살초활성물질 함유 국내 자생식물의 탐색 (I) (Herbicidal activity of Korean native plants (I))

  • 본 연구의 목적은 국내의 자생식물 중 살초활성이 있는 식물 종을 선발하는데 있다. 국내의 자생식물 73과(科) 200종(種) 시료로부터 MeOH 조추출물을 얻은 다음 24-well plate에서 유채 (Brassica napus L.) 에 대한 살초효과를 검정하였다. 실험에 사용한 200종 식물 중 6종 식물 - 고추나무 (Staphylea bumalda), 등나무(Wistaria floribunda), 산마늘(Allium victorialis), 소리쟁이(Rumex crispus), 이팝나무(Chionanthus retusa), 참느릅 나무 (Ulmus parvifolia)는 높은 살초활성을 나타내었고 ($GR_{50}$ 값, < $1,000{\mu}g\;g^{-1}$), 17종 식물 - 갈퀴덩굴(Galium spurium), 느티나무(Zelkova serrata), 능소화(Campsis gradiflora), 두충(Eucommia ulmoides), 마가목(Sorbus commixta.), 물참대(Deutzia glabrata), 박태기나무(Cercis chinensis), 산오리(Alnus hirsuta), 산초나무(Zanthoxylum schinifolium), 상수리나무(Quercus acutissima), 아까시나무(Robinia pseudoacacia), 주엽나무(Gleditsia japonica), 죽단화(Kerria japonica), 쥐똥나무(Ligustrum obtusifolium), 측백나무(Thuja orientalis), 편백(Chamaecyparis obtusa), 할미꽃(Pulsatilla koreana)은 중정도의 살초활성을 나타내었으나($GR_{50}$ 값, $1,000-2,000{\mu}g\;g^{-1}$), 나머지 177종 식물은 살초활성을 나타내지 않았다. 높은 살초활성을 나타낸 식물로부터 분리되는 천연활성물질들은 향후 새로운 제초제 개발을 위한 모화합물 제공, 상호대립억제 작용성을 갖는 작물 품종 개발 혹은 개량에의 활용, 그리고 친환경 유기농업용 잡초방제에의 활용이 기대된다.

  • This study was conducted to investigate the plants status of the woody landscape plants species of 38 elementary, middle, and high school in Iri city, and to obtain some fundamental information for development and use of native species of trees. The results are summarized as follows: 1. Landscape plants species of 38 elementary, middle and high school in Iri city was 55 families, 164 species, and 85, 045 trees and the average of landscape plants species of 17 elementary schools was 50.5 families, 3, 010 trees, on the other hand, middle and high school was 42.4 families, 1, 609 trees. 2. Plants species by type was Deciduous Broadleaved Tree 32.9%, Deciduous Broadleaved Shrub 25.6%, Evergreen Coniferous Tree 14.6%, Evergreen Broadleaved Shrub 9.7%, Vine 4.8%. Evergreen Trees on the other hand, according to the utilization rate 'others' was 32.2%. 3. The rate of plants species of the Evergreen Trees to the Deciduous (except Vine and 'others') was 34:66. on the other hand, rate of quantity was 50.5:49.5 and the rate of the trees to the shrubs(except Vine and 'others') was 58:42. to the plants species and to the quantity of the number of trees was 25.9:74.1. 4. Plants species of fruit plants of 38 elementary, middle and high school in Iri city was 31 species to the 164 species. The rate of frequency was Poncirus trifoliata 27.8%, Lycium chinense 25.1%, Ginkgo biloba 16.7%, to the quantity of the number of trees, fruit plants was 2, 605 trees to the 85, 045 trees and the frequency was 3%. 5. School trees and flowers of 38 elementary, middle and high school in Iri city were appointed 17 elementary school, on the other hand, middle and high schools were only 4 middle and high schools. Plants species of the best of frequency school tree was Zelkova serrata and school flower was Rosa spp.. 6. The rate of the native species to the introduced species was 73.8:26.2. on the other hand, the rate of quantity by the number of trees was 84.7:15.3 to the type, the rate of frequency of plants of native species Evergreen trees was Thuja orientalis, deciduous was Ginkgo biloba, Vine was Wisteria floribunda and 'others' was Sasa borealis. on the other hand, to the introduced species, Evergreen Trees was Juniperus chinensis. Deciduous Trees was Metasequoia glyptostroboides, and Vine was Vitis labrusca and 'others' was Phyllostachys pubescens. 7. As the fundament of the results of above the optimum selecting of woody landscape plants according to native species of trees was 29 families, 58 species. on the basis of 30 points, 28 points was 8 species Ginkgo biloba, Ulmus davidiana var. japonica, Parthenocissus tricuspidata, Elaeagnus umbellata, Fraxinus rhynchophylla, Weigela subsessilis, Koelreuteria paniculata, Lonicera japonica, 26 points was 27 species Thuja orientalis, Zelk ova serrata, Euonymus japonica etc., and 24 points was 23 species Populus tomen tiglandulosa, Juglans sinensis, Sophora japonica etc..

마이크로웨이브를 이용한 측백나무 추출물의 항균 및 항바이러스 특성 (Antibacterial and Antiviral Activities of Microwave-assisted Thuja orientalis Extracts)

  • 본 연구에서는 마이크로웨이브를 이용하여 추출한 측백나무 잎의 성분을 분석하고 세포독성, 항균 및 항바이러스 활성을 평가하였다. 마이크로웨이브 추출법에 따른 주요 성분은 catechin, leucopelargonidin, arecatannin, quinolone 및 kaempferol 유도체 등으로 나타나 플라보노이드와 탄닌 계열 물질을 함유하고 있는 것을 확인하였다. 인간 상피세포 (HaCaT)를 대상으로 한 세포독성 평가에서는 0.11 mg/mL 농도에서 독성이 나타나지 않음을 보였다. 항균 성능은 외용 소독제(의약외품) 효력평가법 가이드라인에 따라 측정하였으며 1.11 mg/mL 농도에서는 그람 음성균인 대장균에 대해 항균 효과가 낮은 반면, 그람 양성균인 황색포도상구균에 대해서는 99.9%의 항균 효율을 나타내었다. 또한, 추출물의 농도를 높이고 균과의 접촉 시간을 증가시킬 경우 대장균에 대해서도 99.9%의 항균 효율을 나타낼 수 있음을 보였다. 항균 활성 외에도 바이러스에 대한 살균제의 활성을 측정하는 표준(ASTM E1052-20)에 따라 평가 결과 인플루엔자 A (H1N1)와 SARS-CoV-2에 대해 99.99% 이상의 항바이러스 활성을 가지고 있음을 확인하였다. 이 결과들은 측백나무 추출물이 항바이러스 소독제나 표면 코팅제, 개인 보호용구 및 방역용 섬유 소재로서 응용될 수 있음을 시사한다.

양식 가물치(Channa argus)에 대한 Saprolegnia sp.의 병리학적 특성과 물곰팡이의 생장을 제어하는 정유의 영향 (Pathological Characteristics of Saprolegnia sp. to Snakehead and Effect of Essential Oils to the Growth of the Water Mold)

  • 양식 가물치로부터 Saprolegnia sp.를 분리하고 그들의 일부 생리학적 특성과 제어를 위한 식물 추출물(精油)을 탐색하였다. 분리된 Saprolegnia sp. 영양체 생장 최적 온도는 $25^{\circ}C$이었고, 최적 pH는 6.0이었으며, 최적 NaCl 농도는 0%이었다. 또한 phosphate 10 mM 농도와 casamino acid 10mg/L농도에서 균사의 생장은 잘되었다. Saprolegnia sp.의 제어 정도를 조사하기 위해 쑥, 편백, 측백나무로부터 정유를 분리하여 10, 100, 500, 1,000, 1,500, 2,000ppm에서 처리한 결과 쑥 정유는 처리농도 10 ppm 이상 농도에서부터 억제 양상이 나타나다가 농도가 높아질수록 더 많은 억제양상을 나타냈다. 측백과 편백나무 정유를 처리한 것에서는 10ppm이상 처리 농도에서부터 억제되기 시작하여 그 이상의 처리 농도에서는 상당한 균사 생장 제어 효과를 나타내다가 500 ppm 이상 처리 농도에서는 완전한 제어 효과를 나타냈다. 또한 Saprolegnia sp.에 감염된 가물치의 조직 병리학적인 조사를 실시한 결과 균사가 감염된 근육조직 부분에는 깊은 곳까지 균사가 침투하였고, 아가미의 세포는 곤봉형태를 나타냈으며, 간조직 세포에서는 일부가 괴사되었고 부분적으로 ceroid 물질이 침착되어 있었다. 또한 비장조직 세포에서는 일부 괴사된 세포를 관찰하였고, ceroid 물질의 침착을 볼 수 있었다. 신장조직은 세뇨관의 상피세포에서 일부 괴사를 관찰하였고, 조혈 조직에는 ceroid 물질의 침착상태가 나타났다.

김해시 국립용지봉자연휴양림 조성예정지의 관속식물상 (Vascular Plants in Sites Reserved for Construction of National Yongjibong Recreational Forest in Gimhae-city)

  • The aim of this study was to investigate the plants in Yongjibong Natural Recreation Forest before construction, to provide basic data that can be used for various purposes. The survey was conducted on the planned recreational forest sites from June to September 2020. The numbers of flora are summarized as follows: 348 taxa, 95 families, 234 genera, 308 species, three subspecies, 30 varieties, and seven forma. The Korean endemic plants were of nine taxa, including Fagus engleriana, Celtis choseniana, and Broussonetia x kazinoki. The rare plants were of six taxa, including Juniperus chinensis var. sargentii, Hylotelephium ussuriense, Thuja orientalis, and F. engleriana; however, Juniperus chinensis var. sargentii, H. ussuriense, T. orientalis, and Iris ensata var. spontanea were planted species. The floristic regional indicator plants (unique plants) found in this survey area were of 33 taxa, comprising three taxa of grade V, four taxa of grade IV, nine taxa of grade III, four taxa of grade II, and 13 taxa of grade I. The naturalized plants were of 26 taxa, including Rumex crispus, Phytolacca americana, and P. esculenta. Among the naturalized plants, it is necessary to prevent the spread of Ambrosia trifida, Bidens frondosa, and Tagetes minuta through continuous monitoring.

Evaluation of Cytotoxic Potential of Natural Products in Cultured Human Cancer Cells

  • In order to discover novel potent antitumor agents, methanolic extracts of approximately 180 herbal medicines were prepared and primarily evaluated for cytotoxic activity in cultured human lung (A549) and colon (Col 2) cancer cells. As a result, 17 natural product extracts were found to be active in the criteria of $IC_{50}$<$20\;{\mu}g/ml$. Especially, the extracts of Aristolochia debilis, Cynanchum ascyrifolium, Cynanchum paniculatum, Daphne genkwa, Euphorbia lathyris, Ipomoea hederacea, Magnolia officinalis, Melia azedarach var. japonica, Solanum nigrum, Thuja orientalis, and Trichosanthes kirilowii showed a strong cytotoxic potential. The flower extract of Daphne genkwa was more selective cytotoxic activity against lung cancer cells $(IC_{50};\;0.2\;{\mu}g/ml)$ compared to colon cancer cells $(IC_{50}>20\;{\mu}g/ml)$. In addition, based on the cytotoxic potential of the root extract of Cynanchum paniculatum, the further fractionation of methylene chloride partition with silica gel column chromatography was performed. Several subfractions were considered to be active, and thus indicating that further studies for the identification of active principles from these fractions might be warranted.

신경안정 생약 추출몰이 in vitro에서 신경전달효소 및 항산화계에 미치는 영향 (Effects of Some Sedative Oriental Medicines on Neurotransmission and Antioxidative System in vitro)

  • The extracts of Euphoria longan, Ziryphus jujuba, Thuja orientalis, Polygala tenuifolia, Acorus gramineus, Cyperus rotundus, Poria cocos, Uncaria rhynchophylla, and Albizzia julibrissin, which have been used as sedative drugs in Korean folk medicine, were evaluated for their effects on neurotransmission and antioxidative system in vitro. Among the tested drugs, Acorus gramineus showed most inhibitory activities on monoamine oxidase, xanthine oxidase, aldehyde oxidase, and lipid peroxidation and Uncaria rhynchophylla also inhibited most effectively GABA transaminase and DPPH radical.

자연산물을 이용한 니켈 독성에 관한 연구 (A Study on Antitoxic Effects of Natural Products Against Nickel Toxicity)

  • This study was performed to find out the detoxication effects of natural products Thuja orientalis, leaf of acorn, aloe ferox mill, pine cone, pine root, lonicerae flos on nickel toxicity. The experimental rats were divided into 3 groups such as nickel alone treatment group, natural products treatment groups before and after nickel treatments. Each group was administered with difference dose of nickel such as 6.6 mg/kg/day, 13.2 mg/kg/day, 19.8 mg/kg/day respectively, for 14 days. Natural products were administered by 400 mg per kilogram body weight rat per day. The weight change of rats, nickel concentration in organs, survival rate of rats, morphology of organs were observed in the experimental groups. The results obtained by the experiment were as follows: 1. The rats in the group treated with natural products had no effect on the weight gain. 2. The mean survival rate of rats in the nickel alone treatment group was 72.5% (6.6 mg/kg/day), 68.9% (13.2 mg/kg/day), 60.2% (19.8 mg/kg/day) respectively. 3. As for the amount of nickel accumulation in organs, it was the lowest amount treated with pine cone, pine root in that order. 4. When kidney and liver tissues were observed with as optical microscope, obvious change were visible in those tissues treated groups with Lonicerae and Aloe.

Co-contamination of Aflatoxins with Ochratoxin A and Zearalenone in Thuja orientalis Semen

  • Korea is representative of a country that consumes herbal medicines; most of the herbal medicines circulating in South Korea have been imported from developing countries in Southeast Asia, such as China and Indonesia. Recently, domestic hygiene and safety are issues that have come to the forefront, because herbal medicines currently in circulation could possibly contain contaminants or residues. Furthermore, the appearance or discovery of harmful new species due to environmental and industrial developments is becoming a social problem. Therefore, it may be necessary to consider and investigate these matters on a continual basis. Recently, mycotoxin contaminations in such foods as cereals, nuts, and powdered red pepper have been reported. They have become a problematic issue; the possibility of contamination in herbal medicines has also been considered. Nevertheless, recognition of and research into mycotoxin contamination in herbal medicines has been scarce because herbal medicine is used in only a few nations. In this research, we identified contamination by aflatoxin which is known to be the most potent mutagenic, carcinogenic, and teratogenic mycotoxin in Thujae Semen, a herbal medicine. We also found co-contaminations involving other mycotoxins, including ochratoxin A and zeraleanone.

장성 치유의 숲에 분포하는 관속식물상 (Vascular Plants Distributed in Jangseong Healing Forest)

  • The purpose of this study is to present the basic data for establishing the plan of usage and management of healing forest by surveying and analysing the vascular plants distributed in Jangseong Healing Forest. The numbers of vascular plants were summarized as 338 taxa including 85 families, 211 genera, 294 species, 3 subspecies, 33 varieties and 8 forms. In the result of life form, hemicryptophytes and geophytes were the most common and hydrophytes were the least. The rare plants were 5 taxa including Abies koreana Wilson(planted), Thuja orientalis L.(planted), Scutellaria insignis Nakai, Tricyrtis macropoda Miq. and Arisaema heterophyllum Blume. The Korean endemic plants were 12 taxa including Philadelphus schrenkii Rupr., Indigofera koreana Ohwi, Lycoris flavescens M.Y.Kim & S.T.Lee and so forth. The specific plants by floristic region were 28 taxa including 2 taxa of grade IV, 3 taxa of grade III, 7 taxa of grade II and 16 taxa of grade I. The naturalized plants were 20 taxa including Fallopia convolvulus (L.) $A.L{\ddot{o}}ve$, Ailanthus altissima (Mill.) Swingle, Veronica arvensis L., Poa pratensis L. and so forth. The invasive alien plants were Rumex acetocella L. and Ambrosia artemisiifolia L..
