Review on the Management for Radioactive Effluent and Methodology for Setting of Derived Release Limits at Pressurized Heavy Water Reactors in Korea (중수로원전 방사성유출물 관리와 유도배출한계 설정방법에 대한 고찰)

  • The radioactive effluents from pressurized heavy water reactors (PHWRs) are relatively larger than those from pressurized water reactors (PWRs). Futhermore, radioactive effluents from PHWRs are released continuously. Thus, the discharge of radioactive effluents is strictly controlled. To do this, radiation detectors are installed at stacks of reactor buildings to monitor the concentration of radioactive effluents in real-time. Derived release limits (DRLs) of annual discharge are also set up for each radionuclide and effluents are rigidly controlled not to exceed those limits. In this paper, the discharge process of radioactive effluents, the standard for establishment of DRL and its methodology, and currents status for PHWRs were reviewed.

Recombinant α and β Subunits of M.AquI Constitute an Active DNA Methyltransferase

  • AquI DNA methyltransferase, M.AquI, catalyses the transfer of a methyl group from S-adenosyl-L-methionine to the C5 position of the outermost deoxycytidine base in the DNA sequence 5'CYCGRG3'. M.AquI is encoded by two overlapping ORFs (termed $\alpha$ and $\beta$) instead of the single ORF that is customary for Class II methyltransferase genes. The structural organization of the M.AquI protein sequence is quite similar to that of other bacterial C5-DNA methyltransferases. Ten conserved motifs are also present in the correct order, but only on two polypeptides. We separately subcloned the genes that encode the $\alpha$ and $\beta$ subunits of M.AquI into expression vectors. The overexpressed His-fusion $\alpha$ and $\beta$ subunits of the enzyme were purified to homogeneity in a single step by Nickel-chelate affinity chromatography. The purified recombinant proteins were assayed for biological activity by an in vitro DNA tritium transfer assay. The $\alpha$ and $\beta$ subunits of M.AquI alone have no DNA methyltransferase activity, but when both subunits are included in the assay, an active enzyme that catalyses the transfer of the methyl group from S-adenosyl-L-methionine to DNA is reconstituted. We also showed that the $\beta$ subunit alone contains all of the information that is required to generate recognition of specific DNA duplexes in the absence of the $\alpha$ subunit.

Ethanol-eluted Extract of Rhus verniciflua Stokes Inhibits Cell Growth and Induces Apoptosis in Human Lymphoma Cells

  • Rhus verniciflua Stokes (RVS) has been used as a traditional herbal medicine. Several earlier studies indicated that an ethanol extract of RVS has both anti-oxidant and anti-tumor properties, although the mechanism for the activity remains to be elucidated. In this report, we prepared a highly purified ethanol extract from RVS, named REEE-1 ($\underline{R}$hus $\underline{e}$thanol $\underline{e}$luted $\underline{e}$xtract-1), and investigated the mechanism involved in its growth-inhibitory effect on the human B and T lymphoma cell lines, BJAB and Jurkat, respectively. Results from tritium uptake proliferation assays showed that the proliferative capacities of both BJAB and Jurkat cells were strongly suppressed in the presence of REEE-1. This was further confirmed through trypan blue exclusion experiments that revealed a dose-dependent decrease in viable cell numbers after REEE-1 treatment. REEE-1-mediated suppression of cell growth was verified to be apoptotic, based on the increase in DNA fragmentation, low fluorescence intensity in nuclei after propidium iodide staining, and the appearance of DNA laddering. In particular, REEE-1 exerted its anti-oxidant activity through the inhibition of hydroxyl radical-mediated degradation by iron ion chelation rather than direct scavenging of hydroxyl radicals. Furthermore, REEE-1 was revealed to be a potential scavenger of superoxide anions. Collectively, our findings suggest that REEE-1 is a natural anti-oxidant that could be used as a cancer chemo-preventive and therapeutic agent.

문경지역 탄산온천수의 지구화학적 및 동위원소적 특성연구

  • The hydrogeochemical and isotopic studies on deep groundwater in the Munkyeong area, Kyeongbuk province were carried out. $CO_2$-rich groundwater (Ca-HC $O_3$ type) is characterized by low pH (5.8~6.5) and high TDS (up to 2,682 mg/L), while alkali groundwater (Na-HC $O_3$ type) shows a high pH (9.I~10.4) and relatively low TBS (72~116 mg/L). $CO_2$-rich water may have evolved by $CO_2$ added at depth during groundwater circulation. This process leads to the dissolution of surrounding rocks and Ca, Na, Mg, K and HC $O_3$ concentrations are enriched. The low Pc $o_2$ (10$^{-6.4}$atm) of alkali groundwaters seems to result from the dissolution of silicate minerals without a supply of $CO_2$. The $\delta$$^{18}$ O and $\delta$D values and tritium data indicate that two types of deep groundwater were both derived from pre-thermonuclear meteoric water. The carbon Isotope data show that dissolved carbon in the $CO_2$-rich water was possibly derived from deep-seated $CO_2$ gas. The $\delta$$^{18}$ S values of dissolved sulfate show that sulfate reduction occurred at great depths. The application of various chemical geothermometers on $CO_2$-rich groundwater shows that the calculated deep reservoir temperature is about 130~175$^{\circ}C$. Based on the geological setting, water chemistry and environmental isotope data, each of the two types of deep groundwater represent distinct hydrologic and hydrogeochemical evolution at depth and their movement is controlled by the local fracture system.m.

  • This study represents the synthetic characteristics of styrene-DVB copolymer which is used as a catalyst support for the removal of tritium in CANDU. To evaluate the effect of solvent on the styrene-DVB polymeric support, the solvating power dependent on various kinds and composition of solvents was calculated. In order to synthesize the macroreticular type polymer bead, the following conditions are needed; solvating power should be higher than 1 for 20% of crosslinkage and higher than 3 for 40% of crosslinkage. Stabilization of organic monomers at about $40^{\circ}C$ for more than 2 hours is prerequisite. Removal of solvent prior to drying is preferred for the post-treatment of the bead. The polymer particle size was increased by lowering concentration of surfactant and stirring speed.

The Role of Adenosine Receptor on Norepinephrine Release from Ischemic-Induced Rat Hippocampus (허혈이 유발된 흰쥐 해마에서 Norepinephrine 유리에 미치는 Adenosine 수용체의 역할)

  • The effects of adenosine analogues on the electrically-evoked norepinephrine(NE) release and the influence of ischemia on the effects were studied in the rat hippocampus. Slices from the rat hippocampus were equilibrated with $0.1{\mu}M$ $[^3H]-norepinephrine$ and the release of the labelled product, $[^3H]-NE$, was evoked by electrical stimulation(3 Hz, 2 ms, 5 $VCm^{-1}$ and rectangular pulses for 90 sec), and the influence of various agents on the evoked tritium-outflow was investigated. Ischemia(15min with 95% $N_2$ +5% $CO_2$) increased both the basal and evoked NE release. These increases were abolished by addition of glucose into the superfused medium, and they were significantly inhibited either by $0.3\;{\mu}M$ tetrodotoxin pretreatment or by removing $Ca^{++}$ in the medium. MK-801$(1{sim}10\;{\mu}M)$, a specific NMDA receptor antagonist, and glibenclamide $(1\;{\mu}M)$, a $K^+-channel$ inhibitor, neither alter the evoked NE release nor affected the Ischemia-Induced increases in NE release. However, polymyxin B(0.03 mg), a specific protein kinase C inhibitor, inhibited the effect of ischemia on the evoked NE release. Adenosine and $N^6-cyclopentyladenosine$ decreased the NE release in a dose-dependent manner in ischemic condition, though the magnitude of inhibition was far less than those in normal (normoxic) condition. Also the treatment with $5{\mu}M$ DPCPX, a potent $A_1-adenosine$ receptor antagonist did not affect the ischemia-effect. These results suggest that the evoked-NE release is potentiated by ischemia, and this process being most probably mediated by protein kinase C, and that the decrease of NE release mediated through $A_1-adenosine$ receptor is significantly inhibited in ischemic state.

A Review and Characteristics for Radioactive Effluents from the Nuclear Power Plants in Korea (국내원전의 방사성유출물 배출현황과 특성에 대한 고찰)

  • As of the end of 2010, 21 nuclear power reactors were operating in Korea. Radioactive effluents from nuclear power plants (NPPs) had been increased continuously and the radioactivity of effluents released in 2010 was 547.12 TBq. From 2001 to 2010, the annual average radioactivity of gaseous and liquid effluents per reactor was 11.61 TBq for pressurized water reactor (PWR) plants and 118.12 TBq for PHWR (pressurized heavy water reactor) plants. Most of the radioactivity from gaseous and liquid effluents was came from $^3H$. Based on the results of release trends and analysis, effluents characteristics was suggested for the management of radioactive effluents from NPPs.

Histochemical Study on the Hemocytes during Metamorphosis (배추흰나비의 變態에 따른 血球細胞의 組織化學的 硏究)

  • Insect hemocytes during metamorphosis were studies by histchemical and autoradiographic methods is Pieris rapae (Lepidoptera). The hemocytes were classified into six types, prohemocytes, plasmatocytes, podocytes, granular hemocytes, spherule cells and oenocytoids on the basis of the transitions in shapes and inclnsions of cytoplasms. Proteins, nucleic acids, polysaccharides and lipids in the hemocytes were detected histochemically from larval to pupal stages to learn the rise and fall of them during growth and metamorphosis. Most of the granules consisted of glycogen, neutral mucopolysaccharides and mucoprotein in addition to some granules of neutral fats and phospholipids were found in the granular hemocytes and spherule cells. Mitotic figures and DNA synthetic activities were observed in every type of hemocytes from 2nd to 5th instars, suggesting the all types of hemocytes originated from the prohemocytes. The cytoplasmic filaments of plasmatocytes and pdocytes extended very long in prepupa and pupa and the vermiform cells were the transformed plasmatocytes due to their further differentiation.

UV-INDUCED POLYMERIZATION OF SIZE-CONTROLLED PLATINUM/POLY[STYRENE-DIVINYLBENZENE-TRI(PROPYLENE GLYCOL) DIACRYLATE] HYDROPHOBIC CATALYST BEADS IN MICROFLUIDICS

  • The catalytic exchange of hydrogen isotopes between hydrogen and water has been known to be a very useful process for the separation of tritium from tritiated water. For the process, a highly active hydrophobic catalyst is needed. This study provides an effective fabrication method of size-controlled platinum/poly[styrene-divinylbenzene-tri(propylene glycol) diacrylate] [Pt/poly(SDB-TPGDA)] hydrophobic catalyst beads with a narrow size distribution. Platinum nanoparticles were prepared by ${\gamma}$-ray-induced reduction in the aqueous phase first, and then uniformly dispersed in SDB-TPGDA comonomer after the hydrophobization of platinum nanoparticles with alkylamine stabilizers. The porous Pt/poly(SDB-TPGDA) hydrophobic catalyst beads were synthesized by the UV-initiated polymerization of the mixture droplets prepared in a capillary-based microfluidic system. The size of as-prepared catalyst beads can be controlled in the range of $200-1,000{\mu}m$ by adjusting the flow rate of dispersed and continuous phases, as well as the viscosity of the continuous phase. Sorbitan monooleate and cyclohexanol were used as coporogens to control the porosities of the catalyst beads.

Study on the Characteristics of Catalyst Reaction for Hydrogen Recovery from Nuclear Fusion Exhaust Gas (핵융합 배가스 중 수소 회수를 위한 촉매반응 특성 연구)

  • In D-T fusion reaction, $D_2$ (duterium) and $T_2$(tritium) are used as fuel gas. The exhaust gas of nuclear fusion includes hydrogen isotopes $Q_2$ (Q means H, D or T), tritiated components ($CQ_4$ and $Q_2O$), CO, $CO_2$, etc. All of hydrogen isotopes should be recovered before released to the atmosphere. This study focused on the recovery of hydrogen isotopes from $CQ_4$ and $Q_2O$. Two kinds of experiments were conducted to investigate the catalytic reaction characteristics of SMR (Steam Methane Reforming) and WGS (Water Gas Shift) reactions using Pt catalyst. First test was performed to convert $CH_4$ into $H_2$ using 6% $CH_4$, 6% CO/Ar feed gas. In the other test, 100% CO gas was used to convert $H_2O$ into $H_2$ at various reaction conditions (reaction temperature, S/C ratio, GHSV). As a result of the first test, $CH_4$ and CO conversion were 41.6%, 57.8% respectively at $600^{\circ}C$, S/C ratio 3, GHSV $2000hr^{-1}$. And CO conversion was 72% at $400^{\circ}C$, S/C ratio 0.95, GHSV $333hr^{-1}$ in the second test.
