TPS-1 배지 및 TYM 배지에서 배양된 질편모충의 생물학적 특성의 변화 (Biological and biochemical modulation of Trichomonas vaginalis KT9 isolate after shifting of culture medium from TPS-1 into TYM)

  • 질편모충의 배양배지인 Diamond의 TPS-1 배지에서 3년간 배양한 질편모충 KT9 국내 분리주를 TYM 배지로 옮겨 1년간 배양하고 양쪽 배지에서 자란 질편모충의 증식, 병원성, 영양형의 크기, 단백질 성상 등을 비교하였다. 각 배지 원충의 generation time은 TYM 배지의 원충은 4.5시간이었고 TPS-1 배지의 원충은 7.1시간으로 TYM 배지에서 더 빨리 자랐다. TYM 배지에서 자란 원충의 크기가 TPS-1 배지에서의 영양형에 비해 유의하게 작았다. TYM 배지에서 배양한 원충은 10마리 마우스 중 9마리에서 피하 농양을 형성한 반면 TPS-1 배지에서 배양한 원충은 2마리에서 피하 농양을 형성하였고, 농양의 크기도 작았다. SDS-PAGE에서 10개의 단백분획이 공통적으로 관찰되었으며 TYM 배지의 원충에서는 136 kDa, 116 Da 및 40 kDa의 분획이, TPS-1 배지의 원충에서는 100 kDa 분획이 특징적으로 관찰되었다. Gelatin SDS-PAGE에서는 200 kDa과 70 kDa 사이에 10개의 gelatin 분획이 공통적으로 관찰되었으며, TYM 배지에서 배양한 원충이 TPS-1 배지에서 배양한 원충에 비해 gelatin을 많이 분해하였다. Bz-Pro-Phe-Arg-Nan를 기질로 사용하여 단백질분해효소활성을 측정하였는데 TYM 배지에서 배양한 원충의 단백질분해효소활성은 TPS-1 배지에서 배양한 원충에 비해 유의하게 높았다. 이 연구결과 질편모충의 배양에서 TPS-1 배지를 TYM 배지로 전환하였을 때 원충의 크기가 커졌고 단백질분해효소 활성이 증가하며 독력 (병원성)이 증가됨이 관찰되었다.

Hydrogenosomal activity of Trichomonas varinalis cultivated under different iron conditions

  • To evaluate whether iron concentration in TYM medium influence on hydrogenosomal enzyme gene expression and hydrogenosomal membrane potential of Trichomonas vaginalis, trophozoites were cultivated in iron-depleted, normal and iron-supplemented TYM media. The mRNA of hydrogenosomal enzymes, such as pyruvate ferredoxin oxidoreductase (PFOR), hydrogenase, ferredoxin and malic enzyme, was increased with iron concentrations in T. vaginalis culture media, measured by RT-PCR. Hydrogenosomal membrane potentials measured with $DiOC_6$ also showed similar tendency, e.g. T. vaginalis cultivated in iron-depleted and iron-supplemented media for 3 days showed a significantly reduced and enhanced hydrogenosomal membrane potential compared with that of normal TYM media, respectively. Therefore, it is suggested that iron may regulate hydrogenosomal activity through hydrogenosomal enzyme expression and hydrogenosomal membrane potential.

Trichomonicidal Activity of Herbal Extracts Used in Traditional Medicine in Korea

  • Trichomonicidal activity of thirty methanolic herbal extracts used in traditional medicine in Korea was evaluated. Trichomonas vaginalis was used as experimental model, and anti-Trichomonas activity was determined over cultures of the parasite in TYM Diamond medium. Six methanolic extracts such as Acanthopanacis Cortex, Agrimoniae Herba, Pulsatillae Radix, Sanguisorbae Radix, Sophorae Radix, and Torilidis Fructus showed more than 50% trichomonicidal activity at the concentration of 200 g/ml. These extracts were further fractionated into n-butanol soluble and aqueous phases. Except for Acanthopanacis Cortex, all of n-butanol soluble phases showed potent trichomonicidal activity, while none of aqueous phases exhibited trichomonicidal activity.

PCR for Diagnosis of Male Trichomonas vaginalis Infection with Chronic Prostatitis and Urethritis

  • The aim of this study was to assess the usefulness of PCR for diagnosis of Trichomonas vaginalis infection among male patients with chronic recurrent prostatitis and urethritis. Between June 2001 and December 2003, a total of 33 patients visited the Department of Urology, Hanyang University Guri Hospital and were examined for T. vaginalis infection by PCR and culture in TYM medium. For the PCR, we used primers based on a repetitive sequence cloned from T. vaginalis (TV-E650). Voided bladder urine (VB1 and VB3) was sampled from 33 men with symptoms of lower urinary tract infection (urethral charge, residual urine sensation, and frequency). Culture failed to detect any T. vaginalis infection whereas PCR identified 7 cases of trichomoniasis (21.2%). Five of the 7 cases had been diagnosed with prostatitis and 2 with urethritis. PCR for the 5 prostatitis cases yielded a positive 330 bp band from bothVB1 and VB3, whereas positive results were only obtained from VB1 for the 2 urethritis patients. We showed that the PCR method could detect T. vaginalis when there was only 1 T. vaginalis cell per PCR mixture. Our results strongly support the usefulness of PCR on urine samples for detecting T. vaginalis in chronic prostatitis and urethritis patients.

Analysis of the Axle Load of a Rice Transplanter According to Gear Selection

  • The objective of this study was to analyze the axle load of a rice transplanter when planting rice seedlings at different working load conditions to select a suitable gear stage and a constant planting depth for rice seedlings. In this study, there are four levels of planting distances (26, 35, 43, and 80 cm) and three planting depths (low, medium, and high) with two gear stages (1.3 and 1.7 m/s). Axle loads and required planting pressures were analyzed statistically. It was observed that axle torques were increased with increasing planting depths for both gear stages, meaning that axle torques were directly proportional to planting depths for both gear stages. It was also observed that required planting pressures had a significant difference between planting distances. Planting pressures also showed significant difference according to gear stage and planting depth. These results indicate that planting pressures were directly proportional to both gear stage and planting depth. Results revealed that the automatic depth control system of a rice transplanter could not guarantee a constant planting depth as supplied pressures were variable. This indicates that a control algorithm is needed to ensure a constant planting depth. In the future, a control algorithm will be developed for an automatic depth control system of a rice transplanter to improve its comprehensive performance and efficiency.
