Bovine piroplasmosis caused by Theileria sergenti is a major cause of economic loss in livestock industry. Five cattle infected with Theileria sergenti showing severe and fatal anemia, confirmed by indirect immunofluorescent assay(IFA), were used in this study. Four cattle were treated with diminazene aceturate and one was not treated as the control. The therapeutic effect of diminazene aceturate against Theileria sergenti infection was monitored by detecting the 32kDa polypeptide specific for Theileria sergenti by the western blotting with both polyclonal and monoclonal antibodies. The 32kDa polypeptide detected at the beginning of diminazene aceturate treatment was not detectable after the treatment. It is postulated that the detection of the 32kDa polypeptide specific for Theileria sergenti may be a good tool for the diagnosis and monitoring the treatment progress of Theileria sergenti infection.
Theileria (T.) buffeli (formerly T. sergenti/T. orientalis) is the major hemo-protozoan distributed in the Far East Asian countries such as Korea, China and Japan. It is responsible for the clinical symptoms of anorexia, ateliosis, anemia, fever and icterus. It also causes abortion and sudden death under severe cases, resulting in economic losses for many livestock farms. The objective of this study was to analyze the genetic diversity of the major surface protein (Msp) gene in T. buffeli in Holstein in Korea, and we characterized the association of the diversification of the Msp gene and its relationship with the pathogenicity of Theileria. For this, complete blood counts and Theileria PCR sequence analysis were performed from 57 Holstein in Jeju Island. A total of 26 PCR positive Holstein (16 anemic and 10 non-anemic) were then randomly selected based on 18s rRNA sequence typing of the Theileria Msp gene. The DNA sequence of the T. buffeli Msp gene in Holstein showed 99.0%, 99.2%, 99.9%, 99.5%, 98.7%, 98.4% and 98.4% homology with T. sergenti, Theileria spp., T. sergenti, Theileria spp., Theileria spp., Theileria spp. and Theileria spp., respectively. The result showed a genetic variation of 57.7% (type I), 3.8% (type II), 15.4% (type III), 7.7% (type IV), 13.5% (type V) and 1.9% (type VI). Type I is the most frequent type in both anemic and non-anemic Holstein while type II was found in only non-anemic Holstein. This results of our study help confirm the diversity of Msp gene types and demonstrate that the gene type distribution of Msp genes varies among Theileria-infected Holstein in Jeju Island.
The objective of this study was to investigate the prevalence of tick-borne pathogens in the Korean water deer (Hydropotes inermis argyropus). Pathogens were identified using PCR which included Anaplasma, Ehrlichia, Rickettsia, and Theileria. Rickettsia was not detected, whereas Anaplasma, Ehrlichia, and Theileria infections were detected in 4, 2, and 8 animals, respectively. The most prevalent pathogen was Theileria. Of the 8 Theileria-positive animals, 2 were mixed-infected with 3 pathogens (Anaplasma, Ehrlichia, and Theileria) and another 2 animals showed mixed-infection with 2 pathogens (Anaplasma and Theileria). Sequencing analysis was used to verify the PCR results. The pathogens found in this study were identified as Anaplasma phagocytophilum, Ehrlichia canis, and Theileria sp. To the best of our knowledge, this is the first report identifying these 3 pathogens in the Korean water deer. Our results suggest that the Korean water deer may serve as a major reservoir for these tick-borne pathogens, leading to spread of tick-borne diseases to domestic animals, livestock, and humans. Further studies are needed to investigate their roles in this respect.
Six different sequences types(A through E and H) and a subtype(Bl) of the small subunit ribosomal RNA(SSUrRNA) gene were found in bovine Theileria isolates from different areas of Korea and Japan. The sequences were aligned and three hypervariable regions were observed in the nucleotide position ranges 212~231, 261~270 and 632~690. Five of the Theileria isolates yielded sequence type A; these were the field isolates KCB, KCN, and KCJ, and the laboratory stock KLS, all from Korea, and a single isolate from Japan (JHS). This sequence type is identical to the SSUrRNA gene sequence listed for Theileria buffeli (GenBank Accession No. Z15106) from Marula, Kenya. The Korean field isolate KKB yielded only a single sequence type (B), but multiple sequence types were found in some isolates. For example, KCB and JHS isolates yielded both types A and B ; isolate KKW showed types B and H; isolate KCN showed types A, C, and D ; and isolate KCJ showed types A, B, E, and a subtype B1. Finding of the multiple sequences SSUrRNA gene sequences suggests that bovine Theileria isolates from both Korea and Japan may consist of mixed populations.
Theileria spp.의 생활환에 대해서는 여러종류의 책을 통해서 잘 소개되어 있다. 그중 Theileria spp.의 분열소체 즉, merozoite는 주로 숙주의 적혈구내에 존재하는데 진드기에 의해 흡혈되지 않으면 더이상 발육하지 못하고 생을 마감한다고 여겨 왔다. 그러나 적혈구내 merozoite가 임파구에 다시 들어가 schizont로 복귀하여 분열 증식된다는 가설은 아직까지 증명된 바 없다. 본 실험은 T sergenti merozoite의 schizont로의 복귀를 입증하고자 수행되었다. T sergenti에 감염되지 않은 3개월령의 송아지를 비장적출시킨 후 T sergenti merozoite에 감염된 순수적혈구를 인공감염시켰다. 인공감염후 경시적으로 혈액과 임파액을 채취하여 적혈구내 T sergenti 감염을 조사하고 백혈구 감별혈구를 계산하였으며 임파구내 schizont 출현을 관찰하여 다음과 같은 결과를 얻었다. 1. 혈구내원충 감염율(parasitemia : PE)은 인공감염후 28일째 최고치인 10.5%를 보였으며 그후 5%이내의 수준을 유지하다가 70일째 다시 8.5%의 상승점을 보였다. 2. 백혈구 감별혈구계산에서는 감염초기에는 호중구가 주종을 이루다가 감염후 19일을 기점으로 임파구(60~80%)가 급격히 증가하여 실험종료 때까지 유지되었다. 3. 인공감염후 19~23일, 59~63일 사이에 말초혈액내 임파구에서 분열 증식하고 있는 schizont를 관찰할 수 있었다. 4. 인공감염후 7일부터 림프액내 임파구의 크기가 커지면서 blast-formation이 진행되었으며 실험종료때까지 유지되었다. 이상의 결과로 보아 적혈구내 merozoite가 임파구에 다시 들어가 schizont로 복귀하여 분열 증식함을 입증하여 기존의 T sergenti 생활사는 수정되어야 된다고 사료된다.
The aim of the current study was to analyze the diversity of the major surface protein (Msp) gene in Theileria buffeli, which is known as the major antigenic protein recognized by the immune system of the host. In addition, we characterized the diversification of the Msp gene and its relationship to with the pathogenicity of Theileria. Complete blood counts (CBC) and Theileria 18S rRNA PCR sequence analysis were performed for 177 Korean indigenous cattle (KIC) in Jeju Island. A total of 28 KIC (16 anemic and 12 non-anemic KIC) were then randomly selected based on 18s rRNA PCR positive samples for sequence analysis of the Theileria Msp gene, which was performed twice for each specimen. The resulting 56 Msp gene sequences were classified into five antigenicity types (type I to V), according to the variable region (517-571 bp), which exhibited high similarity (${\geq}$ 98.9%) to several available GenBank sequences (Theileria spp. from China-EU584237; T. sergenti from China-DQ078264; Theileria spp. from Thailand-AB081329; Theileria spp. from Japan-AB218442; T. sergenti from Japan-AB016280). The 56 Msp sequences consisted of 22, 15, 9, 8, and 2 cases of type I to type V Msp genes, respectively. The most prevalent type in both anemic and non-anemic KIC was type I (37.5% in anemic and 41.7% in non-anemic). Among the remaining types, type II was the most prevalent (37.5%) in anemic KIC, while type IV was the most prevalent (25%) in non-anemic KIC. The results of our study help confirm the diversity of Msp gene types and demonstrate that the gene type distribution of Msp genes varies among Theileria-infected KIC in Jeju Island.
소 림프구내의 Theileria sergenti감염을 확인하기 위하여 T. sergenti감염혈액에서 림프구를 분리한 후 중합효소연쇄반웅을 실시하였다. 또한,분리한 림프구내의 T. sergenti감 염을 증명하기 위하여 IFA test와 acridine orange stain을 실시하였다. 그 결과 다음과 같은 성적을 얻을 수 있었다. T. sergenti 감염헐액의 전혈과 림프구를 각각 생리식염수로 2배율 연 속회석하여 중합효소연쇄반응을 실시한 결과, 림프구내에서는 1,024배 회석배율까지 T. sergenti의 genomic DNA가 중폭되었으며, 전혈내에서는 256배 회석배율까지 증폭되었다. 그리 고 중합효소연쇄반응으로 T. sergenti 감염이 확인된 림프구를 이용하여 IFA test와 acridine orange 염색을 실시한 결과, 림프구내에 T. sergenti가 존재하는 것을 증명할 수있었다. 한편, 중합효소연쇄반응을 이용한 림프구내의 T. sergenti 감염의 진단 유용성을 확인하기 위하여 전 북지역에서 사육중인 소 16두를 대상으로 이들의 혈액으로 PCR 증폭을 실시하였다. 그 결과 전혈에서 genomic DNA를 취한 경우에는 3두(18.8%)만이, 그리고 림프구에서 genomic DNA를 취한 경우에는 11두(68.8%)의 소에서 T. sergenti DNA의 증폭을 관찰할 수 있었다.
Small subunit ribosomal RNA (SSU rRNA) gene nucleotide sequences of bovine ReiLerin isolates from Korea (KLS and KCB) and japan (JHS) were determined. The genes from each isolate were amplified by the polymerase chain reaction and the approxi- mately 1.8 kb product cloned and sequenced by a modified dideoxynucleotide method. Overlapping gene segments produced with a series of primers were sequenced, resoRting in a complete DNA sequence for both forward and reverse strands of the SSU rRNA genes of each isolate. SSU rRNA gene sequences (termed Type A) were identical among the bovine ReiLeri,n isolates from Korea and the isolate from Japan. A GenBank data library homolo- gy search showed the sequence to be the same as that listed as leiLeyia buKeLi isolated from cattle in Marula, Kenya.
The spleen and placenta from the aborted fetuses as well as lymphnodes and placenta of the corresponding dams naturally infected with T sergenti were used to localize the parasite antigens by immunohistochemical staining for the possible congenital transmission of theileriosis. Parasite-specific antigens were detected immunohistochemically by incubating the sections with specific monoclonal antibody prepared against 34KD surface antigen of T sergenti and visualized via the avidin biotin complex(ABC) method. Specific T sergenti antigen was detected in the sections of formalin or acetone-fixed fetal spleens and placenta. Similar antigens were also demonstrated in lymphnodes and placentas of the corresponding dams. It is concluded that this technique will eventually play an important role in specialized diagnostic laboratories in the verification/evaluation of congenital infection with T sergenti.
Immunoblot analysis utilizing bovine sera from naturally or experimentally infected with Theileria sergenti were used to determine the immunodominant polypeptides of T sergenti (Korean isolated. The previously recognized major bands, 18 kDa,29 kDa, 34 kDa and 45 kDa, were excised after electrophoresis and transfer to PnF membrane. The individual bands were sequenced. The 34 kDa polypeptide which was the most antigenic and immunogenic peptide was observed in the Western blot. However, Chou-Fasman prediction sites (antigenic site) for antigen determinants of the 45 kDa, 34 kDa, 29 kDa and 18 kDa polypeptide were 6, 4, 2 and 0, respectively. However, the 45 kDa polypetide showed no reaction with anti- T sergenti hyperimmune serum.
