식물 약재 구성성분과 국내 약전 성분과의 비교 연구 (Comparison of chemical compounds of medicinal plant materials with compounds of domestic pharmacopoeias)

  • Objective : TM-MC was constructed to provide information on the chemical compounds of medicinal plant materials which were listed in Korean, Chinese, and Japanese pharmacopoeias. The aim of this study was to compare of the chemical compounds of TM-MC with those of domestic pharmacopoeias.Methods : First of all, TM-MC was updated on medicinal materials and chemical compounds which were extracted from articles up to the PubMed identifier of 26 millions. After searching for medicinal plant materials with information on their major compounds in Korean pharmacopoeias, we identified that the information on medicinal materials and chemical compounds were also existed in TM-MC. Then, we figured out how much the compounds have been studied by counting the number of articles for those compounds in TM-MC.Results : Information on 9 medicinal materials and about 1,700 compounds was newly added to TM-MC after manually curating 645 articles. Korean pharmacopoeias contained a total of 91compounds from 53 medicinal plant materials, while TM-MC provided information on the 89 compounds from the 53 medicinal materials in Korean pharmacopoeias. The compounds stated in Korean pharmacopoeias has been much studied by comparison with other compounds of each medicinal material in TM-MC.Conclusion : Most of chemical compounds stated in Korean pharmacopoeias were also listed in TM-MC, excluding two compounds. Thus, we concluded that information in TM-MC is mostly consistent with that of Korean pharmacopoeias. In the future, we will periodically update TM-MC with articles added to the MEDLINE.

Expression, Purification and NMR Studies on MC4R-TM2 Mutant

  • Melanocortin-4 receptor (MC4R) subtype is associated with obese humans. Especially, in a patient with severe early-onset obesity, novel heterozygous mutation in the MC4R gene was detected, resulting in an exchange of aspartic acid to asparagine in $90^{th}$ amino acid residue located in the predicted second trans-membrane domain (TM2). Mutations in the melanocortin-4 receptor (MC4R) gene are the most frequent monogenic causes of severe obesity which have been described as heterozygous with loss of function. In order to compare structure difference between MC4R wild type (MC4R-TM2-wt) and mutant (MC4R-TM2-D90N), we designed both MC4R-TM2-wt and MC4R-TM2-D90N construct in pET 21b vector. In this study, we optimized high-yield purification procedure for recombinant TM2-D90N. Eluted recombinant protein was resolubilized under urea condition for thrombin cleavage reaction and we conducted the high-performance liquid chromatography (HPLC) with reverse phase column under 1% acetonitrile, 0.01% TFA buffer solution. The molecular size of purified target peptide was confirmed by Tricine-SDS page analysis. To characterize MC4R-TM2-D90N, we have performed $^{15}N$-isotope labeling of peptide using M9 media and purified labeled target peptide for hetero-nuclear NMR spectroscopy.

Optimization of the experimental conditions for structural studies of the second transmembrane domain from human wild-type & mutant melanocortin-4 receptor

  • Human melanocortin-4 receptor (hMC4R) has a critical role in part of energy homeostasis, and their heterozygous mutations related in genetic cause of severe human obesity. In order to study the structure and function of these membrane proteins, it is important to prepare the samples. However, the preparation of transmembrane peptide is seriously difficult and time-consuming. Overexpression and purification of membrane proteins was reported to be difficult due to their innate insoluble and toxic properties. Among the many difficulties, the most important is the difficulty in obtaining sufficient quantities of purified protein. Recently, we succeed to produce large amounts of the second transmembrane domain from the wild-type hMC4R (wt-TM2) and D90N mutant hMC4R (m-TM2) and proposed the structural difference of them in membrane-like environments. In this paper, we demonstrate the optimization procedures to express and purify wt-TM2 or m-TM2 peptides, and solution NMR studies in different detergents to get high-resolution spectra were also described.

Optimized purification and characterization of expressed hMC4R-TM2

  • Human melanocortin-4 receptor (hMC4R) among MC-Rs, expressed in the brain, is in charge of the control on energy homeostasis and food intake. The structure and function of human MC4R have been studied to understand their essential function and roles. To investigate the structure and function, it is necessary to prepare sufficient amounts of proteins. However, their expression and purification is demanding and time-consuming due to their innate insoluble and toxic properties. The heterozygous mutations of hMC4R, exchange of Asp 90 to Asn located in second transmembrane, cause severe obesity in human. To obtain purified hMC4R wt-TM2 for structural studies, it was first over-expressed and purified by fast protein liquid chromatography (FPLC) and then solution NMR studies were performed to get high-resolution spectra. In here, we established optimized purification scheme to get more purified target peptide.

Comparison of dimensional accuracy between direct-printed and thermoformed aligners

  • Objective: The purpose of this study was to evaluate and compare the dimensional accuracy between thermoformed and direct-printed aligners. Methods: Three types of aligners were manufactured from the same reference standard tessellation language (STL) file: thermoformed aligners were manufactured using Zendura FLXTM (n = 12) and Essix ACETM (n = 12), and direct-printed aligners were printed using Tera HarzTM TC-85DAP 3D Printer UV Resin (n = 12). The teeth were not manipulated with any tooth-moving software in this study. The samples were sprayed with an opaque scanning spray, scanned, imported to Geomagic® Control XTM metrology software, and superimposed on the reference STL file by using the best-fit alignment algorithm. Distances between the aligner meshes and the reference STL file were measured at nine anatomical landmarks. Results: Mean absolute discrepancies in the Zendura FLXTM aligners ranged from 0.076 ± 0.057 mm to 0.260 ± 0.089 mm and those in the Essix ACETM aligners ranged from 0.188 ± 0.271 mm to 0.457 ± 0.350 mm, while in the direct-printed aligners, they ranged from 0.079 ± 0.054 mm to 0.224 ± 0.041 mm. Root mean square values, representing the overall trueness, ranged from 0.209 ± 0.094 mm for Essix ACETM, 0.188 ± 0.074 mm for Zendura FLXTM, and 0.140 ± 0.020 mm for the direct-printed aligners. Conclusions: This study showed greater trueness and precision of direct-printed aligners than thermoformed aligners.

수술가운의 방수성과 방균성에 관한 연구 (The Liquid and Viral Barrier Properties of Reusable and Disposable Surgical Gowns)

  • 최근에 치명적인 바이러스, 특히 에이즈를 이르키는 Human Immunodificiency Virus (HIV), 또는 간에 심각한 병을 이르키는 Hepatitis B Virus (HBV)와 같은 무서운 바이러스가 환자의 피나 분비물을 통하여 의사에게 전염되는 사례가 늘어감에 따라 이제는 환자의 피나 분비물의 침투를 막기 위해 방수성과 방균성을 가진 수술복 착용이 절대적으로 필요하게 되었다. 따라서 본 연구의 목적은 1. 수술가운을 만드는 5가지 대표적인 직물의 방수성과 방균성을 측정하고 2. 세탁시 표백제의 사용유무가 그 직물의 방수성과 방균성에 미치는 영향을 알아보고 3. 반복세탁이 그 직물의 방수성과 방균성에 미치는 영향을 알아 보는데 있다. 이 실험을 위해 3가지 재사용 수술가운과 (Gore-tex$^R$, membrane reinforced; Compel$^{TM}$, fabric reinforced; Acep$^{TM}$, non-reinforced) 두 가지 일회용 가운 (Evolution$^R$ gown, fabric reinforced; Evolution$^R$ Specialty, film reinforced)이 사용되었다. 표백제를 사용한 세탁이 직물의 방수성과 방균성에 미치는 영향을 알아보기 위해 재사용 가운을 세탁, 건조, 살균을 하였다. 세탁과 살균의 빈도수는 1, 20, 40, 50, 60, 70, 80번 이였다. 방수성 측정방법으로 1. Impact Penetration test (AATCC 42-1985), 2. Elbow Lean test, 3. Synthetic Blood Resistance test (ASTM F 23. 40. 01)가 사용되었고 방균성 측정방법으로 Viral Resistance test (ASTM F 23. 40. 02)가 사용되었다. 실험 결과는 다음과 같다. 1. Gore-Tex$^R$ 가운과 Evolution$^R$ Specialty 가운은 방수성과 방균성의 성질을 가졌다. 그러나 표백제를 사용하여 70번 세탁한 Gore-Tex$^R$ 가운은 membrane의 구조가 파괴되어서 방균성을 상실했다. 2. Evolution$^R$ 가운, Acep$^{TM}$ 가운과 Compel$^{TM}$ 가운은 오직 Impact Penetration test만 통과했다. 즉 이 직물들은 큰 압력의 가함이 없는 splash resistance만 가지고있다. 그러나 Acep$^{TM}$ 가운과 Compel$^{TM}$ 가운은 20번과 60번 세탁 후 각각 그들의 splash resistance 마져도 상실했다.

Monte Carlo Algorithm-Based Dosimetric Comparison between Commissioning Beam Data across Two Elekta Linear Accelerators with AgilityTM MLC System

  • Purpose: Elekta synergy® was commissioned in the Seoul National University Veterinary Medical Teaching Hospital. Recently, Chung-Ang University Gwang Myeong Hospital commissioned Elekta Versa HDTM. The beam characteristics of both machines are similar because of the same AgilityTM MLC Model. We compared measured beam data calculated using the Elekta treatment planning system, Monaco®, for each institute. Methods: Beam of the commissioning Elekta linear accelerator were measured in two independent institutes. After installing the beam model based on the measured beam data into the Monaco®, Monte Carlo (MC) simulation data were generated, mimicking the beam data in a virtual water phantom. Measured beam data were compared with the calculated data, and their similarity was quantitatively evaluated by the gamma analysis. Results: We compared the percent depth dose (PDD) and off-axis profiles of 6 MV photon and 6 MeV electron beams with MC calculation. With a 3%/3 mm gamma criterion, the photon PDD and profiles showed 100% gamma passing rates except for one inplane profile at 10 cm depth from VMTH. Gamma analysis of the measured photon beam off-axis profiles between the two institutes showed 100% agreement. The electron beams also indicated 100% agreement in PDD distributions. However, the gamma passing rates of the off-axis profiles were 91%-100% with a 3%/3 mm gamma criterion. Conclusions: The beam and their comparison with MC calculation for each institute showed good performance. Although the measuring tools were orthogonal, no significant difference was found.

Multi-carbohydrase application into energy and amino acid deficient broiler diets: A strategy to enhance performance of broiler chickens

  • The effect of Multi-Carbohydrase (MC) supplementation on growth performance, visceral organ weights, blood metabolites, jejunum morphology, nutrient digestibility, and carcass parameters of broiler chickens fed nutrient-deficient corn soybean-meal based diets containing high levels of non-starch polysaccharides from wheat and wheat by-products was investigated. A total of 378 one-day-old Ross 308 broiler chickens were randomly assigned to one of seven dietary treatments to give six replicates per treatment (nine birds per pen). Dietary treatments were as follows: (1) positive control (PC; commercial standard diet); (2) negative control 1 (NC-1; PC-120 kcal/kg metabolizable energy); (3) NC-2 (PC-3% standardized ileal digestibility [SID] amino acids). The remaining four dietary treatments were formulated with the addition of MC (MC; Superzyme-CSTM) into two negative controls along with two supplementation levels of MC (i.e., 0.025% and 0.05%, respectively). Improved body weight, average daily gain, and feed conversion ratio (p < 0.05) were observed in broiler chickens fed a reduced energy diet supplemented with MC compared to birds fed NC-1 diet from days 1-35. Additionally, birds fed a reduced energy diet with 0.05% MC showed comparable (p > 0.05) growth performance with birds fed PC for 35-day post-hatch. Furthermore, the addition of MC into reduced amino acid diets improved (p < 0.05) growth performance. Broiler chickens fed MC supplemented nutrient-deficient diets showed a greater (p < 0.05) villus height to crypt depth ratio than birds fed diets without MC on days 21 and 35. Similarly, improved (p < 0.05) nutrient digestibility was observed in birds fed reduced energy diets supplemented with MC compared to birds fed NC-1 on days 21 and 35. Our results suggest that MC supplementation into reduced energy or reduced amino acid diets containing wheat and wheat by-products has the potential to improve growth performance and nutrient digestibility while maintaining healthier gut morphology in broiler chickens from 1 to 35 days of age.

오령산 구성약재 성분의 Drug-likeness와 Oral bioavailability (Drug-likeness and Oral bioavailability for Chemical Compounds of Medicinal Materials Constituting Oryeong-san)

  • Objectives : Oryeong-san was composed of Alismatis Rhizoma, Atractylodis Rhizoma Alba, Poria Sclerotium, Polyporus, Cinnamomi Cortex, and known to have hundreds of chemical compounds. The aim of this study was to screen chemical compounds constituting Oryeong-san with the drug-likeness and oral bioavailability from the analysis of their physicochemical properties. Methods : A list of chemical compounds of Oryeong-san was obtained from TM-MC(database of medicinal materials and chemical compounds in Northeast Asian traditional medicine). To remove redundant compounds, the SMILES (Simplified Molecular Input Line Entry System) strings of each compound were identified. All of the physicochemical properties for the compounds were calculated using the DruLiTo(Drug Likeness Tool). Drug-likeness was estimated by QED(Quantitative Estimate of Druglikeness) and OB(Oral bioavailability) was checked based on the Veber's rules. Results : A total of 475 compounds were obtained by eliminating duplication among 544 compounds of 5 medicinal materials. Analysis of the physicochemical properties revealed that the most common values were MW(molecular weight) 200~300 g/mol, ALOGP(octanol-water partition coefficient) 1~2, HBA(number of hydrogen bond acceptors) 0~1, HBD(number of hydrogen bond donors) 0, PSA(polar surface area) 0~50 angstrom, ROTB(number of rotatable bonds) 1, AROM(number of aromatic rings) 0, and ALERT(number of structural alerts) 1. QED had 93% of the values between 0.2 and 0.7, and OB had 90% of the value of TRUE. Conclusions : We in this paper screened the candidate active compounds of Oryeong-san using the QED and Veber's rules. In the future, we will use the screening results to analyze the mechanism of Oryeong-san based on systems pharmacology.
