Purpose: The aim of this study was to investigate the potential effects of mild hypoxia in the mature and immature brain. Methods: We prepared organotypic slice cultures of the hippocampus and used hippocampal tissue cultures at 7 and 14 days in vitro (DIV) to represent the immature and mature brain, respectively. Tissue cultures were exposed to 10% oxygen for 60 minutes. Twenty-four hours after this hypoxic insult, propidium iodide fluorescence images were obtained, and the damaged areas in the cornu ammonis 1 (CA1), CA3, and dentate gyrus (DG) were measured using image analysis. Results: In the 7-DIV group compared to control tissue, hypoxia-exposed tissue showed decreased damage in two regions (CA1: $5.59%{\pm}2.99%$ vs. $4.80%{\pm}1.37%$, P=0.900; DG: $33.88%{\pm}12.53%$ vs. $15.98%{\pm}2.37%$, P=0.166), but this decrease was not statistically significant. In the 14-DIV group, hypoxia-exposed tissue showed decreased damage compared to control tissues; this decrease was not significant in the CA3 ($24.51%{\pm}6.05%$ vs. $18.31%{\pm}3.28%$, P=0.373) or DG ($15.72%{\pm}3.47%$ vs. $9.91%{\pm}2.11%$, P=0.134), but was significant in the CA1 ($50.91%{\pm}5.90%$ vs. $32.30%{\pm}3.34%$, P=0.004). Conclusion: Although only CA1 tissues cultured for 14 DIV showed significantly less damage after exposure to hypoxia, the other tissues examined in this study showed a tendency towards less damage after hypoxic exposure. Therefore, mild hypoxia might play a protective role in the brain.
The present study examined the inhibitory effect of licorice compounds glycyrrhizin and a metabolite $18{\beta}$-glycyrrhetinic acid on the neurotoxicity of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in the mouse and on the 1-methyl-4-phenylpyridinium ($MPP^+$)-induced cell death in differentiated PC12 cells. MPTP treatment increased the activities of total superoxide dismutase, catalase and glutathione peroxidase and the levels of malondialdehyde and carbonyls in the brain compared to control mouse brain. Co-administration of glycyrrhizin (16.8 mg/kg) attenuated the MPTP effect on the enzyme activities and formation of tissue peroxidation products. In vitro assay, licorice compounds attenuated the $MPP^+$-induced cell death and caspase-3 activation in PC12 cells. Glycyrrhizin up to $100{\mu}M$ significantly attenuated the toxicity of $MPP^+$. Meanwhile, $18{\beta}$-glycyrrhetinic acid showed a maximum inhibitory effect at $10{\mu}M$; beyond this concentration the inhibitory effect declined. Glycyrrhizin and $18{\beta}$-glycyrrhetinic acid attenuated the hydrogen peroxide- or nitrogen species-induced cell death. Results from this study indicate that glycyrrhizin may attenuate brain tissue damage in mice treated with MPTP through inhibitory effect on oxidative tissue damage. Glycyrrhizin and $18{\beta}$-glycyrrhetinic acid may reduce the $MPP^+$ toxicity in PC12 cells by suppressing caspase-3 activation. The effect seems to be ascribed to the antioxidant effect.
Radiation exposure may cause tissue damage by ionized or excited atoms. The molecules are transformed from water or they directly interact with cells. Antioxidants such as vitamin E, vitamin C are used to reduce the interference of radiation. Especially, vitamin E inhibits the formation and accumulation of lipid and oxidation and the activity of drug-metabolizing enzymes. After irradiation of 3 Gy to the testis, the entire shape of testis was partly depressed and an irregular vacuole was observed to the edge. The 3 Gy group after treatment with vitamin E showed the low destruction of spermatogonia and partial tissue damage as compared to the 3 Gy group. Vitamin E inhibited the production of vacuoles from the cell death and generated the block to prevent the condensation of the seminiferous tubules.
Extensive research with polarimetry and Mueller matrix has been done for chemical measurements and possible cancer detection. However, the effect of thermally denatured biological tissue on polarization changes is not well known. The purpose of this study is to characterize polarization changes in collagen due to thermal denaturation. The variations in polarized state caused by thermal damage were investigated by obtaining the Mueller matrix elements of collagen sample at multiple thermal damage levels. The changes in birefringence of denatured collagen were also investigated. This information could be used to determine the extent of thermal damage level of clinically heat treated tissues.
Ionizing radiation causes the massive generation of reactive oxygen species, resulting in cellular and tissue damage. The present study was performed to evaluate ${\gamma}$-irradiation induced cellular damage in ICR mice. The mice were divided into four groups with ten mice in each group. Group 1 served as an unexposed control group. Groups 2, 3, and 4 were exposed to 3, 5, and 7 Gy of ${\gamma}$-radiation, respectively. Five mice per group were sacrificed 1 and 7 days after ${\gamma}$-radiation. Exposure to ${\gamma}$-irradiation resulted in hematopoietic damage in a dose-dependent manner when compared with the unexposed control group, which featured a significantly decreased spleen index. However, the exposed mice showed no significant differences in their serum AST, ALT and in the histopathological change of their liver. These results suggest that ${\gamma}$-irradiation is a good tool to prepare a hematopoietic damage model. This animal model can be employed to study the hematopoietic efficacy of biologically active compounds.
Purpose: There is an increasing amount of evidence that S100B could function as a marker of brain damage. However, the cerebral specificity of S100B has been questioned, so the extracerebral sources of S100B have been paid attention. We performed this investigation to show serum S100B levels after extracranial fracture in patients without current head injury and without prior neurological disease. Methods: At the emergency department, we obtained the blood samples within 6 hours from trauma patients hospitalized with extracranial fractures. S100B levels were compared between one fracture and more than two fractures, and analyzed according to the presence of soft tissue damage. Results: Patients with one fracture and those with more than two fractures did not differ by age (mean, 54.70 vs. 47.03, p=0.130), and there was no significant difference in the male-to-female ratio(33:32 vs. 21:12, p=0.226). In patients with one fracture, the mean value of S-100B was $0.56{\mu}g/L$ (95% CI: 0.35-0.77) whereas in those with more than two fractures, the corresponding value was $1.09{\mu}g/L$ (95% CI: 0.46-1.7, p=0.048). The S100B level of patients with soft tissue damage($1.32{\pm}0.38$) was higher than that of patients without soft tissue damage($0.81{\pm}0.21$), whether one fracture or more than two fractures(p=0.049). Conclusion: We present here that S100B levels were raised in 77% of patients with extracranial fractures without cerebral injury who were hospitalized from the emergency room and that the presence of soft tissue damage contributed to the increased S100B rather than the size of the fractured bone size or the number of fracturest. Thus, this study suggests that soft tissue injury may be considered as an important extracerebral source of S100B.
This study was conducted to determine the prevention effect of poly-gamma-glutamic acid (${\gamma}-PGA$) on tissue damage induced by gamma irradiation for development of xenograft. Porcine tendons were treated at various doses of ${\gamma}-PGA$ (0.1, 0.5, 1 and 5%) and then gamma-irradiated (30 kGy). Prevention effects on tissue damage were measured as the result of tensile strength, hydroxyproline contents and viscosity of ${\gamma}-PGA$. Tensile strength was remarkably decrease in gamma-irradiated porcine ligament, but increased by ${\gamma}-PGA$ treated one. Among the ${\gamma}-PGA$ treatment doses, 1% treated group showed the highest values of tensile strength compared to non-treated group. Hydroxyproline contents was significantly increased by gamma irradiation, but decreased by the ${\gamma}-PGA$ treatment. Particularly, 1 and 5% ${\gamma}-PGA$ treated group were exhibited lower values of hydroxyproline contents than other group. In the result of viscosity, gamma-irradiated ${\gamma}-PGA$ (1%) was remarkably increased. Base on the results, it demonstrated that gamma irradiation induces severe alteration of mechanical property and collagen contents on porcine ligament, but ${\gamma}-PGA$ can effectively prevent these tissue damage.
Stress urinary incontinence (SUI) occurs when abdominal pressure increases, such as sneezing, exercising, and laughing. Surgical and non-surgical treatments are the common methods of SUI treatment; however, the conventional treatments still require continuous and invasive treatment. Laser have been used to treat SUI, but excessive temperature increase often causes thermal burn on urethra tissue. Therefore, the optimal conditions must be considered to minimize the thermal damage for the laser treatment. The current study investigated the feasibility of the laser irradiation condition for SUI treatment using non-ablative 980 nm laser from a safety perspective through numerical simulations. COMSOL Multiphysics was used to analyze the numerical simulation model. The Pennes bioheat equation with the Beer's law was used to confirm spatio-temporal temperature distributions, and Arrhenius equation defined the thermal damage caused by the laser-induced heat. Ex vivo porcine urethral tissue was tested to validate the extent of both temperature distribution and thermal damage. The temperature distribution was symmetrical and uniformly observed in the urethra tissue. A muscle layer had a higher temperature (28.3 ℃) than mucosal (23.4 ℃) and submucosal layers (25.5 ℃). MT staining revealed no heat-induced collagen and muscle damage. Both control and treated groups showed the equivalent thickness and area of the urethral mucosal layer. Therefore, the proposed numerical simulation can predict the appropriate irradiation condition (20 W for 15 s) for the SUI treatment with minimal temperature-induced tissue.
To summarize current knowledge regarding mechanisms of radiation-induced normal tissue injury and medical countermeasures available to reduce its severity. Advances in radiation delivery using megavoltage and intensity-modulated radiation therapy have permitted delivery of higher doses of radiation to well-defined tumor target tissues. Injury to critical normal tissues and organs, however, poses substantial risks in the curative treatment of cancers, especially when radiation is administered in combination with chemotherapy. The principal pathogenesis is initiated by depletion of tissue stem cells and progenitor cells and damage to vascular endothelial microvessels. Emerging concepts of radiation-induced normal tissue toxicity suggest that the recovery and repopulation of stromal stem cells remain chronically impaired by long-lived free radicals, reactive oxygen species, and pro-inflammatory cytokines/chemokines resulting in progressive damage after radiation exposure. Better understanding the mechanisms mediating interactions among excessive generation of reactive oxygen species, production of pro-inflammatory cytokines and activated macrophages, and role of bone marrow-derived progenitor and stem cells may provide novel insight on the pathogenesis of radiation-induced injury of tissues. Further understanding the molecular signaling pathways of cytokines and chemokines would reveal novel targets for protecting or mitigating radiation injury of tissues and organs.
In order to investigate a pathogenesis of liver damage induced by skin burn, thermal injury was induced by scald burn on entirely dorsal surface in rats (total body surface area 30%) except for inhalated injury. At 5 and 24 h after scald bum, biochemical assay and morphological changes in skin tissue, serum and liver tissue were examined. The effects of bum injury on the levels of glutathione, lipid peroxide and on the activities of oxygen free radical generating and scavenging enzymes have been determined in association with observing of histologic and ultrastructural changes, measuring the protein concentration in plasma, and counting the number of intravascular polymorphonuclear leukocytes. The activity of xanthine oxidase, an enzyme of oxygen free radical generating system. was elevated (p<0.01) in serum, but not in skin and in liver tissue. Futhermore, thermal injury decreased not only the protein concentration in plasma but also the number of leukocytes, that indicates induction of edema formation with protein exudation and inflammation by neutrophil infiltration into the internal organs. These data suggest that acute dermal scald burn injury leads to liver damage, that is related to elevation of xanthine oxidase activity in serum. Xanthine oxidase may be a key role in the pathogenesis of liver damage induced by skin burn.
