  • Most CCTVs operated by public institutions for crime prevention and parking enforcement are located on roads. The angle of these CCTV's view is often changed for various reasons, such as bolt loosening by vibration or shocking by vehicles and workers, etc. In order to effectively provide AI services based on the collected images, the service target area(ROI, Region Of Interest) must be provided without interruption within the image. This is also related to the viewpoint of effective operation of computing power for image analysis. This study explains how to maximize the application of artificial intelligence technology by setting the ROI based on the marker on the road, setting the image analysis to be possible only within the area, and studying the process of finding the ROI.

Extracellular Acidification Augments NLRP3-Mediated Inflammasome Signaling in Macrophages

  • Inflammation is a series of host defense processes in response to microbial infection and tissue injury. Inflammatory processes frequently cause extracellular acidification in the inflamed region through increased glycolysis and lactate secretion. Therefore, the immune cells infiltrating the inflamed region encounter an acidic microenvironment. Extracellular acidosis can modulate the innate immune response of macrophages; however, its role for inflammasome signaling still remains elusive. In the present study, we demonstrated that macrophages exposed to an acidic microenvironment exhibited enhanced caspase-1 processing and IL-1β secretion compared with those under physiological pH. Moreover, exposure to an acidic pH increased the ability of macrophages to assemble the NLR family pyrin domain containing 3 (NLRP3) inflammasome in response to an NLRP3 agonist. This acidosis-mediated augmentation of NLRP3 inflammasome activation occurred in bone marrow-derived macrophages but not in bone marrow-derived neutrophils. Notably, exposure to an acidic environment caused a reduction in the intracellular pH of macrophages but not neutrophils. Concordantly, macrophages, but not neutrophils, exhibited NLRP3 agonist-mediated translocation of chloride intracellular channel protein 1 (CLIC1) into their plasma membranes under an acidic microenvironment. Collectively, our results demonstrate that extracellular acidosis during inflammation can increase the sensitivity of NLRP3 inflammasome formation and activation in a CLIC1-dependent manner. Thus, CLIC1 may be a potential therapeutic target for NLRP3 inflammasome-mediated pathological conditions.

  • Village-concentrated agricultural regional development projects that aim for increased profits are now being aimed at core agricultural and fishing areas. These agricultural and fishing stimulus projects aim to not only increase the net profit of local citizens but also improve their living conditions. As the industry itself has changed, local rural areas have also changed in various ways. One such aspect is the emergence of rural-urban complexes known as "rurban" areas. These naturally occurring rurban regions are created by a combination of complex urban infrastructure while maintaining the insulated rural communities that agricultural areas are usually so readily identified by. "Jeongeupsi Naejangsnagdong", the target of this research, also specifies the unaltered surrounding natural environment of rural areas while at the same time containing complex central living areas typically found in urban areas. This research suggests that the direction of residents' participation in community-level rurban projects could solve the problems found in existing top-down government development projects. This research also suggests rurban area activation plans to improve living conditions through analysis of both local rurban agricultural characteristics and citizen demands. In order to encourage citizen autonomy and self-governing attitudes, citizen-strengthening workshop programs are proposed, such as citizen workshops or pilot activities. This research was carried out by target area analysis, rudimentary planning, development direction setting, detailed project planning, and finally project processing. This procedure established three goals, which are walking environment improvement, community infrastructure establishment, and good living environment establishments, based on actual site research and citizen demands. This research suggests plans to activate community groups that were already established and reflect citizen needs as the main avenues for local businesses. This research is predicted to promote more active and successful growth through autonomy in stimulating these increasingly emerging rurban agricultural regions.

Time-Series Analysis and Estimation of Prospect Emissions and Prospected Reduction of Greenhouse Gas Emissions in Chungbuk (온실가스 배출량 시계열 분석과 전망 배출량 및 감축 감재량 추정 - 충북을 중심으로 -)

  • In accordance with the enactment of 'the Paris Agreement' in 2015 and 'the Framework Act on Carbon Neutrality and Green Growth for Response to the Climate Crisis' in 2021, each local government has set appropriate reduction target of greenhouse gas to achieve the nationally determined contribution (NDC, the reduction target of 40% compared to 2018) of greenhouse gas (GHG) emissions in 2030. In this study, the current distribution of GHG emissions was analyzed in a time series centered on the Chungbuk region for the period from 1990 to 2018, with the aim of reducing GHG emissions in Chungbuk by 2030 based on the 2030 NDC and scenario. In addition, the prospected reduction by 2030 was estimated considering the projected emissions according to Busines As Usual in order to achieve the target reduction of GHG emissions. Our results showed that GHG emissions in Chungbuk and Korea have been increasing since 1990 owing to population and economic growth. GHG emissions in 2018 in Chungbuk were very low (3.9 %) relative to the national value. Moreover, emissions from fuel combustion, such as cement and lime production, manufacturing and construction industries, and transportation industries, were the main sources. Furthermore, the 2030 target of GHG emission reduction in Chungbuk was set at 40.2% relative to the 2018 value, in accordance with the 2030 NDC and 2050 carbon-zero national scenario. Therefore, when projected emissions were considered, the prospected reduction to achieve the target reduction of GHG emissions was estimated to be 46.8% relative to 2018. The above results highlight the importance of meeting the prospected reduction of GHG emissions through reduction means in each sector to achieve the national and local GHG reduction target. In addition, to achieve the 2030 NDC and 2050 carbon zero, the country and each local government, including Chungbuk, need to estimate projected emissions by year, determine reduction targets and prospect reductions every year, and prepare specific means to reduce GHG emissions.

Pickprimer: A Graphic User Interface Program for Primer Design on the Gene Target Region (픽프라이머 : 유전자 목표 구간 탐색 모듈을 포함한 프라이머 제작 그래픽 프로그램)

  • In genetic and molecular breeding studies of plants, researchers need to design various kinds of primers based on their research purposes. So far many kinds of web- or script-based non-commercial programs for primer design are available. Because most of them do not include user interface for multipurpose usage including gene structure prediction and direct target selection on sequences, it has been a laborious work to design primers targeting on the exon or intron regions of interesting genes. Here we report a primer designing graphic user interface program, Pickprimer, that includes gene structure prediction and primer design modules by combining source codes of the Spidey and Primer3 programs. This program provides simple graphic user interface to input sequences and design primers. Genomic sequence and mRNA or coding sequence of genes can be copy and pasted or input as fasta or text files. Based on alignment of the input sequences using the Spidey module, a putative gene structure is graphically visualized along with exon-intron sequences of color codes. Primer design can be easily performed by dragging mouse on the displayed sequences or input primer targeting position with desirable values of primers. The output of designed primers with detailed information is provided by the Primer3 module. PCR evaluation of 24 selected primer sets successfully amplified single amplicons from six Brassica rapa cultivars. The Pickprimer will be a convenient tool for genetic and molecular breeding studies of plants.

Environmental Impacts Assessment of Elementary School Buildings and Establishment of the Reference Target using Life Cycle Assessment Model (전과정평가 모델을 이용한 초등학교 건축물 환경영향 평가 및 비교기준 수립)

  • In order to determine how much a new green building reduce the environmental impacts, it is necessary to establish the reference target for comparison. Therefore, this study aims to establish the reference target by evaluating the environmental impacts of existing buildings. To ensure this end, this study evaluated the environmental impacts(Global warming potential, ozone layer depletion potential, acidification potential, eutrophication potential, photochemical ozone creation potential, and abiotic depletion potential) of 17 existing elementary school buildings, which are located in Seoul, Busan, Daegu, and Gwangju, by using the hybrid LCA model. As a result, the environmental impacts of the case buildings were clearly distinguished in different regions. Therefore, this study presented the reference targets which are appropriate to each region. For example, the reference targets for global warming potential, which can be used in Seoul, Busan, Daegu, and Gwangju, are 3.76E+03, 1.90E+03, 2.63E+03, $2.81E+03kg-CO_2\;eq./m^2$, respectively. The presented reference targets are expected to be useful for understanding how much environmental impacts can be reduced when a new green school building is constructed.

The vegetation analysis of Northern region at Jungnang riverside - Between two bridges of Wallgae 1 and Sangdo - (서울시 중랑천 북부구간 하천변 식생과 식물상 분석 - 월계1교에서 상도교 구간을 대상으로 -)

  • After the modern industrial revolution, rivers in cities became covered and disappeared due to the pressure to develop them. Likewise, their function which is to serve as the basis of natural ecology system in the cities began to be damaged. This research demonstrated that there are a total of 268 categories when it comes to the list of plants, including 64 families, 179 genera, 230 species, 36 varieties, and 1 subspecies. When the relative abundance of the plants that were found at the target research site was studied, the secondary survey demonstrated Bromus japonicus 22.97, Artemisia princeps var. orientalis 16.76 and Erigeron annuus 15.69 while third survey demonstrated Digitaria ciliaris 26.78, Ambrosia trifida 16.29 and Aster pilosus 14.31. There were 54 species of naturalized plants that appeared. Analysis demonstrated annual plant 23 classification category (43%), perennial 11 classification category (20%), multi-perennation 17 classification category (31%), woody plant 3 classification category (6%) and others. When the naturalized plants that were found at the target research site were analyzed by the place of origin, North America and EU took up 76%, which accounts for 3/4 of the all the naturalized plants. At the target research site, naturalization degree of 5 pertained to 22 classification category (41%), which was the highest, followed by 19 classification category (35%) with naturalization degree of 3, 8 classification category (15%) with naturalization degree of 2 and 5 classification category (9%) with naturalization degree of 4 in the order mentioned. Flora of Jungnangcheon did not manifest any change compared to 10 years ago. Thus, it is necessary to increase of biodiversity efforts to improve SeoulCity's natural environment and cityscape.

Transcriptional Regulation of Genes by Enhancer RNAs (인핸서 RNA에 의한 유전자 전사 조절)

  • Genes in multicellular organisms are transcribed in development, differentiation, or tissue-specific manners. The transcription of genes is activated by enhancers, which are transcription regulatory elements located at long distances from the genes. Recent studies have reported that noncoding RNAs are transcribed from active enhancers by RNA polymerase II (RNA Pol II); these are called enhancer RNAs (eRNAs). eRNAs are transcribed bi-directionally from the enhancer core, and are capped on the 5’ end but not spliced or polyadenylated on the 3’ end. The transcription of eRNAs requires the binding of transcription activators on the enhancer and associates positively with the transcription of the target gene. The transcriptional inhibition of eRNAs or the removal of eRNA transcripts results in the transcriptional repression of the coding gene. The transcriptional procedure of eRNAs causes enhancer- specific histone modifications, such as histone H3K4me1/2. eRNA transcripts directly interact with Mediator and Rad21, a cohesin subunit, generating a chromatin loop structure between the enhancer and the promoter of the target gene. The recruitment of RNA Pol II into the promoter and its elongation through the coding region are facilitated by eRNAs. Here, we will review the features of eRNAs, and discuss the mechanism of eRNA transcription and the roles of eRNAs in the transcriptional activation of target genes.

Development of TaqMan probe-based real-time PCR for rapid identification of beef, pork and poultry meat (소, 돼지, 가금육류의 신속한 동정을 위한 TaqMan probe를 이용한 real-time PCR 개발)

  • Species-specific $TaqMan^{(R)}$ probe-based real-time PCR assays were developed for detection of beef, pork, chicken, duck, goose and turkey. The primer and probe sets used in this study were designed to be complementary to fibroblast growth factor (FGF) for cattle and pig, mitochondrial NADH dehydrogenase (ND) subunit 3 and ND2 for chicken and duck, 12S rRNA for goose and turkey, respectively. As internal positive control we used conserved region in the ribosomal 18S RNA gene to ensure the accuracy of the detection of target DNA by real-time PCR. We confirmed that real-time PCR assays with the primer and probe sets were positive for cattle, pig and chicken intended target animal species with no cross-reactivity with other non-target animal species. Only >50 ng DNA of beef show cross-reactivity in the determination of duck. Using species-specific primer and probe sets, it was possible to detect amounts of 0.1 ng DNA of cattle and pig, 1.0 pg DNA of chicken, duck and turkey, and 0.1 pg DNA of goose for raw samples, respectively. The detection limits were 0.1 ng DNA of cattle, 1.0 ng DNA of pig and 1.0 pg DNA of chicken for DNA mixtures (beef, pork and chicken) extracted from heat-treated ($121^{\circ}C$/5 min) meat samples. In conclusion, it can be suggested that the $TaqMan^{(R)}$ probe-based assay developed in this study might be a rapid and specific method for the identification of meat species in raw or cooked meat products.

Fixed Pattern Noise Reduction in Infrared Videos Based on Joint Correction of Gain and Offset (적외선 비디오에서 Gain과 Offset 결합 보정을 통한 고정패턴잡음 제거기법)

  • Most recent infrared (IR) sensors have a focal-plane array (FPA) structure. Spatial non-uniformity of a FPA structure, however, introduces unwanted fixed pattern noise (FPN) to images. This non-uniformity correction (NUC) of a FPA can be categorized into target-based and scene-based approaches. In a target-based approach, FPN can be separated by using a uniform target such as a black body. Since the detector response randomly drifts along the time axis, however, several scene-based algorithms on the basis of a video sequence have been proposed. Among those algorithms, the state-of-the-art one based on Kalman filter uses one-directional warping for motion compensation and only compensates for offset non-uniformity of IR camera detectors. The system model using one-directional warping cannot correct the boundary region where a new scene is being introduced in the next video frame. Furthermore, offset-only correction approaches may not completely remove the FPN in images if it is considerably affected by gain non-uniformity. Therefore, for FPN reduction in IR videos, we propose a joint correction algorithm of gain and offset based on bi-directional warping. Experiment results using simulated and real IR videos show that the proposed scheme can provide better performance compared with the state-of-the art in FPN reduction.
