  • In the present study, an attempt has been made to assess whether the effect of benzene extract of Ocimum sanctum leaves on the ultrastructural changes in the epithelial cells of the cauda epididymis, its subsequent recovery in the seminiferous epithelium and fertility of male albino rats. Wistar strain male albino rats were orally administered benzene extract of 250 mg/kg body weight of O. sanctum leaves followed by subsequent recovery maintaining suitable controls for 48 days. Results indicate decrease in the weights of testis, epididymis and seminal vesicles. Other accessory organs were not affected. Total count, cell and nuclei diameters of germ cells and Leydig cells were reduced. Cauda epididymis exhibited significant reduction in epithelial height and nuclei diameter of epithelial cells. Cells showed vacuolization with exhibit of signs of degeneration. Ultra study revealed that, in general, the cauda epididymis was affected and in particular, the principal, clear and basal cells were highly disturbed. Further, there was decrease in the size of lipid droplets, mitochondria, Golgi complex, endoplasmic reticulum and accumulation of lysosomal bodies. Fertility performance test showed no implantation in female rats mated with O. sanctum treated rats. Moreover, their recovery after withdrawal of treatment was observed suggesting that the effect of the treatment is transient and reversible. A recovery period resulted in normal spermatogenesis and fertility, suggesting reversible antispermatogenic and antifertility effects of the plant.

  • These studies were carried out to investigate the properties of H-Y antigen purified by immunoaffinity chromatography using monoclonal H-Y antibody. Immunoaffinity column was prepared by the coupling of monoclonal antibody to the Aminolink Coupling Gel. Murine testis supernatant was applied onto the column and eluted by O.lM glycine-HCl buffer and 31${\mu}g$ of H-Y Ag was eluted from one testis. Purified H-Y Ag strongly reacted with Con A and lentil from 6 different kinds of lectins tested, which may indicate that sugar moiety of H-Y Ag is composed of glucose, mannose and their derivatives. Con A-sepharose affinity column was used to purified H-Y Ag based on that H-Y Ag is glycoprotein. The fraction eluted by 0.2M Me-${\alpha}$-D-mannoside from the column loaded with murine testis supernatant was identified to be H-Y Ag by dot blot test. Molecular weight of the purified H-Y Ag was estimated by Sepharose G-75 gel filtration and SDS-PAGE, and showing that it was about 67,000 dalton. In fluorescence test, the ratio of XY embryos and XX embryos was 1:1.

  • Boar taint, an unpleasant odor of pork, is associated with two substances including androstenone and skatole. Testosterone is a steroid hormone as well as a strong predictor for androstenone secretion. Isoflavones of soy origin play a role in modulating the metabolism of sex hormones. Although several methods responsible for reducing boar taint are under investigation, the precise mechanism by which isoflavones reduce testosterone has not yet been identified. The objective of the current study was to investigate the effects of isoflavones extracted from a soy by-product on the concentration of serum testosterone in mouse. A total of 24 mice were supplemented with basal diet (control), daidzin plus genistin mix (T1), or isoflavone extracts (T2). After 11 days of treatment, size and weight of testis, as well as the concentration of sex hormones, including testosterone and estrogen, were analyzed. There was no difference in size or weight of testis from mice among control, T1, and T2. Serum testosterone levels were significantly decreased (p < 0.05) both in T1 and T2 when compared with the control group. Furthermore, estrogen concentration in blood was increased (p < 0.05) in T2 (numerically increased in T1) compared with the control group. Taken together, the use of isoflavones extracted from soy by-products would be a plausible strategy for reducing testosterone level, ultimately reducing boar taint without castration of piglets.

  • These experiments were undertaken in order to find out the useful clinicopathological diagnostic methods of cadmium poisoning in dogs. Twenty-one dogs were divided into a control group and 6 experimental groups. The experimental groups were adminstered orally 5, 10, 15, 30, 60 and 120mg of cadmium per kg of body weight for 56 days. All dogs were examined for clinical signs, and weekly changes in hematological and blood chemical values. All dogs were necropsied on 57th days of experiment. Tissue samples including hair, skin, muscle, lung, liver, kidney, spleen, pancreas, testis, ovary, uterus, and bone were collected and analyzed for cadmium, zinc, iron and copper contents using atomic absorption spectrophotometer. From these experiments following results were obtained : 1. All experimental dogs showed vomitting, salivation, anorexia, decreased water-intake, dehydration, and marked weight loss. The dogs received 30mg/kg or more of cadmium died during the period from 2nd to 7th week after administration. 2. Hematologically, all experimental dogs showed decrease in erythrocyte count, hemoglobin concentration, and packed cell volume. The anemia was identified as normocytic and regenerative morphologically. 3. No significant differences in serum glutamic oxaloacetic transminase, glutamic pyruvic transaminase, blood urea nitrogen, and cholosterol value were obseved between the control and experimental dogs. 4. The cadmium contents in various tissues of experimental dogs were estimated as $37.8{\sim}201.8{\mu}g/g$ in bone, $14.1{\sim}49.5{\mu}g/g$ in liver, $13.2{\sim}53.1{\mu}g/g$ in kidney, $0.4{\sim}35.2{\mu}g/g$ in pancreas, $0.8{\sim}35.4{\mu}g/g$ in spleen, $0.9{\sim}30.1{\mu}g/g$ in hair, $0{\sim}7.1{\mu}g/g$ in lung, $0{\sim}5.1{\mu}g/g$ in skin, and $0{\sim}3.6{\mu}g/g$ in muscle, respectively. However, the serum, testis, ovary and uterus showed no cadmium accumulation. Two contol dogs showed cadmium accumulation only in bone. 5. Significant differances in zinc, iron, and copper contents in tissue samples were observed between the control and experimental groups.

  • Extra-hypothalamic growth hormone-releasing hormone (GHRH) plays an important role in reproduction. To study the treatment effect of Grin (a novel hGHRH homodimer), the infertility models of 85 male Chinese hamsters were established by intraperitoneally injecting 20 mg/kg of cyclophosphamide once in a week for 5 weeks and the treatment with Grin or human menopausal gonadotropin (hMG) as positive control was evaluated by performing a 3-week mating experiment. 2-8 mg/kg of Grin and 200 U/kg of hMG showed similar effect and different pathological characteristics. Compared to the single cyclophosphamide group (0%), the pregnancy rates (H-, M-, L-Grin 26.7, 30.8, 31.3%, and hMG 31.3%) showed significant difference, but there was no difference between the hMG and Grin groups. The single cyclophosphamide group presented loose tubules with pathologic vacuoles and significant TUNEL positive cells. Grin induced less weight of body or testis, compactly aligned tubules with little intra-lumens, whereas hMG caused more weight of body or testis, enlarging tubules with annular clearance. Grin presented a dose-dependent manner or cell differentiation-dependentincrease in testicular GHRH receptor, and did not impact the levels of blood and testicular GH, testosterone. Grin promotes fertility by proliferating and differentiating primitive cells through up-regulating testicular GHRH receptor without triggering GH secretion, which might solve the etiology of oligoasthenozoospermia.
