Alteration of MRP2 expression and the graft outcome after liver transplantation

  • Purpose: Multidrug resistance-associated protein (MRP) 2 is a glutathione conjugate in the canalicular membrane of hepatocytes. Early graft damage after liver transplantation (LT) can result in alteration of MRP2 expression. The purpose of this study was to evaluate the relationship between the pattern of MRP2 alteration and graft outcome. Methods: Forty-one paraffin-embedded liver graft tissues obtained by protocol biopsy within 2 months after LT; these were stained using monoclonal antibodies of MRP2. We selected 15 live donor biopsy samples as a control, that showed homogenous canalicular staining for MRP2. The pattern of canalicular MRP2 staining of graft was classified into 3 types: homogenous (type C0), focal (type C1), and no (type C2,) staining of the canaliculi. Results: In total, 17.1% graft tissues were type C0, 36.6% were type C1, and 46.3% were type C2. The median operation time was longer in patients with type C2 (562.6 minutes) than in patients with type C0 (393.8 minutes) (P = 0.038). The rates of posttransplant complications were higher in patients with type C2 (100%) than in patients with type C0 (42.9%) and C1 (73.3%) (P < 0.001). Conclusion: MRP2 expression pattern was altered in 82.9% after LT. The pattern of MRP2 alteration was associated with longer operation time and higher rates of post-LT complications.

Longitudinal Tracking of Alteration Pattern on Trabecular Bone Microarchitecture at Tibial Epiphysis Induced by Post Traumatic Osteoarthritis Over Time (외상성 관절염 진행에 따른 경골 골단 해면골에서의 골 미세구조 변화 패턴 추적 관찰)

  • This study aims to track the longitudinal alteration pattern on the trabecular bone microarchitecture at tibial epiphysis induced by T-OA over time using in vivo micro computed tomography (${\mu}CT$). Ten SD rats were divided into control (n = 5) and T-OA (n = 5) groups. Anterior cruciate ligament transaction was performed for the T-OA group. The results showed that the alteration pattern on the trabecular bone microarchitecture at tibial epiphysis in the T.OA group was definitely different compared with that in the CON group from 0 to 8 weeks (approximately 4-16%, P > 0.05). In particular, a difference was observed in the bone formation and density distributions over time (from 0 or 4 to 8 weeks; approximately 5.15%, P < 0.05). An improved understanding of the alteration pattern on the trabecular bone microarchitecture at tibial epiphysis may assist in developing more targeted treatment interventions for T-OA.

Effect of Trichostatin A on CNE2 Nasopharyngeal Carcinoma Cells - Genome-wide DNA Methylation Alteration

  • Trichostatin A (TSA) is a histone deacetylase (HDAC) inhibitor. We here investigated its effects on proliferation and apoptosis of the CNE2 carcinoma cell line, and attempted to establish genome-wide DNA methylation alteration due to differentially histone acetylation status. After cells were treated by TSA, the inhibitory rate of cell proliferation was examined with a CCK8 kit, and cell apoptosis was determined by flow cytometry. Compared to control, TSA inhibited CNE2 cell growth and induced apoptosis. Furthermore, TSA was found to induce genome-wide methylation alteration as assessed by genome-wide methylation array. Overall DNA methylation level of cells treated with TSA was higher than in controls. Function and pathway analysis revealed that many genes with methylation alteration were involved in key biological roles, such as apoptosis and cell proliferation. Three genes (DAP3, HSPB1 and CLDN) were independently confirmed by quantitative real-time PCR. Finally, we conclude that TSA inhibits CNE2 cell growth and induces apoptosis in vitro involving genome-wide DNA methylation alteration, so that it has promising application prospects in treatment of NPC in vivo. Although many unreported hypermethylated/hypomethylated genes should be further analyzed and validated, the pointers to new biomarkers and therapeutic strategies in the treatment of NPC should be stressed.

On a Pitch Alteration Method Compensated with the Spectrum for High Quality Speech Synthesis (스펙트럼 보상된 고음질 합성용 피치 변경법)

  • The waveform coding are concerned with simply preserving the wave shape of speech signal through a redundancy reduction process. In the case of speech synthesis, the wave form coding with high quality are mainly used to the synthesis by analysis. However, because the parameters of this coding are not classified as either excitation and vocal tract parameters, it is difficult to applying the waveform coding to the synthesis by rule. In this paper, we proposed a new pitch alteration method that can change the pitch period in waveform coding by using scaling the time-axis and compensating the spectrum. This is a time-frequency domain method that is preserved in the phase components of the waveform and that has a little spectrum distortion with 2.5% and less for 50% pitch change.

  • In the area of the speech synthesis techniques, the waveform coding methods maintain the intelligibility and naturalness of synthetic speech. In order to apply the waveform coding techniques to synthesis by rule, we must be able to alter the pitches of synthetic speech. In this paper, we propose a new pitch altering method that compensates phase distortion of the cepstral pitch alteration method with time scaling method in the time domain. This method can remove some spectrum distortion which is occurred in conjunction point between the waveforms. For performance test the spectrum distortion rate was used as objective criterion and the MOS(Mean Opinion Score) was used as subjective criterion. As a result, the spectrum distortion and MOS are obtained by 0.66% and 3.9, respectively.

In the case of speech synthesis, the waveform codings with high quality are mainly used to the synthesis by analysis. Because the parameters of this coding are not classified as both excitation and vocal tract, it is difficult to apply the waveform coding to the synthesis by rule. Thus, in order to apply the waveform coding to synthesis by rule, it is necessary to alter the pitches. In this paper, we proposed a new pitch alteration method that can change the pitch period in waveform coding by dividing the speech signals into the vocal tract and excitation parameters. This method is a time-frequency domain method preserving the phase component of the waveform in time domain and the magnitude component in frequency domain. Thus, it is possible that the waveform coding is carried out the synthesis by rule in speech processing. In case of using the algorithm, we can obtain spectrum distortion with $2.94\%$. That is, the spectrum distortion is decreased more $5.06\%$ than that of the pitch alteration method in time domain.

  • Preventing the forgery and alteration of test reports is important, in order to maintain the reliability of military supplies. Based on a survey of reports on testing institutions and a case study of established electronic document systems in the public sector, we propose a system architecture, which helps prevent the forgery and alteration of test reports with legal force. The proposed architecture takes advantage of both the time stamp that records the time a document is received and a server synchronized with the testing institution. Using the proposed system architecture, the user is able to request a test and receive reports from the testing institution without delay. Also, a defense agency is able to conveniently prove the authenticity of the test reports and utilize the statistical data collected by the architecture. Therefore, we expect that the proposed system architecture will help defense agencies to prevent the forgery and alteration of test reports and ensure their reliability and quality.

A Study of Driver Brain Wave Characteristics through Changes in Headlamp Brightness

  • In this study, tests of brain waves were carried out to investigate the physiological characteristics of drivers during a change of headlight brightness. The participants were 20 males in their 20s. Twenty-three different conditions combining the waveform of light, voltage, and alteration time were used. The measurement of brain waves was performed by an internationally standardized 10-20 method using LXE3232-RF. The results were as follows. 1. From the results of the brain wave map analysis, it was suggested that waveform A increases mental stress and waveform B affects mental and visual stress. The longer the stimulation time, the more stress level was detected. 2. The voltage alteration time of the B waveform should be kept to less than 1500msec, while the voltage should not fall below 11.5[V].

The participants were 20 men in their 20s and twenty-three different conditions in combinations of waveform of light, voltage, and alteration time were used. The measurement of the brain waves was performed by internationally standardized 10-20 method using LXE3232-RF. The results were analyzed by Power Spectrum Analysis using alpha-, and beta-wave and by the analysis of different brain domains using Brain wave Map. The results were as follows. 1. From the results of the Brain wave Map analysis, it was suggested that A waveform increases mental stress and B waveform affects mental and visual stress. The longer the stimulation time, the more stress level was detected. 2 The voltage alteration time of B waveform should be kept less than 1500msec, and the voltage should not fall below 11.5V.
