Effects of Takju intake and moderate exercise training on brain acetylcholinesterase activity and learning ability in rats

  • Takju is a Korean alcoholic beverage made from rice, and is brewed with the yeast Saccharomyces cerevisiae. This study was conducted to evaluate the effects of exercise training and moderate Takju consumption on learning ability in 6-week old Sprague-Dawley male rats. The rats were treated with exercise and alcohol for 4 weeks in six separate groups as follows: non-exercised control (CC), exercised control (EC), non-exercised consuming ethanol (CA), exercised consuming ethanol (EA), non-exercised consuming Takju (CT), and exercised consuming Takju (ET). An AIN-93M diet was provided ad libitum. Exercise training was performed at a speed of 10 m/min for 15 minutes per day. Ethanol and Takju were administered daily for 6-7 hours to achieve an intake of about 10 ml after 12 hours of deprivation, and, thereafter, the animals were allowed free access to deionized water. A Y-shaped water maze was used from the third week to understand the effects of exercise and alcohol consumption on learning and memory. After sacrifice, brain acetylcholinesterase (AChE) activity was analyzed. Total caloric intake and body weight changes during the experiment were not significantly different among the groups. AChE activity was not significantly different among the groups. The number of errors for position reversal training in the maze was significantly smaller in the EA group than that in the CA and ET groups, and latency times were shorter in the EA group than those in the CC, EC, CT, and ET groups. The latency difference from the first to the fifth day was shortest in the ET group. The exercised groups showed more errors and latency than those of the non-exercised groups on the first day, but the data became equivalent from the second day. The results indicate that moderate exercise can increase memory and learning and that the combination of exercise and Takju ingestion may enhance learning ability.

  • This study was conducted to investigate the effects of Takju lees hot water extracts on the blood pressure in spontaneously hypertension rats (SHR). Twenty eight male SHR were grouped by their blood pressure and fed a control diet or experimental diets containing 1% (G1), 2% (G2), or 4% (G4) Takju lees extracts for 4 weeks. Food intake was not significantly different among the groups. However, body weight gain was significantly lower in groups fed the Takju lees extract than the control group. The systolic blood pressure was significantly lower in the Takju lees extract containing groups (especially in G4 group) than the control groups. In addition, mean blood pressure {(systolic+dyastolic)/2} decreased with an increase in the amount of Takju lees extract in the diet and feeding period. Takju lees extract decreased angiotensin I converting enzyme (ACE) activity in a dose-dependent manner. These results suggest that the Takju lees extract exert an antihypertensive effect by decreasing ACE activity.

  • The purpose of this study was to develop measures for establishing and enforcing legal nutrition labeling for Takju based on results from consumer awareness surveys, statistical model development, and evaluation of nutrients. The statistical model developed with consumer survey results showed that consumers would like to know the nutrients they intake from drinking Takju, as their awareness about Takju was low. Specifically, consumers would like to see information regarding alcohol content, calories, carbohydrates, and saccharides on the label. Structural equations from the research model showed that consumers who had some knowledge of Takju also had positive thoughts of the nutrition fact labels for Takju. Evaluation of nutrients in Takju showed that the starch sources and other ingredients used in Takju fermentation did not influence nutrient facts, and nutrient concentrations also varied among the different Takju. In addition, this research suggests methods for consumers to make reasonable selections and to inform them of the nutrition fact labeling for Takju. Benners and pop-up were manufactured to promote voluntary participation of companies and to provide nutrition facts from Takju. Eventually, a measure was suggested to establish and enforce nutrition labeling, using results from consumer and nutrient surveys of Takju.

탁주 주박의 섭취가 스트렙토조토신으로 당뇨를 유발시킨 흰쥐의 혈당수준에 미치는 효과 (Effects of Takju(Korean turbid rice wine) Lees on the Serum Glucose levels in Streptozotocin-induced Diabetic Rats)

  • This study was performed to determine the effect of Takju(Korean turbid rice wine) lees on the serum glucose level in streptozotocin-induced diabetic rats. 24 Sprague-Dawley male rats were divided into three groups: normal control(NC), diabetic control(DC) and diabetic rats(DS) were fed on experimental diet and water ad libitum for 4 weeks. DS diet was containing 20% Takju lees. Body weight gain and food Efficiency Ratio(FER) were significantly lower in DC and DS than NC. DS tended to have higher weight, weight gain and FEF than DC nevertheless food intake. Therefore Takju lees could possibly complement casein as a protein source. Gastrointestianl transit time in DS significantly decreased than NC while not significantly than DC. Serum lipid profiles and AST. ALT and amylase were not significantly different between diabetic DC and DS. Blood glucose was measured at fasting state and 30, 60, 90 and 120 minute by oral glucose tolerance test, DS tended to lower the mean(${\pm}$ SE) incremental blood glucose concentrations than DC and was significantly low at 120 min. But incremental AUG(area under the curve) of postprandial glucose response was not significantly different. In conclusion, in spite of high contents of carbohydrate Takju lees perhaps have a benefit effect on the diabetes.

  • We investigated the effect of Takju lees extract on blood glucose levels in the db/db mice (a murine model of type 2 diabetes mellitus). We fed 40 male db/db mice a control diet (G0, AIN93G) and experimental diets containing 1% (G1), 2% (G2), or 4% (G4) Takju lees extract for 4 weeks. We found no difference in food intake and body weight gain among the animal groups. In the G1 and G2 groups, plasma glucose levels decreased significantly between Days 10 and 21 compared with the G0 group. However, we found no difference in plasma glucose levels between groups G4 and G0. The change in insulin concentrations was not significant among these animal groups, and we found no significant difference in glucose transporter 4 (GLUT4) expression in the soleus muscle. These results suggest that the Takju lees extract has a beneficial effect in animals with type 2 diabetes.

Exposure Assessment of Ethyl Carbamate in Alcoholic Beverages

  • Ethyl carbamate, a by-product produced naturally during fermentation and contained in fermented foods and beverages, is a carcinogen. Thus, due to the high consumption of alcoholic beverages in Korea, the ethyl carbamate concentrations in popular alcoholic beverages were determined, and the daily intake of ethyl carbamate through alcoholic beverages was estimated. The major Korean alcoholic beverages, Soju, beer, and Takju, with the highest market share were sampled and their ethyl carbamate concentrations determined by GC/MS/SIM. The ranges of ethyl carbamate contained in Soju, beer, and Takju was 0.83-10.07, 0.45-0.77, and 0.40-0.93 ppb, respectively. These results and data on the average daily intake of alcoholic beverages were then used to calculate the average and maximum estimated daily intakes (EDI) of ethyl carbamate through alcoholic beverages. As a result, a relatively high EDI of ethyl carbamate through alcoholic beverages was found for Korean males over 30 years old, indicating the need to reduce the ethyl carbamate content in alcoholic beverages.

  • Levels of ethyl carbamate, by-product produced naturally during fermentation, in Korean alcoholic beverages were determined by Gas Chromatography/mass spectrometry/selected ion mode (GC/MS/SIM), and their daily intake by Korean adult group was estimated. In GC/MS/SIM analysis 0.8-10.1, 0.5-0.8, 0.4-0.9, 3.5-689.9, 8.4-30.3, 13.9-30.0, and 1.7-11.7 ppb of ethyl carbamate were detected in soju, beer, takju, fruit wine, cheongju, whiskey, and grape wine, respectively. Maximum daily exposure of ethyl carbamate through alcoholic beverage consumption was 7.41 ng/kg body weight/day for average Korean male, with one soju brand and two fruit wine brands showing high ethyl carbamate level.

국내 유통 주류 중 중금속 실태조사 (Monitoring of Heavy Metal Content in Alcoholic Beverages)

  • 한국인의 주류 섭취로 인한 중금속 위해 영향 여부를 파악하기 위하여 국내산 및 수입산 주류를 대상으로 납, 카드뮴, 비소 및 수은을 분석하고, 일일식품섭취량을 고려하여 이 들 식품 섭취에 대한 위해 평가를 수행하였다. 시료 분석결과로 발효주류 중 납 함량은(${\mu}g/kg$) 11.1(N.D~66.5), 카드뮴 7.6(N.D~47.1), 비소 24.6(1~140.0), 수은 $0.7\;{\pm}\;1.2$(N.D~10.6), 증류주류 중 납 함량은(${\mu}g/kg$) 9.1(N.D~33.4), 카드뮴 1.8(N.D~19.6), 비소 14.9(N.D~209.7), 수은 0.3(N.D~2.3)이었으며 기타주류 중 납 함량은(${\mu}g/kg$) 14.0(N.D~37.8), 카드뮴 10.3(0.6~17.4), 비소 33.6(0.5~103.3), 수은 0.3(0.1~0.5)이었다. 본 연구에서 얻어진 탁주, 약주, 청주, 맥주, 과실주, 소주, 위스키, 브랜디, 일반증류주, 리큐르, 기타주류 중 중금속 함량은 국내 외 문헌들과 유사하거나 낮게 확인 되었으며, 식품별 1일섭취량을 고려한 납, 카드뮴, 비소 및 수은의 주간섭취량은 FAO/WHO의 PTWI와 비교할 때 각각 0.03%, 0.06%, 0.01%, 0.01%이하로 안전한 수준으로 판단된다.
