산화물 표면의 U(VI) 흡착에 미치는 살리실산과 피콜린산의 영향 (Effect of Salicylic and Picolinic Acids Acids on the Adsorption of U(VI) onto Oxides)

  • 세 종류의 산화물($TiO_2$(아나타제), $SiO_2$(비결정성) 및 $Al_2O_3$(비결정성)) 표면에 U(VI)이 흡착될 때 유기산이 미치는 영향을 연구하였다. 유기산으로는 살리실산과 피콜린산을 사용하였다. 유기산의 존재 여부에 따라 달라지는 U(VI)의 흡착률 변화를 pH 함수로 측정하였다. 또한 U(VI)의 존재 여부에 따라 달라지는 유기산의 흡착량을 pH 함수로 측정하였다. $TiO_2$의 경우, 살리실산과 피콜린산이 U(VI)과 수용성 착물을 형성함으로써 U(VI)의 흡착률을 저하시킨다. $SiO_2$의 경우, 살리실산은 U(VI) 흡착에 영향을 주지 않지만, 피콜린산은 오히려 U(VI) 흡착을 증가시킨다. 이 현상을 삼성분 표면착물(ternary surface complex) 생성으로 해석하였으며 U(VI) 흡착에 의존하는 피콜린산의 흡착량 변화, 그리고 흡착된 U(VI)의 형광 특성 변화로 이를 확인하였다. $Al_2O_3$의 경우, 살리실산과 피콜린산 모두 U(VI) 흡착과 무관하게 높은 흡착량을 보였으나 U(VI) 흡착을 감소시키지는 않았다. 따라서 삼성분 표면착물 생성을 배제할 수 없으나 이를 확인하기 위해서는 분광 분석과 같은 추가 연구가 필요하다.

R3V6 Amphiphilic Peptide with High Mobility Group Box 1A Domain as an Efficient Carrier for Gene Delivery

  • The R3V6 peptide includes a hydrophilic arginine stretch and a hydrophobic valine stretch. In previous studies, the R3V6 peptide was evaluated as a gene carrier and was found to have low cytotoxicity. However, the transfection efficiency of R3V6 was lower than that of poly-L-lysine (PLL) in N2A neuroblastoma cells. In this study, the transfection efficiency of R3V6 was improved in combination with high mobility group box 1A domain (HMGA). HMGA is originated from the nuclear protein and has many positively-charged amino acids. Therefore, HMGA binds to DNA via charge interaction. In addition, HMGA has a nuclear localization signal peptide and may increase the delivery efficiency of DNA into the nucleus. The ternary complex with HMGA, R3V6, and DNA was prepared and evaluated as a gene carrier. First, the HMGA/DNA complex was prepared with a negative surface charge. Then, R3V6 was added to the complex to coat the negative charges of the HMGA/DNA complex, forming the ternary complex of HMGA, R3V6, and DNA. A physical characterization study showed that the ternary complex was more stable than the PLL/DNA complex. The HMGA/R3V6/DNA complex had a higher transfection efficiency than the PLL/DNA, HMGA/DNA, or R3V6/DNA complexes in N2A cells. Furthermore, the HMGA/R3V6/DNA complex was not toxic to cells. Therefore, the HMGA/R3V6/DNA complex may be a useful gene delivery carrier.

Label-free and sensitive detection of purine catabolites in complex solutions by surface-enhanced raman spectroscopy

  • Purine catabolite screening enables reliable diagnosis of certain diseases. In this regard, the development of a facile detection strategy with high sensitivity and selectivity is demanded for point-of-care applications. In this work, the simultaneous detection of uric acid (UA), xanthine (XA), and hypoxanthine (HX) was carried out as model purine catabolites by surface-enhanced Raman Spectroscopy (SERS). The detection assay was conducted by employing high-aspect ratio Au nanopillar substrates coupled with in-situ Au electrodeposition on the substrates. The additional modification of the Au nanopillar substrates via electrodeposition was found to be an effective method to encapsulate molecules in solution into nanogaps of growing Au films that increase metal-molecule contact and improve substrate's sensitivity and selectivity. In complex solutions, the approach facilitated ternary identification of UA, XA, and HX down to concentration limits of 4.33 𝜇M, 0.71 𝜇M, and 0.22 𝜇M, respectively, which are comparable to their existing levels in normal human physiology. These results demonstrate that the proposed platform is reliable for practical point-of-care analysis of biofluids where solution matrix effects greatly reduce selectivity and sensitivity for rapid on-site disease diagnosis.

뵈마이트 표면의 인산염 및 Eu(III) 수착: 인(P) X-선 흡수분석(EXAFS)에 의한 연구 (Phosphate Sorption on Boehmite with Eu(III): P K-edge EXAFS Fingerprinting)

  • 액티나이드(actinide)의 지질물질로의 수착은 핵에너지와 핵무기 개발로 인하여 인위적으로 자연환경에 노출된 핵종 원소의 이동성과 생물이용가능성을 낮추어 줄 수 있을 것으로 기대된다. 3가의 액티나이드 이온은 3가의 란타나이드(lanthanide) 이온과 유사한 화학적 성질을 띠므로, 3가 액티나이드의 산화광물 표면수착량은, 중성 또는 약한 산성의 pH 상태에서 3가의 란타나이드처럼, 인산염 이온(${PO_4}^{3-}$) 이 수착된 상태에서 크게 증가될 것으로 사료된다. 본 연구에서는 3가액티나이드 이온의 화학적 동족체인 3가 유로피움 이온($Eu^{3+}$)이 인산염이 수착된 상태의 뵈마이트 (${\gamma}$-AlOOH; boehmite) 표면에 수착되는 삼성분 수착계를 X선 흡수분석(EXAFS)을 통하여 관찰하였다. Eu X-선 흡수분석은 Eu-$PO_4$-뵈마이트 삼성분 수착계에서 뵈마이트 표면에 $EuPO_4$ 표면침전물이 형성되는 것을 지시하여 준다. 인산염이 뵈마이트 표면에서 $EuPO_4$ 표면침전물을 형성할 뿐 아니라 두자리 단핵 표면착물을 형성한 것을 P X-선 흡수분석을 통하여 확인하였다.

합성 Goethite에 의한 인산이온, 황산이온 및 구리이온의 혼합용액에서의 흡착특성 (Adsorption Characteristics by Synthesized Goethite in the Mixed Solution Systems of Phosphate, Sulfate, and Copper Ions)

  • Adsorption on goethite of individual component from a solution containing phosphate, sulfate, or copper ion was investigated. Competitive adsorption in the binary and ternary solution systems composed of phosphate, sulfate, and copper ions was also investigated. In competitive adsorption systems with phosphate and sulfate ions, the presence of phosphate ion reduced the adsorption of sulfate ion largely. On the other hand, the presence of sulfate ion caused only a small decrease in phosphate adsorption. This result suggests that phosphate ion is a stronger competitor for adsorption on goethite than sulfate ion, which is consistent with the higher affinity of phosphate for the surface compared to sulfate ion. Compared to the results from single-sorbate systems, adsorption of copper ion in the binary system of sulfate ion and copper ion was found to be enhanced in the presence of sulfate ion. Addition of sulfate ion to the binary system of copper ion and phosphate ion resulted in a small enhancement in copper sorption. This result implies that the presence of sulfate ion promotes adsorption of the ternary complex FeOHCuSO$_4$. The adsorption isotherms could be well described by the Langmuir adsorption equation.

An Experimental Study on the Sorption of U(VI) onto Granite

  • The sorption of U(Vl) on a domestic granite is studied as a function of experimental conditions such as contact time, solution-solid ratio, ionic strength, and pH using a batch procedure. The distribution coefficients, $K_{d}$'s, of U(VI) are about 1-100mL/g depending on the experimental conditions. The sorption of U(VI) onto granite particles is greatly dependent upon the contact time, solution-solid ratio, and pH, but very little is dependent on the ionic strength. It is noticed that an U(VI)-carbonate ternary surface complex can be formed in the neutral range of pH. In the alkaline range of pH above 7, U(VI) sorption onto granite particles is greatly decreased due to the formation of anionic U(VI)-carbonate aqueous complexes.s.

Dynamic lipopolysaccharide transfer cascade to TLR4/MD2 complex via LBP and CD14

  • Toll-like receptor 4 (TLR4) together with MD2, one of the key pattern recognition receptors for a pathogen-associated molecular pattern, activates innate immunity by recognizing lipopolysaccharide (LPS) of Gram-negative bacteria. Although LBP and CD14 catalyze LPS transfer to the TLR4/MD2 complex, the detail mechanisms underlying this dynamic LPS transfer remain elusive. Using negative-stain electron microscopy, we visualized the dynamic intermediate complexes during LPS transfer-LBP/LPS micelles and ternary CD14/LBP/LPS micelle complexes. We also reconstituted the entire cascade of LPS transfer to TLR4/MD2 in a total internal reflection fluorescence (TIRF) microscope for a single molecule fluorescence analysis. These analyses reveal longitudinal LBP binding to the surface of LPS micelles and multi-round binding/unbinding of CD14 to single LBP/LPS micelles via key charged residues on LBP and CD14. Finally, we reveal that a single LPS molecule bound to CD14 is transferred to TLR4/MD2 in a TLR4-dependent manner. These discoveries, which clarify the molecular mechanism of dynamic LPS transfer to TLR4/MD2 via LBP and CD14, provide novel insights into the initiation of innate immune responses.

NMR Structure of Syndecan-4L reveals structural requirement for PKC signalling

  • Syndecans, transmembrane heparan sulfate proteoglycans, are coreceptors with integrin in cell adhesion process. It forms a ternary signaling complex with protein kinase C and phosphatidylinositol 4,5 bisphosphate (PIP2) for integrin signaling. NMR data indicates that cytoplasmic domain of syndecan-4 (4L) undergoes a conformational transition in the presence of PIP2, forming oligomeric conformation. The structure based on NMR data demonstrated that syndecan-4L itself forms a compact intertwined symmetric dimer with an unusual clamp shape for residues Leu$^{186}$ -Ala$^{195}$ . The molecular surface of the syndecan-4L dimer is highly positively charged. In addition, no inter-subunit NOEs in membrane proximal amino acid resides (Cl region) has been observed, demonstrating that the Cl region is mostly unstructured in syndecan-4L dimmer. However, the complex structure in the presence of PIP2 induced a high order multimeric conformation in solution. In addition, phosphorylation of cytoplasmic domain induces conformational change of syndecan-4, resulting inhibition of PKC signaling. The NMR structural data strongly suggest that PIP2 promotes oligomerization of syndecan-4 cytoplasmic domain for PKC activation and further induces structural reorganization of syndecan for mediating signaling network in cell adhesion procedure.

비소의 적철석 표면 흡착에 토양유기물이 미치는 영향: 화학종 모델링과 흡착 기작 (Effect of Soil Organic Matter on Arsenic Adsorption in the Hematite-Water Interface: Chemical Speciation Modeling and Adsorption Mechanism)

  • 본 연구는 이성분계의 화학종 모델링과 삼성분계의 흡착 모델링으로부터 As(III)와 As(V)의 적철석 표면 흡착에 휴 믹산의 영향과 그 결합기작을 고찰하였다. 비소와 휴믹산의 유기 결합의 모델링은 음이온 사이의 정전기적인 반발력 과 비소의 유기 결합을 위한 결합금속의 영향을 고려한 결합 모델이 적합하였다. 삼성분계의 흡착 실험 자료와 비교 할 때 이성분계의 고유상수를 사용한 음이온 경쟁 모델이 음이온 경쟁에 따른 비소의 흡착량과 일치하였다. 반면, 비 소의 유기 결합량의 감소와 휴믹산과의 음이온 흡착경쟁이 흡착량을 감소시키기 때문에 단순합모델은 양이온 중금속 과는 반대로 모델의 편차를 보였다. 반응 기작의 관점에서 휴믹산의 공존하에 비소 유기결합 화학종과 중성화학종의 As(III) 및 음이온의 As(V)가 속착물을 형성하며, 경쟁적으로 적철석 표면으로 이동하여 흡착하게 된다.
