Induction and Culture of Hairy Roots of Crotalaria sessiliflora L. (활나물(Crotalaria sessiliflora L.)로부터 모상근의 유도 및 배양)

  • The hairy roots of Crotalaria sessiliflora were induced from the tissue segments infected with Agrobacterium rhizogenes ATCC 15834. The induced hairy roots were subjected to paper electrophoresis fro the detection of opine-positive clones which were considered to have been transformed. Mannopine and agropine were presented in hairy root clones while mannopine was presented in two hairy root clones. Eight hairy root clones were selected and cultured in MS, B5 and WP media. Each of hairy root clones was showed a difference in branch pattern and growth rate. The best culture medium and culture conditions of hairy roots were in $\frac{1}{2}$MS(3% sucrose, pH 5.7) liquid medium at 25$^\circ C$, 70 rpm under dark, the growth rate in $\frac{1}{2}$MS liquid medium was increased with 210-fold more than that of inoculated hairy roots and with 2-fold more than that in MS liquid medium. Also, the adequate condition for hairy root growth was such that concentration of KH$_2PO$_4 was 1.25mM and the ratio of NH${_4}{^+}$ : NO${_3}{^-}$ was 1 to 3 in MS medium. The presence of pyrrolizidine alkaloids, monocrotaline, in the hairy roots was detected by TLC.

Quality Monitoring of Distributed Herbal Medicine, Scutellariae Radix (유통생약 황금의 품질모니터링 연구)

  • This study was investigated that quality inspection of distributed Scutellariae Radix in Korea. To evaluate the quality of these herbal medicines, we carried out TLC pattern, foreign matter in purity, loss on drying, ash, acid-insoluble ash, oil content, dilute ethanol-soluble, water-soluble, ether-soluble extracts contents, quantitative analyses, residual $SO_2$, individual heavy metals and organochlorine pesticides. To measure contents of baicalin, baicalein and wogonin, we were quantitative analyzed using HPLC. The average contents of baicalin, baicalein and wogonin were detected by 13.28 (${\pm}$0.43)%, 1.17 (${\pm}$0.04)% and 0.40 (${\pm}$0.02)%, respectively. Each average contents As, Cd, Hg and Pb was 0.059 (${\pm}$0.054) mg ${\cdot}$ $kg^{-1}$, 0.019 (${\pm}$0.010) mg ${\cdot}$ $kg^{-1}$, 0.017 (${\pm}$0.057) ppm and 0.242 (${\pm}$0.084) mg ${\cdot}$ $kg^{-1}$, respectively.

Method for Supplementing Lecithin to Ginseng Extract (레시틴이 강화된 인삼 추출물 제조 방법)

  • This study was carried out to develop the method of preparing lecithin-fortified ginseng extract. Firstly, soybean lecithin was mixed with soybean oil (LCS) in varying ratio (2.5%, 5%, 10% and 20%). Then, one part volume of LCS was mixed with three parts volume of ginseng extract with 10% solid matter content and the mixture was vortexed vigorously. Finally, the mixture was spinned at the speed of 3,000 rpm for 30 minutes to separate oil and aqueous ginseng extract layer (AG). AG was then subjected to qualitative and quantitative analysis of phospholipids and ginsenosides. Fatty acid composition and crude fat content before and after LCS was determined. Stability of lecithin in ginseng extract was determined by analyzing phospholipid content in the one third upper and lower layer of the concentrated AG in Falcon tubes while storing the LCS treated concentrated AG in 4, 25 and 40oC for 6 months. Ratio of lecithin transferred to AG increased with the increase in lecithin content of soybean oil. There was no significant change in fatty acid composition and crude fat content, and ginsenoside content in the ginseng extract before and after LCS treatment. TLC and HPLC pattern of saponin fraction before and after treating the ginseng extract with LCS demonstrated no observable difference. There was no change in lecithin content in the upper and lower one third layer of ginseng extract in the tubes after storing the concentrated AG in 4, 25 and $40^{\circ}C$ for 6 months. Ginsenosides HPLC pattern was not changed when stored the LCS-treated ginseng extract in those conditions for six months, indicating satisfiable stability of the LCS-treated concentrated ginseng extract. From these results, it can be concluded that treatment of the ginseng extract with lecithin containing soybean oil is a labor effective method with satisfiable stability to fortify lecithins to ginseng extract.

A Novel in Vitro Method for the Metabolism Studies of Radiotracers Using Mouse Liver S9 Fraction (생쥐 간 S9 분획을 이용한 방사성추적자 대사물질의 새로운 체외 측정방법)

  • Purpose: Usefulness of mouse liver S9 fraction was evaluated for the measurement of the metabolites in the in vitro metabolism study of $^{18}F$-labeled radiotracers. Materials and Methods: Mouse liver S9 fraction was isolated at au early step in the course of microsome preparation. The in vitro metabolism studies were tarried out by incubating a mixture containing the radiotracer, S9 fraction and NADPH at $37^{\ciirc}C$, and an aliquot of the mixture was analyzed at the indicated time points by radio-TLC. Metabolic defluorination was further confirmed by the incubation with calcium phosphate, a bone mimic. Results: The radiotracer $[^{18}F]1$ underwent metabolic defluorination within 15 min, which was consistent with the results of the in vivo method and the in vitro method using microsome. Radiotracer $[^{18}F]2$ was metabolized to three metabolites including $4-[^{18}F]fluorobenzoic$ acid within 60 min. It is likely that the one of these metabolites at the origin of radio-TLC was identical with the one that obtained from the in vivo and in vitro (microsome) method. Compared with the in vitro method using microsome, the method using S9 fraction gave a similar pattern of the metabolites but with a different ratio, which can be explained by the presence of cytosol in the S9 fraction. Conclusion: These results suggest that the findings of the in vitro metabolism studies using S9 fraction can reflect the in vivo metabolism of novel radiotracers in the liver. Moreover, this method can be used as a tool to determine metabolic defluorination along with calcium phosphate absorption method.

Preservation of Washed Fresh Ginsengs by Gamma Irradiation (방사선(放射線) 조사(照射)에 의한 수세된 수삼(水蔘)의 저장(貯藏))

  • The washed fresh ginsengs packed with air, vaccum and nitrogen gas were irradiated at the levels of 1,2 and 3kGy gamma radiation and then stored at $4{\sim}5^{\circ}C$ for 90days to investigate the effects of gamma radiation on microbial inactivation, eelworm disinfestation and physicochemical changes. After a 90 day storage, $2{\sim}3kGy$ irradiated groups showed 20% of weight loss and 10% of rot while non-irradiated group 100% and 20% or more, respectively. Also the irradiated groups showed somewhat lower values of specific gravity, color density and hardness immediately after irradiation, thereafter higher value of them with storage time than those of non-irradiated group. The irradiation increased the yields of ginseng extract and crude saponins but no effects on the proximate composition and TLC and HPLC patterns of saponin. The food-borne microorganisms decreased in viable cell counts by $2{\sim}3$ log cycles with $2{\sim}3kGy$ radiation and the eelworms were completely disinfested with 1 kGy radiation.

Ginsenoside derivatives and quality characteristics of fermented ginseng using lactic acid bacteria (유산균을 이용한 발효인삼의 ginsenoside 유도체 및 품질특성)

  • This study was done in order to investigate the bioconversion of ginsenoside, as well as the quality characteristics of fermented ginseng, by using lactic acid bacteria. Quality characteristics such as the thin layer chromatography(TLC) pattern, ginsenosides, total phenolic content, electron donating ability, and total sugar of fermenting ginseng and red ginseng were analyzed. The ginsenoside Rg2r, Rh2s and Rh2r of the fermented ginseng and red ginseng for 65 hours at a temperature of $37^{\circ}C$ were not detected. The ginsenoside Rg1 and Re contents have decreased, while the Rh1, Rg2s, Rd, Rg3r, and Rg3s have increased due to fermentation. The ginsenoside Rg3 of the fermented red ginseng has increased and the contents were $114.83{\sim}131.68{\mu}g/mL$ (control $104.56{\mu}g/mL$). The total phenolic content and electron donating ability of the red ginseng have totally decreased after 7 days of fermentation. The total phenolic contents of the fermented ginseng and red ginseng with different lactic acid bacteria did not show any tendency as different strains. The electron donating ability of the fermented ginseng has increased; however, the electron donating ability of the red ginseng has decreased. The total sugars of the fermented ginseng and red ginseng with different lactic acid bacteria have also decreased.

A Study on the Composition of Polar Lipid in Adlay (율무의 극성지질 조성에 관한 연구)

  • The compositional characteristics of polar lipids in the brown and polished adlay were studied. Total lipids of brown and polished adlay were extracted, purified and fractionated into neutral and polar lipids by silicic acid column chromatography. The glycolipid(GL) and phospholipid(PL) of polar lipids were seperated by thin layer chromatography and quantitated by scanner. The fatty acid composition in polar lipids were determined by gas chromatographr. The contents of GL and PL in brown adlay were 5.67% and 1.83%, and their contents on polished adlay were 5.49% and 1.78%, respectively. Of the GL in the brown and polished adlay, monogalactosyl diglycerides+esterified steryl glycosides and digalactosyl diglycerides were the major components, but cerebrosides+steryl glycosides were also found as the minor component of GLs. Of the PLs, phosphatidyl choline and phosphatidyl ethanolamine were the major components. Phosphatidyl inositol, lysophosphatidyl choline and phosphatidyl serine were also present in the PLs. The major fatty acids of GLs were linoleic, oleic and palmitic acids. The fatty acid composition in the PLs were similar to the pattern in the GLs, but PLs contained the lower percentage of linoleic acids than the GLs.

Changes in Total Fatty Acids, Total Number of Fatty Acid Acyl Carbon Atoms and Species of Triglycerides from Human Milk Lipids during the Course of Lactation (수유 기간의 경과에 따른 인유 트리글리세리드의 지방산 조성, 아실 탄소수 및 종의 변화)

  • The fatty acid composition, total number of fatty acid acyl carbon atoms and species of triglycerides from human milk samples obtained during 70 days of lactation from 39 mothers were determined by argentation thin-layer and gas chromatographic procedures. The medium- and long-chain saturated fatty acids(8:0, 1O:0, 12:0 and 14:0) which are formed exclusively by synthesis within mammary gland increased significantly from colostrum to mature milk. Long-chain saturated fatty acids(16 : 0, 22 : 0 and 24: 0) were significantly higher than tile levels found in transitional and mature milk. The precursors of w C- and w 3-series, 18:2 w 6 and 18:3 w 3, were increased slightly in progressing lactation. Colostrum contained significantly higher proportions of 18:1 w 9 and w 6- and w 3- derived long-chain polyunsaturated fatty acids than transitional milk, and these levels were further reduced in mature milk. The triglycerides of human milk lipids which were made up of 30-60 acyl carbon atoms showed a pattern with major contributions made by the glycerides with 44-52 acyl carbon atoms. The levels of triglycerides with less than 46 acyl carbon atoms increased significantly with the elapse of lactation period, whereas those with more than 50 acyl carbon atoms decreased significantly. The fully saturated trig1ycerides increased significantly as the lactation proceeded, but the dienoic triglycerides declined significantly.

Serum Mineral and Haematobiochemical Profile of Microfilariae Infected Cattle in India: Its Effects on Production and Therapy

  • A survey was under taken of six district of Northern India viz. Bareilly, Pilibhit, Udham Singh Nagar, Nainital, Almora and Rampur. The age, breed, sex and physiological status recorded. A total number of 854 cattle examined out of which lactating (274 cases), non lactating (302 cases) heifers (128 cases), calves (82 cases) and adult male (68 cases) were examined. An incidence of 4.92 percent (42) of microfilarisis was recorded. The highest prevalence was observed in Rudrapur District of Udham Singh Nagar (33.33%, 4/12), followed by Lalkaun in Nanital District (21.74%, 10/46), Rampur (12.50%, 2/16), Bareilly (8.16%, 8/98) and Pilibhit (1.22%, 1/82). No infection was observed in Almora region. Amongst 854 cattle of different group incidence was highest in adult male (12.20%, 10/82), followed by non lactating (3.82%, 12/314) and lactating (2.70%, 2/74), (7.64%, 12/157) was found in Heifers. For haemeto-biochemical, serum minerals estimations and therapeutic study 32 animals suffering from filariasis and 18 healthy animals were taken. 16 animals were treated with ivermectin $@200{\mu}g/kg $ body weight. Effect of this disease on production has also been estimated for which body weight and milk production was observed. The main clinical manifestations observed were anaemia, loss of appetite, debility, oedematous swelling especially in the abdominal region, increased heart rate, and respiration rate. Haematological changes indicated decrease in hemoglobin, total erythrocyte count, packed cell volume, erthrocyte fragility and neutrophil, whereas there was significant increase in erythrocytes sedimentation rate (ESR), total leukocyte count (TLC), lymphocyte and eosinophils. Biochemical changes showed significant reduction in the values of serum albumin, A : G ratio, where as there was significant increase in blood glucose, blood urea nitrogen (BUN), globulin, total lipid, total cholesterol, phospholipids, serum bilirubin. Serum mineral profile also altered markedly, which indicate a significant decrease in Ca, Cu, Fe, Zn, and Mn with increase value of Na and Cl. There was no significant change in P and K values. Enzyme pattern in micro filaria infected animal indicated increased level of AST, ALT, alkaline phosphatase, ornithine carbamyl transferase, sorbitol dehydrogenase, glutamate dehydrogenase, isocitric dehydrogenase and lactate dehydrogenase. In blood gas values and acid/base balance, there was an increase in $PVCo_2$ and $PVo_2$. It has been observed that microfilaria infected cattle showed decrease in body weight and milk production. Animal treated with ivermectin showed the return of these above values toward normalcy.

  • Among 12 kinds of ginsenosides in ginseng saponin, ginsenoside-Rb$_1$was contained the most abundantly. But ginsenoside-Rd which is similar to ginsenoside-Rb$_1$in structure, was known to be superior to ginsenoside-Rb$_1$pharmaceutically. In order to convert ginsenoside-Rb$_1$into ginsenoside-Rd by microbial enzyme treatment, a Rhizopus sp. was selected among various strais of molds found in rotten ginseng roots. Enzyme was prepared from the extract of wheat bran koji culture by ammonium sulfate precipitation (1.0 sat'd) and succeeding ammonium sulfate fractionation method (0.6-0.9 sat'd). For the purpose of use as substrate, saponins were purified by the several purification steps from alcohol extract of red ginseng roots. We obtained the total saponin which was composed of 36.5% of ginsenoside Rb$_1$, 12.2% of ginsenoside-Rd and other ginsenosides. For increase of ginsenoside-Rb$_1$ component ratio, we also obtained further purified ginsenoside-Rb group saponin containing 54.5% of ginsenoside-Rb$_1$, 1.1% of ginsenoside- Rd and other ginsenosides from purified the total saponin. In the enzymatic reaction system including the total saponin or the ginsenoside-Rb group saponin, we confirmed the specific conversion of ginsenoside-Rb$_1$to ginsenoside-Rd proportionally and no change of any other ginsenoside patterns by thin layer chromatography and high performance liquid chromatography.
