Krigia속의 과피의 해부학적 구조와 분류학적 의미 (Fruit wall anatomy of the genus Krigia (Asteraceae, Lactuceae) and their taxonomic implications)

  • Krigia속 7종 모두와 근연속인 Nothocalais cuspidata 1종에 대해 과피 해부학적 형질을 조사하여 분류학적 유연관계를 추론하였다. 과피 해부학적 형질 중 과피의 모양, 늑의 수는 Krigia속 모든 종이 같은 형질을 공유하고 있으나, costa 의 모양, costa와 intercosta에서 중과피를 이루는 libriform 섬유세포와 fiber-sclereid 세포층의 수와 이들 세포층의 발달정도가 종간에서 차이점이 나타났다. Krigia biflora, K. cespitosa, K. occidentalis와 K. wrightii는 costa와 intercosta에서 libriform 섬유세포가 잘 발달되어있는 반면에 K. dandelion, K. montana와 K. virginica는 잘 발달되어있지 않고, 대부분 fiber-sclereid 세포로 이루어져있다. 이전 분류체계에서 Krigia 절에 속했던 K. biflora는 과피 해부학적 형질상의 차이로 볼 때 Cymbia절에 더 많은 유사성을 가지고 있다.

Intestinal Microbial Dysbiosis in Beagles Naturally Infected with Canine Parvovirus

  • Canine parvoviral enteritis (PVE) is an important intestinal disease of the puppies; however, the potential impact of the canine parvovirus (CPV) on the gut microbiota has not been investigated. Therefore, the aim of this study was to evaluate the gut microbial shifts in puppies naturally infected with CPV. Fecal samples were collected from healthy dogs and those diagnosed with PVE at 4, 6, 8, and 12 weeks of age. The distal gut microbiota of dogs was characterized using Illumina MiSeq sequencing of the bacterial 16S rRNA genes. The sequence data were analyzed using QIIME with an Operational Taxonomic Unit definition at a similarity cutoff of 97%. Our results showed that the CPV was associated with significant microbial dysbiosis of the intestinal microbiota. Alpha diversity and species richness and evenness in dogs with PVE decreased compared to those of healthy dogs. At the phylum level, the proportion of Proteobacteria was significantly enriched in dogs with PVE while Bacteroidetes was significantly more abundant in healthy dogs (p < 0.05). In dogs with PVE, Enterobacteriaceae was the most abundant bacterial family accounting for 36.44% of the total bacterial population compared to only 0.21% in healthy puppies. The two most abundant genera in healthy dogs were Prevotella and Lactobacillus and their abundance was significantly higher compared to that of dogs with PVE (p < 0.05). These observations suggest that disturbances of gut microbial communities were associated with PVE in young dogs. Evaluation of the roles of these bacterial groups in the pathophysiology of PVE warrants further studies.

Fungal Community Analyses of Endophytic Fungi from Two Oak Species, Quercus mongolica and Quercus serrata, in Korea

  • Fungal endophytes have been recorded in various plant species with a richness of diversity, and their presence plays an essential role in host plant protection against biotic and abiotic stresses. This study applied the Illumina MiSeq sequencing platform based on the amplification of fungal ribosomal ITS2 region to analyze fungal endophytic communities of two oak species (Quercus mongolica and Q. serrata) with different oak wilt disease susceptibilities in Korea. The results showed a total of 230,768 sequencing reads were obtained and clustered at a 97% similarity threshold into 709 operational taxonomic units (OTUs). The OTUs of Q. serrata were higher than that of Q. mongolica with the number of 617 OTUs and 512 OTUs, respectively. Shannon index also showed that Q. serrata had a significantly higher level of fungal diversity than Q. mongolica. Total of OTUs were assigned into 5 fungal phyla, 17 classes, 60 orders, 133 families, 195 genera, and 280 species. Ascomycota was the dominant phylum with 75.11% relative abundance, followed by Basidiomycota with 5.28%. Leptosillia, Aureobasidium and Acanthostigma were the most abundant genera detected in Q. serrata with the average relative abundance of 2.85, 2.76, and 2.19%, respectively. On the other hand, Peltaster, Cladosporium and Monochaetia were the most common genera detected in Q. mongolica with the average relative abundance of 4.83, 3.03, and 2.87%, respectively. Our results indicated that fungal endophytic communities were significantly different between two oak species and these differences could influence responses of host trees to oak wilt disease caused by Raffaelea quercus-mongolicae.

A report of 43 unrecorded bacterial species within the phyla Bacteroidetes and Firmicutes isolated from various sources from Korea in 2019

  • In 2019, 43 bacterial strains were isolated from food, soil, marine environments, human, and animals related sources from the Republic of Korea. Based on the analysis of 16S rRNA gene sequence, these isolates were allocated to the phyla Bacteroidetes and Firmicutes as unrecorded species in Korea. The 10 Bacteroidetes strains were classified into the families Bacteroidaceae, Chitinophagaceae, Cytophagaceae, Flavobacteriaceae, and Prolixibacteraceae (of the orders Bacteroidales, Chitinophagales, Cytophagales, Flavobacteriales, and Marinilabiliales, respectively). The 33 Firmicutes strains belonged to the families Bacillaceae, Paenibacillaceae, Planococcaceae, Staphylococcaceae, Clostridiaceae, Lachnospiraceae, Peptostreptococcaceae, Enterococcaceae, Lactobacillaceae, Leuconostocaceae, and Streptococcaceae (of the orders Bacillales, Clostridiales, and Lactobacillales). These unrecorded bacteria were determined based on taxonomic criterion (>98.7%; 16S rRNA gene sequence similarity). In addition, their phylogenetic affiliation, as well as cell and colony morphologies, staining reactions, and physiological and biochemical properties were investigated. Therefore, we report 43 isolates as unrecorded species, and described basic features, isolation source, and locations of these strains.

Mucilaginibacter aquariorum sp. nov., Isolated from Fresh Water

  • A Gram-stain-negative, rod-shaped bacterial strain, JC4T, was isolated from a freshwater sample and determined the taxonomic position. Initial identification based on 16S rRNA gene sequences revealed that strain JC4T is affiliated to the genus Mucilaginibacter with a sequence similarity of 97.97% to Mucilaginibacter rigui WPCB133T. The average nucleotide identity and digital DNA-DNA hybridization values between strain JC4T and Mucilaginibacter species were estimated below 80.92% and 23.9%, respectively. Strain JC4T contained summed feature 3 (C16:1 ω7c and/or C16:1 ω6c) and iso-C15:0 as predominant cellular fatty acids. The dominant polar lipids were identified as phosphatidylethanolamine, one unidentified aminophospholipid, one unidentified phospholipid, and two unidentified lipids. The respiratory quinone was MK-7. The genomic DNA G+C content of strain JC4T was determined to be 42.44%. The above polyphasic evidences support that strain JC4T represents a novel species of the genus Mucilaginibacter, for which the name Mucilaginibacter aquariorum sp. nov. is proposed. The type strain is JC4T (= KCTC 92230T = LMG 32715T).

A report of 20 unrecorded bacterial species isolated from the coastal area of Korean islands in 2022

  • Bacterial communities inhabiting islands play a vital role in the functioning and formation of a unique, isolated ecosystem. Nevertheless, there has been a lack of systematic research on the indigenous microbiological resources of the islands in Korea. To excavate microbial resources for further studies on the metabolism and biotechnological potential, a standard dilution plating was applied to coastal seawater samples collected from islands along the west coast of the Korean Peninsula, including Deokjeokdo, Baengnyeongdo, and Daebudo in 2022. A total of 2,007 bacterial strains were isolated from the samples as single colonies and identified using 16S rRNA gene sequence analyses. A total of 20 strains, with ≥98.7% 16S rRNA gene sequence similarity to bacterial species having validly published names but not reported in Korea, were designated as unrecorded bacterial species in Korea. The unrecorded bacterial strains were phylogenetically diverse and belonged to four phyla, five classes, 12 orders, 17 families, and 18 genera. The unreported species were assigned to Algimonas, Amylibacter, Notoacmeibacter, Roseibium, and Terasakiella of the class Alphaproteobacteria; Alteromonas, Congregibacter, Marinagarivorans, Marinicella, Oceanospirillum, Psychromonas, Thalassotalea, Umboniibacter, and Vibrio of the class Gammaproteobacteria; Lutibacter and Owenweeksia of the class Flavobacteriia; Paenibacillus of the class Bacilli; and Pelagicoccus of the class Opitutae. The taxonomic characteristics of the unreported species, including morphology, biochemistry, and phylogenetic position are provided in detail.

Description and Genomic Characteristics of Weissella fermenti sp. nov., Isolated from Kimchi

  • A Gram-positive, non-motile, and non-spore-forming lactic acid bacterium, designated as BK2T, was isolated from kimchi, a Korean traditional fermented vegetable food, and the taxonomic characteristics of strain BK2T, along with strain LMG 11983, were analyzed. Both strains optimally grew at 30℃, pH 7.0, and 1.0% NaCl. Cells of both strains were heterofermentative and facultatively anaerobic rods, demonstrating negative reactions for catalase and oxidase. Major fatty acids (>10%) identified in both strains were C18:1 ω9c, C16:0, and summed feature 7 (comprising C19:1 ω6c and/or C19:1 ω7c). The genomic DNA G+C contents of both strains were 44.7 mol%. The 16S rRNA gene sequence similarity (99.9%), average nucleotide identity (ANI; 99.9%), and digital DNA-DNA hybridization (dDDH; 99.7%) value between strains BK2T and LMG 11983 indicated that they are different strains of the same species. Strain BK2T was most closely related to Weissella confusa JCM 1093T and Weissella cibaria LMG 17699T, with 100% and 99.4% 16S rRNA gene sequence similarities, respectively. However, based on the ANI and dDDH values (92.3% and 48.1% with W. confusa, and 78.4% and 23.5% with W. cibaria), it was evident that strain BK2T represents a distinct species separate from W. confusa and W. cibaria. Based on phylogenetic, phenotypic, and chemotaxonomic features, strains BK2T and LMG 11983 represent a novel species of the genus Weissella, for which the name Weissella fermenti sp. nov. is proposed. The type of strain is BK2T (=KACC 22833T=JCM 35750T).

Discovery of 18 previously unrecorded bacterial species in the coastal areas surrounding Korean islands in 2023

  • Bacterial communities residing on islands have a significant impact on the functioning and establishment of a unique isolated ecosystem. Notwithstanding, systematic research on the indigenous microbial resources of domestic islands has been lacking. In order to understand the biodiversity and potential bioresources, we conducted sampling in 2023 from coastal waters from various islands off the west coast of the Korean Peninsula, including Baengnyeongdo, Daebudo, Deokjeokdo, Jangbongdo, Yeonpyeongdo Islands, along with Somaemuldo Island along the south coast. The coastal seawater samples were used to unearth microbial resources through the standard dilution plating. In total, approximately 1,600 bacterial strains were isolated from the samples as single colonies and identified using 16S rRNA gene sequence analyses. Eighteen strains, exhibiting ≥98.7% 16S rRNA gene sequence similarity to bacterial species with validly published names but not previously reported in Korea, were categorized as unrecorded bacterial species in Korea. These unrecorded bacterial strains displayed phylogenetic diversity, representing three phyla, four classes, 9 orders, 13 families, and 18 genera. The unrecorded species were assigned to the classes Alphaproteobacteria (Aliiroseovarius, Kiloniella, Maritalea, Palleronia, and Roseobacter), Gammaproteobacteria (Aliamphritea, Aliivibrio, Enterovibrio, Francisella, Leucothrix, Pseudoalteromonas, Psychrobium, Shewanella, and Vibrio), Flavobacteriia (Aquimarina, Pseudofulvibacter, and Tenacibaculum), and Verrucomicrobiae (Roseibacillus). This study presents comprehensive descriptions of the taxonomic attributes of these unrecorded species, covering morphology, biochemistry, and phylogenetic position.

향나무속(屬)의 분류학적(分類學的) 연구(硏究) (Taxonomic Studies of Genus Juniperus)

  • 남한(南韓)에 자생(自生) 또는 도입재배(導入栽培)되고 있는 향나무속(屬) 종류(種類)들의 분류학적(分類學的) 문제(問題)들을 해결(解決)하고 재정비(再整備)할 목적(目的)으로 자생종(自生種) 7종류(種類)와 조입종중(導入種中)에서 많이 재배(栽培)되고 유용(有用)하게 이용(利用)되고 있는 원예종(園藝種) 4종류(種類)를 재료(材料)로 하여 각종류(各種類)의 꽃의 구조(構造), 구과(毬果), 종자(種子), 가지습성(習性), 기본수형(基本樹形) 및 잎의 형태적(形態的) 특징(特徵)을 조사(調査)해서 속(屬)과 종(種)의 검사표(檢査表)를 작성(作成)했으며 외부형태적(外部形態的) 특징(特徵)으로는 종간(種間)의 식별(識別)이 모호(模糊)한 것을 명확(明確)히 구명(究明)하기 위(爲)해서 근단(根端)을 재료(材料)로 하여 염색체(染色體)의 기본수(基本數)와 핵형분석(核型分析)을 하고 그 결과(結果)로 각(各) 종간(種間)의 차이점(差異點)을 비교분석(比較分析)하였던바 전종(全種)의 염색체장(染色體長)은 $9.0{\sim}15{\mu}m$, 향나무, 심향나무, 가이즈가향나무, 섬향나무(일본산(日本産))은 4배체(倍體)(4n=44), 옥향나무, 눈향나무, 뚝향나무, 연필향나무, 노간주나무, 좀노간주나무, 해변노간주나무는 2배체(倍體)(2n=22)이고 향나무절(節)의 종(種)들은 염색체(染色體)길이 및 동원체(動原體)의 위치(位置)의 변이(變異)가 대단(大端)히 심(甚)하였다. 노간주나무절절(節)의 종(種)들은 11번 염색체(染色體)가 t형(型)이고, m형(型) 염색체(染色體)가 많은 비교적(比較的) 균일(均一)하고 단순(單純)한 염색체(染色體) set를 이루는 것이 향나무절(節)과 뚜렷이 다른 세포학적(細胞學的)인 특징(特徵)이다. 향나무절(節)에서는 대도시(大都市)에 여러 종류(種類)들이 혼식(混植)되어 잇는 곳에서는 동종(同種)의 지역품종(地域品種) ecotype간(間), 때로는 기본종(基本種)과 변종간(變種間)에 교배(交配)가 일어나고 있기 때문에 향나무 종내개체(種內個體)들 간(間)에는 형태적(形態的)으로 차이(差異)가 다양(多樣)하다. 상기(上記) 결과(結果)들을 종합(綜合)하여 향나무속(屬)의 특성(特性)을 세포학적(細胞學的)으로 명확(明確)히 식별(識別)하고 종간(種間)의 고유특성(固有特性)을 밝히고 종(種)의 특징(特徵)을 기재(記載)하였다.

  • 한국산 기름종개 3 아종의 분류학적 의문점을 해결하기 위하여 전국 23개 집단을 대상으로 체측반문을 비롯한 형태형질 분석과 전기영동법에 의한 유전자 분석을 실시하였다. 3 아종의 체측반문 변이 분석 결과 기름종개와 점줄종개에서는 각 아종 의 특정인 점열형 (A, B type: 14.5%)의 개체보다 중간형 (C, D type: 51.0%)의 빈도가 월등히 높았다. 기름종개 3 아종은 모두 암수간 체측반문 형태가 달리 발현되는 성적이형 현상이 뚜렷하였다. 형태계측 및 집단 판별분석에서 집단간 및 아종간의 뚜렷한 차이는 발견되지 않았다. 전기영동법에 의한 유전자 분석결과 3 아종중 줄종개 5개 집단의 평균 유전적 변이가 A=1.48. P=31.2%. HD= 0.082 및 HQ=0.099로서 기름종개(A=1.37. P=24.7%. HD=0.058 및 HQ = =0.065) 및 점줄종개(A=1.43, P=27,0%. 매=0.066 및 HQ=0.079)에 비하여 높은 변이를 나타내었다 각 아종의 유전적 근연관계에 있어 기름종개와 줄종개 및 점줄종개의 평균 유전적 근연치는 S = O.64로서 일반적인 담수어류의 종간차이를 보였고, 줄종개와 점줄종개간은 S=0.82로서 아종 수준의 유전적 근연관계를 나타내었다. 체측반문 변이, 형태분석 결과 및 유전자 분석결과 기름종개는 타 두 아종과 약 200만년전에 분화된 별종으로 사료된다. 기름종개 (C. t. taenia)의 학명사용에 있어 이미 보고된 자료(핵형, 형태, 분포 등)들을 비교할 때 중국산 C. Sinensis와 많은 차이가 있어 한국산 기름종개를 중국산 C. sinensis Sauvage a and Dabry 의 synonym으로 보는 것은 타당성이 없다. 그러나 줄종개와 점줄종 개는 형태 및 유전적 특정등을 고려할 때 아종으로 취급하는 것이 타당하리라 여겨 지며 앞으로 이들에 대한 학명의 재검토가 요구된다.
