A Study on Major Components of Bee Venom Using Electrophoresis (전기영동법(Electrophoresis)을 이용한 봉약침의 주요 성분에 관한 연구)

  • This study was designed to study on major components of various Bee Venom(Bee Venom by electrical stimulation in Korea; K-BV I, Bee Venom by Microwave stimulation in Korea; K -BV II, 0.5rng/ml, Fu Yu Pharmaceutical Factory, China; C-BV, 1mg /ml, Monmouth Pain Institute, Inc., U.S.A.; A-BV) using Electrophoresis. The results were summarized as follows: 1. In 1:4000 Bee Venom solution rate, the band was not displayed distinctly usmg Electrophoresis. But in 1: 1000, the band showed clearly. 2. The results of Electrophoresis at solution rate 1:1000, K-BV I and K-BVII showed similar band. 3. The molecular weight of Phospholipase $A_2$ was known as 19,000 but its band was seen at 17,000 in Electrophoresis. 4. Protein concentration of Bee Venom by Lowry method was different at solution rate 1:4000 ; C-BV was $250{wmu}g/ml,\;K-BV\;I\;was\;190{wmu}g/ml,\;K-BVII\;was\;160{wmu}/ml\;and\;C-BV\;was\;45{wmu}/ml5$. Electrophoresis method was unuseful for analysis of Bee Venom when solution rate is above 1:4000 but Protein concentration of Bee Venom by Lowry method was possible. These data from the study can be applied to establish the standard measurement of Bee Venom and prevent pure bee venom from mixing of another components. I think it is desirable to study more about safety of Bee Venom as time goes by.

Long Term Effect of Allii tuberosi semen on Muscle Fatigue and Muscular Strength (구자를 장기간(長期間) 투여(投與) 했을 때 생쥐의 근피로(筋疲勞)와 근력(筋力)에 미치는 영향(影響))

  • Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Allii tuberosi semen after long term administration. Materials and Methods: 4-6 weeks old ICR mice were used in this study and we administered the water soluble extracts of Allii tuberosi semen in the concentration of 1, 10 and 100mg/0.3ml to each 5 mice (ATS group) and same volume of normal saline was administered to 5 mice (control group) once a day for 90 or 120 days. After the administration we performed the swimming exercise test and the grip strength test. And we measured the concentration of glucose and the activity of lactate dehydrogenase in plasma regarded as biochemical indicator related with the muscular fatigue. Results: In swimming exercise test, all ATS groups showed increased time compared with relative control group after 90 days administration. But after 120 days adminstration 10 and 100mg/0.3ml ATS groups showed increased results. In grip strength test, all ATS group increased grip strength after 90 days administration and 10mg/0.3ml ATS group showed increased result after 120 days administration. All the results of plasma glucose didn't showed statistically significant difference after 90 and 120 days administration. In the plasma concentration of activity level of lactate dehydrogenase, 1mg/0.3ml and 10mg/0.3ml ATS groups showed decreased concentration compared with relative control group after 90 days administration. But after 120 days adminstration 1mg/0.3ml ATS groups showed decreased results. Conclusion: From this study we could find the anti-fatigue effect of Allii tuberosi semen in the muscle strength related test.

Restoring Ampicillin Sensitivity in Multidrug-Resistant Escherichia coli Following Treatment in Combination with Coffee Pulp Extracts

  • Escherichia coli, particularly multidrug-resistant (MDR) strains, is a serious cause of healthcare-associated infections. Development of novel antimicrobial agents or restoration of drug efficiency is required to treat MDR bacteria, and the use of natural products to solve this problem is promising. We investigated the antimicrobial activity of dried green coffee (DGC) beans, coffee pulp (CP), and arabica leaf (AL) crude extracts against 28 isolated MDR E. coli strains and restoration of ampicillin (AMP) efficiency with a combination test. DGC, CP, and AL extracts were effective against all 28 strains, with a minimum inhibitory concentration (MIC) of 12.5-50 mg/ml and minimum bactericidal concentration of 25-100 mg/ml. The CP-AMP combination was more effective than CP or AMP alone, with a fractional inhibitory concentration index value of 0.01. In the combination, the MIC of CP was 0.2 mg/ml (compared to 25 mg/ml of CP alone) and that of AMP was 0.1 mg/ml (compared to 50 mg/ml of AMP alone), or a 125-fold and 500-fold reduction, respectively, against 13-drug resistant MDR E. coli strains. Time-kill kinetics showed that the bactericidal effect of the CP-AMP combination occurred within 3 h through disruption of membrane permeability and biofilm eradication, as verified by scanning electron microscopy. This is the first report indicating that CP-AMP combination therapy could be employed to treat MDR E. coli by repurposing AMP.

Effect of Guar gum levels in Backsulgies on Plasma Glucose and Insulin in Healthly Men (백설기에 첨가된 Guar gum 수준이 혈장 포도당과 인슐린에 미치는 영향)

  • Backsulgies containing various levels of guar gum(0g, 2.5g or 5g) were ingested by ten healthy male subjects after 12-h fast. Guar backsulgies was given a time in a week a time in a week for 3 weeks with 75g glucose. The Sensory qualities of backsulgies containing guar gum powder of different levels(0g, 2.5, 5g) were also evaluated using a hedonic scoring technique. On each test day 4ml samples of venous blood were taken from the fasted subjects. Further 4ml post-prandial blood samples were taken from the subjects 30, 60, 90 and 120min after guar backsulgies had commenced. Result were as follows: Plasma glucose and insulin levels were not sifnificantly reduced according to guar gum levels in backsulgies. But plasma glucose and insulin levels of the case which ingested 5g guar backsulgies tended to be lower than those of the case which ingested 0g or 2.5g guar backsulgies. Therefore it is recommended that 5g guar backsulgies is most effective snack of diabetics. KEY WORDS: guar backsulgies plasma glucose, plasma insulin, snack.

Anti-proliferative and Apoptosis Inducing Effect of Momordin I on Oral Carcinoma (KB) Cells

  • Treatment of oral cancers with chemotherapeutic agents become evaluated as an effective method to reduce cancer cell proliferation. Anti-proliferative and anti-oral cancer activities of momordin I on oral cancer cells were evaluated in this study. Momordin I was originally purified from a natural product, Ampelopsis radix and showed the antiproliferative activity against oral carcinoma, KB cells. Obtained $IC_{50}$ value was approximately $10.4{\mu}g/ml$. Time-and dose-dependent chromosomal DNA fragmentations were observed in momordin I-treated KB cells. Flow cytometry analysis showed time-dependent apoptotic cell appearance after treatment of momordin I. Approximately 18.6% apoptotic cells were observed at 72 hours after $20{\mu}g/ml$ of momordin I treatment. These observation were consistent with the results obtained in DNA fragmentation analysis. These data suggest that momordin I has anti-proliferative effect and induces cell death in KB cells through apoptosis.

The Effect of Circadian Rhythm on the Pharmacokinetics of Acebutolol after Intravenous Administration to Rabbits (가토에서 정맥투여시 생체리듬이 아세부토롤의 약물동태에 미친영향)

  • The effect of circadian rhythm on the pharmacokinetics of acebutolol was studied in rabbits administered intravenous 5 mg/kg dose of acebutolol at 09:00 in the morning (a.m) and 22:00 in the evening (p.m). A significant effect of circadian rhythm of pbarmacokinetic parameters as a function of time of day was noted in rabbits, showing lower total body clearance (CLt), higher plasma concentration and the area under the plasma concentration time curve (AUC) when acebutolol was given in the evening. The plasma concentration of acebutolol was increased significantly (p<0.05) at 12-24 hr after dosing in the evening. The AUC was greater in the evening $(111\%)$ than that in the morning and $CL_t$, was higher when acebutolol was given in the morning ($1.12\pm0.24$ ml/hr) versus in the evening ($1.01\pm0.22$ ml/hr), but those were not significant. Therefore, It is reasonable to consider individual circadian rhythm for effective dosage regimen of acebutolol in clinical chronotherapeutics.

Optimization of Antibacterial Activity by Gold-Thread (Coptidis Rhizoma Franch) Against Streptococcus mutans Using Evolutionary Operation-Factorial Design Technique

  • This study was conducted to find the optimum extraction condition of Gold-Thread for antibacterial activity against Streptococcus mutans using The evolutionary operation-factorial design technique. Higher antibacterial activity was achieved in a higher extraction temperature ($R^2=-0.79$) and in a longer extraction time ($R^2=-0.71$). Antibacterial activity was not affected by differentiation of the ethanol concentration in the extraction solvent ($R^2=-0.12$). The maximum antibacterial activity of clove against S. mutans determined by the EVOP-factorial technique was obtained at $80^{\circ}C$ extraction temperature, 26 h extraction time, and 50% ethanol concentration. The population of S. mutans decreased from 6.110 logCFU/ml in the initial set to 4.125 logCFU/ml in the third set.

  • This paper proposes an efficient detection algorithm, which is composed of an interference nulling-and cancelling-based detection algorithm and a maximum likelihood (ML) detection algorithm having reduced numbers of signal points to be tested, for the quasi-orthogonal space-time code with four transmit antennas. When high-level modulation schemes are employed, the algorithm enables the quasi-orthogonal code to achieve near ML performance with a significant reduction in the computational load.

  • The enhancing effect of excreted cell factors on the production of somatic embryos from suspension cultures of Daucus carota was studied as a function of factors including molecular size, harvesting time, injection period, and concentration of the extracellular compounds. The production of late-stage embryos was increased up to 1, 500 embryos/ml compared with control cultures when high molecular size and extracellular factors, extracted from newly established embryo culture at early stationary phase, were added at the starting time. The stimulating effect on the production of somatic embryos can be attributed to the presence of high molecular size(> 10 kDa) compounds.

Maximum Likelihood and Signal-Selective TDOA Estimation for Noncircular Signals

  • This paper addresses the issue of time-difference-of-arrival (TDOA) estimation for complex noncircular signals. First, under the wide-sense stationary assumption, we derive the maximum likelihood (ML) estimator and the Cramer-Rao lower bound for Gaussian noncircular signals in Gaussian circular noise. The ML estimator uses the second-order statistics information of a noncircular signal more comprehensively when compared with the cross-correlation (CC) and the conjugate CC estimators. Further, we present a scheme to modify the traditional signal-selective TDOA methods for noncircular signals on the basis of the cyclostationarity of man-made signals. This scheme simultaneously exploits the information contained in both the cyclic cross-correlation (CCC) and the conjugate CCC of a noncircular signal.
