전해수 생성온도에 따른 그람양성균과 그람음성균의 살균 효과 (Bactericidal Effect of Electrolyzed Activated Water Prepared at Different Water Temperatures on Gram-Positive and Gram-Negative Bacteria)

  • 본 연구에서는 다양한 온도의 물로 제조한 전해수를 이용하여 그람양성균과 그람음성균에 대한 살균력을 확인하였다. 전해수의 물성은 물의 온도가 높아질수록 pH와 유효 잔류염소 농도 값이 높아졌으나, 산화환원전위 값은 감소하였다. 4, 22, $40^{\circ}C$의 물을 이용하여 생성한 전해수에 Escherichia coli O157:H7을 15분 동안 처리하였을 때 각각 1.18, 4.47, 5.46 log CFU/mL 감소하였으며, Staphylococcus aureus는 각각 0.72, 4.90, 5.54 log CFU/mL 감소하였다. 이를 통해 전해수를 생성하는 물의 온도가 증가할수록 살균 효과가 높아지는 것을 알 수 있다. 4, $40^{\circ}C$의 물을 이용하여 생성한 전해수에 균주를 처리한 경우, Listeria monocytogenes의 D 값(일정한 온도에서 90 %의 미생물이 사멸하는 데 걸리는 시간)이 각각 6.60, 1.57분으로 나타나 가장 낮았으며, $22^{\circ}C$ 전해수에서는 Salmonella Enteritidis의 D 값이 2.92분으로 가장 낮은 값을 나타내었다. 그람양성균과 그람음성균을 비교하였을 때 모든 온도에서 D 값에 대한 유의적인 차이는 나타나지 않았다(P>0.05). 본 연구 결과는 전해수를 제조할 경우 높은 온도의 물을 이용하여 제조하는 것이 미생물 살균 효과가 높다는 것을 보여주고 있다. 따라서 전해수를 생성할 때 물의 온도를 고려하여 높은 살균력을 나타내는 전해수를 생성할 수 있으며, 이는 물의 온도 외에도 물의 경도나 물의 종류 등을 고려하여 최적의 살균 전해수를 제조함으로써 식품산업에 적용하기 위한 자료로 활용될 것으로 기대된다.

수입 원산지별 도토리묵의 저장기간에 따른 겔화 특성 (Gel Properties of Mook Manufactured from Acorns Harvested in Various Countries according to Storage Period)

  • 물결합능력은 북한산이 중국산과 국내산에 비해 높은 것으로 나타났으며 3개 시료간의 원산지별 유의적 차이가 있었다(p<0.05). X-선 회절도의 강도 및 피크의 위치나 모양은 거의 비슷하게 관찰되었으나 원산지별 뚜렷한 차이는 보이지 않았다. 장방출주사전자현미경으로 관찰한 조전분은 중국산과 북한산에서 일부 호화가 관찰되어 원산지별 뚜렷한 차이를 보였으나 겔전분은 원산지별 차이를 보이질 않았다. 겔의 이수율에서는 국내산과 중국산의 경우 10일차 저장기간 중 2, 3일차에서 북한산은 3, 4일차에서 가장 높은 이수율을 보였다. 3개 시료 간 저장 1, 2, 3일차에서는 유의적 차이를 나타내었으나(p<0.05), 4일차부터 10일차까지는 유의적 차이가 없었다. 시차주사열량계에 의한 호화특성피크에서는 저장 1일차 시료에서 국내산이 $55.28^{\circ}C$로 중국산의 $54.45^{\circ}C$와 북한산의 $54.12^{\circ}C$보다 높았다. 저장일수가 지날수록 점차 감소하였으며 호화개시온도 및 최대호화온도에서 비슷한 경향을 보였다. 호화엔탈피값(${\Delta}H$)은 시료의 저장일수에 따라 증가하였으며, 국내산 북한산 중국산으로 높았다. 물성의 특성치를 나타내는 조직감의 경도에서는 북한산이 국내산, 중국산보다 높은 값을 보였다. 저장일수별 1, 3, 5, 7일차에서 3개 시료간의 원산지별 유의적 차이가 있었다(p<0.05). 도토리묵의 외관적 품질을 결정하는 표면의 색도 변화는 L값, a값에서 북한산, 중국산, 국내산으로 높았으며, 저장일수 1, 3, 5, 7, 9일차에서 시료간의 유의적 차이를 나타내었다(p<0.05). b값은 국내산이 가장 높았으며 그 다음 북한산, 중국산으로 높았다. b값에서도 시료간의 유의적 차이가 있었다(p<0.05). 관능검사 전체항목에서는 3개 시료간의 유의성은 없는 것으로 나타났다.

Direct Effect of a Hot Environment on Ruminal Motility in Sheep

  • The aim of this research was to clarify the direct effects of a hot environment on ruminal motility in sheep fed twice a day. In the first experiment, in order to equalize variable factors excluding the ambient temperature between the thermoneutral environment ($23^{\circ}C$, relative humidity 80%) and the hot environment ($32^{\circ}C$, relative humidity 80%), sheep were fed equal amounts of the same quality feed twice a day. The sheep were allowed free access to water for the duration of the two one-hour feeding periods (10:00 am-11:00 am, 5:00 pm-6:00 pm). On the fourth day after exposure to the hot environment, the frequency and strength of ruminal contractions were continuously recorded between 9:30 am and 11:00 pm. Prior to the exposure to a hot environment the frequency and strength of ruminal contractions were recorded in a thermoneutral environment during the period 9:30 am-11:00 pm. In the second experiment, in order to maintain the stomach content of the sheep at equal levels in both environments, the sheep were fed equal amounts of the same quality feed twice a day. Following the completion of the two one-hour feeding periods, a fixed amount of warm water was infused into the rumen. Rumen motility was then recorded during the same period as for the first experiment (9:30 am-11:00 pm). In the first experiment, when the frequency of ruminal contractions prior to (24, 24 frequency/15 min), during (48, 47 frequency/min) and after (22, 19 frequency/min) both the morning and afternoon feeding in a hot environment was compared with the values from the thermoneutral environment (20, 22; 50, 50; 21, 20 frequency/min), there was found to be no difference. However, the strength of ruminal contractions after morning and afternoon feeding (3.7, 3.1 mm Hg) in the hot environment decreased significantly in comparison with the thermoneutral environment (4.3, 3.8 mm Hg). In the second experiment, the frequency of ruminal contractions in the hot environment was not significantly different from that in the thermoneutral environment. The strength of ruminal contractions after ruminal infusion of warm water in the hot environment (morning: 4.6, afternoon: 4.5 mm Hg) was significantly lower than that in the thermoneutral environment (morning: 5.6, afternoon: 5.0 mm Hg). The results suggest that a hot environment acts directly on the strength of ruminal contractions in sheep fed twice a day rather than on the frequency.

Effects of Rumen Undegradable Protein and Minerals Proteinate on Early Lactation Performance and Ovarian Functions of Dairy Cows in the Tropics

  • A 90 d study was designed to investigate the effects of rumen undegradable protein (RUP) and a mixture of Cu, Zn and Mn proteinate (CZMP) on milk yield and composition and ovarian functions during rainy months. Twenty four Holstein${\times}$ indigenous cows in their 2nd and 3rd lactation were randomly allocated to total mixed rations (TMR) containing soy bean meal (SBM) as a source of rumen degradable protein (RDP), SBM plus CZMP, and formalin treated SBM (FSBM) as a source of RUP. Maximum and minimum temperature humidity index during the experimental period were 83.6-84.7 and 75.4-76.1. There were no differences (p>0.05) in intakes of dry matter, crude protein and net energy and in contents of butterfat, lactose and minerals. Cows on TMR containing FSBM not only lost less weight (-278, -467 and -433 g/d) with more intake of RUP (0.92, 0.58 and 0.59 kg/d) but also produced more milk (19.27, 18.23 and 18.13 kg/d) and 4% fat corrected milk (18.57, 17.57 and 17.51 kg/d) with more protein (3.06, 2.81 and 2.80%), solids-not-fat (8.69, 8.38 and 8.38%) and less milk urea N (9.3, 15.4 and 15.0 mg/dl) compared with those on TMR containing SBM and SBM+CZMP, respectively (p<0.01). However, cows on TMR containing SBM and SBM+CZMP did not differ in these respects (p>0.05). Whereas incidence of cystic ovaries at 20 and 90 d pospartum was less (p<0.01) in cows on TMR containing SBM+CZMP (37.3 and 12.5%) than those on TMR containing SBM (62.5 and 25%), it was nil for cows on TMR containing FSBM. Cows in all three group differed (p<0.01) from each other for the recurrence of first observed estrus with those on TMR containing FSBM having least days (22, 36 and 47 d) compared with their counterpart on TMR containing SBM+CZMP and SBM, repectively. The results suggest that RUP is one of the limiting factors affecting milk yield and its composition and ovarian functions during early lactation of dairy cows in the tropics.

Effect of Short-term and Long-term Preservation on Motion Characteristics of Garole Ram Spermatozoa: A Prolific Microsheep Breed of India

  • Garole is a prolific, rare, less known and small size Indian sheep breed found in low and humid Sunderban region of West Bengal. Although information on stored Garole ram liquid semen upto 24 h is available, but there is a need to further investigate the short-term and long-term preservability of Garole ram semen for extensive utilization of this valuable germplasm by artificial insemination. The aim of the present study was to apply computer-assisted sperm analysis technique for assessing the motion characteristics of Garole ram semen stored (i) in liquid state at refrigeration temperature for short-term preservation upto 48 h and (ii) in frozen state at $-196^{\circ}C$ for long-term preservation after packaging in mini straws. Short-term preservation had a significant effect on motility (p<0.01) as the motility progressively decreased from 90.1% at 0 h to 85.5% and 73.2% after 24 and 48 h of storage, respectively. Although the decline in rapid moving sperms was also significant (p<0.01) on storage but the decrease was more pronounced at 48 h as compared to 24 h of storage period. Storage of chilled semen had also a significant effect on % linearity (p<0.05), % straightness (p<0.01), sperm velocities (p<0.01), amplitude of lateral head displacement (p<0.01) and beat frequency (pO.Ol) of spermatozoa. The replication had a significant effect for all the variables except average path and straight line velocity. However, the interactions of short-term storage and replication were non-significant for most of the variables except % of medium moving sperms, sperm velocities and beat frequency. On long-term preservation of Garole ram spermatozoa under controlled conditions the mean post-thaw recovery of 70.4 and 71.4% motile spermatozoa was achieved having 48.8 and 48.9% of rapidly motile spermatozoa, respectively in both the replicates. The effect of replication on cryopreservation was significant (p<0.05) on amplitude of lateral head displacement and beat frequency, but there was no significant effect on motility, rapidly motile spermatozoa, linearity, straightness and sperm velocities of frozen-thawed spermatozoa. It can be concluded from these results that an average 70% motility can be achieved on storage of Garole ram semen in chilled liquid state upto 48 h or in liquid nitrogen after freezing under controlled conditions in straws. However, further studies are required to evaluate the fertility of short-term and long-term preserved Garole ram semen for extensive use of this prolific sheep breed.

Effect of Transportation at High Ambient Temperatures on Physiological Responses, Carcass and Meat Quality Characteristics in Two Age Groups of Omani Sheep

  • The aim of this study was to determine the effects of short road transportation in an open truck during hot season on live weight shrink, physiological responses, and carcass and meat quality of Omani sheep at 6 and 12 months of age. Thirty-six male sheep, 18 of each age group, were used. Age groups were assigned randomly to transported and not-transported groups. The transported group was transported to the slaughterhouse the day of slaughter in an open truck covering a distance of approximately 100 km. The average temperature during transportation was $37^{\circ}C$. The not-transported group was kept in a lairage of a commercial slaughterhouse with ad libitum feed and water for 48 h prior to slaughter. Blood samples were collected from sheep before loading and prior to slaughter via jugular venipuncture to assess their physiological response to transport in relation to hormonal levels. Animals were weighed just before loading onto a truck and after transport to assess shrinkage. Muscle ultimate pH, expressed juice, cooking loss percentage, WB-shear force value, sarcomere length and colour L*, a*, b* were measured on samples from longissimus dorsi, biceps femoris and semitendinosus muscles collected at 24 h postmortem at $1-3^{\circ}C$. Live weight shrinkage losses were 1.09 and 1.52 kg for 6 and 12 month transported sheep, respectively. The transported sheep had significantly (p<0.05) higher cortisol, adrenaline, noradrenaline, and dopamine concentration levels prior to slaughter at both ages than the not-transported sheep. Transportation significantly influenced meat quality characteristics of three muscles. Muscle ultimate pH and shear force values were significantly higher, while CIE L*, a*, b*, expressed juice and cooking loss were lower in transported than not-transported sheep. Age had a significant effect on meat quality characteristics of Omani sheep. These results indicated that short-term pre-slaughter transport at high ambient temperatures can cause noticeable changes in physiological and muscle metabolism responses in sheep.

Recycling of Fermented Sawdust-based Oyster Mushroom Spent Substrate as a Feed Supplement for Postweaning Calves

  • The objective of this study was to find the way to prolong the storage time of sawdust-based oyster mushroom (Pleurotus osteratus) spent substrate (OMSS) by fermenting with potential probiotic microorganisms to recycle the otherwise waste of mushroom farms. To this purpose, lactic acid bacteria (LAB) were screened to select the best lactic acid-producing strains. Three strains of LAB (Lactobacillus plantarum Lp1', Pediococcus acidilacticii Pa193, L. plantarum Lp2M) were selected and in mixture they lowered the pH of the fermented OMSS to 3.81. fOMSS (fermented sawdust-based oyster mushroom spent substrate) could be stored at room temperature for at least 17 days without any deterioration of feed quality based on the pH, smell, and color. In dry matter disappearance rate in situ, commercial TMR (total mixed ration), OMSS and OMMM (oyster mushroom mycelium mass) showed no significant differences between the samples after 6, 12 and 24 h incubation except for 48 h. Two separate field studies were performed to test the effects of fOMSS supplement on the growth performance of postweaning Holstein calves. Field trials included groups of animals feeding calf starter supplemented with: Control (no supplement), AB (colistin 0.08% and oxyneo 110/110 0.1%), fOMSS (10% fOMSS) and fConc (10% fermented concentrate) and DFM (direct-fed microbials, average $10^9$ cfu for each of three LAB/d/head). Growth performance (average daily gain and feed efficiency) of the fOMSS supplement group was higher than that of AB followed by fConc and DFM even though there was no statistically significant difference. The Control group was lower than any other group. Various hematological values including IgG, IgA, RBC (red blood cell), hemoglobin, and hematocrit were measured every 10 days to check any unusual abnormality for all groups in trial I and II, and they were within a normal and safe range. Our results suggest that sawdust-based OMSS could be recycled after fermentation with three probiotic LAB strains as a feed supplement for post-weaning calves, and fOMSS has the beneficial effects of an alternative to antibiotics for a growth enhancer in dairy calves.

Molecular cloning, purification, expression, and characterization of β-1, 4-endoglucanase gene (Cel5A) from Eubacterium cellulosolvens sp. isolated from Holstein steers' rumen

  • Objective: This study was conducted to isolate the cellulolytic microorganism from the rumen of Holstein steers and characterize endoglucanase gene (Cel5A) from the isolated microorganism. Methods: To isolate anaerobic microbes having endoglucanase, rumen fluid was obtained from Holstein steers fed roughage diet. The isolated anaerobic bacteria had 98% similarity with Eubacterium cellulosolvens (E. cellulosolvens) Ce2 (Accession number: AB163733). The Cel5A from isolated E. cellulolsovens sp. was cloned using the published genome sequence and expressed through the Escherichia coli BL21. Results: The maximum activity of recombinant Cel5A (rCel5A) was observed at $50^{\circ}C$ and pH 4.0. The enzyme was constant at the temperature range of $20^{\circ}C$ to $40^{\circ}C$ but also, at the pH range of 3 to 9. The metal ions including $Ca^{2+}$, $K^+$, $Ni^{2+}$,$Mg^{2+}$, and $Fe^{2+}$ increased the endoglucanase activity but the addition of $Mn^{2+}$, $Cu^{2+}$, and $Zn^{2+}$ decreased. The Km and Vmax value of rCel5A were 14.05 mg/mL and $45.66{\mu}mol/min/mg$. Turnover number, Kcat and catalytic efficiency, Kcat/Km values of rCel5A was $96.69(s^{-1})$ and 6.88 (mL/mg/s), respectively. Conclusion: Our results indicated that rCel5A of E. cellulosolvens isolated from Holstein steers had a broad pH range with high stability under various conditions, which might be one of the beneficial characteristics of this enzyme for possible industrial application.

Evaluation of high nutrient diets on litter performance of heat-stressed lactating sows

  • Objective: The present study investigated the litter performance of multiparous sows fed 3% and 6% densified diets at farrowing to weaning during summer with mean maximum room temperature of $30.5^{\circ}C$. Methods: A total of 60 crossbred multiparous sows were allotted to one of three treatments based on body weight according to a completely randomized design. Three different nutrient levels based on NRC were applied as standard diet (ST; metabolizable energy, 3,300 kcal/kg), high nutrient level 1 (HE1; ST+3% higher energy and 16.59% protein) and high nutrient level 2 (HE2; ST+6% higher energy and 17.04% protein). Results: There was no variation in the body weight change. However, backfat thickness change tended to reduce in HE1 in comparison to ST treatment. Dietary treatments had no effects on feed intake, daily energy intake and weaning-to-estrus interval in lactating sows. Litter size, litter weight at weaning and average daily gain of piglets were significantly greater in sows in HE1 compared with ST, however, no difference was observed between HE2 and ST. Increasing the nutrient levels had no effects on the blood urea nitrogen, glucose, triglyceride, and creatinine at post-farrowing and weaning time. The concentration of follicle stimulating hormone, cortisol and insulin were not affected by dietary treatments either in post-farrowing or weaning time. The concentration of blood luteinizing hormone of sows in ST treatment was numerically less than sows in HE2 treatment at weaning. Milk and colostrum compositions such as protein, fat and lactose were not affected by the treatments. Conclusion: An energy level of 3,400 kcal/kg (14.23 MJ/kg) with 166 g/kg crude protein is suggested as the optimal level of dietary nutrients for heat stressed lactating sows with significant beneficial effects on litter size.

Isolation and characterization of a novel gossypol-degrading bacteria Bacillus subtilis strain Rumen Bacillus Subtilis

  • Objective: The aim of the study was to isolate gossypol-degrading bacteria and to assess its potential for gossypol degradation. Methods: Rumen liquid was collected from fistulated cows grazing the experimental pasture. Approximately 1 mL of the rumen liquid was spread onto basal medium plates containing 2 g/L gossypol as the only source of carbon and was then cultured at $39^{\circ}C$ to isolate gossypol-degrading bacteria. The isolated colonies were cultured for 6 h and then their size and shape observed by microscope and scanning electron microscope. The 16S rRNA gene of isolated colonies was sequenced and aligned using National Center for Biotechnology Information-Basic Local Alignment Search Tool. The various fermentation conditions, initial pH, incubation temperature, inoculum level and fermentationperiod were analyzed in cottonseed meal (CSM). The crude protein (CP), total gossypol (TG), and free gossypol (FG) were determined in CSM after fermentation with isolated strain at $39^{\circ}C$ for 72 h. Results: Screening results showed that a single bacterial isolate, named Rumen Bacillus Subtilis (RBS), could use gossypol as a carbon source. The bacterium was identified by 16S rDNA sequencing as being 98% homologous to the sequence of Bacillus subtilis strain GH38. The optimum fermentation conditions were found to be 72 h, $39^{\circ}C$, pH 6.5, moisture 50%, inoculum level $10^7cell/g$. In the optimum fermentation conditions, the FG and TG content in fermented CSM decreased 78.86% and 49% relative to the control. The content of CP and the essential amino acids of the fermented CSM increased respectively, compared with the control. Conclusion: The isolation of a gossypol-degrading bacterium from the cow rumen is of great importance for gossypol biodegradation and may be a valuable potential source for gossypol-degradation of CSM.
