Detection of Tobamoviruses and Survey on Contamination Rate in Commercial Pepper Seeds Using Gelatin Particle Agglutination Test (젤라틴입자응집반응법을 이용한 국내 시판 고추종자의 Tobamovirus 검출 및 오염률 조사)

  • Gelatin particle agglutination test (GPAT) was optimized for detection of Tobamovirus and contamination of the virus in commercial pepper seeds was evaluated. The optimum concentration of ${\gamma}$-globulin G, specific to tobacco mosaic virus pepper strain, was 100 ug/ml. The sensitivity of GPAT for the detection of Tobamovirus in pepper seeds was as high as enzyme-linked immunosorbent and dot immunoblotting assays. Optimum dilution ranges of the seed extract for GPAT was 5-25 folds. Using the optimized GPAT with above conditions, the rate of Tobamovirus contamination in seeds was turned out to be average of 79.1%.

Effects of Proton Beam Irradiation on Germination and Growth of Tobacco and Rice Plants (담배와 벼의 발아와 생장에 대한 Proton 빔조사의 영향)

  • Effects of proton beam irradiation on seed germination and growth pattern of tobacco (Nicotiana tabacum L. cv. BY-4; N. plumbaginifolia) and rice (Oryasativa L.) plants were estimated to develop the efficient conditions of irradiation. Seed germination rate was decreased by increasing the proton beam the current and the beam irradiation time in both tobacco and rice seeds. The beam irradiation conditions showing $50\%$ germination were over 60 sec at 10 nA, approximately 5 sec at 100 nA and at 500 nA beam current in tobacco seeds. And the conditions of $50\%$ germination were 60 sec at 10 nA, and 100 nA and 30 sec at 500nA in rice (cv. Dongjin 1) seeds. The growth of irradiated plants was decreased, but significant difference in morphological changes was not observed by the proton beam treatment. The proton beam is able to use as a mutagen, but some of the factors including beam size and beam detector-system must be established for efficient usage of the beam.

  • Influence of fungi living on the endocarp surface of depulped seeds of Panax ginseng on the dehiscence was investigated with a fungicidal treatment of the seeds and sterilization of the sand at the beginning of stratification. 1. Isolation frequency of the fungi living on the endocarp surface of de-pulped seed was reduced and hardness of the endocarp did not change significantly in seed treated with a fungicide Captan wp. 50%. A significant negative correlation $(r=-0.984^**)$ was found between the frequency of fungi isolation and the hardness of the endocarp. 2. Increase of water content in the seed treated with the fungicide was delayed 20days compared to the untreated. 3. Growth of the embryo and dehiscence of the seed was suppressed by the fungicide treatment. The length of the embryo was inversely proportional to the hardness of the seed. It is suggested that the fungi facilitate the softening of the endocarp thereby enhancing the supply of oxygen and water necessary for the embryo development, therefore, accelerate the growing of embryo and cause the dehiscence.

  • Anatomical changes in the forming and germinating processes of tobacco seeds were investigated to obtain basic information on the ecological characteristics of tobacco seeds. Seed development studied through the longitudinal section of the fertilized ovule clarified that the cell division of the zygote was initiated after 7 days of flowering. After 12 days of flowering, perfect seed constituents such as cotyledon, epicotyle and radicle were formed and those were expressed to recognizable level of germinability. After 15 days of flowering germination rate reached higher than 30% and 17 and 21 days after flowering a perfect seed which have 70% or higher germinability were produced. Seed size was ranged between 0.3-0.6 mm and varietal differences were noted in the given seed size range. Under the light treatment, the morphological changes were observed by elongation of radicle after 2 days of imbibition and apparent germination after 3 days of imbibition. But no responses of the seeds imbibed 6 days under the dark condition were observed.

  • Though higher plants car not metabolize D-amino acid, many prokaryotes and eukaryotes have the D-amino acid metabolism. Therefore, we transformed tobacco plants with D-amino acid oxidase (DAO), which can metabolize D-amino acid, and confirmed that transgenic tobacco plants might metabolize D-amino acid. Transgenic tobacco plants were survived a high concentration of D-serine, however non-transgenic plants were not grown on D-serine medium. From Southern and Northern blot analysis, transgenic tobacco plants selected on D-serine medium were confirmed by insert and expression of transgene. $T_{1}$ tobacco seeds derived $T_{0}$ tobacco plants selfing were grown on D-serine medium and showed normal phenotype compared to wild tobacco plants. Transgenic tobacco plants displayed the metabolic capability of D-serine. Therefore, we suggested that DAO is useful selectable marker gene for plant transformation.

Development of Basta Resistant Tobacco Using Artificial Phosphinothricin Acetyltransferase Gene (인공합성 Phosphinothricin Acetyltransferase 유전자에 의한 Basta 내성 연초식물체의 개발)

  • This experiment was conducted to introduce phosphinothricin acetyl -transferase(PAT) gene, resistant to basta and non-selective herbidide, into tobacco(Nicotiana tabacum cv.BY4). For shoot formation,tobacco leaf disks were placed on the MS medium supplemented with 2.0mg/L BA and 0.1mg/L NAA. In this medium condition, tobacco leaf disces were cocultivated with A. tumefaciens MP90 containing NPT IIand PAT resistant to kanamycin and Basta, respectively. Shoots were obtained in the medium containing antibiotics, and those were transferred to rooting medium supplemented with 0.1mg/L NAA and antibiotics. The plants obtaining roots were transplanted into soil. Phenotype of transgenic tobacco plant was mostly as normal plant. However, about 5% was abnormal plant, which did not set seeds. PCR analysis and southern blot were performed to determine transformation. As the results, it was confirmed that PAT gene was stably integrated into tobacco genome.When herbicide, basta, was sprayed to the plants confirmed by PCR, the transgenic plants showed normal growth, whereas normal plants died. Therefore, the result of this experiment show that tobacco transformation for the resistance to basta, non-selective herbicide, was successful because PAT gene was stably integrated into tobacco.

  • The development of selectable markers for transformation has been a major factor in the successful genetic manipulation of plant. We established a new selectable marker system for tobacco transformation using chimeric adenosine deaminase (ADA) gene, which confers resistance to cytotoxic adenosine analogues, 9-$\beta$-D-arabinofuranosyl adenine(Ara-A) and cordycepin. The transformants with the chimeric ADA gene in tobacco grew in the presence of normally lethal level of cytotoxic adenosine analogues, 100 $\mu$M Ara-A and 50 $\mu$M cordycepin. We successfully distinguished transformed shoot from non-transformed shoot on the same selectable media with cytotoxic adenosine analogues. In this selectable media, we were able to select seeds with/ without ADA gene from transgenic tobacco seeds. Theses results show that the mammalian ADA gene may serve as a new selectable marker for tobacco transformation.

  • Nicotiana Tabacum L., cv. NC2326, Burley2l and Sohyang seeds were coated with different five materials - zeolite, serpentine, coal briquet ash, serpentine+zeolite and coal briquet ash+zeolite - to investigate the effects of coating on germination. The coating materials had different characteristics and gave varied effects on germination. Among three tobacco varieties, coated seeds of Burley2l showed the most sensitive responses in germination rate of decreasing and delaying at low temperature on seedbed and laboratory tests.

Resistance Characteristics of Flue-cured Tobacco Plants Transformed with CDNA of Potato Virus Y Replicase Gene (감자 바이러스 Y 복제유전자 cDNA로 형질전환된 황색종 담배의 저항성 특성)

  • A flue-cured tobacco variety (Nicotiana tabacum cv. Wisconsin) was used for Plant transformation with the complementary DNA (cDNA) of potato virus Y-necrosis strain (PVY-VN) replicase gone (Nb) which was synthesized through reverse-transcription Primed with oligo(dT) and Polymerization using RNase H-digested template. The cDNA was cloned into Plant expression vector Plasmid (PMBP2), and introduced into tobacco plants by co-culturing tobacco leaf disks with Agrobacterium tumefaciens LBA4404 containing the plasmid before Plant regeneration. Eight Plants, in which the inserted cDNA fragment was detected by Polymerase chain reaction (PCR), out of 70 putative transformants inserted with sense-oriented Mb cDNA showed no symptom at 3 weeks after inoculation, while the other 62 plants, and all plants with vector gone only and antisense-oriented NIb cDNA had susceptible vein-necrosis symptoms. However, only 2 of the 8 resistant plants were highly resistant, which remained symptomless up to 10 weeks after inoculation. Among the first progenies (T1) from self-fertilized seeds of the two resistant transgenic plants, less than 10 % of 71 plants appeared highly resistant (with no symptom), 70% moderately resistant (with mild symptoms on 1 - 2 leaves), and about 20% susceptible (with susceptible symptoms on 3 or more leaves) at 3 weeks after inoculation. These results suggest that the PVY resistance was inherited in the 71 generation. Key words : potato virus Y. viral replicase gene, transgenic tobacco Plants, resistance.

Effects of Pre-sowing Treatments of Tobacco Seed on Germination and Seedling Uniformity (담배종자의 파종 전처리가 발아 및 묘의 균일성에 미치는 영향)

  • Pre-sowing treatments of tobacco seeds (cv. NC82) were evaluated as a means of improving the speed of emergence, early seedling growth and uniformity of the seedlings. Seeds were treated through the use of polyethylene glycol 6000(PEG), solid matrix priming(SMP), chilling and dark-preincubati(DPI). Under the laboratory condition of 15$^{\circ}C$, all the treated seeds germinated more rapid and the time to 50% germination(T50) reduced than control. Among the treatments, SMP was the best to reduce the T50 for germination. All the treatments did not affect the final germination percentage, while the chilling showed of little benefit for germination and T50. Under the greenhouse condition, seedling from SMP emerged earlier and more uniformity and also greater mean fresh and dry weight showed than other treatments. The advantage of T50 from SMP was affected more in temperature than in water content of Agro-Lig during the treatment.
