Trichostatin A (TSA) is a histone deacetylase (HDAC) inhibitor. We here investigated its effects on proliferation and apoptosis of the CNE2 carcinoma cell line, and attempted to establish genome-wide DNA methylation alteration due to differentially histone acetylation status. After cells were treated by TSA, the inhibitory rate of cell proliferation was examined with a CCK8 kit, and cell apoptosis was determined by flow cytometry. Compared to control, TSA inhibited CNE2 cell growth and induced apoptosis. Furthermore, TSA was found to induce genome-wide methylation alteration as assessed by genome-wide methylation array. Overall DNA methylation level of cells treated with TSA was higher than in controls. Function and pathway analysis revealed that many genes with methylation alteration were involved in key biological roles, such as apoptosis and cell proliferation. Three genes (DAP3, HSPB1 and CLDN) were independently confirmed by quantitative real-time PCR. Finally, we conclude that TSA inhibits CNE2 cell growth and induces apoptosis in vitro involving genome-wide DNA methylation alteration, so that it has promising application prospects in treatment of NPC in vivo. Although many unreported hypermethylated/hypomethylated genes should be further analyzed and validated, the pointers to new biomarkers and therapeutic strategies in the treatment of NPC should be stressed.
PES/DMF/TSA/PVP 고분자용액을 물에 침지시 시키는 상전이 공정을 통하여 비대칭성이 향상된 PES 멤브레인을 제조하였다. PES (polyethersulfone) 용액에 응고제 (p-toluenesulfonic acid, TSA)와 습윤제 고분자 (polyvinylpyrrolidone, PVP)의 첨가와 습윤 상태에 노출 시간으로 일어나는 멤브레인 표면의 pre-coagulation 현상은 멤브레인의 구조적 특성과 투과량 특성에 있어 중요한 역할을 한다. PES 용액은 폴리에스터 필름에 코팅된 후 약 $72{\sim}144$초 동안 80%의 습도하에서 공기에 노출된 후 응고조에 침지되었다. Capillary flow porometer, FE-SEM과 수투과 평가장치를 통하여 멤브레인의 특성을 살펴보았다. TSA 20 wt%와 PVP 10 wt%가 첨가된 11 wt%의 PES 용액에서 멤브레인 구조의 비대칭성이 크게 향상되었으며 최소 기공충의 두께도 얇아짐을 볼 수 있었다. 그 결과로 수투과량에 큰 증가를 가져옴을 볼 수 있었다.
The esterification of the reactants of Jatropha oil and methanol added by propyleneglycol was done using p-TSA catalyst. And then the emulsification of triglyceride and methanol was conduced by 1.0vol% GMS. The emulsified reactants were transesterified at $65^{\circ}C$ using TMAH and mixed catalyst (50wt%-TMAH+50wt%-NaOH) respectively. The esterification conversion at the 1:8 molar ratio of free fatty acid to methanol using 8.0wt% p-TSA was 94.7% within 80min. The overall conversion at the 1:8 molar ratio of mixed fat(50wt% Beef Tallow) to methanol and $65^{\circ}C$ using mixed catalyst was 95.4% The cloud point of Biodiesel decreased with the addition of petroleum diesel.
The esterification of free fatty acid in Jatropha oil added by propylene glycol using p-TSA catalyst was done, and then the transesterification of Jatropha oil added by 1.0vol% GMS as an emulsifier using TMAH, and mixed catalyst(60wt%-TMAH+ 40wt%-KOH) respectively was followed at $60^{\circ}C$. The esterification conversion at the 1:8 molar ratio of free fatty acid to methanol using 8.0wt% p-TSA was 94.7% within 60min. The overall conversion at the 1:8 molar ratio of Jatropha oil to methanol and $60^{\circ}C$ using mixed catalyst was 95.4%. The kinematic viscosity of Biodiesel using TMAH and mixed catalyst in 24h met the ASTM D-6751 above $30^{\circ}C$, and showed a little more than its criterion.
Adipic acid(AA)와 diethylene glycol(DEG) 사이에서 일어나는 반응의 속도에 대한 유기나노점토(Cloisite 30B)의 영향을 규명하기 위하여 383K와 423 K에서 Br${\phi}$nsted acid인 p-toluene sulfonic acid(p-TSA)와 Lewis acid인 butylchlorotin dihydroxide를 촉매로 사용하여 에스테르 반응을 수행하였다. 촉매의 양은 전체반응물의 0.14 mol%만큼 사용하였다. 반응 생성물의 산가로부터 계산된 전환율 vs. 시간의 자료를 사용하여 반응속도를 해석하였다. [OH]/[COOH] 초기 몰비가 2인 상태에서 수행되는 polyester-diol 합성반응의 속도는 p-TSA 촉매하에서는 AA에 대하여 2차 반응으로, butylchlorotin dihydroxide 촉매하에서는 AA에 대하여 1차 반응으로 잘 해석되었으며 두 반응의 활성화 에너지는 각각 42.2 kJ/mol, 63.8 kJ/mol이었다. AA와 DEG에 Cloisite 30B를 5 wt% 첨가시키면 p-TSA 촉매는 활성이 크게 줄어 반응속도가 매우 느려졌으며, 이 때 활성화 에너지는 72.9 kJ/mol로 높게 계산되었다. Butylchlorotin dihydroxide 촉매는 Cloisite 30B의 첨가에 관계없이 거의 일정한 활성을 유지하였다. Butylchlorotin dihydroxide 촉매하에서 Cloisite 30B를 첨가한 반응의 활성화 에너지는 61.8 kJ/mol로 Cloisite 30B를 첨가하지 않은 반응의 활성화 에너지보다 약간 낮았다.
New asymmetric dimers, N,N'-dialkyl-4'-hydroxy-4-oxo-2,2',3,3'-tetrahydro-2,2'-diphenyl-4,4'-quinolones 3a-f were synthesized through the dehydration and dea1coholation of N-alkylanilines and ethyl benzoylacetate. Dimers 3a-f were identified by NMR, IR and GC-MS. A series of dimer 3a-f has been synthesized using acid-catalyzed one-pot reaction that involved the condensation, cyc1ization and dimerization. Similarly, the 6,6'-methoxy (or 7,7'-methoxy) substituted dimers were prepared from N-alkyl-meta-(or para)-anisidines. Formation of dimers was undertaken with p-toluenesulfonic acid (p-TSA) at 90∼11$0^{\circ}C$ in toluene for 2∼6 hours over the Dean-Stark apparatus.

Histone acetylation plays a critical role in the regulation of transcription by altering the structure of chromatin, and it may influence the resistance of some tumor cells to tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) by regulating the gene expression of components of the TRAIL signaling pathway. In this study, we investigated the effects and molecular mechanisms of trichostatin A (TSA), a histone deacetylase inhibitor, in sensitizing TRAIL-induced apoptosis in Caki human renal carcinoma cells. Our results indicate that nontoxic concentrations of TSA substantially enhance TRAIL-induced apoptosis compared with treatment with either agent alone. Cotreatment with TSA and TRAIL effectively induced cleavage of Bid and loss of mitochondrial membrane potential (MMP), which was associated with the activation of caspases (-3, -8, and -9) and degradation of poly (ADP-ribose) polymerase (PARP), contributing toward the sensitization to TRAIL. Combined treatment with TSA and TRAIL significantly reduced the levels of the cellular Fas-associated death domain (FADD)-like interleukin-$1{\beta}$-converting enzyme (FLICE) inhibitory protein (c-FLIP), whereas those of death receptor (DR) 4, DR5, and FADD remained unchanged. The synergistic effect of TAS and TRAIL was perfectly attenuated in c-$FLIP_L$-overexpressing Caki cells. Taken together, the present study demonstrates that down-regulation of c-FLIP contributes to TSA-facilitated TRAIL-induced apoptosis, amplifying the death receptor, as well as mitochondria-mediated apoptotic signaling pathways.
KSMBR process is dynamic state advanced wastewater treatment applied with Trisectional Aeration (TSA) mode combined with membrane. TSA was remodeled conventional intermittent aeration which was operated nonaeration-aeration. TSA operates nonaeration ($N_1$) - aeration (A) - nonaeration ($N_2$) in Trisectional Aeration Reactor (TAR). Organics of influent could be nearly consumed to denitrification without influence by remained DO in TAR and it could be operated about sludge return ratio of 1Q (influent base). The purpose of this study was to apply KSMBR to the full-scale plant and to evaluate efficiency of nitrogen and phosphorus removal and TSA operation. The result of this study, average CODcr/T-N and CODcr/T-P ratio were 7.8 and 59.6, respectively. BOD, TCODcr, SS, T-N, T-P, E-coli removal efficiency were 98.4, 95.2, 73.0, 69.6, 99.95 %, respectively. KSMBR obtained high removal efficiencies of C, N and P when it applied full-scale plant.

Background: Physical therapy (PT) plays an important role in the recovery of function following anatomic total shoulder arthroplasty (aTSA). While several PT protocols have been published for these patients, there is no standardized protocol for aTSA rehabilitation. This lack of standardization may lead to confusion between patients and physicians, possibly resulting in suboptimal outcomes. This study examines how PT protocols provided by academic orthopedic surgery programs vary regarding therapeutic goals and activities following aTSA. Methods: PT protocols for aTSA available online from the Accreditation Council for Graduate Medical Education accredited orthopedic surgery programs were included for review. Each protocol was analyzed to evaluate it for differences in recommendation of length of immobilization, range of motion (ROM) goals, start time for and progression of therapeutic exercises, and timing for return to functional activity. Results: Of 175 accredited programs, 25 (14.2%) had protocols publicly available, programs (92%) recommended sling immobilization outside of therapy for an average of 4.4±2.0 weeks. Most protocols gave recommendations on starting active forward flexion (24 protocols, range 1-7 weeks), external rotation (22 protocols, range 1-7 weeks), and internal rotation (18 protocols, range 4-7 weeks). Full passive ROM was recommended at 10.8±5.7 weeks, and active ROM was 13.3±3.9 weeks, on average. ROM goals were inconsistent among protocols, with significant variations in recommended ROM and resistance exercise start times. Only 13 protocols (52%) gave recommendations on resuming recreational activities (mean, 17.4±4.4 weeks). Conclusions: Publicly available PT protocols for aTSA rehabilitation are highly variable. 본 연구는 VOC 제거 기술인 TSA 공정에서 제올라이트 13X와 활성탄이 채워진 두 종류의 탑을 사용하여 원료농도, 질소 유량, 수증기 유량, 탑 온도 등 조업조건의 영향을 분석하였다. 본 연구의 TSA 사이클은 흡착단계, 수증기 탈착단계, 건조 및 냉각단계로 구성되었다. 2% 벤젠 농도에서 제올라이트 13X와 활성탄의 사이클 당 전체 흡착량은 각각 4.44 g과 3.65 g으로 활성탄보다 충전밀도가 큰 제올라이트 13X가 더 많은 양의 벤젠을 흡착할 수 있었다. 수증기 탈착의 결과에서 수증기 유량을 증가시키고 탑의 외부 가열로 온도를 높이면 탈착시간이 짧아지고 배출되는 벤젠의 농도가 높아지는 것으로 나타났다. 2% 벤젠 농도에서 수증기 유량을 75 g/hr로 증가시키면 탈착시간이 1 hr에서 최대 33 min까지 단축되어 상대적으로 건조 및 냉각단계의 시간이 늘어나 수증기 제거와 탑 냉각을 충분히 진행할 수 있었다. 탑 온도를 높이면 탈착량이 증가하나 $150^{\circ}C$ 이상에서는 에너지소비는 증가하는 반면 탈착량은 거의 일정했다. 연속 사이클 조업에서 재생단계 완료 시 잔존하게 되는 벤젠의 비율이 늘어나면 흡착제 working capacity 감소의 원인이 될 수 있다. 제올라이트 13X를 이용해 연속 사이클 공정실험을 수행한 결과 탑 내부에 잔존하는 벤젠의 비율이 4번째 사이클 이후 일정한 값으로 유지되었다.
