly as the closure level of white water system is increased and the quality of raw material deteriorates. To establish a process efficient in removing micro stickies is strongly required. In this study, the surface characteristic of micro stickies was investigated with contact angle measurement. Surface energies of MCC, PSA film and model micro stickies were 53.37 mN/m, 29.75mN/m, and 29.63mN/m, respectively. This indicates that the surface characteristic of MMS is very similar to PSA. Thus, solvent coating of PSA and evaporation of the solvent provided excellent model micro stickies for flotation experiment.

  • Military material-supply problem is one of the most important logistic problems under conscription system. Formally, two approaches were followed lowed to this problem. (1) Needs for material per soldier is estimated by past experience. The total demand for the material is estimated by multiplication of this coefficient and the number of soldiers given in the governmental programme. (2) The total demand for the material is estimated by the forcast based on the past statistics. The material supply system based on these estimates, however, relies too much on past statistics ;lack of flexibility is feared to adapt itself to changes in conscription programme, life-time of materials and so on. In this paper, the author has followed new approach : The conscription system itself is a linear input-output system, in which sequences of enlistment and dischargement are regarded as input and output. And the sequencial demands for the material are related by another linear transformation to the former sequences. In this regard z-transformation is applied to construct to transfer functions associated with this system. With these transfer functions, methods are established to determine the material demand corresponding to conscription programme and life-time distribution. Numerical methods by computers are also prepared.

  • This research provides practical data to develop UV protection masks and dust protection masks. It surveyed uncomfortable factors and significant characteristics for masks. It used 653 subjects 20-79 years old and performed the research from April to May in 2014. Wearing masks at outdoor activities is not widely popular yet, but golf players, mountaineers and old people were quite interested in masks. People mainly wore a basic shape mask, but main age group of each mask shape was different. People usually prioritized the protection function and comfort of wearing over design; however, women, young generation and people attending to outdoor activities longer than 1 hour considered design quite important. People going on picnic or camping consider mask characteristics most important. Therefore, various mask shapes and protective functions should be developed to reflect consumer needs. Meanwhile, the discomfort levels of masks were not too high, but old people felt more discomfort than young people. The most discomfort factors were 'other's eye', 'distracting face movement' and 'feeling of foreign object'. They also mentioned falling problem of C shape mask and short-rib shape mask, and asked for a 'tighter fit' at the ear.

4:2:1 compromise plans using Min-Max method (Min-Max 방법을 적용한 4:2:1 절충적 계획)

  • Testing high reliability devices under nomal operating condition is difficult, because the devices are not likely to fail in the relatively short time available for tests. For most applications it is necessary to accelerate the causes of failure by increasing a stress above its nomal value. Previous accelerated life test(ALT) plans have shown how to find optimum allocation, lowest stress and sample size subject to minimizing the variance of mean life estimator. In these ALT plans, the highest acceptable test-stress was assumed to be specified in advance by the experimenter but there is no guidance for selecting it. This assumption is, however, inappropriate for many applications. Testing devices at too-high stress levels can invalidate the extrapolation model, or introduce failure mechanisms that are not anticipated under nomal operating conditions. In this paper, we propose new 4:2:1 compromise plans using Min-Max method to minimize this risk and present minimized test-stress levels(max, middle, min), and find sample allocation based on Min-Max 4:2:1 compromise plans. In result, we compare previous 4:2:1 compromise plans specified maximum test-stress with Min-Max 4:2:1 compromise plans minimized maximum test-stress.

Study of Appropriate Meal Duration at an Italian Restaurant (이탈리안 레스토랑의 적절한 Meal Duration 선정에 관한 연구)

  • This study examined the different meal duration expectations and dining characteristics of customers each stage of service (order, cook, check), and investigated the impact of perceived meal duration on customers' satisfaction. The procedures consisted of four phases. During the first phase, the different meal duration expectations and dining characteristics of customers of an Italian restaurant in Seoul were examined by survey at each stage of service. The second phase investigated the impact of perceived meal duration on customer' satisfaction. The third phase compared perceived meal duration and actual meal duration. The last phase suggested appropriate meal duration for maximization of customer' satisfaction. We also examined the effects of meal time (lunch or dinner) on meal duration preferences. Customer satisfaction was decreased with longer perceived meal duration for the order and check stages and too short perceived meal duration for the cook stage. To determine appropriate meal duration for each service stage, the relationship between perceived and actual meal durations was examined. This study confirms that customers dining at restaurants preferred a longer cook time and that a shorter meal duration at dinner decreased satisfaction ratings more when compared to lunch.

The Recognition and Improvement of the Hwangnamppang Available to University Students in the Gyeongju Area (경주 지역 대학생의 황남빵에 대한 인식 및 개선 방향)

  • The purpose of this study was to investigate the recognition and need for improvement of the Hwangnamppang provided to university students in the Gyeongju area of Korea. To accomplish this, we surveyed 195 university students by questionnaire. The consumption frequency rate revealed that 49.7% of students consumed Hwangnamppang $1{\sim}2$ times a year, while 32.8% consumed it $2{\sim}3$ times per six months and 16.4% consumed it $1{\sim}3$ times a month. Furthermore, the results revealed that 45.6% of the students purchased Hwangnamppang to give as a present. Moreover, 55% of the respondents thought that Hwangnamppang was a suitable gift, but that it was too expensive. In addition, 47.7% of the respondents felt that there were not enough varieties of the product and 26.7% for factor that do not increase Hwangnamppang's consumption respond. Finally, 90% of the respondents felt that there was a need to diversify the red-bean paste used in Hwangnamppang, and felt that the best pieces contained chestnuts (3.66/5 score) or sweet potato (3.62/5 score). Overall, the results of this study revealed that Hwangnamppang requires further development to increase the varieties available and improve the overall acceptability.

A Study on the Cathodic Protection Design Optimization of Steel Piles for LNG Storage Tanks by Numerical Analysis (수치해석에 의한 LNG 저장탱크용 강관파일 전기방식 설계 최적화 연구)

  • For the longer service life of steel pile, cathodic protection is selected sometimes at corrosive environment. The cathodic protection design improvement was investigated in this study. The current demand for cathodic protection was calculated from the potentiostatic current monitoring of the steel specimen in the deaerated soil samples. In this study, the current distribution was studied using the Boundary Element Method (BEM) and the Finite Element Method (FEM) numerical analysis methods. The optimum layout of the anode was developed and confirmed by numerical analysis. Under the conventional design of the anode, the length of the anode hole is same as the pile length. We found that, at the bottom end of the pile, the current density is too high. When the anode hole length was 80% of the pile length, the current consumption at the end was reduced. The construction cost of anode hole drilling was decreased about 20%, as compared to the conventional design. Furthermore, the life of the anode materials could be extended by reducing the current consumption at the end section. Using this approach, the construction cost was reduced significantly without any under-protection area on the steel piles.

Chinese College Students Perception and Preference of Korean Kimchi (김치에 대한 중국 남녀 대학생의 인식 및 기호도 조사)

  • The purposes of this study are to investigate perception and preference of Chinese college students for Korean Kimchi by a survey and sensory evaluation and to globalize and industrialize Kimchi in the world. A questionnaire survey was conducted on 588 students residing in Beijing. About 80% of all the participants knew what Kimchi was and 63.4% had eaten Kimchi before the survey . On thier first impression of Kimchi, 43.6% of respondents, answered the nationality of Kimchi is impression after eating Kimchi was 'good'. 51.4% of respondents, answered the nationality of Kimchi is 'Korea'. The most liked Kimchi is Baechu Kimchi, and they preferred the white stem of the Chinese cabbage. On their perception of Kimchi,'Kimchi is a good side dish with cooked rice' was totally received point. The most well-known cooking method of Kimchi turned out to be Kimchi Chigae. As the results of sensory evaluation on several types cabbage Kimchies at room temperature, the most preferred Kimchi by the students was two-day old and made out of 5g red pepper. And two-day old Kimchi lOg red pepper and 2g shangchai added was preferred, too. But the six-day old Kimchi was disliked. The two-day old Kimchi made out of lOg red pepper preserved in the refrigerator was the most-liked. The dishes using Kimchi as the main ingredient are listed according to the students preference as follows Kimchi Hundun, Kimchi Ddungchai, Kimchi Bokum, and Kimchi Ramen, and so on.
