Focused electrospray (FES) deposition method is presented for matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. FES ion optics consists of two cylindrical focusing electrodes capped with a truncated conical electrode through which an electrospray emitter passes along the cylindrical axis. A spray of charged droplets is focused onto a sample well on a MALDI target plate under atmospheric pressure. The shape and size distributions of matrix crystals are visualized by scanning electron microscope and the mass spectra are obtained by time-of-flight mass spectrometry. Angiotensin II, bradykinin, and substance P are used as test samples, while $\alpha$-cyano-4-hydroxycinnamic acid and dihydroxybenzoic acid are employed as matrices. FES of a sample/matrix mixture produces fine crystal grains on a 1-3 mm spot and reproducibly yields the mass spectra with little shot-to-shot and spot-to-spot variations. Although FES greatly stabilizes the signals, the space charge due to matrix ions limits the detection sensitivity of peptides. To avoid the space charge problem, we adopted a dual FES/FES mode, which separately deposits matrix and sample by FES in sequence. The dual FES/FES mode reaches the detection sensitivity of 0.88 amol, enabling ultrasensitive of peptides by homogeneously depositing matrix and sample under atmospheric pressure.

The pixelated phase grating has been studied as a kind of diffraction gratings splitting and input beam into multiple spots. It consists of regular size cells which produce phase delays, and each cell provokes the phase delay up to sixteen levels. We have compared and analyzed the characteristics of multi-level phase gratings, laying streess on efficiency and resulted pattern. Experimental resutls obtained form fabricated grating have been presented, and the real-time method using a liquid-crystal spatial light modulator has been demonstrated through experiments. Gratings making meams with specific intensities have been designed and optical images have been generated by them. The purpose of this study was to investigate the therapeutic effects of DA-9801, an optimized extract of Dioscorea species, on diabetic peripheral neuropathy in a type 2 diabetic animal model. In this study, db/db mice were treated with DA-9801 (30 and 100 mg/kg, daily, p.o.) for 12 weeks. DA-9801 reduced the blood glucose levels and increased the withdrawal latencies in hot plate tests. Moreover, it prevented nerve damage based on increased nerve conduction velocity and ultrastructural changes. Decrease of nerve growth factor (NGF) may have a detrimental effect on diabetic neuropathy. We previously reported NGF regulatory properties of the Dioscorea genus. In this study, DA-9801 induced NGF production in rat primary astrocytes. In addition, it increased NGF levels in the sciatic nerve and the plasma of type 2 diabetic animals. DA-9801 also increased neurite outgrowth and mRNA expression of Tieg1/Klf10, an NGF target gene, in PC12 cells. These results demonstrated the attenuation of diabetic peripheral neuropathy by oral treatment with DA-9801 via NGF regulation. DA-9801 is currently being evaluated in a phase II clinical study.

Background: Worldwide, breast cancer is the most common cancer diagnosed among women and a leading cause of cancer deaths. The age of onset in Iran has become reduced by a decade for unknown reasons. Herceptin, a humanized monoclonal antibody, is a target therapy for breast cancer cells with over expression of HER2-neu receptors, but it is an expensive drug with only 20% beneficial rate of survival. This study introduces a novel approach to enhance the efficacy of this drug through immunoconjugation of the antibody to botulinum toxin. Decreasing the cost and adverse effects of the antibody were secondary goals of this study. Materials and Methods: Botulinum toxin was conjugated with Herceptin using heterobifunctional cross linkers, succinimidyl acetylthiopropionate (SATP) and sulfo-succinimidyl-4-(N-maleimidomethyl) cyclohexane-1-carboxylate (SMCC) according to the supplier's guidelines and tested on two breast cancer cell lines: SK-BR-3 and BT-474. Toxin and Herceptin were also used separately as controls. The cytotoxicity assay was also performed using the new bioconjugate on cultured cells with Alamar blue and a fluorescence plate reader. Results: Herceptin-Toxin bioconjugation significantly improved Herceptin efficacy on both breast cancer cell lines when compared to the control group. Conclusions: Toxin-Herceptin bioconjugation can be a potential candidate with increased efficiency for treating breast cancer patients with over expression of the HER2 receptor.

In this paper, a Kriging metamodel-based multi-objective optimization strategy in conjunction with an NSGA-II(non-dominated sorted genetic algorithm-II) has been employed to optimize the valve-plate shape of the axial piston pump utilizing 3D CFD simulations. The optimization process for minimum pressure ripple and maximum pump efficiency is composed of two steps; (1) CFD simulation of the piston pump operation with various combination of six parameters selected based on the optimization principle, and (2) applying a multi-objective optimization approach based on the NSGA-II using the CFD data set to evaluate the Pareto front. Our exploration shows that we can choose an optimal trade-off solution combination to reach a target efficiency of the axial piston pump with minimum pressure ripple.
This study investigates the feasibility of using the results of the UPV (ultrasonic pulse velocity) test to assess the UCS (unconfined compressive strength) of unsaturated soil. A series of laboratory tests was conducted on samples of unsaturated lateritic soils of northern Taiwan. Specifically, the unconfined compressive test was combined with the pressure plate test to obtain the unconfined compressive strength and its matric suction (s) of the samples. Soil samples were first compacted at the designated water content and subsequently subjected to the wetting process for saturation and the following drying process to its target suction using the apparatus developed by the authors. The correlations among the UCS, s and UPV were studied. The test results show that both the UCS and UPV significantly increased with the matric suction regardless of the initial compaction condition, but neither the UCS nor UPV obviously varied when the matric suction was less than the air-entry value. In addition, the UCS approximately linearly increased with increasing UPV. According to the investigation of the test results, simplified methods to estimate the UCS using the UPV or matric suction were established. Furthermore, an empirical formula of the matric suction calculated from the UPV was proposed. From the comparison between the predicted values and the test results, the MAPE values of UCS were 4.52-9.98% and were less than 10%, and the MAPE value of matric suction was 17.3% and in the range of 10-20%. Thus, the established formulas have good forecasting accuracy and may be applied to the stability analysis of the unsaturated soil slope. However, further study is warranted for validation.
In order to create a worker-friendly environment for institutional foodservice, facilities operating with a dry kitchen system have been recommended. This study was designed to compare the work safety and work environment of foodservice between wet and dry kitchen systems. Data were obtained using questionnaires with a target group of 303 staff at 57 foodservice operations. Dry kitchen facilities were constructed after 2006, which had a higher construction cost and more finishing floors with anti-slip tiles, and in which employees more wore non-slip footwear than wet kitchen (76.7%). The kitchen temperature and muscular pain were the most frequently reported employees' discomfort factors in the two systems, and, in the wet kitchen, "noise of kitchen" was also frequently reported as a discomfort. Dietitian and employees rated the less slippery and slip related incidents in dry kitchens than those of wet kitchen. Fryer area, ware-washing area, and plate waste table were the slippery areas and the causes were different between the functional areas. The risk for current leakage was rated significantly higher in wet kitchens by dietitians. In addition, the ware-washing area was found to be where employees felt the highest risk of electrical shock. Muscular pain (72.2%), arthritis (39.1%), hard-of-hearing (46.6%) and psychological stress (47.0%) were experienced by employees more than once a month, particularly in the wet kitchen. In conclusion, the dry kitchen system was found to be more efficient for food and work safety because of its superior design and well managed practices.

In the case of power plant facilities, seismic stability of non-structural elements is important. In particular, Cabinet structures to which electrical equipment is attached, should have functional safety against earthquakes. Therefore, in this study the dynamic characteristics of non-structural elements inside existing power plant buildings were identified and the response level generated during dynamic behavior was analyzed. In addition, The steel plate dampers were adopted and adjusted to suit the size of the target equipment. In order to derive the optimal seismic reinforcement, the variables according to the attachment location were set and the responses were analyzed by the seismic movement input before and after reinforcement.
Bacterial endospores, especially those of Bacillus and Clostridium genera, are the target of sterilization in various foods. We used Bacillus subtilis ATCC 6633 spores to screen novel antimicrobial substances against spores from medicinal plants. We collected 79 types of plant samples, comprising 42 types of herbs and spices and 37 types of medicinal plants used in traditional medicine in Korea and China. At a concentration of 1%(w/v), only 14 of the ethanol extracts exhibited antimicrobial activity against B. subtilis spores of at least 90%. Crude extracts of Torilis japonica, Gardenia jasminoides, Plantago asiatica, Fritllaria, and Arctium lappa showed particularly high sporicidal activities, reducing the spore count by about 99%. Consideration of several factors, including antimicrobial activity, extraction yields, and costs of raw materials, resulted in the selection of T. japonica, G. jasminoides, A. lappa, and Coriandrum sativum for the final screening of novel antimicrobial substances. Verification tests repeated 10 times over a 4-month period showed that the ethanol extract of T. japonica fruit reduced aerobic plate counts of B. subtilis spores the most, from $10^7$ to $10^4\;CFU/mL$ (99.9%) and with a standard deviation of 0.21%, indicating that this fruit is the most suitable for developing a novel antimicrobial substance for inactivating B. subtilis spores.

Non-contacting capacitive sensors, based on principle of the cross capacitor, for measuring small displacement less than $1.95{\pm}0.5\;mm$ have been fabricated and characterized. To overcome disadvantages of the existed capacitive sensors with 2-electrodes and 3-electrodes, the new sensor is consisted of 4-electrodes which are formed two electrode(high, low) and 2 guard electrodes on a sapphire plate with diameter 17 mm and thickness 0.7 mm, and are symmetrically situated with a constant gap of 0.2 mm between the electrodes. This sensor can be used for measuring both metallic and non-metallic target without ground connection, and is evaluated to the correlation coefficient of 0.9987 for the range of $1.95{\pm}0.5\;mm$ and that of 0.9995 for $1.95{\pm}0.25\;mm$ range.
