Production and Properties of Mannanase and Xylanase by a Bacillus subtilis Isolate (Bacillus subtilis 분리균의 Mannanase와 Xylanase 생산성과 효소 특성)

  • A bacterial strain capable of hydrolyzing xylan and locust bean gum (LBG) was isolated from the Saemangeum tideland of Korea. Based on the biochemical properties and the 16S rRNA gene sequence, the isolate YB-30 was identified as Bacillus subtilis. Xylanase productivity was increased effectively when B. subtilis YB-30 was grown in the presence of wheat bran, while mannanase productivity was increased drastically when grown in the presence of konjac or LBG. Particularly, maximum mannanase and xylanase activities were detected in the culture filtrate of media containing 3.5% konjac and 1% wheat bran. Both enzyme productivities reached maximum levels in the stationary growth phase. The culture filtrate exhibited the highest activity at 60℃ and pH 6.0 for mannanase and at 55℃ and pH 5.5 for xylanase, respectively. Both enzymes were not stable at high temperatures and xylanase was less stable than mannanase. In addition, wheat bran was hydrolyzed to liberate reducing sugar to a greater extent than rice bran by the culture filtrate because the wheat bran contained more arabinoxylan than the rice bran. Hence, xylanase and mannanase produced by B. subtilis YB-30 have a potential use as feed additive enzymes.

Determining Irrigation Requirements and Water Management Practices for Normal Growth of Dry Field Crops in Reclaimed Tidelands (간척지 밭작물의 정상생육을 위한 관개용수량 및 물 관리방법의 결정)

  • This study was carried out in order to determin optimum irrigation requirements and water management practices for normal growth of dry field crops in reclaimed tidelands, and apply m planning of the irrigation projects. Desalinization experiments were performed by water management practices in the experimental field with high salt concentration, and growth experiments were conducted by irrigation point treatments using tomato and beet with relatively high salt tolerance. The results obtained from this study were summarized as follows : 1. Leaching or rinsing-leaching method was found to be effective in desalinizing the reclaimed tideland with rather high permeability. In this case, the water requirement for desalinizing the root zone layer of 40cm in depth, was estimated to be 1,200mm in depth. 2.The gypsum treatment in the desalinization of reclaimed tidelands, was ineffective in water requirements ; however, it could produce the desired effect in the facility of desalinization and the shortening of desalinization period with the sustaining permeability, in case of the desalinization by leaching method. 3.The optimum irrigation point which maintains the salt concentration within salt tolerance and maximizes the crop yield in reclaimed tidelands of silt loam soil, was found to be pF 1.6 in tomato and pF 1.8 in beet. The interval of irrigation date within 2 days was proved to he effective in both cases. 4.The optimum irrigation requirement and the water reguirement for the prevention of salt rise during the growing period after transplanting, were estimated to be 602mm(6.7mm/day) and 232mm for tomato, respectively. 5.The optimum irrigation requirement and the water requirement for the prevention of salt rise during the growing period after transplanting, were estimated to be 261mm(3.7mm/day) and 66mm for beet, respectively.

Survey for Farmland Development in Western Coast of North Korea Using Satellite Image Data (인공위성 화상데이터를 이용한 북한 서해안지역의 농지기반조성 현황조사)

  • The aim of this study was to find out and confirm the project formulation, feasibility, scale and locations on the farmland development projects such as planned and ongoing tideland reclamation and irrigation facilities along the western coast of North Korea using satellite image data, Landsat TM, JERS OPS and SPOT PAN and aged maps. In order to apply to the study, remote sensing technologies such as geometric correction. digital mosaicking, image merging, linear extraction and land cover classification were studied. As the results of the study, the reclaimable tidal flats are recognized at about 178,000 ha equivalent to 59% of announced 300,000ha. and 16,000 ha of completed, 17,000 ha of ongoing project areas although 27,000 ha were revealed to be completed during 1987-1993. Almost planned projects are appeared to be shortage of water supply due to their small watersheds, however, most projects are connected with 2000 mile canal system.

  • This experiment was carried out to select the optimum forage cropping system at the Dae-Ho reclaimed tideland, Korea from March, 2004 to October, 2006. Emergency rate of summer crop such as sorghum ${\times}$ sudangrass was low ($23%{\sim}30%$). Winter crop such as Italian ryegrass after continually cultivating wholecrop rice were highest (91%). Wintering rate of Italian ryegrass after continually cultivating wholecrop rice was highest and lowest in wholecrop barley after continually cultivating sorghum${\times}$sudanuass. In cropping system, fresh and dry matter yield of annually wholecrop rice+Italian ryegrass (50,807kg, 15,065kg) were highest and lowest in annaually serghum${\times}$sudangrass+wholecrop barley (17,2471kg, 5,209kg), respectively.

  • The purposes of this study are to investigate the perception of renewable energy technology among elementary school teachers, and confirm whether elementary school teachers have basic knowledge about renewable energy sources, including solar, wind, and tidal power generation. We conducted preliminary interviews to gather information related to other studies about renewable energy. We developed the last interview question about the perception and knowledge of elementary school teachers regarding renewable energy. This study analyzed the transcribed responses of 10 elementary school teachers in Siheung-city, Gyeonggi-do, following 30-minute interviews. The study's findings are as follows. First, elementary school teachers recognize that they are unfamiliar with concepts and they have only shallow content knowledge about renewable energy. And they tended to distorted to other concepts, and analyze to different meanings. Second, elementary school teachers thought that knowledge about renewable energy should be part of a well-rounded education. And they felt positively about solar energy and wind power energy generation but they had a negative view towards tidal power generation because it destroys tideland. Third, teachers tended to confuse solar heat energy and geothermal energy, they tend to think this two energy sources the same. Teachers had generally correct concepts about wind power energy generation. In the case of tidal power generation, elementary school teachers answered mechanically that it is possible on the western sea, and that 'the difference between the rise and fall of the tide' grows. But they could not talk in depth about 'the difference between the rise and fall of the tide' and the force of waves. This suggests that they are answering by simple memorization and without deep understanding.

Influence of Gypsum, Popped Rice Hulls and Zeolite on Contents of Ca2+, Mg2+, Na+, K+ in Reclaimed Tideland Soils in Kyehwado (계화도 간척지에서 석고, 팽화왕겨 및 제올라이트 처리가 토양 중 양이온 함량에 미치는 영향)

  • The effect of application of gypsum (G), popped rice hulls (PRH), and zeolite (Z) in exchangeable cations concentrations of reclaimed tideland soil in Kyehwado was investigated for 3 years from 2004 to 2006 in a pot experiment with bermuda grass (Cynodon dactylon). Treatments with three soil conditioner and with three applications were established with three replications; G1 (1,550 kg $10a^{-1}$), G2 (3,100), and G3 (6,200) for gypsum, H1 (1,000), H2 (2,000), and H3 (3,000) for PRH, and HZ1 (200), HZ2 (400), and HZ3 (800) for co-application of zeolite with PRH at the 1,500 kg $10a^{-1}$. At 60, 90, 120 days after treatment (DAT), exchangeable cations ($K^+$, $Na^+$, $Mg^{2+}$, and $Ca^{2+}$) were analyzed Gypsum application significantly decreased $k^+$, $Na^+$, $Mg^{2+}$ in the soil probably due to exchange and subsequent leaching of these cations by $Ca^{2+}$ from the gypsum applied. Overall, $K^+$ concentration was gradually decreased by continuous application of soil conditioners and was in the order of 2004>2005>2006 regardless of the kinds and application rate of soil conditioners. Comparing $K^+$ concentrations among the soil conditioners in the same year, its concentration was in the order of gypsum$Na^+$ concentration; i.e. $Na^+$ concentration was in the order of gypsum$\ll$PRH$Mg^{2+}$ also showed a similar pattern to $Na^+$. Gypsum application significantly increased $Ca^{2+}$ concentration and in the gypsum treated soil $Ca^{2+}$ concentration increased with years.

  • Soil conditioner, such as $CaSO_4{\cdot}2H_2O$ (gypsum), popped rice hulls (PRH), and PRH with zeolite, were treated to the silt loam of Mangyeong in Saemangeum tideland reclaimed as 1550 (G1), 3100 (G2) and 6200 (G3) of gypsum kg/10 a, 1000(H1), 2000(H2), and 3000 (H3) of PRH kg/10 a, and 200 (HZ1), 400 (HZ2), 800 (HZ3) of zeolite kg/10 a added to 1500 PRH kg/10 a, respectively, each year until 2006 from 2004 for soil aggregation. Under these conditions with growing bermuda grass (Cynodon dactylon) it was analyzed cations in soil, such as $K^+$, $Na^+$, $Mg^{2+}$, and $Ca^{2+}$, at 60, 90, and 120 days after treatment (DAT) to research how soil conditioners influenced to change those contents in soils, respectively. The change of cations in soil was almost the same things as fine sandy loam that gypsum treated decreased remarkably contents of $K^+$, $Na^+$, $Mg^{2+}$ in soil. The change of $K^+$ content in soil by continuous using soil conditioners was gradually decreased in the order of 2004>2005>2006, regardless of the sorts and levels of soil treated conditioners, and $K^+$ content was high in the order of gypsum$Na^+$ content was high in the order of gypsum$Mg^{2+}$ content in soil was increased in the order of gypsum$Ca^{2+}$ content in soil was remarkably increased with continuous treatment of gypsum, and its level was in the order of 2004<2005<2006.

A Study on the Surface Erosion by the Development of Cropland on the Hillslope in the West Coast Area of North Korea Using Quick Bird Satellite Images (Quick Bird 영상을 이용한 북한 서해안 구릉지 개간에 따른 지표 침식 분석)

  • The study deals with surface erosion patterns due to the development of cropland toward hillslope and hilltop in the Oncheon-gun, pyeongbuk province and Nampo city of west coast area in the North Korea, using Quick Bird satellite images with 60cm resolution. In North Korea, for national economic difficult after 1980 year, newly developed croplands have been along the gentle hillslope, in which it is possible for individual man power different from the tideland which needs large scaled man-power and equipment. The new croplands are named Darakbat(terraced farm with embankment) and Bitalbat(titled farm developed on the original hill slope), neighboring with orchard and grouped settlement in lower valley. For supplying agricultural water, irrigation ditches and temporal crop storages have been constructed, connecting Darakbat, Bitalbat and orchard. These cropland developments have caused surface erosion composed of 3 types such as pit, linear and headward erosion, together with rill and gully. Owing to poor management of cropland and irrigation system, topsoil erosion and, collapse and sedimentation of ditch and pool, caused the decrease of agricultural productivity. These analysis using Quick Bird images can suggest original raw data about geographical facts on North Korea agriculture and help to recover their agricultural system and plan future national unified land.

Soil Problems and Agricultural Water Management of the Reclaimed Land in Korea (한국의 간척지에서 토양 문제와 농업 용수 관리)

  • Soil problems and agricultural water management of the reclaimed land in Korea were reviewed through research results conducted in RDA and ADC. According to the Korean Soil Classification and Soil Survey(NIAST, 2002), the 5 soil orders with the 45 soil series were distributed on the fluvio-marine or marine deposit of the west and south coastal plains. Yeompo, Munpo, Hasa, Gwangwhal, and Poseung soil series were most commonly distributed soil on the fluvio-marine deposits, associated with tideland of the sea coast. Former 4 soils were Entisols, and the latest one was the Inceptisols. Buyong soil associated with Poseung series was an Alfisols. Extent of Myeongji soil, a Molisols, and Yongho soil, a Histosol, were minor. Salinity control and management problems were closely related with high water table and low percolation rate due to plow-pan layer developed during the leaching process in the silty textured soil. For evaluation of field salinity, use of an electromagnetic inductance, EM38, with GPS was helpful to understand salinity status and field variability. Deep plowing, subsoiling and drainage improvement by tile drainage might be effective in paddy with plow-pan. New technology such as variable rate fertilization might save fertilizers and thus reduce environmental impact of agriculture on water quality. Water quality of agricultural water resources in reclaimed land was less adequate than that of inland water resources. Proper crop management is necessary depended upon quality for crop growth as well as to match with water quality target.
