Comparison of Digital PCR and Quantitative PCR with Various SARS-CoV-2 Primer-Probe Sets

  • The World Health Organization (WHO) has declared the coronavirus disease 2019 (COVID-19) as an international health emergency. Current diagnostic tests are based on the reverse transcription-quantitative polymerase chain reaction (RT-qPCR) method, which is the gold standard test that involves the amplification of viral RNA. However, the RT-qPCR assay has limitations in terms of sensitivity and quantification. In this study, we tested both qPCR and droplet digital PCR (ddPCR) to detect low amounts of viral RNA. The cycle threshold (CT) of the viral RNA by RT-PCR significantly varied according to the sequences of the primer and probe sets with in vitro transcript (IVT) RNA or viral RNA as templates, whereas the copy number of the viral RNA by ddPCR was effectively quantified with IVT RNA, cultured viral RNA, and RNA from clinical samples. Furthermore, the clinical samples were assayed via both methods, and the sensitivity of the ddPCR was determined to be equal to or more than that of the RT-qPCR. However, the ddPCR assay is more suitable for determining the copy number of reference materials. These findings suggest that the qPCR assay with the ddPCR defined reference materials could be used as a highly sensitive and compatible diagnostic method for viral RNA detection.

  • Structural information has been a major concern for biological and pharmaceutical studies for its intimate relationship to the function of a protein. Three-dimensional representation of the positions of protein atoms is utilized among many structural information repositories that have been published. The reliability of the torsional system, which represents the native processes of structural change in the structural analysis, was partially proven with previous structural alignment studies. Here, a web server providing structural information and analysis based on the backbone torsional representation of a protein structure is newly introduced. The web server offers functions of secondary structure database search, secondary structure calculation, and pair-wise protein structure comparison, based on a backbone torsion angle representation system. Application of the implementation in pair-wise structural alignment showed highly accurate results. The information derived from this web server might be further utilized in the field of ab initio protein structure modeling or protein homology-related analyses.

  • The purpose of this study was to analyze of Hospital Coordinator based on the DACUM(Developing A Curriculum) method. The contents of this study were to extract the duties, tasks consisting of job of Hospital Coordinator and to investigate levels of importance, difficulty, frequency and entry level on each task, and to make out a job model of Hospital coordinator. A DACUM committee was composed to analyze job of Hospital Coordinator and the committee members were total 17, a facilitator, 15 hospital administrator and a recorder. The major findings of this study were as the followings ; first, duties in job of Hospital Coordinator were total 10, which were organization of Hospital Coordinator affairs, customerfacing services, consultation support, customer counsel, customer management, financial management, medical service planning, medical service marketing, organizational management, image making management, and self-development. And total tasks in job of Hospital coordinator were 76. Second, the tasks which were important, difficult, frequent and essential in entry step of occupation were counseling before consultation, explaining treatment plan after consultation, checking progress of consultation, answering teleconsultation, and finding out customer's consultation information. Third, a job model of Hospital Coordinator was constructed based on the results of DACUM job analysis.

The Analysis of segmented Group Characteristics about Beer Customer's Purchase Motivation (맥주 소비자의 구매동기에 따른 시장세분화)

  • This study explore the market segmentation based on beer consumers' purchase motivation 1) to analyze beer consumers and markets more closely and 2) to demonstrate the characteristics of each market segment and contribute to marketing strategies with beer consumer segment. Using -administrated questionnaires asked questions recognizable beer purchase within 6 months by over 20-years-olds, this study 201 surveys online 22 days. The results as follows: beer consumers' purchase motivation consisted of three factors enhancement, social orientation, and value enhancement. Cluster analysis based on beer purchase showed that there were three groups motivation multi-motivation and motivation group based on average value of beer purchase motive factors and relative load per factor. demographic,beer drinking characteristics and group difference according to cluster gender and monthly average income, and beer drinking characteristics also significantly different by preferred beer, preferred beer type, occasion and drinking place.

Evaluation Study of Home Health Care Program Operated in G Public Health Center (인천광역시 일 보건소 대도시 방문보건사업의 평가 연구)

  • Purpose: The purpose of this study was to evaluate the achievements of the project, and also to find out its strengths and weaknesses. Method: This evaluative study employed system theory and analytic techniques by using criteria which were relevance, appropriateness, adequacy, progress, effectiveness and satisfaction for input, process, output, and outcome of program. Study subjects were participants in the home health care program implemented in G public health center in Inchon metropolitan city in 2003. Results: Input factors including recruitment personnel, and support organization development were not adequately met for the program. However. the goal and objectives of the project were really appropriate for the community needs as well as government's policy. For the Process evaluation, home health care record form and computer data base had not progressed as scheduled, but overall program activities were finished on time. However, cooperation between organizations in the health center during service activity were not supportive. Managerial ability of program charged personnel about coordination and integration of team members was not affirmative. Output and outcome evaluation showed that people improved self care ability were 221 (17.5%). and 71 (5.62%) of subjects were moved into category of possible self-care. Client's satisfaction for this project showed a high degree. Conclusions: Based on the above results support organization and staff personnel for this program should be developed. Also, a community network of resources should be established and case management services should be focused continuously in community based home health care.

The Comparison of Apartment Management System Between Korea and Japan (한국과 일본의 공동주택 관리제도 비교)

  • This research is carried out to inspect the Apartment Management System of Japan, to examine the similarities and differences between the Korean Apartment Management System and the Japanese Apartment Management System. First, as the basis law of apartment management, there exists the Building Unit Ownership Act, the Promotion Law fur Adequate Mansion Management of Japan, the Housing Law, Housing Execution Law and Rule of Korea. Second, the Association of Apartment Owners, an organization of owners of apartments in Japan and the Commission of the Representatives of the Occupants in Korea become the subject of maintenance. The Japan structure is made of the Assembly, the Director and the President. The Korean structure is made of Regular and Temporary Conferences and elected Officers(1 president, at least 2 directors and at least 1 inspector). The Commission of the Representatives of the Occupants and the Organization of Owners of Apartments make bylaws and diverse maintenance rules. Third, the foremost reason why the Korean structure of maintenance of apartments is less efficient than Japan is because of the small number of people dispatched to the living-environment maintenance team and their short terms. It is necessary to grow professionals related to this sector and to have enough public servants that specialize in this. Fourth, although it is compulsory to make long term plans for maintenance based on the Housing Law, because the reserve fund is decided by the maintenance rule(with no compulsory standards of reservation) of each apartment, it is difficult to reserve an adequate amount of reserve fund. So as in the example of the state of Hawaii in America, based on long term plans for maintenance, an execution rule of the Housing Law should be made which enforces to reserve at least 50 percent of future maintenance expenses.

Understanding Collaborative Working Processes within Construction Project Teams Using Agent-Based Modeling and Simulation (에이전트기반 시뮬레이션을 활용한 건설프로젝트 조직 내 협업과정의 이해)

  • Collaborative processes among team members including communication, coordination, and information-handling processes either during pre-construction or project execution are required in order to accomplish the objectives of construction projects. However, current construction management practice does not explicitly take the effect of organizational aspects on project performance into account. This paper introduces a method to understand collaborative processes in an explicit and systematic fashion. An agent-based simulation of collaborative working processes within construction project teams was designed from game theory perspective and implemented. The simulation produced both individual behavior and network dynamics. Individuals represented as agents made efforts to improve performance by communication and coordinating with other members, and overall team network was emerged as a result of interactions among members. Interestingly, it was found that the tendency of forming cohesive subgroups increased when sustaining relations with between-group partners incurs higher cost. The primary contribution of this paper is that it presented an explicit approach to examining collaborative working processes in construction project teams and it extended existing computational organization and network studies by integrating individual behavior models and network models.

노인복지시설의 호스피스 운영모형 개발

  • The purpose of this study is to develop a management model to effectively provide hospice service of nursing home. In a method of study to achieve this purpose, a positive research and literature study have been performed at the same time. Concerning contents to be examined as a definition of notions and patterns about hospice for the aged, foreign hospice and structural elements of hospice management, a literature study to support a theoretical background has presented leading studies as an analysis form of this study through analyzing domestic and foreign literature, thesis, journal and so on. In order to attain the purpose of this study, the positive research is to carry out a survey of demand and desire of people who have already got hospice service in the first place as a fact-finding survey related to the subject of hospice service, investigate organizations to operate nursing home and hospice and relevant experts in the second place as a survey of supply system to provide hospice service, and develop an appropriate management model to connect the demand and supply of nursing home through it. Therefore, this study has developed a delivery system and a chart of the practical trend to be done minutely, presenting a model to manage a hospice of nursing home based on the above results. The management model externally has connected medical team of hospital with hospice team of home based on a community, and internally is composed centering on five elements such as administrative organization, manpower, program and contents of service, finances. In practical model of hospice management of nursing home, in order to plan a program, provide service and give a high-quality service to the aged, this study has presented a trend chart of service management on the basis of five structural elements, an analysis form of this study. In conclusion, this study could present data to develop a hospice in the field of nursing home for the aged by showing a model concerning a hospice management of nursing home, but in my opinion, a following study to be detailed and developed even more than this study should be continued.

Second, the members are aware of all the rules and principles of a united whole is accepted and leads to good performance. In addition, the ability to share sensitive business activities for self-development and also lead to work to make this a regular activity to create a team that can collaborate to help the environment and the atmosphere. Third, a systematic construction of the online collaboration system is made efficient and rapid task. According to Student team and A corporation we knew that Cloud services and social media, low-cost, high-efficiency services could achieve. The introduction of the latest information technology changes, the members of the organization's systems and active participation can take advantage of continuing education must be made. Fourth, the company to inform people both inside and outside of the organization to communicate actively to change the image of the company activities, the creation of corporate performance is very important to figure. Reflects the latest trend to actively use social media to communicate the effort is needed. For development of systematic collaboration promoting model steps to meet the organizational role. First, the Chief Executive Officer to make a firm and clear vision of the organization members to propagate the faith, empathy gives a sense of belonging should be able to have. Second, middle managers, CEO's vision is to systematically propagate the organizers rules and principles to establish a system would create. Third, general operatives internalize the vision of the company stating that the role of outside companies must adhere. The purpose of this study was well done in collaboration organizations promoting factors for strategic alignment model based on the golden circle and collaboration to understand and reflect the latest trends in information technology tools to take advantage of smart work and business know how student teams through case analysis will derive the success factors. This is the foundation for future empirical studies are expected to be present.

Validity of patient-derived xenograft mouse models for lung cancer based on exome sequencing data

  • Patient-derived xenograft (PDX) mouse models are frequently used to test the drug efficacy in diverse types of cancer. They are known to recapitulate the patient characteristics faithfully, but a systematic survey with a large number of cases is yet missing in lung cancer. Here we report the comparison of genomic characters between mouse and patient tumor tissues in lung cancer based on exome sequencing data. We established PDX mouse models for 132 lung cancer patients and performed whole exome sequencing for trio samples of tumor-normal-xenograft tissues. Then we computed the somatic mutations and copy number variations, which were used to compare the PDX and patient tumor tissues. Genomic and histological conclusions for validity of PDX models agreed in most cases, but we observed eight (~7%) discordant cases. We further examined the changes in mutations and copy number alterations in PDX model production and passage processes, which highlighted the clonal evolution in PDX mouse models. Our study shows that the genomic characterization plays complementary roles to the histological examination in cancer studies utilizing PDX mouse models.
