The Japanese common squid Todarodes pacificus is one of the fish species within the total allowable catch (TAC) system which requires further investigation. In this study, the acoustic survey method was used to analyze the distribution of the Japanese common squid Todarodes pacificus across all the seas of South Korea. The sea area within Korea was investigated using the research vessels 20, 21, and 22 of the National Institute of Fisheries Science. The acoustic surveys were carried out from July to September 2019 and February to May 2020. The acoustic systems used in the survey had frequencies of 38 kHz and 120 kHz (EK60, EK80, Simrad, Norway) of the split-beam scientific echosounder. The results showed that, in spring, 277 m2/nmi2 was the highest in the east sea area, and the same in the summer season 880 m2/nmi2 was the highest in the east sea area. In autumn, the highest nautical area scattering coefficient (NASC) value was observed in the coastal portion of the south sea, and in winter, the NASC values were generally low in all the sea area.
We have proposed a method for estimating the size-dependent mortality (M) of short-lived semelparous cephalopod species. To estimate the size-dependent mortality of a winter cohort of the common squid Todarodes pacificus, a commercially important cephalopod species in the North Pacific, we used the data and results of previous studies. On the basis of the bigger-is-better the steady-state hypothesis, we derived a theoretical mortality curve by assuming M as an inverse function of mantle length (ML). The derived constant for size-specific instantaneous mortality (q), assuming an embryonic survival rate of 86.6%, was 0.413 day-1 mm in ML. Estimates of life-stage-specific M ranged from 0.0280 to 0.435 day-1 for paralarvae, 0.00278 to 0.0269 day-1 for juveniles, 0.00197 to 0.00275 day-1 for pre-spawning adults, and 0.0913 to 0.0920 day-1 for post-spawning adults. Sensitivity analyses showed that the derived size-dependent M did not significantly change with varying embryonic survival rates, from 48%, the lowest reported value, to 100%. Additionally, comparison of the length frequencies derived from our simulations with those derived from catch data did not show a significant difference, suggesting that our approach and procedures are reliable for stock assessment and management of the common squid.
We investigated the effects of enrichment with oil or bacteria on the fatty acid composition of rotifers and Artemia as live prey. One enrichment(oil source) was mainly composed of squid Todarodes pacificus liver oil; the other(photosynthetic-bacterial source) was primarily made up of Schizochytrium sp. The enrichments were intended to enhance the nutritional value of the live prey, such as their EPA, DHA and n-3 HUFA contents. The lipid content as EPA and DHA of rotifers was higher when enriched with the oil source rather than the photosynthetic-bacterial source. The DHA content of Artemia nauplii after enrichment differed significantly, depending on the type of enrichment used(P<0.05). When the Artemia nauplii were enriched with the oil source, the DHA content was increased to 16.8%, whereas it increased only to 1.1% when enriched with the photosynthetic-bacterial source. These results indicate that selection of the enrichment is important for Artemia nauplii but not for rotifers.
Data on squid catches, water temperature, and climatic factors collected for the Northwest and subtropical North Pacific were analyzed to examine the influence of oceanic and climatic conditions in spawning grounds on catches of Japanese common squid, Todarodes pacificus, in the East (Japan) Sea. The main spawning ground was divided into four sub-areas: the South Sea of Korea (R1), the southern waters off Jeju, Korea (R2), the southwestern part of Kyushu, Japan (R3), and the northern part of Okinawa, Japan (R4). Interannual and decadal fluctuations in water temperatures correlated well with squid catches in the East/Japan Sea. In particular, water temperatures at a depth of 50 to 100 m in sub-areas R3 and R4 showed higher correlation coefficients (0.54 to 0.59, p<0.01) in relation to squid catches in the East/Japan Sea than for R1 and R2, which had correlation coefficients of 0.40 or less (p>0.05). Air temperature and wind velocity fluctuations in each sub-area are correlated with water temperature fluctuations and were closely connected with variations in the surface mixed layers. Water, air temperatures and wind velocities at the main spawning grounds are linked to the Southern Oscillation Index (SOI) with higher signals in the ca. 2-4-year band. Strong changes in a specific band and phase occurred around 1976/77 and 1986/87, coincident with changes in squid catches.
Cephalopods, such as the Japanese flying squid Todarodes pacificus and golden cuttlefish Sepia esculenta, are an important food source for all toothed whales, particularly bottlenose dolphins in costal and offshore waters around the Korean Peninsula. A controlled laboratory experiment was conducted to investigate the broadband acoustic backscattering from live individuals of these two cephalopod species using linear chirp signals (100-200 kHz). The backscattered echo signal was measured at about 1° intervals from -45° (head down) to +45° (head up) in the dorsal plane. The species-specific, frequency-dependent scattering characteristics were investigated by comparing the relationship between the wavelength-normalized backscattering cross-section (σ/λ2) and the wavelength-normalized fish length L/λ for each species. The estimated σ/λ2 value for Japanese flying squid was 9.51 at an L/λ range of 12.79-30.27 (mean, 21.26). This was approximately 7% of the σ/λ2 value (136.1) for golden cuttlefish at an L/λ range of 9.07-25.49 (mean, 15.77).
Analysis of stomach content for common squid was carried out to investigate the difference in seasonal and ontogenetic feeding behavior. Totals of 1368 common squid were collected monthly off Busan from September 2004 to August, 2005. Size range of common squid was 8.5-31.8 cm mantle length (ML) during the study period. Of the 1368 stomachs examined, 599 specimens (43.8%) were empty. Stomach contents indicated that common squid was carnivore: Large portion of stomach contents consisted of fish and mollusca (mainly common squid). The stomach contents index (SCI) seemed to increase with size, though there was no statistical difference between size classes significantly. Cannibalism was minimum in size range of 15-20 cm ML, but was steadily increased with size. Cephalopods were the major prey during spring and summer when common squid were small, but fish replaced it during autumn and winter when they became large.
In this study, an angiotensin I-converting enzyme (ACE) inhibitor from squid skin was purified and characterized. Squid (Todarodes pacificus) skin protein isolates were hydrolyzed using six commercial proteases: alcalase, ${\alpha}$-chymotrypsin, neutrase, papain, pepsin, and trypsin. The peptic hydrolysate had the highest ACE inhibitory activity. The ACE inhibitory peptide was purified using Sephadex G-25 column chromatography and reverse phase high-performance liquid chromatography (HPLC) with a $C_{18}$ column. The purified ACE inhibitory peptide was identified and sequenced, and found to consist of seven amino acid residues: Ser-Ala-Gly-Ser-Leu-Val-Pro (657Da). The $IC_{50}$ value of the purified ACE inhibitory peptide was 766.2 ${\mu}M$, and Lineweaver-Burk plots suggested that the purified peptide acts as a noncompetitive ACE inhibitor. These results suggest that the ACE inhibitory peptide purified from the peptic hydrolysate of squid skin may be of benefit in developing antihypertensive drugs and functional foods.
