단감 잎 추출물의 생리 활성 (Physiological activities of water extracts from sweet persimmon leaves)

  • 단감연구소에서 출하 시기 조절을 위해 육성된 중생종 단감 신품종 홍추와 기존 재배품종 상서조생, 서촌조생, 부유 3품종의 잎 추출물을 제조하여 생리활성을 비교하였다. 그 결과, 홍추의 잎 추출물의 총페놀 함량, Vit. C 함량, DPPH 및 ABTS 라디칼 소거능, ADH 활성에 미치는 영향이 다른 추출물에 비하여 유의하게 높다는 결과를 얻을 수 있었다. ADH 활성도는 부유의 잎 추출물에서 다른 품종에 비해 비교적 높게 나타났다. 이상의 결과는 신품종 단감 홍추의 잎을 이용한 고품질 감잎차의 제조가 가능함을 시사한다.

Improving productivity of mulberry trees and silkworm, Bombyx mori L., using vermicompost application

  • Vermicompost, manure, compost and organic fertilization are ecofriendly. Nowadays, many products resulted from sericulture consumed by humans such as mulberry leaves, fruits, mulberry tea, silk and natural silk cosmetics. Soil applications of three treatments with vermicompost (0.5, 1 and 2 tons per 0.42 hectare) and recommended rate of mineral fertilizers of nitrogen phosphorus potassium were used for investigation. Impact of fertilization on mulberry plant traits of moisture, number of shoots/tree, total shoots length/tree, number of leaves/shoot, number of leaves/ (100g), leaf yield/tree and leaf yield of fadden/season were recorded. In addition the effect of fertilization on larval and cocoon characters of young instar duration, fifth instar duration, total larval duration, larval mortality percentage, weight of third instar larvae, weight of fourth instar larvae, weight of fifth instar larvae, fresh cocoon weight, fresh shell weight, pupae weight, cocoon shell ratio, silk productivity, cocooning percentage, pupation ratio, number of cocoons/ liter, crop cocoons by number, crop cocoons by weight, fecundity and fertility. Using vermicompost treatment was enhancing plant characters. Treatments of V3, V2 and V1 were shortage young, fifth and larvae durations. Mostly feeding silkworm during the whole larval duration on treated mulberry leaves with vermicompost improving the traits average. Using vermicompost for fertilization by rate of V3 and V2 is better than others for cocoon characters for females and males.V3 and V2 of vermicompost per 0.42 hectare is recommended for rearing mulberry silkworm instead of mineral fertilization.

모싯잎의 육성 계통별 성분 함량, 항산화 및 ACE 저해 활성 (Component Content, Antioxidant and ACE Inhibitory Activity of Ramie (Leaf Boehmeria nivea M.) according to Breeding Lines)

  • Ramie (Boehmeria nivea M.) has been used for fiber materials in Korea traditionally, but in recent years, the concern with ramie leaves for the food industry such as tteok (a kind of Korean rice cake) industry has been increasing, so a study for eatable ramie is required for the expansion of ramie consumption. Moreover, the ramie varieties for the food industry are not established, so the natural species are cultivated in general; therefore, it is very important to select the ramie varieties for the food industry such as rice cakes, tea, beverage and so on. This study was undertaken to compare the physiochemical properties among 9 ramie lines selected in the Yeonggwang-gun Agricultural Technology Center to select the eatable ramie varieties for the food industry. The contents of the protein among 9 ramie lines was 6.21~7.56% and had the highest content in the YG55. The folic acid (folate) and vitamin C content had varying differences among the 9 lines; the content of folate showed $771.52{\sim}1,978.84{\mu}g%$, that of vitamin C showed 149.42~275.34 mg%. The ACE inhibitory activity appeared to be the highest in YG88 (21.5%) among the 9 ramie varieties tested.

고온에서의 브로콜리 부위별 추출물의 항산화성 및 항균성 (Antioxidant Activity and Antibacterial Activities from Different Parts of Broccoli Extracts under High Temperature)

  • 브로콜리의 잎, 꽃, 꽃-줄기,줄기 그리고 껍질을 에탄올, 아세톤 및 증류수로 추출하여 그 추출물의 라디칼소거 활성을 조사하였다 각각의 시료는DPPH 라디칼에 대한 라디칼 소거 활성 (FRSA)을 나타내었다. 다섯 가지의 시료 중, 줄기 추출물, 꽃 추출물 및 잎 추출물에서 dot-blot 분석을 통해 라디칼소거 활성을 확인하였다. 브로콜리의 에탄을 추출물 에서는 $pH\;2\~6$의 산성영역 및 $60\~80^{\circ}C$의 온도 범위에서 강력한 라디칼소거 활성을 보였다. 녹차 추출물과 쑥 추출물에서는 Staphylococcus aureus에 대한 항균성을 보였고, 다섯 가지의 추출물중에서 단지 꽃 추출물 및 꽃-줄기 추출물이 고온 하에서 Bacillus arnyloliquefaciens에 대해 강력한 항균성을 나타내었다.

내한 초다수성 차나무 신품종 "한광" (New Tea Plant Variety, "Hankwang", with High Yield and Cold Tolerance)

  • 전국 차나무 자생지에서 차나무를 수집하여 조성된 클론보존원의 630클론 중 생육이 왕성하고 내한성이 높은 차나무 31계통을 선발하였다. 선발된 31계통 중 최종적으로 3계통을 선발하여 2006년부터 2008년까지 전국 6개 지역에서 지역적응성 및 영양계 비교시험을 실시하였다. 그 결과 내한성이면서 다수성을 가진 영양계 차나무 "한광"을 최종 선발하였다. "한광"은 생육이 왕성하여 새싹수는 대비품종인 야부기다에 비하여 27.6% 많았고 백아중은 47.9 g으로 대비품종보다 99.6% 무거웠다. 수량도 대비품종보다 152% 증수된 182 kg/10a으로 초다수성이었다. 또한 "한광"은 내한성이 매우 우수하여 겨울철에 적고현상 발생이 적고 탄저병에도 매우 강하게 나타났다. 탄닌, 카테킨, 총질소, 카테킨 등의 기능성분 함량도 비교적 높게 나타났다. "한광"의 아미노산 함량은 2,567 mg%로 대비품종보다 112.7% 높았다.

Sasa quelpaertensis Leaf Extracts Induce Apoptosis in Human Leukemia HL-60 Cells

  • In east Asia, the leaves of various Sasa species have been used in folk medicine for centuries. The effects of the methanolic extract and its subsequent fractions derived from the leaves of Sasa quelpaertensis Nakai on cell proliferation in human leukemia HL-60 cells were evaluated. The ethyl acetate fraction of this extract (ESQL) significantly reduced cell viability in a dose-dependent manner ($0-250\;{\mu}g/mL$). ESQL ($IC_{50}=24.8\;{\mu}g/mL$) exhibited growth inhibition comparable to the main constituent of green tea, epigallocatechin ($IC_{50}=26.2\;{\mu}g/mL$), which was used as a positive control. ESQL treatment induced apoptosis, which was confirmed by the presence of nuclear condensation and annexin V-staining. These results demonstrate that ESQL contains chemopreventive phytochemicals that may be useful in neutraceutical applications.

Activity of Essential Oils Against Bacillus subtilis Spores

  • Alternative methods for controlling bacterial endospore contamination are desired in a range of industries and applications. Attention has recently turned to natural products, such as essential oils, which have sporicidal activity. In this study, a selection of essential oils was investigated to identify those with activity against Bacillus subtilis spores. Spores were exposed to 13 essential oils, and surviving spores were enumerated. Cardamom, tea tree, and juniper leaf oils were the most effective, reducing the number of viable spores by 3 logs at concentrations above 1%. Sporicidal activity was enhanced at high temperatures ($60^{\circ}C$) or longer exposure times (up to 1 week). Gas chromatography-mass spectrometry analysis identified the components of the active essential oils. However, none of the major oil components exhibited equivalent activity to the whole oils. The fact that oil components, either alone or in combination, did not show the same level of sporicidal activity as the complete oils suggested that minor components may be involved, or that these act synergistically with major components. Scanning electron microscopy was used to examine spores after exposure to essential oils and suggested that leakage of spore contents was the likely mode of sporicidal action. Our data have shown that essential oils exert sporicidal activity and may be useful in applications where bacterial spore reduction is desired.

소아 성장 한의약건강증진프로그램 개발 (Development of Korean Medicine Health Promotion Program for Short Stature Children)

  • Objectives : The aim of this study was to introduce the development process of Korean medicine health promotion program for short stature children that is suitable for applied at public health center. Methods : The draft of health promotion program was developed through literature search of previous similar programs and advice of several experts. A small conference targeted public health Korean medicine doctors was also held to introduce the developed program and discuss for improvements. Results : The details of 12-week of Korean medicine health promotion program for children growth are as follows: 4-week of taking herbal medicines (Yukmijihwang-tang or Yukgunja-tang), contactless counseling with Korean medicine doctor, self-care home kit consisting of Sogeonjung-tang tea leaf, Sogeonjung-tang with Cervi Parvum Cornu extract, ear acupressure stick, finger chuna manual, growth diary, and jump rope. Conclusions : This health promotion program can help to increase the height of children and self-esteem of children with short stature as well as decrease the stress of parents.

Comparison of in vitro antioxidant capacities of Phragmites communis Trin. and Phragmites japonica Steud.

  • The rhizome of Phragmites communis Trin. is used for vomiting and belching by clearing stomach and the sprout is used as tea. Phragmites japonica is similar with P. communis except the color of sheath is purple. This study is aimed to compare the in vitro antioxidant activity, total polyphenol and flavonoids contents of P. communis and P. japonica. The antioxidant activities of fractions from the two Phragmites plants were evaluated by 1,1-diphenyl-2- picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assay. The antioxidant activity varied with plant parts and extract solvents. The fractions of leaf extract from the two Phragmites plants (4.06±1.32-16.47±1.28%) showed higher antioxidant activity by DPPH assay compared with rhizome fractions of two Phragmites plants (0.00±0.00-14.15±0.07%), these are lower compared with ascorbic acid and butylhydroxyanisole (BHA). The highest ABTS radical scavenging activity was found for rhizome ether fraction, namely 74.95±0.56% and 73.04±1.85% for P. communis and P. japonica, these are higher than BHA. The total polyphenol and flavonoids contents were different with plant parts and extract solvents, likewise antioxidant activity. A significant correlation was shown between DPPH and ABTS radical scavenging activity. Considering the results of this study, the leaves and stems of P. communis and P. japonica are expected to be used as natural antioxidants.

차나무 잎과 배 배양에 있어서 식물 생장조절물질이 형태형성에 미치는 영향 (Effect of exogeneous plant growth regulators on morphogenetic response in vitro by embryo and leaf cultures of Camellia sinensis(L.) O. Kuntze)

  • 한국 야생차나무의 잎절편과 배배양에서 생장조절물질의 첨가에 따른 형태형성 과정의 변이를 조사하였다. 그 결과 접합자배는 싸이토키닌을 5-20$\mu\textrm{M}$을 첨가한 MS 배지에서 직접적인 체세포 배와 부정아 및 액아 발생률이 높았으며 옥신의 함량이 높아질수록 형태형성율이 급격히 저하되었다. 1/2 MS 및 1/4 SH배지에 $10\mu\textrm{M}$ IBA가 첨가된 배지에서 모든 줄기가 발근 되었다. MS배지에 2,4-D를 첨가한 배지에서 미성숙 접합자 배를 배양한 결과 체세포배성 캘러스가 유발되었다. 성숙된 접합자 배를 발아시킨 후 어린잎을 채취하여 고농도의 옥신 (IBA와 NAA) 또는 싸이토키닌 (BAP)이 함유된 MS배지에 배양한 결과 체세포배 형성 캘러스가 발생되었으며 또한 직접적인 체세포배가 발생하였다. 그러나 뿌리와 줄기 형성에는 각각 요구하는 옥신의 농도와 종류가 각각 달랐다.
