Antibacterial Effects and Cellular Responses of Imipenem-resistant Pseudomonas aeruginosa Exposed to Green Tea Polyphenols (녹차 폴리페놀에 노출된 Imipenem 내성 Pseudomonas aeruginosa의 항균효과 및 세포반응)

  • The aim of this work was to investigate the synergically bactericidal effects and cellular responses of tea polyphenols (TPP) and imipenem on imipenem-resistant Pseudomonas aeruginosa. Imipenem-resistant Ps. aeruginosa was isolated from patient in hospital. The bactericidal effects of TPP and imipenem were evaluated on the basis of its minimum inhibitory concentrations (MIC). The combined use of TPP and imipenem resulted in 16-fold and 8-fold reductions in the MICs of imipenem for the imipenem-susceptible and imipenem-resistant Ps. aeruginosa, respectively. The bactericidal effects of the imipenem and TPP against the Ps. aeruginosa was evaluated using the time-kill assay. The synergetic effects of the combinations of TPP and imipenem against Ps. aeruginosa were confirmed. Western blot using anti-DnaK and anti-GroEL monoclonal antibodies was performed to investigate the expression of stress shock proteins (SSPs) in imipenem-susceptible and imipenem-resistant strains exposed to TPP. The amount of SSPs were induced as the exposure time increased and decreased. The molecular weights of DnaK and GroEL were 70 kDa and 60 kDa, respectively. SDS-PAGE with silver staining revealed that the amount of lipopolysaccharides (LPS) increased or decreased in the strain treated to different concentrations and exposing periods of TPP. Scanning electron microscopic analysis demonstrated the presence of umblicated and wrinkled surfaces for cells treated with TPP or imipenem.

  • Trans-Pacific Partnership started as a small scale 4 countries regional trade agreement. It became a grand free trade agreement when the United States announced its participation and Japn considers to join. Korea still hasn't decided the stance however, considering the economic and political relationship with the United States and the impact of TPP, it looks like that Korea is going to join TPP. US led the TPP agreement is expected to be made based on the previous FTA agreements of the US. In the promising E-Commerce part, it is likely that the e-commerce leading US would suggest favorable agreement. If Korea joins the TPP, it's necessary to suggest what Korea wants and needs based on the previous KORUS FTA to promote Korean e-commerce for the future.

  • Purpose: Lipophilic cations including tetraphenylphosphonium (TPP) salts penetrate the hydrophobic barriers of the plasma and mitochondrial membranes, resulting in accumulation in mitochondria in response to the negative inner transmembrane potentials. The development of radiolabeled phosphonium cations as a noninvasive imaging agent may serve as a new molecular "voltage sensor" probe to investigate the role of mitochondria in the pathophysiology and diagnosis of cancer. Materials and Methods: We have synthesized a reference compound (4-fluorophenyl)triphenylphosphonium (TPP) and a labeled compound $[^{18}F]$TPP via two step nucleophilic substitution of no-carrier-added $[^{18}F]$fluoride with the precursor, 4-iodophenyltrimethylammonium iodide, in the presence of Kryptofix-2.2.2 and $K_2CO_3$. Result: The reference compound (4-fluorophenyl)triphenylphosphonium (TPP) was synthesized in 60% yield. The radiolabeled compound $[^{18}F]$TPP was synthesized in $10\sim15%$ yield. The radiochemical purity of the $[^{18}F]$TPP was $95.57{\pm}0.51%$ (n=11). Conclusion: $[^{18}F]$TPP was successfully synthesized that might have a potential to be utilized as a novel myocardial or cancer imaging agent for PET. However, it is required to improve the radiochemical yield to apply $[^{18}F]$TPP in preclinical or clinical researches.

  • The catalytic oxidations of several cycloolefins in $CH_2Cl_2$ were been investigated using Mn(III)-, Fe(III)-porphyrin complexes as a catalyst and sodium hypochlorite as a terminal oxidant. Porphyrins were $(p-CH_3O)TTP,\;(p-CH_3)TTP,$ TPP, (p-F)TPP, (p-Cl)TPP and $(F_{20})TPP$ (TPP = tetraphenylporphyrin), and olefins were cyclopentene, cyclohexene and cycloheptene. The substrate conversion yield was discussed according to the substituent effects of metalloporphyrin. The conversion yield of substrate by changing the substituent of TPP increased in the order of $p-CH_3O$ < $p-CH_3$ < H < p-F < p-Cl, which was consistent with the sequence of $4{\sigma}$ values of TPP. The conversion of cycloalkene followed the order of $C_5\;<\;C_6\;<\;C_7$.

  • The catalytic oxidation reaction of several cycloolefins in $CH_2Cl_2$ have been investigated using non-redox metalloporphyrin(M = Ga(III), In(III) and TI(III) complexes as a catalyst and sodium hypochlorite as a terminal oxidant. Porphyrins were $(p-CH_3O)$TPP, $(p-CH_3)$TPP, TPP, (p-F)TPP, (p-Cl)TPP and $(F_{20})$TPP (TPP=5,10,15,20-tetraphenyl-21H,23H-porphyrin) and olefins were cyclopentene, cyclohexene, cycloheptene and cyclooctene, The substrate conversion yield(%) was investigated according to the radius effect of non-redox metal ion, substituent effect and hindrance effect of metalloporphyrin. The conversion yield of cycloolefin was in the following order : $C_5$ < $C_6$ < $C_7$ = $C_8$.

Thermodynamic and Kinetic Study on the Protonation of Free Base Tetraphenylporpyrin Derivatives in Solution

  • The protonation of tetraphenylporphyrin (TPP) in acidic organic solutions was analyzed by acid titrimetric and temperature-dependent absorption measurements. Competition between the protonation of free base TPP $(TPPH_2)$ and the solvation of proton by near solvent molecules determines the equilibrium of the diprotonated TPP $(TPPH_4^{2+})$ formation. The diprotonated TPP exists as an ion pair complex with the acid counterions, which are found to affect the degree of red shift of the Soret band. The rotation of the phenyl rings also plays an important role in the diprotonation, as suggested by the decrease in the degree of diprotonation for the fluorophenyl TPP derivatives whose phenyl ring rotation is significantly hindered relative to normal TPP. The difference of fluorescence lifetime between $TPPH_2 \;({\pi}_{FL}=19.6\;ns)\; and\; TPPH_4^{2+} \;({\pi}_{FL}=2.1 \;ns)$ was used advantageously to measure the rate of protonation in the excited state. The protonation of TPPH2 are found to occur much slower than the diffusion of protons from bulk solution to the porphyrin ring. The monoprotonated TPP is suggested to be the transient species for the diprotonation process.

Cure Kinetics of Self-Extinguishing Epoxy Resin Systems with Charge Transfer Complex Type Latent Catalyst for Semiconductor Encapsulation (전하전이착체형 잠재성 촉매를 사용한 반도체 성형용 자소성 에폭시 수지 시스템의 경화 반응속도 연구)

  • The cure properties of self-extinguishing epoxy resin systems with different charge transfer type latent catalysts were investigated, which are composed of YX4000H as a biphenyl epoxy resin, MEH-7800SS as a hardener, and charge transfer type latent catalysts. We designed and used five kinds of charge transfer type latent catalyst and compared to epoxy resin systems with Triphenylphosphine-Benzoquinone(TPP-BQ) as reference system. The cure kinetics of these systems were analyzed by differential scanning calorimetry with an isothermal approach, the kinetic parameters of all systems were reported in generalized kinetic equations with diffusion effects. The epoxy resin systems with Triphenylphosphine-Quinhydrone(TPP-QH), Triphenylphosphine-Benzanthrone(TPP-BT) and Triphenylphosphine-Anthrone(TPP-AT) as a charge transfer type latent catalyst showed a cure conversion rate of equal or higher rate than those with TPP-BQ. These systems with TPP-QH and Triphenylphosphine-Tetracyanoethylene(TPP-TCE) showed a critical cure reaction conversion of equal or higher conversion than those with TPP-BQ. The increases of cure conversion rates could be explained by the decrease of the activation energy of these epoxy resin systems. It can be considered that the increases of critical cure reaction conversion would be dependent on the crystallinity of the biphenyl epoxy resin systems.

Fibrinolytic, Immunostimulating, and Cytotoxic Activities of Microbial Strains Isolated from Kochujang (고추장 분리 균주의 혈전용해능, 면역활성능 및 세포독성 효과 조사)

  • This study was carried out to investigate the functional activities of microorganisms isolated from kochujang, such as fibrinolytic, immunostimulating, and cytotoxical activities, and to apply these microorganisms to kochujang products. Ninety-one microbial strains with proteolytic activity were selected from 294 strains isolated from traditional and commercial kochujang. Three strains (TPP 0014, TPP 6013, and TPP 6015) with high fibrinolytic activity were tested for their immunostimulating and cytotoxical activities. For the assessment of macrophage activation, cytokines such as tumor necrosis factor, $interleukin-1{\alpha}$ and nitrogen oxide were measured with the murine macrophage cell line RAW 264.7. In addition, the cytotoxical activities of the three strains were examined by MTT assay on the colon cancer cell line SNU-C4 and normal cell line CHO-K1. Using an API identifying kit, two of the microbial strains (TPP 0014 and TPP 6015) were identified as Bacillus stearothermophilus and the other strain (TPP 6013) was identified as B. amyloliquefacience.

Thermal Properties and Microencapsulation of a Phosphate Flame Retardant with a Epoxy Resin (에폭시 수지를 이용한 인계 난연제의 마이크로캡슐화 및 열적 특성 연구)

  • The microcapsules containing triphenyl phosphate (TPP), a flame retardant, were prepared by phase-inversion emulsification technique using the epoxy resin (Novolac type) with excellent physical properties and network structure. This microencapsulation process was adopted for the protection of TPP evaporation and wetting of polymer composite during the polymer blend processing. The TPP, epoxy resin and mixed surfactants were emulsified to oil in water (O/W) by the phase inversion technology and then conducted on the crosslinking of epoxy resin by in-situ polymerization. The capsule size and size distribution of TPP capsules was controlled by mixed surfactant ratio, concentration and TPP contents, The formation and thermal property of TPP capsules were measured by differential scanning calorimetry and thermogravimetric analysis. The morphology and size of TPP capsules were also investigated by scanning and transmission electron microscopies. As the surfactant concentration increased, the TPP capsules were more spherical and mono-dispersed at the same weight ratio of mixed surfactants (F127: SDBS).

Health Effects of the Offensive Odor in Residents Living Near the Petrochemical Industries Complex Area and the Thermoelectric Power Plant (석유화학공단과 화력발전소 주변지역 주민들이 인식하는 악취발생과 건강영향의 관련성 연구)

  • This study investigated the health effects of offensive odor in residents living near the petrochemical industry complex area(PICA) and the thermoelectric power plant(TPP) by using questionnaire. Residents who felt the offensive odor were 58.3% at PICA, 50.9% at TPP and 24.4% at classical fishing and agrarian villages (CFAV)(p=0.000). People who answered that the offensive odor was sever at CFAV were 95.2% only on summer, but at PICA and TPP, were 44.1% and 57.3% on Spring, 62.4% and 68.8% on Summer, 22.0% and 31.7% on Autumn, and 21.7% and 25.7% on Winter, respectively. Average days that the odor occurred were 4.4 days/month at CFAV, but 12.0 and 9.5 days/month at PICA and TPP, respectively. People who experienced the sleep disturbance were 28.0% and 27.1% at PICA and TPP, respectively. The most frequently subjective symptoms were headache(0.953), frequently sneezing(0.825), itchy eyes(0.766), and stimulating eyes(0.709) at PICA, and headache(1.082), itchy eyes(0.931), itchy skin(0.826), and frequent sneezing(0.674) at TPP, respectively. At PICA and TPP, the occurrence rates of diseases in respondents' families were 15.4% and 15.6% for asthma, 12.4% and 9.2% for respiratory diseases, 27.8% and 31.2% for skin diseases, and 9.1% and 6.9% for nervous diseases, respectively. In conclusion, many residents who living near the PICA and TPP experienced the offensive odor during four seasons, especially high on summer, the most frequently subjective symptoms such as headache, itchy and stimulating eyes, frequently sneezing, and some diseases among their families such as asthma, respiratory diseases, skin diseases, and nervous diseases.
