Silibinin is the major active component of silymarin, extracted from the medicinal plant Silybum marianum. Silibinin has potent antibacterial activity; however, the exact mechanism underlying its activity has not been elucidated. Here, we investigated the novel mechanism of silibinin against Escherichia coli. Time-kill kinetic assay showed that silibinin possess a bactericidal effect at minimal inhibitory concentration (MIC) and higher concentrations (2-and 4-fold MIC). At the membrane, depolarization and increased intracellular $Ca^{2+}$ levels were observed, considered as characteristics of bacterial apoptosis. Additionally, cells treated with MIC and higher concentrations showed apoptotic features like DNA fragmentation, phosphatidylserine exposure, and caspase-like protein expression. Generally, apoptotic death is closely related with ROS generation; however, silibinin did not induce ROS generation but acted as a scavenger of intracellular ROS. These results indicate that silibinin dose-dependently induces bacterial apoptosis-like death, which was affected by ROS depletion, suggesting that silibinin is a potential candidate for controlling bacteria.
Combination therapy of antibiotics is leading to improved efficacy or safety profiles with decrease emergence of bacterial resistance. Because of this benefit, many of antibacterial combinations have been used in veterinary practice for the past few decades. The purpose of this study was to examine the in vitro activity of an amoxicillin alone and in combination with other antibiotics against pathogenic bacteria of fish origin. Based on the fractional inhibitory concentration (FIC) index (FIC $$\leq_-$$ 0.5), a synergistic interaction was shown in combination of florfenicol with amoxicillin or cefuroxime. The combination of florfenicol and amoxillin showed higher antibacterial activity than that of florfenicol and cefuroxime. Ratio of amoxicillin and florfenicol in combination was 1 : 1, which showed the antibacterial activity against bacterial isolates of fish as compared with other ratios. A synergetic effect of the combination (amoxicillin and florfenicol) was further confirmed in the time-kill curve study. The study showed a better in vitro antibacterial activity of a 1 : 1 combination of amoxicillin and florfenicol than the individual antibacterial against bacterial isolates of fish. In conclusion, the combination of florfenicol and amoxicillin may serve as a potential antibacterial therapy in fishes infected pathogenic bacteria.
Vibrlo vulnificus가 병원성을 일으키는 한가지 요인으로서 iron의 역할을 본 연구에서 알아보고 V. vulnificus의 성장에 미치는 사람과 토끼 혈청에서의 iron과 $CCl_4$의 영향도 조사하였다. 쥐에 iron을 주사한 결과 50% 치사량이 감소하였고 감염 후 치사 시간도 감소하였으며 $CCl_4$,로 감염된 간에 의해서 혈청 중 iron준위는 증가하였다. 쥐를 치사시키는데 필요한 접종량은 혈청 중 iron준위와 직접적으로 상호 관련성이 있었다. Iron은 이 미생물이 포유류 혈청에서 살아남거나 성장하는 능력에 있어서 제한 요인으로 나타났다. 시험관 내에서와 생체 내에서 이들 결과는 iron이 V. vulnificus의 병원성에 있어서 중요한 역할을 한다는 강한 증거를 제공해주었다.

Staphylococcus aureus is a major human pathogen, implicated in both community and hospital acquired infections. The therapy for methicillin-resistant Staphylococcus aureus (MRSA) infections is becoming more difficult because of multidrug resistance and strong biofilm-forming properties. Schiff bases have attracted attention as promising antibacterial agents. In this study, we investigated the in vitro activity of taurine-5-bromosalicylaldehyde Schiff base (TBSSB) against MRSA. The minimum inhibitory concentrations (MIC) and minimum bactericidal concentration (MBC) were determined using a microtiter broth dilution method. TBSSB effectively inhibited planktonic MRSA, with an MIC of $32{\mu}g/ml$. The time-kill curve confirmed that TBSSB exhibited bactericidal activity against MRSA. TBSSB was also found to significantly inhibit MRSA biofilm formation at 24 h, especially at $1{\times}MIC$ and sub-MIC levels. Furthermore, scanning electron microscopy and transmission electron microscopy showed remarkable morphological and ultrastructural changes on the MRSA cell surface, due to exposure to TBSSB. This study indicated that TBSSB may be an effective bactericidal agent against MRSA.
박물관에서 질소나 아르곤과 같은 불활성 가스를 사용한 저산소 농도 살충처리법은 메틸브로마이드나 유독성 살충제의 대체법으로써 성공적으로 사용되어 왔다. 저산소 농도 살충법의 살충 효과는 해충의 종, 산소 농도, 온도, 습도, 가스의 종류 등에 따라 영향을 받으며, 박물관에서의 문화재 보존 환경(온도 $15{\sim}25^{\circ}C$, 습도 40~60%)에서 산소 농도를 0.03% 이하로 유지함으로써 1개월 이내에 가장 내성이 강한 문화재 해충의 살충이 가능하다. 그리고 살충 처리 대상 문화재의 수량이나 크기에 따라 소형 중형 백, 대형 텐트, 버블, 챔버 등 다양한 시스템을 적용할 수 있다.
Large scale of fish kills by red tides has been occurred every year in coastal water fisheries of Korea. To suppress red tide spreading out over the south coastal water of the Korean Peninsula large amount of loess has been applied every year because loess is known to be effective in removing red tide organisms. Effects of loess application in ecosystem of underwater near sea shore were investigated with some physical characteristics of loess. Loess used for the red tide reduction consisted of very fine particles, of which size was mostly less than 0.1 mm. Particles of loess blocked light penetration, which is essential for the underwater ecosystem. Loess also pushed pH down by the hydrolysis activities of aluminium and iron It was found that underwater ecosystems where loess was applied near sea shore were devastated. Sea plants such as sea weeds were gone leaving only their roots. Clams and snails were dead under the loess dust blanket. And fishes were not found at all where loess has been sprayed lot long time. It was found that even if loess has some capacity to reduce red tide temporarily, loess application should be slopped to protect underwater ecosystems.
Alveolar epithelial cells have been functionally implicated in Mycobacterium tuberculosis infection. This study investigated the role of ursolic acid (UA)-a triterpenoid carboxylic acid with potent antioxidant, anti-tumor, anti-inflammatory, and anti-tuberculosis properties in mycobacterial infection of alveolar epithelial A549 cells. We observed that M. tuberculosis successfully entered A549 cells. Cytotoxicity was mediated by nitric oxide (NO). A549 toxicity peaked along with NO generation 72 h after infection. The NO generated by mycobacterial infection in A549 cells was insufficient to kill mycobacteria, as made evident by the mycobacteria growth indicator tube time to detect (MGIT TTD) and viable cell count assays. Treatment of mycobacteria-infected cells with UA reduced the expression of inducible nitric oxide synthase, NO generation, and eventually improved cell viability. Moreover, UA was found to quench the translocation of the transcription factor, nuclear factor kappa B (NF-${\kappa}B$), from the cytosol to the nucleus in mycobacteria-infected cells. This study is the first to demonstrate the cytotoxic role of NO in the eradication of mycobacteria and the role of UA in reducing this cytotoxicity in A549 cells.

선박에서 발생되는 다양한 형태의 디지털 정보를 그 특성에 따라 적절하게 처리하기 위해서는 용도에 적합한 네트워크의 구성이 필요하다. 이러한 필요성에 부합하는 표준으로 NMEA 2000과 IEC 61162-4가 있다. NMEA 2000은 실시간 데이터의 처리에 적합한 표준이고, IEC 61162-4는 데이터의 통합 관리에 적합한 표준이다. 따라서, NMEA 2000 데이터를 통합하여 관리하기 위해서는 IEC 61162-4 포맷으로 변환하여야 한다. 본 논문에서는 이러한 변환을 위한 참조화일 및 알고리즘을 제시하고 그 구현 결과를 기술한다.
Objectives The aim of this study was to investigate antimicrobial activity of Huanggeumjakyak- tang water extract against MRSA. Methods The antibacterial activities of Huanggeumjakyak-tang were evaluated against 3 strains of Methicillin-resistant Staphylococcus aureus (MRSA) and 1 standard Methicillinsusceptible S. aureus (MSSA) strain by using the disc diffusion method, minimal inhibitory concentrations (MICs) assay, colorimetric assay using MTT test, checkerboard dilution test and time-kill assay was performed under dark. Results The MIC (minimum inhibitory concentration) of Huanggeumjakyak-tang water extract against S. aureus strains ranged from 1,000 to $2,000{\mu}g/ml$. So we confirmed that it has a strong antibacterial effect. Also the combinations of Huanggeumjakyak-tang water extract and conventional antibiotics exhibited improved inhibition of MRSA with synergy effect. Conclusions The results obtained in this study suggest that Huanggeumjakyak-tang water extract showed antibacterial effect against MRSA, and it also showed reducing effect on the side-effect problems that are the major weak points of traditional antibiotics.
Aircraft survivability is determined by the susceptibility and the vulnerability. The aircraft susceptibility and vulnerability depend upon the hardware and software factors. Each of the hardware and software factors consisted of the qualitative and quantitative attributes varies according to the time of the mission. In order to establish the mathermatical model to analyze and evaluate the aircraft survivability, qualitative factors have to be transformed into quantitative factors. Even if many researches in the area of dynamic concept analysis and conversion of qualitative factors into the quantitative factors has been insufficient. This research enhances these insufficient area by developing a reliable aircarft survivability analysis method. The major areas of this research are as follows. First, a method for the conversion of the qualitative factors into the quantitative factors is developed by combining the Fuzzy Set Theory concept and the Delphi Technique. Second, by using the stochastic network diagram for the dynamic survivability analysis, the aircraft survivability and the probability of kill are calculated from the state probability for the situation during mission. The advantage of the analysis technique developed in this research includes ease of use and flexibility. In other words, in any given aircraft's mission execution under any variable probability density function, the developed computer program is able to analyze and evaluate the aircraft survivability.
