  • The level of oxidative tissue damage caused by free radicals generated from ethanol oxidation was determined in the myocardium of chronic ethanol fed-rats and the protective action of various radical scavenging enzymes was monitored, also. Adult male Sprague-Dawley rats were given ethanol in an amount of 36% of total calories via Lieber-DeCarli liquid diet for 6 weeks. Control group was pair-fed with the diet containing isocaloric amount of dextrin-maltose instead of ethanol. Chronic ethanol administration resulted in the increased amount of myocardial thiobarbituric acid reactive substance(TBARS), th parameter of lipid peroxidation, under our experimental condition. Chronic ethanol ingestion did not cause any change in activities of either glutathione peroxidase or glutathione reductase and glucose-6-phosphate dehydrogenase were decreased after ethanol treatment. Therefore, chronic ethanol administration seemed to cause considerble changes in cellular defense function against oxidative tissue damage in rat myocardium through glutathione utilizing system and radical generation system. However the ultimate net result of chronic ethanol inestion on the myocardium of rat was the oxidative tissue damage revealed by increased TBARS content.

새우젓 발효 중 이화학적 특성 및 색도 변화 (Changes in the Physicochemical Properties and Color Values of Salted and Fermented Shrimp)

  • This study was performed to examine the effects of salt addition level on changes in the physicochemical properties and color values of salted shrimp during fermentation. Fresh shrimp were salted with different concentrations (8, 18, 28%) of Chunil salt and fermented at $20^{\circ}C$ for 160 days. In the 8% salted shrimp treatment, volatile basic nitrogen (VBN), amino nitrogen (AN), and trimethylamine (TMA) contents rapidly increased during the fermentation period, while in the 18 and 28% salt groups these increases were reduced throughout the 160 days of fermentation. Thiobarbituric acid (TBA) values increased rapidly for 120 days of fermentation and then decreased. The thiobarbituric acid values of the 18, and 28% salted and fermented shrimp groups were lower than the that of the 8% group. In addition, Hunter's color L, a, and b values decreased as the fermentation period increased.

  • Antioxidative properties of fifteen anthraquinone derivatives, including eleven atural anthraquinones isolated from the roots of Morinda elliptica and four from synthetic origin were evaluated using thin layer chromatography (TLC), ferric thiocyanate (FTC) and thiobarbituric acid (TBA) methods. Five of the compounds, nordamnacanthal, damnacanthal, 2-formyl-1-hydroxyanthraquinone, morindone and alizarin showed higher antioxidative activity than standard natural antioxidant, ${\alpha}-tocopherol$, on the FTC assay. Morindone and alizarin showed the strongest antioxidant activity. The results from the bioassay using TBA method correlated well with the results of the FTC method.

Effects of Oxygen Scavenging Package on the Quality Changes of Processed Meatball Product

  • Processed meatball products were packaged in a passive package without oxygen scavenger as 1 control and 3 active packages of which have PP-based oxygen scavenger master batch materials (OSMB) of 40, 80, and 100%(w/w) in the middle layer and stored at 23 and $30^{\circ}C$ up to 9 months. Quality changes of packaged products were evaluated by measuring the oxygen concentration of the headspace in containers, thiobarbituric acid (TBA), color, and flavor. The oxygen concentration of the package having 100% OSMB was lower than those of 40 and 80%. The color changes and TBA values of the meat ball in the package containing 100% OSMB were the least among the treatments. Using principal component analysis (PCA), the control showed more flavor change than the packages containing oxygen scavenger. As a result, all active packages could extend the shelf life of the meatball products compared with that of the passive package.

경옥탕(瓊玉湯)의 항산화작용(抗酸化作用)에 대한 실험적(實驗的) 연구(硏究) (Experimental study on antioxidant action of Kyungoktang)

  • In order to evaluate the effectiveness of KOK(kyungokko) experimentally used for longevity hematology, erythrocyte hemolysis and TBA(thiobarbituric acid) were measured, after LKOK(low concentration KOK) and HKOK(high concentration KOK) were administrated to mice pretreated with AAPH (2,2'-Azobis(2-amidinopropane), hydrocholoride, free radical inducer. In hematology WBC and hemoglobin were reduced significantly, while the number of platelet was significantly incresed as compared with control. Erythrocyte hemolysis by damage of free radical on cell membrane of RBC and TBA(Thiobarbituric acid) were significantly inhibited in both sample groups as compared with control. These results suggest KOK had antioxidant effect.

Effects of Coptidis Rhizoma on Lowering Lipid and Oxidative Stress

  • The effects of hwangryun(Coptidis Rhizoma) on lowering lipid and oxidative stress in the induced obesity rat was observed. The concentration of plasma triglyceride in hwangryun treatment groups showed the low values compared to the control group, and as the increased hwangryun, the concentration of triglyceride decreased. The concentration of plasma total cholesterol decreased in all hwangryun treatment groups. However the concentration of plasma HDL-cholesterol and LDL-cholesterol showed no significantly difference in all the treatment groups. Thiobarbituric acid reactive substance(TBARS) concentration in plasma and liver showed low values in all hwangryun treatment groups compared to the control group. Glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) activities showed no significantly difference in all the treatment groups. However catalase(CAT) activity showed a tendence to increase in hwangryun groups, and in 200mg/kg hwangryun treatment group showed significantly a high value than the control group. Summarizing the results above, hwangryun has the functional materials that lowering lipid and works with oxidative stress.

Detectio of Malonaldehyde-thiobarbituric Acid (MA-TBA) Complex by High Performance Liquid Chromatography(HPLC) in a Model System

  • Various concentrations of malonaldehyde (MA) produced upon hydrolysis of 1, 1, 3,3-tetraethoxypropane (TEP) were reacted with 2-thiobarbituric acid (TBA)and th e contents of MA-TBA complex were measured both with spectrophotometer and high performance liquid chromatography (HPLC). As the concentrations of MA-TBA increased, their absorbances and the corresponding HPLC peak areas increased. The correlation coefficient between absorbances and HPLC peak areas of MA-TBA peaks from the other compounds and butanol extraction of the complex increased its recovery its recovery by 29.4% . Measurement of the content of MA-TBA complex for monitoring the development of lipid oxidation was proven to be successful with the use of high performance liquid chromatography.

가열쇠고기 지방질 산화에 대한 김치종류별 항산화작용 (Antioxidative Effect of Different Kinds of Kimchi on the Lipid Oxidation of Cooked Meat)

  • This study was carried out to investigate the antioxidative effect of different kinds of kimchi on the lipid oxidation of cooked meat in model systems. Model systems of cooked ground meat(CGM), CGM-Chinese cabbage kimchi(CK), CGM-radish kimchi(RK), and CGM-mustard leaf kimchi(MLK) were prepared and their oxidation were evaluated during the storage at 4$^{\circ}C$ for 5 weeks. Thiobarbituric acid(TBA) values of CGM significantly increased with the storage at 4$^{\circ}C$ for 5 weeks. Thiobarbituric acid(TBA) values of CGM significantly increased with the storage time, however, TBA value of CGM-CK, CGM-RK, and CGM-MLK lowered and that of CGM-MLK was lowest. Antioxidative effect of CGM-MLK increased with the addition levels of kimchi in the system. And also in the model systems which were prepared with CGM and MLK in different fermentation periods, the antioxidative effect was highest in the properly fermented-kimchi.

Glutathione suppresses lipid oxidation of Clanis bilineata larvae meat during frozen storage

  • The lipids of Clanis bilineata larvae meat (CBLM) are susceptible to oxidation, and thus the commercial and consumption values of CBLM decrease during frozen storage. In the present study, peroxide values (PV), thiobarbituric acid-reactive substances (TBARS), free fatty acid (FFA) content, fatty acid composition, and likeness (palatability) score of CBLM were determined to investigate the effect of glutathione on the oxidation of CBLM lipids. Glutathione decreased the PV, TBARS, and FFA content, maintained fatty acid composition, and increased the likeness (palatability) score of the CBLM, indicating that glutathione can be used as a cryoprotectant to extend the shelf life of CBLM.

방사선 조사 사료를 섭취한 Mouse의 혈장, 간, 소장 점막의 과산화지질과 림프구 DNA의 산화적 손상 (Plasma, Tissue Thiobarbituric Acid Reactive Substance and Lymphocyte Oxidative DNA Damage in Mouse Fed Gamma Irradiated Diet)

  • Food irradiation has been steadily increasing in many countries in line with increasing international trade and concerns about naturally occurring harmful contaminants in food. Although irradiation provides an excellent safeguard for the consumer by destroying almost 100% of harmful bacteria, it is necessary to ensure the safety of irradiated foods. This study was performed to investigate the effect of an irradiated diet on lipid peroxidation in the plasma, liver, small intestinal mucosa, and lymphocyte DNA damage in mice. Eight-week old ICR mice were assigned to two groups to receive either non-irradiated or irradiated (10 kGy) diets containing 20.38% fish powder and 6.06% sesame seeds for 4 weeks. The resulting changes in the degrees of lipid peroxidation were evaluated based on the level of plasma and liver thiobarbituric acid reactive substance (TBARS), transmission electron micrograph of jejunal mucosa, and free radical-induced oxidative DNA damage in lymphocytes, as measured by alkaline comet assay (single cell gel electrophoresis). The peroxide values of the gamma irradiated diet were measured every week, and the sample for comet assay was taken at the end of the four week experimental period. There was no significant difference in food efficiency ratio between the two groups. The peroxide values of the diet were immediately increased to 35.5% after gamma irradiation and kept on increasing during storage. After 4 weeks, no differences in tissue or plasma TBARS value were observed between the two groups, but epithelial cells of jejumum showed osmiophillic laminated membranous structures, considered as myelin figures,. The oxidative DNA damage expressed as tail moment (TM) increased 30% in the blood lymphocytes of the mice fed the irradiated diet. In conclusion, the comet assay sensitively detected differences in lymphocyte DNA damage after feeding with the irradiated diet for 4 weeks. However, in order to ensure the safety of irradiated foods, it would be more useful to conduct a long-term feeding regimen using an irradiated diet and examine the level of lipid peroxidation and the state of oxidative stress in a greater range of organs.
