Anti-inflammatory Effect of Dactyloquinone B and Cyclospongiaquinone-1 Mixture in RAW264.7 Macrophage and ICR Mice

  • Sesquiterpene-quinone is a class of secondary metabolites frequently encountered from marine sponge. The present study was designed to examine the anti-inflammatory action of sponge-derived dactyloquinone B (DQB) and cyclospongiaquinone-1 (CSQ1) mixture using lipopolysaccharide (LPS)-induced inflammatory responses. We measured the production of nitric oxide (NO), tumor necrosis factor-alpha ($TNF-{\alpha}$), $interleukin-1{\beta}$ ($IL-1{\beta}$), and interleukin-6 (IL-6) and expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein. $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6 production, which increased by treatment with LPS, were significantly inhibited by DQB and CSQ1 mixture. It also decreased the production of NO production, and iNOS and COX-2 expression. Furthermore, it reduced 12-O-tetradecanoylphorbol 13-acetate (TPA)-induced ear edema of ICR mice. These results demonstrate that sesquiterpene-quinone, DQB and CSQ1 mixture, might serve as a chemical pipeline for the development of anti-inflammatory agent.

Structure-Activity Relationships of Polyhydroxyursane-type Triterpenoids on the Cytoprotective and Anti-inflammatory Effects

  • Eleven polyhydroxyursane triterpenoids (PHUTs) were tested to determine their cytoprotective, immunosuppressive and anti-inflammatory effects. To compare the bioactivities of $19{\alpha}$-hydroxyursane-type triterpenoids {23-hydroxytormentic acid (6), its methyl ester (7), tormentic acid (8), niga-ichigoside $F_1$ (9),euscaphic acid (10) and kaji-ichigoside $F_1$ (11)} of the Rosaceae crude drugs (Rubi Fructus and Rosa rugosae Radix) with PHUTs possessing no $19{\alpha}-hydroxyl$ of Centella asiatica (Umbelliferae), the four PHUTs, asiaticoside (1), madecassoside (2), asiatic acid (3), and madecassic acid (4) were isolated from C. asiatica and 23-hydroxyursolic acid (5) from Cussonia bancoensis. Cytoprotective effects were assessed by measuring cell viabilities against cisplatin-induced cytotoxocity in $LLC-PK_1$, cells (proximal tubule, pig kidney) to determine whether these agents have protective effects against nephrotoxicity caused by cisplatin. The inhibitory effect of 11 PHUTS on nitric oxide (NO) and prostaglandin $E_2\;(PGE_2)$ were evaluated by measuring nitrite accumulation in lipopolysaccharide (LPS)-induced macrophage RAW 264.7 cells, and their anti-inflammatory effects were tested in 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mouse ear edema model. Six MHUTs (compounds 1, 2, 4, 6, 10, and 11) exhibited higher cell viabilities during cisplatin-induced cytotoxicity testing even at a concentration of $200\;{\mu}g/ml$ than cisplatin only-treated group, suggesting that ese compounds have the potentcytoprotective efffcts. Compounds 1 and 3 of the C. asiatica and niga-ichigoside $F_1$ exhibited no inhibitory effect on NO and/or $PGE_2$ production whereas other PHUTs produced mild to significant NO and/or $PGE_2$ production.The four compounds (2, 5, 9, and 10) potently inhibited mouse ear edema induced by TPA whereas two compounds (1 and 3) had no activity in this test. These results suggest that many PHUTs are potentchemopreventives. Structure-activity relationship (SAR) was also discussed in each assay with regard to the significant role of OHs at the position of 2, 3, 6, 19, and 23 and to the glycoside linkage at the 28-carboxyl.

The Indian Magical Herb 'Sanjeevni' (Selaginella bryopteris L.) - A Promising Anti-inflammatory Phytomedicine for the Treatment of Patients with Inflammatory Skin Diseases

  • Objectives: Selaginella bryopteris L. (family: Selaginaceae), is often used in traditional Indian systems of medicine for the prevention and cure of several disorders and for the treatment of patient with spermatorrhoea, venereal disease, constipation, colitis, urinary tract infections, fever, epilepsy, leucorrhoea, beri-beri and cancer. It is also used as a strength tonic. This study aimed to evaluate the mechanisms underlying the anti-inflammatory effects of topically administered aqueous, polar and non-polar methanolic fractions ($10mg/20{\mu}L$) of Selaginella bryopteris. Methods: An acute oral toxicity study of Selaginella bryopteris at doses from 250 to 2,000 mg/kg body weight (bw) was performed. Aqueous, polar and non-polar methanolic extracts ($10mg/20{\mu}L$) applied topically for 5 days were evaluated for their anti-inflammatory effects against 12-tetra-O-decanoyl phorbol acetate (TPA)- induced inflammation by using the redness in the ear, the ear's weight (edema), oxidative stress parameters, such as lipid-peroxide (LPO) and nitric oxide (NO), and the pro-inflammatory cytokines involved in inflammation, such as tumour necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$ and IL-6. Indomethacine ($0.5mg/20{\mu}L$) was used for the positive control. Results: Selaginella bryopteris produced no mortalities when administered orally at doses from 250 to 2,000 mg/kg bw. Topical treatment with the non-polar methanolic fraction ($10mg/20{\mu}L$) significantly suppressed redness ($2.4{\pm}0.5$) and edema ($30.4{\pm}1$) and effectively reduced the LPO level ($32.3{\pm}3.3$). The NO level was ($8.07{\pm}0.55$), and the $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6 levels were decreased to $69.6{\pm}15.5$, $7.7{\pm}4.8$ and $82.6{\pm}5.9$, respectively. Conclusion: This study demonstrated for the first time the mechanisms underlying the anti-inflammatory effect of medicinal plants like Selaginella bryopteris and quantified the pharmacological interactions between them. The present study showed this herbal product to be a promising anti-inflammatory phytomedicine for the treatment of patients with inflammatory skin diseases.

통밀가루 첨가량에 따른 설기떡의 품질 특성 (Quality Characteristics of Sulgidduk with Whole Wheat Flour)

  • 식이섬유가 풍부한 WWF를 우리 전통 음식인 설기떡에 첨가하여 수분함량, 색, 기계적 조직감 특성, 노화도, 관능적 특성 등 품질 특성을 규명하였으며, 품질과 영양적 가치를 고려한 적절한 첨가 수준을 찾고자 하였다. 시료들 간 유의적인 수분 함량의 차이는 없었으며, 색도 중 L(명도)값은 WWF의 첨가량이 증가할수록 감소하는 값을 나타내었다. TPA 검사에서 hardness는 제조 직후 시료들 간 유의적인 차이가 없었으나, 저장 후에는 WWF의 함량이 많아질수록 그 값이 유의적으로 감소하여, WWF의 첨가가 설기떡의 노화 지연에 효과가 있음을 알 수 있었다. 또한 저장 기간 중 WWF 첨가량이 증가할수록 springiness와 chewiness가 대조군에 비해 낮은 값을 나타내었다. 그 밖의 조직감 특성은 시료 간 유의적인 차이를 보이지 않았다. Avrami 방정식의 시간상수(1/k)값은 WWF 함량이 증가할수록 증가하여 WWF의 노화지연 효과를 확인하였다. 관능적 품질특성은 맛과 향, 조직감에서 5% 대체군이 전반적으로 높게 평가되었으며 전반적인 기호도까지 고려하면 5% WWF 대체가 맛과 향 및 조직감에 긍정적인 영향을 미칠 수 있음을 알 수 있었다.

세몰리나 첨가가 쌀국수의 품질특성에 미치는 영향 (Effects of Semolina on Quality Characteristics of the Rice Noddles)

  • 쌀국수 제품의 품질을 향상시키기 위한 방법으로서 기존의 쌀국수에 파스타가 지닌 물성 및 조리 상의 장점을 접목하고자 파스타의 제조원료인 세몰리나를 글루텐이 소량 첨가된 쌀가루에 대해 함량을 달리하여(0-20%, w/w) 대체하여 생면을 제조하였다. 제조된 생면을 끓인 조리면의 품질을 측정한 결과 전분노화가 다소 억제되며, 씹는 맛("ldente")이 증가하고, 끓이는 과정에서 일어나는 전분유출이 감소하여 전반적으로 제품의 품질이 향상되었다. 생면의 제조과정에서는 반죽의 점성 이 커져 반죽이 잘 뭉치고 단단해져 면대형성이 향상되는 효과가 기대되었다. RVA 측정에 따른 전분혼합액의 호화개시온도(pasting time)는 10% 첨가까지는 거의 영향이 없다가 20% 첨가구에서 최고 1.2 증가한 반면 최고점도(peak viscosity)는 첨가량 15% 이상부터 큰 폭으로 감소하였다. 전분의 breakdown값 역시 세몰리나 첨가량이 늘수록 급격히 줄어 shear thinning이 줄어들며 10% 이상 첨가구에서 최종점도(final viscosity)와 setback값 차이가 크게 줄어 노화억제에 의해 조리면이 딱딱해지는 정도가 감소하는 효과가 예상되었다. 생면을 끓일 때 국수에 의한 수분흡수가 낮아져 조리면의 중량 및 부피 변화가 감소되었으며 국물의 탁도가 증가되지 않아 전분침출 억제기능이 확인되었다. 조리면의 색도에 명도(L)는 감소하였으며 적색도(a)와 황색도(b)가 증가하였으나 일정 수준(15%) 이상이 되면 그 영향이 적게 나타났다. 적은 양(5%)의 세몰리나 첨가에 의해서도 Texture Analyzer로 측정된 경도(hardness), 탄력성(springiness), 응집성(cohesiveness), 검성(gumminess) 및 씹힘성(chewiness)과 같은 모든 텍스쳐 특성치 및 인장강도가 소량(5%)의 첨가에 의해서도 현저히 증가하였으나 10% 이상에서는 변화폭이 줄어는 공통적인 변화양상을 보였다. 이상의 결과를 토대로 세몰리나를 첨가한 쌀국수를 제조하는 경우 원료비용 및 기능성을 고려할 때 쌀가루:세몰리나 최적 첨가비율은 9:1(10%, w/w)로 제시되었다.
