Optimal Threonine:Lysine Ratio for Growing Pigs of Different Sexes

  • This study was conducted to investigate the effects of threonine:lysine ratios on growth performance, apparent nutrient digestibility and blood urea nitrogen (BUN) concentration, and to estimate the optimal threonine:lysine ratios for growing barrows and gilts. A total of 150 pigs (Landrace${\times}$Yorkshire${\times}$Duroc, $16.75{\pm}0.42kg$ average body weight, 75 barrows and 75 gilts) was randomly allotted into six treatments in a $2{\times}3$ factorial design. Six diets were formulated to contain 1.12% lysine for barrows and 1.33% lysine for gilts with three threonine:lysine ratios (50, 60 and 70%) for both barrows and gilts. Throughout the whole experimental period (16 to 56 kg body weight), there was no interaction between sex and dietary threonine:lysine ratio in average daily gain (ADG), average daily feed intake (ADFI) and feed conversion rate (FCR). Between sexes, there was a clear sex-effect showing better growth performance of barrows. Barrows consumed more feed (p<0.01) and grew faster (p<0.01) than gilts. For barrows, there was a trend to improved ADG and FCR with increasing threonine:lysine ratio. For gilts, there was a trend to improved ADG and FCR up to threonine:lysine ratio of 60%, but not significant. There was no interaction between sex and threonine:lysine ratio in nutrient digestibilities of growing pigs except for crude ash (CA). Between sexes, there were differences in nutrient digestibilities, except for calcium for which gilts showed higher a digestibility (p<0.01). Among dietary threonine:lysine ratios, there were no differences in nutrient digestibilities. Mean values of essential amino acids (EAA), non-essential amino acids (NEAA) and total amino acids (TAA) digestibilities were not affected by sex and dietary threonine:lysine ratio. There was no evidence of an interaction between sexes and dietary threonine:lysine ratio. Between sexes, total BUN concentration was lower in gilts than barrows (p<0.05). It was concluded that a 70 and 60% dietary threonine:lysine ratio for barrows (1.12% lysine) and gilts (1.33% lysine) tended to result in better growth performances and nutrient utilization and lower BUN concentration than other threonine:lysine ratios.

Effects of Crystalline Lysine, Threonine and Tryptophan Supplementation of Diets Containing Reduced Protein Levels on Performance of Growing Pigs

  • Three hundred and fifty-two growing pigs were utilized in three growth trials to investigate the effects of crystalline amino acid supplementation on growing pigs' growth performance and feed consumption. In Exp. 1, diets were formulated on the basis of digestible amino acids. Utilization of rapeseed meal plus cottonseed meal (R + C) in diet supported similar average daily gain (ADG), feed/gain (F/G) and cost per gain (p > 0.05) to group of soybean meal (SBM). In Exp. 2, 14% CP diets containing equal amount of cottonseed meal and rapeseed meal were formulated with the addition of threonine and/or tryptophan. Supplementation with threonine improved ADG and F/G (p < 0.05). Supplementation with only tryptophan made no benefits for ADG of growing pigs. In Exp. 3, diets with different threonine level were formulated. Increasing dietary threonine to 0.54% improved (p < 0.05) ADG and F/G compared to 0.45% dietary threonine. Increase in dietary threonine did not improve ADG (p > 0.05) but decreased F/G (p < 0.05) because of a decrease in average daily feed intake. Pigs fed diet with dietary threonine/lysine ratio of 67.5% supported optimum growth performance.

  • Controlling the residual glucose concentration is important for improving productivity in $\text\tiny{L}$-threonine fermentation. In this study, we developed a procedure to automatically control the feeding quantity of glucose solution as a function of ammonia-water consumption rate. The feeding ratio ($R_{C/N}$) of glucose and ammonia water was predetermined via a stoichiometric approach, on the basis of glucose-ammonia water consumption rates. In a 5-L fermenter, 102 g/l $\text\tiny{L}$-threonine was obtained using our glucose-ammonia water combined feeding strategy, which was then successfully applied in a 500-L fermenter (89 g/l). Therefore, we conclude that an automatic combination feeding strategy is suitable for improving $\text\tiny{L}$-threonine production.

형질전환된 Escherichia coli변이주에서 Sodium citrate를 이용한 고농도 L-Threonine 생산 (Hyperproduction of L-Threonine by Adding Sodium Citrate as Carbon Source in Transformed Escherichia coli Mutant.)

  • 유용성이 확보된 L-threonine의 효율적인 발효생산을 위하여 생산균주 E. coli MT201를 유전자재조합을 통하여 개량하고 적절한 탄소원을 발굴하여 전체적인 생산량 증대를 도모하였다. 먼저, 5 liter발효조에서 유가식 배양을 통하여 생산균주 E. coli MT201이 균체량이 52($OD_{660}$)일때 57 g/1의 생산량을 보이는 것을 확인하였다. L-Threonine의 생산성 향상을 위하여 균체 내에서 생합성 전구물질인 oxaloacetate를 충분하게 공급하기 위해 C. glutamicum 유래의 pyruvate carboxylase의 유전자를 plasmid pPYC의 형태로 E. coli MT201에 도입하였다(E. coli MT/PYC). 그렇지만 E. coli MT/PYC을 배양한 결과로부터 E. coli MT201와 비교할 때 균체증식 및 생산량이 모두 감소하는 경향을 보였으며, 이를 해결하기 위하여 플라스크배양을 통하여 포도당과 sodium citrate를 1.5:3.5의 비율로 배지 중에 첨가하였을 때 이들 문제가 개선되는 것을 관찰하였다. 상기 비율의 탄소원 조건하에서 5liter 발효조를 이용한 유가식 배양에서 배양 75시간째에 L-threonine의 생산량 및 균체량($OD_{660}$)이 각각 75.7 g/l와 48로 효율적으로 향상되는 것을 알 수 있었다. 이는 과도한 anaplerosis에 의한 TCA 회로의 불균형을 중간산물인 citric acid를 sodium citrate의 형태로 공급함으로써 E. coli MT/PYC에서 균체증식이 정상화됨을 의미한다.

Effect of increasing levels of threonine relative to lysine on the performance and meat quality of finishing pigs

  • Objective: The present study aimed to evaluate the effects of varying standardized ileal digestible lysine:threonine (SID Lys:Thr) ratio in the diet on the performance and meat quality of finishing pigs. Methods: In total 192 crossbred pigs ([Landrace×Yorkshire]×Duroc, 17 weeks old), with an initial body weight (BW) of 70.6±3.9 kg were used in an 8-wk trial. Pigs were randomly allotted to one of six dietary treatments based on their initial BW and sex (8 replications; 4 pigs per pen, 2 barrows and 2 gilts). The pigs in the 6 treatments were fed diets having different SID Lys:Thr ratios such as 1:0.65, 1:0.66, 1:0.67, 1:0.68, 1:0.69, and 1:0.70. Results: A linear increment (p<0.05) in average daily gain (ADG) and trends in reduction in feed conversion ratio (FCR) were observed during day 29 to 56 of the experiment and the apparent total tract digestibility (ATTD) of dry matter tended to increase linearly (p = 0.094) at the end of the experiment (day 56) with the increase in the dietary SID Lys:Thr ratios. The backfat thickness and lean percentage increased (linear effect, p<0.05) on day 28. In addition, at day 56, a linear (p<0.05) increment in lean percentage was observed. Significant quadratic responses (p = 0.02) for pH and drip loss at day 7 (p = 0.02), a linear increase (p<0.05) in cooking loss and drip loss at day 7, and a trend in quadratic response (p = 0.07) in the lightness of meat color (L*) were observed, whereas other meat quality indices were unaffected by varying the SID Lys:Thr ratios. Conclusion: The SID Lys:Thr ratio for maximum ADG, minimum FCR and enhanced digestibility was found to be 0.70. However, for carcass trait and meat quality, the SID Lys:Thr ratio of 0.65 was enough.

Estimation of the Optimal Ratio of Standardized Ileal Digestible Threonine to Lysine for Finishing Barrows Fed Low Crude Protein Diets

  • Two experiments were conducted to determine the standardized ileal digestible (SID) lysine (Lys) requirement and the ideal SID threonine (Thr) to Lys ratio for finishing barrows. In Exp. 1, 120 barrows with an average body weight of $72.8{\pm}3.6$ kg were allotted to one of six dietary treatments in a randomized complete block design conducted for 35 d. Each diet was fed to five pens of pigs containing four barrows. A normal crude protein (CP) diet providing 15.3% CP and 0.71% SID Lys and five low CP diets providing 12% CP with SID Lys concentrations of 0.51, 0.61, 0.71, 0.81 and 0.91% were formulated. Increasing the SID Lys content of the diet resulted in an increase in weight gain (linear effect p = 0.04 and quadratic effect p = 0.08) and an improvement in feed conversion ratio (FCR) (linear effect p = 0.02 and quadratic effect p = 0.02). For weight gain and FCR, the estimated SID Lys requirement of finishing barrows were 0.71 and 0.71% (linear broken-line analysis), 0.79 and 0.78% (quadratic analysis), respectively. Exp. 2 was a 26 d dose-response study using SID Thr to Lys ratios of 0.56, 0.61, 0.67, 0.72 and 0.77. A total of 138 barrows weighing $72.5{\pm}4.4$ kg were randomly allotted to receive one of the five diets. All diets were formulated to contain 0.61% SID Lys (10.5% CP), which is slightly lower than the pig's requirement. Weight gain was quadratically (p = 0.03) affected by SID Thr to Lys ratio while FCR was linearly improved (p = 0.02). The SID Thr to Lys ratios for maximal weight gain and minimal FCR and serum urea nitrogen (SUN) were 0.67, 0.71 and 0.64 using a linear broken-line model and 0.68, 0.78 and 0.70 using a quadratic model, respectively. Based on the estimates obtained from the broken-line and quadratic analysis, we concluded that the dietary SID Lys requirement for both maximum weight gain and minimum FCR was 0.75%, and an optimum SID Thr to Lys ratio was 0.68 to maximize weight gain, 0.75 to optimize FCR and 0.67 to minimize SUN for finishing barrows.

Phosphorylation of the Nucleocapsid Protein of Bovine Coronavirus Expressed with a Recombinant Baculovirus Vector

  • Post-translational modifications of the nucleocapsid protein of bovine coronavirus (Quebec strain) were investigated. Coronavirions were radiolabelled in vivo with inorganic $[^{32}P]$orthophosphate and analysed by SDS-PAGE, followed by autoradiography. A single polypeptide with a migration rate of 55 KDa was identified by metabolic phosphate labelling, demonstrating that the nucleocapsid protein of bovine coronavirus was a phosphoprotein. A gene encoding the nucleocapsid protein was inserted immediately downstream from the polyhedrin promoter of Autographa californica nuclear polyhedrosis baculovirus. Spodoptera frugiperda cells infected with this recombinant baculovirus synthesized a 55 KDa polypeptide, as demonstrated by immunoprecipitation with anti-nucleocapsid monoclonal antibody. The recombinant nucleocapsid protein synthesized in Spodoptera cells could also be labelled by $[^{32}P]$orthophosphate. Phosphoamino acid analysis showed that both serine and threonine residues were phosphorylated in authentic, as well as in recombinant nucleocapsid proteins, with a relative phosphorylation ratio of 7:3. Our studies demonstrated that the nucleocapsid protein of bovine coronavirus was a serine and threonine-phosphorylated protein and that Spodoptera insect cells were able to properly phosphorylate the relevant foreign proteins.

오징어를 이용한 조미 건조포의 개발 (Development of Seasoned and Dried Squid Slice)

  • 주요 수산가공자원의 하나인 오징어의 소비 촉진을 위하여 오징어포의 제조를 시도하였다. 오징어의 효율적 이용 측면에서 조미 오징어포의 제조는 오징어를 70% 이상으로 첨가하는 것이 적절하리라 판단되었다. 오징어와 수리미를 70:30의 비율을 사용하여 제조한 조미 오징어포는 조미 수리미포에 비하여 총 아미노산의 함량이 높았으며, 주요 아미노산으로는 glutamic acid, leucine, lysine 및 threonine 등이었다. 또한, 조미 오징어포의 칼슘 및 인 함량은 조미 수리미포에 비해 높았으며, 조미 오징어포의 유리아미노산의 총함량 및 taset value는 각각 3,199.1 mg/100g 및 402.00이었고 맛에 지대하게 영향을 미치는 아미노산은 glutamic acid 및 aspartic acid이었다.

Effects of Amino Acid Supplementation on Growth Performance for Weanling, Growing and Finishing Pigs

  • Four feeding trials with 260 pigs were conducted to evaluate the effects of supplementing the diet with different amino acids on growth performance and blood metabolites for weanling, growing and finishing pigs. One hundred twenty weanling pigs (Exp. 1, BW 8 kg), eighty growing pigs (Exp. 2. BW 20 kg), thirty growing pigs (Exp. 3, BW 29 kg) and thirty finishing pigs (Exp. 4, BW 50 kg) were randomly allotted to different dietary treatments according to sex and body weight. Pigs weight and feed consumption were measured at initiation and termination of each trial with 4 weeks. At the end of trial, blood samples from three pigs selected in each pen (Exp. 1) and each pig (Exp. 2) were obtained to determine the level of blood urea nitrogen, glucose, insulin and cortisol in the serum. In Exp. 1, pigs fed diet supplemented both with lysine and methionine had the best feed conversion ratio (p < 0.05), but no significant differences (p > 0.05) were observed in ADG and ADFI. Pigs receiving control diet obtained the obtained the optimal ADG (p < 0.05), ADFI (p < 0.05) and F/G for the whole period. No differences were detected in serum glucose, insulin and cortisol concentrations. In Exp. 2, pigs receiving the control diet exhibited the lowest serum urea nitrogen (p < 0.05), ADG, F/G and serum insulin concentration increased linearly (p < 0.05) with the inclusion of lysine, methionine, threonine and tryptophan in diets. No significant differences (p > 0.05) were detected for glucose and cortisol content in pigs serum among dietary treatments. In Exp. 3 and 4, pigs growth rate increased linearly (p < 0.01), and feed conversion efficiency was also improves by addition of lysine, methionine, threonine and tryptophan. In conclusion, pigs fed diets supplemented with lysine, methionine, threonine and tryptophan together obtained optimal growth performance in growing and finishing periods.

키토산과 견사 단백질 복합체의 특성 (Characteristics of Chitosan-Silk fibroin composites)

  • 기능성 다이어트 식품을 개발을 목적으로, 키토산 과견 피브로인의 복합체를 만들고 성질을 조사하였다. 키토산은 대동새우로부터 추출하였고, 견사 피브로인 섬유는 누에로부터 추출하였다. 견사의 특성을 알아보기 위하여 용해도를 조사한 결과, 용해도는 8M Libr 이상의 농도와 4$0^{\circ}C$ 이상에서 가장 높았다. 견사 피브로인의 아미노산 조성은 천연견사 및 용해시킨 피브로인과 같았다. 키토산, 견사 피브로인 복합체의 아미노산 조성은 glycine, alanine, serine, tyrosine, threonine 및 glutamic acid 순서로 많았다. 또한 IR 스펙트럼 결과, 키토산.견사 피브로인 복합체는 배합비율에 따른 스텍트럼상의 차이는 거의 없었다.
