Effect of GC Content on Target Hook Required for Gene Isolation by Transformation-Associated Recombination Cloning (Transformation-associated recombination cloning에 의한 유전자 분리에 사용되는 target hook에 대한 GC content의 영향)

  • Transformation-associated recombination (TAR) cloning is based on co-penetration into yeast spheroplasts of genomic DNA along with TAR vector DNA that contains 5'- and 3'-sequences (hooks) specific for a gene of interest, followed by recombination between the vector and the human genomic DNA to establish a circular YAC. Typically, the frequency of recombinant insert capture is 0.01-1% for single-copy genes by TAR cloning. To further refine the TAR cloning technology, we determined the effect of GC content on target hooks required for gene isolation utilizing the $Tg\cdot\AC$ mouse transgene as the targeted region. For this purpose, a set of vectors containing a B1 repeated hook and Tg AC-specific hooks of variable GC content (from 18 to 45%) was constructed and checked for efficiency of transgene isolation by radial TAR cloning. Efficiency of cloning decreased approximately 2-fold when the TAR vector contained a hook with a GC content ~${\leq}23$% versus ~40%. Thus, the optimal GC content of hook sequences required for gene isolation by TAR is approximately 40%. We also analyzed how the distribution of high GC content (65%) within the hook affects gene capture, but no dramatic differences for gene capturing were observed.

  • The purpose of this paper is to design high-order (or radial) surface gradient coil (SGC), which can provide multi-dimensional spatial selection. Although the spatial Selection with High-Order gradienT (SHOT) can provide a 2-D selection with only one selective RF pulse, the high-order gradient pro- duced by conventional cylindrical-shape coils has not been clinically useful due to the large selection size caused by the limited radial gradient intensity. However, by using the proposed high-order SGCs located near the imaging region, the size of volume selection can be reduced to a clinically useflll size of 1-2 cm in diameter by applying stronger radial gradient field with much less gradient driving power. So far radial SGCs have been designed by using the field component method and may cause distortion in the selection shapes. In this paper, by using the target field approach for the coil design, selected volumes became almost circular. A 40 cm-by-40 cm $z^2$_surface gradient coil has been designed and implemented by using the target field approach. Phantom and volunteer studies have been performed Experimental results using spatially localized MRI show good agreement to the theoretically predicted behavior.

A Study on Prospective Plan Comparison using DVH-index in Tomotherapy Planning (토모 테라피 치료 시 선량 체적 히스토그램 표지자를 이용한 치료계획 비교에 관한 연구)

  • Purpose: We proposed the method using dose-volume Histogram index to compare prospective plan trials in tomotherapy planning optimization. Materials and Methods: For 3 patients in cranial region, thorax and abdominal region, we acquired computed tomography images with PQ 5000 in each case. Then we delineated target structure and normal organ contour with pinnacle Ver 7.6c, after transferred each data to tomotherapy planning system (hi-art system Ver 2.0), we optimized 3 plan trials in each case that used differ from beam width, pitch, importance. We analyzed 3 plan trials in each region with isodose distribution, dose-volume histogram and dose statistics. Also we verified 3 plan trials with specialized DVH-indexes that is dose homogeneity index in target organ, conformity index around target structure and dose gradient index in non-target structures. Results: We compared with the similarity of results that the one is decide the best plan trial using isodose distribution, dose volume histogram and dose statistics, and the another is using DVH-indexes. They all decided the same plan trial to better result in each case. Conclusion: In some of case, it was appeared a little difference of results that used to DVH-index for comparison of plan trial in tomotherapy by special goal in it. But because DVH-index represented both dose distribution in target structure and high dose risk about normal tissue, it will be reasonable method for comparison of many plan trials before the tomotherapy treatments.

A Study on the Flounder Consumer Market in the US (미국의 넙치 소비 시장에 관한 연구)

  • Flounder was selected as one of the 10 strategic export aquaculture products for seafood export expansion in 2013. The flounder aquaculture industry has promoted export market diversification and product diversification from live to processed goods as a it's main strategy. The purpose of this study is to find an improvement plan for export expansion to the United States, as it emerged as a new target export market for the flounder. A summary of the key findings is as follows. First, the western region of the United States prefers to consume live and fresh flounder, whereas the eastern region prefers to consume fresh flounder. Second, because of it's high quality, Korean flounder is favored in the western region of the United States despite it's high price, whereas in the eastern region of the United States, where production volume is high, Korean flounder has to compete with US flounders because of it's high price. Third, according to the survey results, US consumers tend to enjoy seafood, as well as flounder cuisines. Fourth, the main consumption place of flounders by US consumers are restaurants, and they prefer to consume them in the form of sashimi and sushi. Fifth, 70% of US consumers expressed willingness to consume flounder when eating out. which shows great market potential. However, the high price of Korean flounder and limited size of the live fish market act as major obstacles to expanding export volume. To expand exporting Korean flounder, continuos efforts such as price reduction, exporting highly fresh fish, the co-development of processed food with the US are needed.

A Hydroacoustic Survey Analyzing Fish Populations and Their Distribution Upstream and Downstream of Changzhou Dam, China, Based on Spillway Conditions (중국 Changzhou댐의 방수로 운용에 따른 어류밀도와 분포에 관한 음향조사)

  • Hydroacoustic surveys were performed to clarify the density and spatial distribution of fish upstream and downstream of Changzhou Dam in the Pearl River, China, with regard to the condition of the spillways. Fish were densely distributed in the forebay of the upstream dam and the average fish density was 0.22 individuals/$m^3$ under open spillways on 24 June 2010, but when the spillways were closed on 25 June 2010, the fish in the upstream dam dispersed and the average fish density decreased to 0.007 individuals/$m^3$. Meanwhile, the average fish density in the downstream region was 0.28 individuals/$m^3$ before the spillway was opened on 24 May 2010, but it decreased to 0.08 individuals/$m^3$ on 26 June, just after the spillway was closed. The vertical distribution of fish upstream of the dam was not consistent. The target strength (TS) of fish upstream of the dam was larger than that of fish in the downstream region, although the distribution of TS was similar between the upstream and downstream regions. Therefore, we concluded that while numerous fish could swim to the upstream region while the spillways were open, closed spillways obstructed fish migrating upward from the downstream region.

Active Object Tracking based on stepwise application of Region and Color Information (지역정보와 색 정보의 단계적 적용에 의한 능동 객체 추적)

  • An active object tracking algorithm using Pan and Tilt camera based in the stepwise application of region and color information from realtime image sequences is proposed. To reduce environment noises in input sequences, Gaussian filtering is performed first. An image is divided into background and objects by using the adaptive Gaussian mixture model. Once the target object is detected, an initial search window close to an object region is set up and color information is extracted from the region. We track moving objects in realtime by using the CAMShift algorithm which enables to trace objects in active camera with the color information. The proper tracking is accomplished by controlling the amount of pan and tilt to be placed the center position of object into the middle of field of view. The experimental results show that the proposed method is more effective than the hand-operated window method.

Analysis of a Simulated Optical GSO Survey Observation for the Effective Maintenance of the Catalogued Satellites and the Orbit Determination Strategy

  • A strategy is needed for a regional survey of geosynchronous orbits (GSOs) to monitor known space objects and detect uncataloged space objects. On the basis of the Inter-Agency Debris Committee's recommendation regarding the protected region of geosynchronous Earth orbit (GEO), target satellites with perigee and apogee of $GEO{\pm}200km$ and various inclinations are selected for analysis. The status of the GSO region was analyzed using the satellite distribution based on the orbital characteristics in publicly available two-line element data. Natural perturbation effects cause inactive satellites to drift to two stable longitudinal points. Active satellites usually maintain the designed positions as a result of regular or irregular maneuver operations against their natural drift. To analyze the detection rate of a single optical telescope, 152 out of 412 active satellites and 135 out of 288 inactive satellites in the GSO region were selected on the basis of their visibility at the observation site in Daejeon, Korea. By using various vertical view ranges and various numbers of observations of the GSO region, the detection efficiencies were analyzed for a single night, and the numbers of follow-up observations were determined. The orbital estimation accuracies were also checked using the arc length and number of observed data points to maintain the GSO satellite catalog.

A Study on the Development of a Robot Vision Control Scheme Based on the Newton-Raphson Method for the Uncertainty of Circumstance (불확실한 환경에서 N-R방법을 이용한 로봇 비젼 제어기법 개발에 대한 연구)

  • This study aims to develop a robot vision control scheme using the Newton-Raphson (N-R) method for the uncertainty of circumstance caused by the appearance of obstacles during robot movement. The vision system model used for this study involves six camera parameters (C1-C6). First, the estimation scheme for the six camera parameters is developed. Then, based on the six estimated parameters for three of the cameras, a scheme for the robot's joint angles is developed for the placement of a slender bar. For the placement of a slender bar for the uncertainty of circumstances, in particular, the discontinuous robot trajectory caused by obstacles is divided into three obstacle regions: the beginning region, middle region, and near-target region. Then, the effects of obstacles while using the proposed robot vision control scheme are investigated in each obstacle region by performing experiments with the placement of the slender bar.

  • A weapon control algorithm equipped on a fighter is closely related to the mission accomplishment and fighter survivability during the engagement. In the case of a air-to-surface missile, the weapon control algorithm typically provides a pilot the target shoot-down possible region known as launch acceptability region(LAR) in the multi function display(MFD). LAR is produced by the range table(RT) through computation of an engagement range. In this paper, the operation system of AGM-84 and AGM-88 air-to-surface missiles is introduced. And the engagement range computation and LAR algorithm based on the real-time pseudo 6-DOF simulation are proposed. In order to verify the performance of the algorithm, numerical engagement simulations of air-to-surface missiles to produce LAR have been done.

  • WNT11 is a member of the non-canonical Wnt family and plays a crucial role in tumor progression. However, the regulatory mechanisms underlying WNT11 expression are unclear. Tumor necrosis factor-alpha (TNFα) is a major inflammatory cytokine produced in the tumor microenvironment and contributes to processes associated with tumor progression, such as tumor invasion and metastasis. By using site-directed mutagenesis and introducing a serial deletion in the 5'-regulatory region of WNT11, we observed that TNFα activates the early growth response 1 (EGR1)-binding sequence (EBS) in the proximal region of WNT11 and that the transcription factor EGR1 is necessary for the TNFα-induced transcription of WNT11. EGR1 bound directly to the EBSs within the proximal 5'-regulatory region of WNT11 and ectopic expression of EGR1 stimulated WNT11 promoter activity, whereas the knockdown of EGR1 expression by RNA interference reduced TNFα-induced WNT11 expression in T47D breast cancer cells. We also observed that mitogen-activated protein kinases (MAPK), extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 kinase mediated TNFα-induced transcription of WNT11 via EGR1. Our results suggest that EGR1 directly targets WNT11 in response to TNFα stimulation in breast cancer cells.
