Immunocytochemistry utilizes the specificity of the antigen-antibody reaction to localize specific antigens in cells or cellular organelles. Here we report the use of monoclonal antibodies, in conjunction with gold-labeled second antibodies to study the ultrastructural localization and tissue distribution of the Mr 98, 000 anionic peroxidase and other wall antigens. The antibody specific for this wall peroxidase, mWP3, labeled mainly the cell wall area. At the tissue level, the Mr 98, 000 peroxidase is located predominantly in the leaf mesophyll, internal coleoptile and sieve elements, but not in the root, as assayed with these procedures. The coleoptile walls were less heavily stained than the walls of leaf mesophyll cells. At the subcellular level, it is localized mainly in intercellular regions of the cell walls. A similar staining pattern was revealed by mWP19, one of anti-$\beta$ glucosidase antibody, though it looked less heavily stained than one with mWP3. In order to serve as a control wall staining using IgM monoclonal antibodies, mWP18 was used. Most of the label is localized over wall regions of cells of the young leaf mesophyll and coleoptile.

Tissue transglutaminase (TGII, $G{\alpha}h$) belongs to a family of enzymes which catalyze post-translational modification of proteins by forming isopeptides via $Ca^{2+}$-dependent reaction. Although TGII-mediated formation of isopeptides has been implicated to play a role in a variety of cellular processes, the physiological function of TGII remains unclear. In addition to this Tease activity, TGII is a guanosine triphosphatase (GTPase) which binds and hydrolyzes GTP It is now well recognized that the GTPase action of TGII regulates a receptor-mediated transmembrane signaling, functioning as a signal transducer of the receptor. This TGII function signifies that TGII is a new class of GTP-binding regulatory protein (G-protein) that differs from "Classical" heterotrimeric G-proteins. Regulation of enzyme is an important biological process for maintaining cell integrity. This review summarizes the recent development in regulation of TGII that may help for the better understanding of this unique enzyme. Since activation and inactivation of GTPase of TGII are similar to the heterotrimeric G-proteins, the regulation of heterotrimeric G-protein in the transmembrane signaling is also discussed.

A new method for micro-quantification of abscisic acid from plant tissue is described. GLC-ECD method of abscisic acid pertafluorobenzyl ester synthesized by reaction of plant extract with pentafluorobenzyl bromide showed higher sensitivity than that of abscisic acid methyl ester, and the detection limit of abscisic acid by the described method was as low as 5 pg.

Determination of sex origin of cattle meat by fast and reliable molecular methods is an important measure to ensure correct allocation of export refunds particularly in European countries and also female cattle (cow) slaughter is legally banned in India because of religious beliefs. Based on the DEAD box protein gene located on the X and Y chromosomes, 2 pair of primers were designed and the system of PCR was optimized. Upon PCR amplification, male tissue showed 2 bands, while female tissue resulted in only one band. The accuracy and specificity of the primers was assessed using DNA template extracted from cattle meat of known sex. The protocol was subjected to a blind test and showed 100% concordance, proving its accuracy and reliability.

The bone scan reflects the metabolic reaction of bone to a disease process, whether neoplastic, traumatic or inflammatory. In normal bone tissue, the labeled diphosphate complexes accumulate in the perivascular fluid next to the marrow cavity at the interface between uncalcified and calcified bone matrix. HMDP has the most rapid plasma clearance among the commonly used diphosphonate followed by MDP and then HEDP. I have studied about bone scan time by use of $^{99m}Tc-MDP$, and got 336 images from 112 patients. The result obtained as follow; 1. Accumulation rate of $^{99m}Tc-MDP$ is higher than other age groups in below 30-year old group. 2. Accumulation rate, in 10year old group, is 75.85% on 120min. but other groups are most high on 180 min. 3. The density differants between bone and soft tissue increased with time. 4. Image contrast is good in younger group than old.

This experiment was carried out to establish the immunohistochemical diagnostic method for infectious pancreatic necrosis in the monolayers of CHSE-214 cell cultures and paraffin-embedded tissue sections from rainbow trout infected with infectious pancreatic necrosis virus(IPNV). Specific identification of IPNV antigens was often demonstrated in the pancreatic exocrine cells, and less in the intestinal mucous epithelia and the renal hemopoietic tissues by the use of monoclonal antibodies against capsid protein VP2. The specific reaction was seen as a distinct red cytoplasmic color, often as small granules of various sizes. The result showed that streptavidin alkaline phosphatase immunohistochemisry specifically identified IPNV antigens in both infected cell cultures and tissue sections.

Three mature layer chickens from a farm in which chickens showed green diarrhea, cyanosis, lethargy, loss of appetite were pathologically examined. Grossly, multiple variable sized caseous nodules were detected in the liver, intestinal serosa and mesentery. In addition, parathypoid nodules in the liver and fibrous serositis on the several peritoneal organs and tissues were noticed. One of spleens had multiple infarction areas. Histologically caseous nodules consisted of central caseous core and peripheral epithelioid cells overlying the fibrous connective tissue. Multinucleated giant cells were scattered between the epithelioid cells and fibrous connective tissue. In these nodules Gram negative cocobacilus bacterial colonies were present, whereas Periodic Schiff reaction and Ziehl-Neelsen stain detected neither fungi nor acid fast bacteria. From these results multiple granulomas might be induced by Escherichia coli. In addition, severe Ascafdiodf and Salmonellosis were coinfected in these chickens.

Various tissue and follicular components were analyzed for the determination of lactate dehydrogenase(LDH) isozyme patterns by electrophoretic technique with chromogen reaction in the pig. Optimum conditions for the tissue homogenate and the storage were finally established. Small quantities of follicular components were analysed for typing of LDH isozymes by microelectrophoresis. Microelectrophoretic analysis showed that only LDH-1 was visible in the oocytes, all isozymes in cumulus masses, and LDH-1, 2 and 3 in follicular fluid. The results provide critical information on the LDH activity of various tissues and follicular components. Furthermore, t he developed methods should be useful the analysis of LDH in the small quantity of samples, especially in the oocyte, and easily applicable to the oocyte and early embryos of other domestic species.

An experimental study was performed to observe the epithelial-mesenchymal interacion in the developing odontogenic tissue of 10-18 days rat embryos. Several results were obtained with the histochemical study of this experimentss. 1. Through invagination of dental lamina, P A S mucicarmine staining of middle and basal cells was shown to have weak reaction. but in mesenchymal cells, these staining was reacted with diffus form. 2. In accordance with the differentiation of enamel organ, these odontogenic tissue was being to form mesenchymal condensation, it was alse shown that the intermesenchymal cell was stained deeply with colloidal iron and alcian blue.

60 cases of swine and bovine ovaries were investigated for detecting the homosiderin pigment by means of Perl's prussian blue reaction. The results abtained were as follows; 1. Among the 30 cases of normal adult swine ovaries, 20 cases (66.6%) were presented as positive. 2. The hemosiderin pigment was detected in corpus albicans, connective tissue of corpus luteum, theca externa of follicles and interstitial tissue of ovary in swine. 3. The morphological characteristics of the hemosiderin pigment of swine ovaries were fine and coarse granules or amorphous and homogenous pigment. 4. In 30 cases of bovine ovaries, the hemosiderin pigment was not detected.
