This study examined the effects of environmental conditions and the presence of refractory organic matter on oxidation rates of total organic carbon (TOC) measurements based on high temperature combustion and ultraviolet-sulfate methods. Spectroscopic indices for prediction of oxidation rates were also explored using the UV spectra and fluorescence excitation-emission matrix (EEM) of humic acids. Furthermore, optimum TOC instrument conditions were suggested by comparing oxidation rates of a standard TOC material under various conditions. Environmental conditions included salts, reduced ions, and suspended solids. Salts had the greatest influence on oxidation rates in the UV-sulfate method. However, no effect was detected in the high temperature combustion method. The UV-sulfate method showed lower humic substance oxidation rates, refractory natural organic matter, compared to the other methods. TOC oxidation rates for the UV-sulfate method were negatively correlated with higher specific-UV absorbance, humification index, and humic-like EEM peak intensities, suggesting that these spectroscopic indices could be used to predict TOC oxidation rates. TOC signals from instruments using the UV-sulfate method increased with increasing chamber temperature and increasing UV exposure durations. Signals were more sensitive to the former condition, suggesting that chamber temperature is important for improving the TOC oxidation rates of refractory organic matter.

The occurrence of exotic weeds and their influx into farmlands due to climate change poses many problems. Therefore, it is necessary to generate a prediction model for the occurrence pattern of these exotic weeds based on scientific evidence and devise prevention measures. The photosynthetic apparatus is known as the most temperature-sensitive component of a plant cell and its initial response to temperature stress is to inhibit the activation of photosystem II. This study investigated the potential of OJIP transients in assessing temperature stress in exotic weeds. The four exotic weeds currently flowing into Korean farmlands include Amaranthus spinosus, Conyza bonariensis, Crassocephalum crepidioides, and Amaranthus viridis. These weeds were treated at 5℃, 10℃, 15℃, 20℃, 25℃, 30℃, 35℃, and 40℃ and the OJIP curves and JIP parameters were measured and analyzed. The results showed that heat and chilling stress affected the photosystem II(PSII) electron transport of A. spinosus, whereas C. crepidioides and A. viridis were more affected by high-temperature stress than by low-temperature stress. Lastly, C. bonariensis showed resistance to both high and low-temperature stress. The results of this study suggest that OJIP transients and JIP parameters can be used to analyze damage to the photosynthetic apparatus by temperature stress and that they can serve as sensitive indicators for the occurrence pattern of exotic weeds.
In the present wort the Laser-induced Fluorescence (LIF) technique and Confocal Laser Scanning Microscopy (CLSM) have been combined to measure the temperature distribution across a micro-scale liquid layer as a direct and non-invasive method. Only the fluorescent light emitted from a very thin volume around a focal plane can be selectively detected, and it enables us to measure the liquid temperatures even at the close vicinity of the walls. As an experimental verification, a test section consists of two flat plates (for heating and cooling, respectively) separated by about 240 microns was made, and the methanol mixed with a temperature-sensitive dye, Rhodamine B, was filled in the gap between them. The measured temperature distribution across the gap showed good linearity, which is a typical characteristic of conduction heat transfer through a thin liquid layer. In result, the CLSM-LIF technique proposed in the present study was found to be a promising method to measure the local temperatures in the liquid flow field in microfluidic devices.
A simple, selective and sensitive fluorescence quenching method was developed to the determination Cr(VI). The method is based on the oxidation of $I^-\;to\;{{I_3}^-}$ by Cr(VI) in sulfuric acid solution followed by immediate formation of ion association compound between I3 − and rhodamine 6G in Tween-80 micellar media at room temperature. The influence of several surfactants on rhodamine 6G fluorescence signal was studied; particular attention was paid in the aggregation behavior of rhodamine 6G–Tween-80 system. The experimental parameters (e.g., type of surfactant, reagent concentration) were studied and the optimal conditions were established. The linear calibration graph was obtained in the range 2.0 - 100.0 ng m$L^{-1}$ Cr(VI). The detection limit of the method was 0.37 ng m$L^{-1}$. The relative standard deviation (R.S.D.) is less than 5% (n = 5). The efficiency of the method for the determination of Cr(VI) in the presence of Cr(III) in the sample was investigated. The method was applied successfully to the determination of Cr(VI) and total Cr in water, and liver tissue samples.
5-methylthioninhydrin (5-MTN) is an amino acid sensitive reagent used for the development of latent fingermarks deposited on porous surfaces such as paper and wood. The present study demonstrates that the 5-MTN can be used as a latent footwear impression enhancement reagent, by reacting with trace multivalent metal ions, which are the main components of the latent footwear impression. 5-MTN and L-alanine complex (MTN-ALA) used for the latent footwear impression development was prepared, by mixing $4.5{\times}10^{-3}M$ 5-MTN (in methanol) and $4.5{\times}10^{-3}M$ L-alanine (in methanol) in 1:1 ratio, and keeping undisturbed at room temperature for 24 h. The latent footwear impressions were deposited on white and black non-porous surfaces (glass plate, polyethylene panel, polypropylene panel, acryl panel, polyvinyl chloride (PVC) panel, poly(methyl methacrylate) (PMMA) panel, acrylonitrile-butadiene-styrene (ABS) panel, tile), and a semi-porous surfaces (painted wood). The latent footwear impressions on these surfaces were treated with MTN-ALA complex by spraying. The fluorescence of footwear impressions (occurred due to the reaction between MTN-ALA and metal complexes) was observed under a 505 nm forensic light source and an orange barrier filter. The enhancement of latent footwear impression was achieved from black surfaces without any blurring. However, the fluorescence (enhancement) of footwear impression was not observed on the white PVC, PMMA, and ABS surfaces, because the incident light interfered and reflected on the surface. The sensitivity of MTN-ALA was superior to 2,2'-dipyridil, which is a representative non-fluorescing footwear impression enhancement reagent, and similar to 8-hydroxyquinoline, which is a representative fluorescing footwear impression enhancement reagent.
Plants possess the ability to dissipate the excitation energy for the protection of photosynthetic apparatus from absorbed excess light. Heat dissipation is regulated by xanthophyll cycle in thylakoid membranes of chloroplasts. We investigated the mechanistic aspects of xanthophyll cycle-dependent photoprotection against low-temperature photoinhibition in plants. Using barley and rice as chilling-resistant species and sensitive ones, respectively, chilling-induced chlorophyll fluorescence quenching, composition of xanthophyll cycle pigments and mRNA expression of the zeaxanthin epoxidase were examined. Chilled barley plants exhibited little changes in chlorophyll fluorescence quenching either of photochemical or non-photochemical nature and in the photosynthetic electron transport, indicating low reduction state of PS II primary electron acceptor. In contrast to the barley, chilled rice showed a marked decline in those parameters mentioned above, indicating the increased reduction state of PS II primary electron acceptor. In addition, barley plants were shown to have a higher capacity to elevate the pool size of xanthophyll cycle pigments in response to cold stress compared to rice plants. Such species-dependent regulation of xanthophyll cycle activity was correlated with the gene expression level of cold-induced zeaxanthin epoxidase. Chilled rice plants depressed the gene expression of zeaxanthin epoxidase, whereas barley increased its expression in response to cold stress. We suggest that chilling-induced alterations in the pool size of xanthophyll cycle pigments related to its capacity would play an important role in regulating plant's sensitivity to chilling stress.
The absorption and fluorescence spectral properties of piroxicam (PRX) in the hydrogenbonding solvents show the most sensitive dependence on the concentration ranging from 8 x 10$^{_5}$ to 2 x10$^{_5}$ M. These are attributed to both the solvent-mediated ground-state intermolecular proton transfer (GSIerPT) leading to formation of the ground state anion and the excited-state intmmolecular proton transfer (ESIraPT). The concentration dependences of the time-resolved emission kinetics at both room temperature and 77 K have also been investigated. It is shown that in the excited state, the ESIraPT of PRX is the dominant process to form a keto tautomer at the high concentration, whereas at the low concentration the excited-state conformational change of the anion is an additional process leading to formation of a zwitterion. The ESI~PT of PRX in the hydrogenbonding solvent is coupled with the ultrafast excited-state solvent reorganization.

Purpose: Poloxamer 407 (P407) thermo-sensitive hydrogel formulations were developed to enhance the retention time in the urinary bladder after intravesical instillation. Materials and Methods: P407 hydrogels (P407Gels) containing 0.2 w/w% fluorescein isothiocyanate dextran (FD, MW 4 kDa) as a fluorescent probe were prepared by the cold method with different concentrations of the polymer (20, 25, and 30 w/w%). The gel-forming capacities were characterized in terms of gelation temperature (G-Temp), gelation time (G-Time), and gel duration (G-Dur). Homogenous dispersion of the probe throughout the hydrogel was observed by using fluorescence microscopy. The in vitro bladder simulation model was established to evaluate the retention and drug release properties. P407Gels in the solution state were administered to nude mice via urinary instillation, and the in vivo retention behavior of P407Gels was visualized by using an in vivo imaging system (IVIS). Results: P407Gels showed a thermo-reversible phase transition at $4^{\circ}C$ (refrigerated; sol) and $37^{\circ}C$ (body temperature; gel). The G-Temp, G-Time, and G-Dur of FD-free P407Gels were approximately $10^{\circ}C-20^{\circ}C$, 12-30 seconds, and 12-35 hours, respectively, and were not altered by the addition of FD. Fluorescence imaging showed that FD was spread homogenously in the gelled P407 solution. In a bladder simulation model, even after repeated periodic filling-emptying cycles, the hydrogel formulation displayed excellent retention with continuous release of the probe over 8 hours. The FD release from P407Gels and the erosion of the gel, both of which followed zero-order kinetics, had a linear relationship ($r^2=0.988$). IVIS demonstrated that the intravesical retention time of P407Gels was over 4 hours, which was longer than that of the FD solution (<1 hour), even though periodic urination occurred in the mice. Conclusions: FD release from P407Gels was erosion-controlled. P407Gels represent a promising system to enhance intravesical retention with extended drug delivery.

The aim of this study was to investigate the sensitivity to environmental stress, and changes in the photosynthesis capacity in Abies koreana seedlings by age and to suggest the most effective age for restoration. To identify these physiological characteristics of A. koreana, the chlorophyll fluorescence and photosynthetic capacity of 1-, 2-, 3-, 5- and 6-year-old A. koreana seedlings were observed from June 2020 to June 2021. The maximum quantum efficiency of Photosystem II (Fv/Fm), a chlorophyll fluorescence measurement parameter, was strongly positively correlated with the monthly average temperature (1-year-old seedling: r=0.8779, 2-year-old seedling: r=0.8605, 3-year-old seedling: r=0.8697, 5-year-old seedlings: r=0.8085, and 6-year-old seedlings: r=0.8316). The Fv/Fm values were the lowest in winter (November 2020-March 2021). In addition, the Fv/Fm values of 1-, 2-, and 3-year-old seedlings in winter were lower than that of 5- and 6-year-old seedlings, while the Fv/Fm values in summer were relatively higher than those in winter. Further, the Fv/Fm values of seedlings of all ages decreased in August 2020, when the monthly average temperature was the highest. In particular, 1-year-old to 3-year-old seedlings showed Fv/Fm values less than 0.8. Further, the photosynthetic capacity measured in August 2020 increased with increasing seedling age. The analysis of variance results for summer Fv/Fm values showed significant differences in age-specific averages (p<0.05), and Duncan's multiple range test showed significant differences between 5- and 6-year-old seedlings and 1-, 2-, and 3-year-old seedlings (p<0.05). These results suggested that the 5- and 6-year-old seedlings were less sensitive to environmental stress and showed better photosynthetic capacity than the 1-, 2-, and 3-year-old seedlings. Therefore, 5-year-old or older A. koreana seedlings can be used as restoration materials because they can show increased adaptability and stable growth during transplantation due to their relatively high environmental resistance and photosynthetic capacity.
Light-chilling effects were investigated in chilling-sensitive cucumber (Cucumis sativus L. cv. Ilmichungjang) and chilling-resistant pea (Pisum sativum L. cv. Giant) leaf discs in relation to possible damage in D1 protein. In both plants, dark-chilling did not cause any noticeable changes in (Fv)m/Fm and lincomycin did not affect the decrease in (Fv)m/Fm caused by light-chilling. This result suggests that the de novo synthesis of D1 protein did not occur actively during light-chilling. In pea light-chilled for 6 h. the decreased (Fv)m/Fm was partly recovered in the dark, and almost complete recovery was observed in the light. In cucumber light-chilled for 3 h. the reduced (Fv)m/Fm decreased further for the initial 2 h recovery process in the light regardless of the treatment of lincomycin and recovered very slowly. In both plant species, the treatment of lincomycin inhibited the recovery process in the light, but did not significantly inhibit the process in the dark. In cucumber leaves pulse-labeled with $[^{35}S]Met$, the labeled band intensities of isolated pigment-protein complexes were almost the same during the 6 h light-chilling, but significant decreases in band intensities were observed during the 3 h recovery period. This result suggests that the irreversibly damaged D1 protein was degraded during the recovery period. However, no noticeable changes were observed in the pea leaves during the 12 h chilling and 3 h recovery period. The polyacrylamide gel electrophoresis of the pigment-protein complexes showed that the principal lesion sites of light-chilling were different from those of room temperature photoinhibition.
