We observed the frequency of chromosome aberrations induced by UVB irradiations, and the suppressing effect of tannic acid on chromosome aberrations induced by UVB irradiations in CHL cells, which is a phenolic compound, a hydrolysate of tannin and a components of green tea. UVB doses used for the frequency of chromosome aberrations were from 0.2 to 1.6 KJ/m$^2$ and tannic acid concentrations were from 1.16 $\mu$g/ml to 37.50 $\mu$g/ml. For the observation of suppressing effect of tannic acid on UVB-induced chromosome aberrations, UVB dose was 1.6 KJ/m$^2$ and tannic acid concentrations were 1.0, 2.0, 4.0 $\mu$g/ml. In our study, tannic acid was treated for 24 hours in CHL, cells after UVB irradiation without S9 mix or for 6 hours with S9 mix. From this study, we obtained the following results : (1) The frequency of chromosome aberrations UVB induced were dose-dependently increased. (2) The tannic acid did not induce chromosome aberrations in cultured Chinese hamster cells. (3) UVB-induced chromosome aberrations were suppressed by tannic acid at every concentration from 1.0 $\mu$g/ml to 4.0 $\mu$g/ml with or without metabolic activation. These results suggest that the tannic acid acts as an inhibitor to UVB-induced clastogenicity of the cultured cell.

Using mouse tissue, we studied electron opacity effect of tannic acid for transmission electron microscopic staining. Tannic acid-glutaraldehyde in 0.1M phosphate buffer was used as a fixative. To compare with this we have tested another method consisting of heavy metal staining after treatment of tannic acid in sodium tetraborate (borax) on glutaraldehyde-fixed sections. We have achieved equally consistent electron opacity in both methods. The elastin, collagen, basal lamina of skin and gap junctions of the epithelial cells gave excellent results, while it was good for glycogen, cilia, and plasma. Also fat cells and lipid droplets gave good preservation when tannic acid was added in the fixative. However, prolonged fixation in tannic acid-added fixative was hazardous for further processing, i.e., sectioning problem and deep electron opacity background.

Inhibitory effects of tannic acid on skin toxicity and heat shock protein induced by UVB were investigated. Tannic acid was administered either topically or orally for 3 days to hairless mice, which were previously irradiated with UVB. UVB was found to cause skin erythema . However, the skin erythema was decreased when tannic acid was administered either topically or orally. The heat shock proteins, Hsp-78 kDa and 70 kDa, were induced by UVB irradiation, but the induction was decreased by treatment of tannic acid in both topically and orally administered groups. The hsp induction was more prominent in orally administered groups than in topically administerd groups. However, the difference between two groups was not statistically significant. The route of administrations, topical and oral, does not affect the activity of tannic acid. In the skin tissue observation, tannic acid regenerated the epithelial cells with 7-9 cell layers which were injured by UVB. In conclusion, tannic acid has an ability to protect against UVB irradiation and regenerate the skin.
Inhibitory effects of tannic acid on the lipid peroxidation induced by UVB were investigated. Tannic acid was administered either topically or orally for 3 days to hairless mice, which were previously irradiated with UVB, and inhibitory effects of tannic acid were measured. The UVB was found to cause skin erythema and hemolysis. When tannic acid was administered either topically or orally, hemolysis was decreased. After the skin was irradiated by UVB, the production of malondialdehyde was significantly decreased in erythrocyte and skin tissue, and the activities of SOD and catalase were significantly increased in plasma and skin tissue. In case of oral treatment, catalase activity was not significantly increased. The inhibitory effects of tannic acid on malondialdehyde production, SOD inhibition and catalase inhibition were more prominent in orally administerd groups than in topically administerri groups. However, the difference between two groups was not statistically significant. In conclusion, tannic acid decreased lipid peroxidation possibly by free radical scavenger action. The route of administrations, topical or oral, did not affect the antioxidative activity of tannic acid.
The present study was undertaken to investigate depressor action of tannic acid and the mechanism underlies it in the rabbit. The changes in arterial blood pressure were studied after intravenous administration of tannic acid in normal rabbits and the animals pretreated with atropine, propranolol, dibenamine, and hexamethonium. The results obtained were as follows; 1) Following administration of 1.5 mg/kg, 3.0 mg/kg, and 5.0 mg/kg of tannic acid into rabbits the maximum depressor responses observed were $12.0{\pm}0.9\;mmHg$, $23.4{\pm}1.0\;mmHg$, and $34.0{\pm}1.8\;mmHg$ respectively and generally depressor responses increased in proportion to dosage of tannic acid. 2) Since there were no changes in depressor responses to tannic acid in animals pretreated. separately with atropine, propranolol, dibenamine, and hexamethonium, the depressor responses appeared to be resulting from direct vasodilator action of tannic acid on the vascular smooth muscle. Comparing tannic acid and acorn extract in their mechanisms of depressor responses, it is strongly indicated that in acorn there might exist another depressor substance. 3) After administration of large doses of tannic acid, in addition to respiratory changes, the mean arterial blood pressure decreased markedly and was never recovered throughout the experiment. comma Therefore it is also suggested that the lethal action of tannic acid resides in a drastic decline of arterial blood pressure and in respiratory changes as well.
The effect of tannic acid on GAs and cyclic-AMP promoted amylase induction in barley aleurone layers was examined. Of a variety of adenine compounds, only cyclic-AMP and ADP showed significant activity, and these activities were promoted by addition of theophylline to the incubation medium. When aleurone layers of barley endosperm tissues were incubated with GAs in the presence of tannic acid, the amylase activity in the incubation medium was reduced. Cyclic-AMP induced amylase activity was also reduced by additiion of tannic acid. The cyclic-AMP response promoted was more sensitive to tannin inhibition than GAs response. The inhibitory effect of tannic acid shwoed reversibility by addition of higher concentration of GAs or cyclic-AMP. The tannic acid effect on GAs response was also recovered by addition of a higher concentration of cyclic-AMP. Experiment with polyacrylamide disc electrophoresis showed different isozyme patterns according to the additions in the incubation medium. Inhibitory effects of decursinol and coumarin was compared with that of tannic acid. They showed different zymogram patterns.
The tannins represent a highly heterogeneous group of water-soluble plant polyphenols that may play an important role in antimutagenic and antioxidant properties. We investigated the antioxidant function of tannic acid in comparison to other phenolic compounds including catechin, chlorogenic acid, cinnamic acid, ellagic acid, and gallic acid for their ability to scavenge several stable radicals and reactive oxygen species (ROS) such as ${\bullet}DPPH^+$, ${\bullet}ABTS^+$, hydrogen peroxide, hydroxyl radical, and superoxide radical. The ability of tannic acid to decrease paraquat-induced lipid oxidation in mouse liver and lung through its antioxidant properties was also assessed. The results showed that almost all the tested compounds have stable radical scavenging activity except cinnamic acid. Tannic acid, gallic acid, and ellagic acid demonstrated remarkable ROS scavenging properties toward $H_2O_2$, ${\bullet}OH^-$, ${\bullet}O_2^-$ and especially only tannic acid could inhibit paraquat-induced lipid peroxidation effectively in mouse liver and lung. Based on these results, it appears that increased number of galloyl and ortho-hydroxyl groups enhances the antioxidant activity of phenolic compounds and tannic acid is evaluated as the most effective antioxidant among all the tested compounds. These results suggest that the tannins, especially tannic acid, can be used as therapeutic agent for various diseases caused by ROS.
Weighting of silk fabrics have long been practiced in silk fabric trading based on the primary consideration of price-weight, and secondary one of quality improvement in handle, luster, and drape properties. Recent trend of practicing weighting of silk fabrics is, however, focused on the improvement of the handle, luster, drape, and other properties. During the finishing processes of synthetic fiber, nylon, comprising amide structure, include the use of tannic acid, especially in the dyeing. A multitude of studies are being implemented in terms of improving fastness to washing, fastness to light of dyed nylon product, or the light fastness of nylon 6 itself. In this study, the effects of various tannic acid treatments on the physical properties related to the handle of nylon 6 and silk fabrics are examined and reviewed. The effects of treatment condition of the tannic acid, e.g., the concentration of the aqueous tannic acid solution, treatment time, and temperature were investigated. As the concentration of the aqueous solution of tannic acid increased, the bending rigidity values of the silk and nylon 6 fabrics increased. The treated fabrics felt stiff to the touch. Within the mild conditions of bending employed in the bending measurement of KES, nylon 6 treated fabric specimen exhibited a trend of improvement of bending resiliency within the range of small bending deformation. The weight of treated fabrics have all increased. The air-permeability values decreased as the treatment concentration increased. However, the decrease tendency of air permeability values may be alleviated by adjusting the fabric count during the tentering or expanding processes, either by tension adjustment or heat treatment. Optimum conditions of the treatment for nylon 6 are 1.25% tannic acid concentration, bath temperature of $85^{\circ}C$, pH 3.1, and those for silk fabric treatment are 1.25% tannic acid concentration, bath temperature of $85^{\circ}C$, pH 3.1. The treatment conditions will lead to the improvement in the properties of fabrics for summer.
