Stable Expression of TMV Resistance and Responses to Major Tobacco Diseases in the Fifth Generation of TMV CP Transgenic Tobacco

  • TMV resistant lines (TRLs) originated from the Blo plant of Nicotiana tabacum cv. NC82 transformed with TMV coat protein cDNA which initially showed delayed disease symptom were selected for increased resistance in each subsequent generation. The result of field experiment of the transgenic tobacco lines in the fifth generation for TMV resistance and their response to other tobacco diseases (black shank, bacterial wilt, and powdery mildew) is described in this report. When fifteen TRLs of the fifth generation were tested for TMV resistance by mechanically inoculating the individual plants, over 95 percent of the plants of 6 lines showed complete resistance even 8 weeks after the inoculation. Average frequency of the resistant plants in TRLs of the fifth generation 8 weeks after the inoculation was 87%. Stable insertion and expression of TMV coat protein cDNA in the fifth generation of the transgenic tobacco plant were confirmed by PCR and immunoblot hybridization, respectively. All TRLs were resistant to the black shank but were susceptible to the bacterial wilt disease and the powdery mildew to the same degree as non-transgenic NC82 was. Therefore, it was indicated that the phenotypes related at least to disease resistance were not changed in the transgenic tobacco. Disease Occurrence on Tobacco Plants Surveyed in the Northern Kyeongbuk Area from 1993 to 1994 (1993~1994년도 경북북부지역 담배병해)

  • Main tobacco diseases were surveyed on tobacco plants in the northern Kyeongbuk area to evaluate the effectiveness of current disease control techniques from 1993 to 1994. The major disease in seedlings was the anthracnose (Colletotrichum tabacum) but the mosaic (TMV) in adult plants. Wild fire (Psedomonas syringae pv. tabaci) and hollow stalk (Erwinia carotovora subsp. carotovora) were more severe in 1993 than those in 1994 because of more rain. Establishment of drain-outlet at the bottom of the temporary transplanting pot gave a significant reduction in anthracnose severity. Most farmers have ignored the protection effect of using milk during transplanting and the removal of diseased roots from the fields on mosaic (TMV). It was considered that the educating program on disease diagnosis and fungicide application might help farmers for the proper use of chemicals.

  • The significance of soil and/or rhisosphere populations of Erwinia carotovora sobsp. carotovora (Ecc) as a source of primary inoculum for tobacco hollow stalk disease has been demonstrated conclusively. The survival of Ecc in field soils fter overwintering was estimated by using the enrichment technique. The population number of pectolytic erwinia (PE) in field soils relatively decreased at the rate of 102-104 colony forming unit(CFU) per g of soil after overwintering. Higher level of PE population overwintered in the rhlzosphere foils of tobacco stubbles and detected more frequently in rhizosphere soils of weed plants than in those of bare fields. All of the tobacco stubbles collected from fields where tobacco had been grown the previous year contained Ecc. The more survived population number of PE at the 30cm depth of artifitiany infested soils than at the upper of those by introducing with diseased tobacco plant tissue after overwintering. Ecc overwintered effectively in rhizosphere soils of tobacco stubbles, overwintered weeds and tobacco debris in field soils.

  • Major diseases of tobacco and farmer's control practices were surveyed in the Kyeongbuk province area from 2005 to 2006. Mosaic caused by potato virus Y and bacterial wilt caused by Ralstonia solanacearum were most severe during the harvest season. Compared with the disease occurrence of ten years ago, the damage by tobacco mosaic virus reduced but bacterial wilt increased. These changes in the disease occurrences may probably be due to releasing the resistant tobacco cultivar to the mosaic virus but susceptible to the bacterial wilt pathogen. More than thirty percentage of the farmers have misused fungicides and also have applied the continuous mono-cropping system for more than ten years, and have chosen the incorrect crops for the rotation.

  • For crops that are planted in a lattice layout, sampling designs can be made to take advantage of this regular arrangement. In order to select which tobacco plants to be examined in a survey to estimate disease loss in tobacco a method of, so called, bent plots was devised based on the regularity of plantings in the tobacco fields. We will first describe this sample selection and measurement method and then provide estimators and their bias and variance properties.

Additive effect of tobacco waste in substrates on yield and mycelial growth of oyster mushroom, Pleurotus spp. and Trichoderma disease (느타리버섯 배지에 담배가루의 처리가 버섯균과 푸른 곰팡이병원균의 균사생장과 수량에 미치는 영향)

  • This study was carried out to investigate effect of adding tobacco waste to promote oyster mushroom, Pleurotus ostreatus, mycelial growth and control of Trichoderma disease. Tobacco waste was treated on the surface of rice straw inoculated with oyster mushroom strains. The mycelial growth treated with 0.1g tobacco waste was somewhat faster than non-treatment. In the case of mixed media with 1.0g tobacco waste, mycelial growth was increased compare to control, but adding 2.0 g tobacco waste decreased the mycelial growth. We also treated the tobacco waste on Trichoderma disease. All of the tobacco waste treatment force down the growth of Trichoderma disease. We treated tobacco waste on the surface of rice straw substrates. ASI 2072 mycelial growth was decreased regardless of amount of tobacco waster and method of treatment. Mycelial growth of ASI 2180 was somewhat decreased with treatment of 0.1g and 0.2g In the case of media mixed, mycelial growth of ASI 2072 was decreased in the overall treatment. Mycelial growth of other strains were decreased as only 2.0g treatment. Field test of mycelial growth was conducted using rice straw and waste cotton with substrates. Rice straw culture with treated 3% tobacco waste and cotton waste culture with treated 6% tobacco waste showed the highest yield. There was no effect to Trichoderma disease.

Quitline Activity in China

  • In order to help smokers quit easier, China has started to provide quitline service since 2004. There are two models for Chinese quitline service-the National Quitline Model, which provides only cessation service to smokers, and the 12320 Hotline Model, which integrates cessation counseling into public health hotline service and is currently adapted in public health hotlines in 28 provinces. A protocol of 4 counseling calls is used by 12320 Hotline. Three-month abstinence rate for clients is about 20%. The fact that most smokers who attempted quit don't seek cessation help or quitline service is not well known by the public are major constraints for quitline service in China. Effective advocating campaign should be implemented to propagate quitline. Diverse protocols targeting different subpopulation will also need to be developed to better service the public.

  • Tobacco abuse is a major preventable cause of premature death and disease, including various cancers. The Global Adult Tobacco Survey India (GATS) 2009-10 revealed that more than one-third of adults use tobacco in one form or the other. Nearly two in five smokers and smokeless tobacco users made attempts to quit the habit in the past 12 months. Tobacco dependence is a chronic condition characterized by susceptibility of relapse over years. It can be well handled by sustained professional support from health care providers mainly through behavioral counseling and pharmacotherapy. Dental professionals can play a pivotal role in diagnosing and effectively managing tobacco dependence. Dental Institutions have rapidly grown in last two decades across the country and so has the curriculum been adapted to improve student competencies to accommodate changing disease patterns and technological advances, but not in regard to tobacco cessation. Untapped dental manpower like undergraduates, dental hygienists and other paramedical staff need effective training to be more penetrative. The present review paper explores the potential role of dental training institutions and recommends various approaches to counter public health jeopardy of tobacco related diseases.

  • This experiment was conducted to examine characteristics of agronomic characters and estimate of gene effect for several mutant characters. The genetic populations were derived from cross between 83H-5 and Hicks. There were significant difference for plant height, stlk height, leaf shape and bacterial wilt disease index except leaf number, leaf length, and what is more, F3 variance is more than Bl and B2 generation from cross 83H-5 X Hicks. Gene actions for stalk height and bacterial wilt disease were estimated by 3-parameter, and by 6- parameter model for all characters except above two characters but stalk height and bacterial wilt disease index are not significant in the additive and dominance effects. Dominant$\times$dominant epitasis for plant height, dominant and dominant$\times$dominant epistasis for leaf length, additive and additive$\times$additive and dominant$\times$dominant epistasis for leaf width, and additive and additive$\times$dominant epistasis for days to flower were appeared significant in gene action.

Stueies on the Occurrence of Seedling Rot of Burley Tobacco Transplants Caused By Pyhium spp. and Its Control Measures in Field. (버어리종 담배 포장에서의 가칭 담배 묘썩음병(Pythim spp.) 발생 및 방제에 관한 연구)

  • Seedling rot occurred on burley tobacco at the early growing stage after transplanting was surveyed in the Chonbuk province, the main area for burley tobacco production, during the 3 crop years from 1986. The incidence of disease was about 0.6-0.7% every year and major causal organisms were Pythium spp. More diseased plants were observed in the fields in which tobacco planted with improved mulching system than in that with any other mulching system. The temperature and relative humidity in the planting hole covered with polyethylene film for 11 days after transplanting were recorded by 27-45$^{\circ}C$ and 75~95%, respectively. The punched polyethylene film mulching field showed less diseased plants than that with improved mucking. In improved mulching system the longer period of mulching caused higher incidence of seedling rot(r=0.74**). Soil drench treatment of 200m1 of aqueous Metalaxyl Solution(125ppm) was effective to control the disease.
