Monitoring of Commercial Red Pepper Powders for Their Irradiation Status (물리적 확인시험법을 이용한 시판 유통 중인 고춧가루의 방사선 조사여부 판별 모니터링)

  • Ten commercially available red pepper powders were investigated using photostimulated-luminescence (PSL), thermoluminescence (TL) and electron spin resonance (ESR) analyses to confirm their irradiation status. The application of PSL, TL, and ESR analyses was also confirmed by in-house irradiation. In PSL-based screening, all samples gave negative photon counts (<700 PCs). The PSL calibration dose (1 kGy) showed a low sensitivity of 4 samples, while the others provided reliable screening results. TL glow curves demonstrated maximum peaks after $250^{\circ}C$ for the 6 samples; however 4 samples gave complex TL glow curves with maximum peaks in the range of $185-260^{\circ}C$ (radiation-specific), which could be the effect of an irradiated component in low concentration as the TL ratios of all samples were <0.1. Radiation-specific ESR features were absent in the all commercial samples. Variable irradiation detection properties were found; where the TL analysis showed the possible presence of an irradiated component in 4 samples requiring further monitoring and investigation.

Lamellar Body Counts in Fetal Rabbits' Experimental Diaphragmatic Hernia and Tracheal Ligation (기도결찰을 시행한 토끼 태자의 실험적 횡격막탈장에서 Lamellar body counts)

  • Experimental tracheal ligation (TL) has been shown to reverse the pulmonary hypoplasia associated with congenital diaphragmatic hernia (CDH) and to normalize gas exchange. The purpose of this study was to determine whether the TL would correct the surfactant deficiency present in the fetal rabbit model of CDH by using lamellar body count. Lamellar bodies are synthesized and secreted by the type II pneumocytes of fetal lung. The phospholipids present in these bodies constitute the major component of pulmonary surfactant. Twenty-one pregnant New Zealand rabbits underwent hysterotomy and fetal surgery on gestational day 24. Two fetuses of each pregnant rabbit were operated. In the fetus of one end of bicornuate uterus, left DH was created by excision of fetal diaphragm through open thoracotomy (DH Group). In the fetus of the other end of bicornuate uterus, left DH and TL were created (TL Group). The fetuses were delivered by Cesarean section on gestational day 31. Fourteen in control group, 12 in the DH group and 13 in TL group were born alive. En bloc excision of lungs, bronchi and trachea was done in all newborn rabbits. A five Fr catheter was inserted through trachea and repeated irrigations with 10 cc normal saline were done. The irrigated fluid was centrifuged at $280{\times}g$ for 5 minutes and the lamellar bodies were counted with the upper level fluid in platelet channel of electronic cell counter. The average lamellar body counts were $37.1{\pm}14.2{\times}10^3/{\mu}L$ in control group, $11.5{\pm}4.4 {\times}10^3/{\mu}L$ in DH group, and $6.5{\pm}0.9{\times}10^3/{\mu}L$ in TL group. Lamellar body count in DH group was lower than in control group and did not increase after TL. This study shows TL has no therapeutic effect on decreased surfactant level of CDH and the pregnant rabbit is appropriate for the animal model of CDH.

Algorithm to determine injection position in olive flounder, Paralichthys olivaceus

  • This study aimed to develop an algorithm to determine the optimal injection position in olive flounder, Paralichthys olivaceus when a vaccine is injected into the fish by using a vision-based automatic equipment measruing the total length (TL), width and weight of the fish. Over a 5-month period, 500 olive flounders were examined to analyze the relation of the fish size and to the shape and location of its abdomen, using radiography, a ruler plate and scale. There were significant correlations between the TL and the shape and location of the abdomen. The abdomen was located 0.232TL - 2.7221 mm ($R^2$=0.8787) from the end of the mouth of the fish. The height and width of the abdomen in the fish were 0.1292TL + 1.8768 mm ($R^2$=0.7935) and 0.183TL-5.9791 mm ($R^2$=0.8641), respectively. The injection point in the abdominal region avoiding organs was determined by calculating the center of gravity of the abdomen volume. This can be expressed as g (0.2759TL - 2.0965, 0.1295TL - 4.2325) on the basis of TL line coordinates. The injection point by the expressed coordinates had errors for the x and y axes as 12.15 mm and 8.28 mm, respectively. These were enough to use the algorithm to injection for the equipment. This automated method to determine the position of any part in the fish can also be used for other purposes, for example, intramuscular injection or auto-tagging of fish.

Target strength estimation by tilt angle and size dependence of rockfish (Sebastes schlegeli) using ex-situ and acoustic scattering model (현수법과 모델을 이용한 조피볼락의 유영자세각과 체장에 따른 음향 후방산란강도)

  • Rockfish was a commercially important fish specie in marine ranching areas in Korea. To estimate density and biomass of rockfish using acoustic method, target strength (TS) information is required on the species. This study measured TS dependence on tilt angle and size on 14 live rockfish individuals at 38, 70, and, 120 kHz by ex-situ measurement (tethered method) and acoustic scattering model (Krichhoff ray mode, KRM). The swimbladdered angle ranged from 18 to $30^{\circ}$ ($mean{\pm}s.d.=26{\pm}4^{\circ}$). The mean TS for all individuals was highest -35.9 dB of tilt angle $-17^{\circ}$ at 38 kHz, -35.4 dB of tilt angle $-25^{\circ}$ at 70 kHz, and -34.9 dB of tilt angle $-22^{\circ}$ at 120 kHz. The ex-situ TS-total length (TL, cm) relationships were $TS_{38kHz}=20log_{10}(TL)-67.1$, $TS_{70kHz}=20log_{10}(TL)-68.6$, and $TS_{120kHz}=20log_{10}(TL)-69.9$, respectively. The model TS-total length (TL, cm) relationships were $TS_{38kHz}=20log_{10}(TL)-66.4$, $TS_{70kHz}=20log_{10}(TL)-67.0$, $TS_{120kHz}=20log_{10}(TL)-67.0$. The two measurements between the ex-situ TS and KRM model for TS-tilt angle and fish size were found to be significantly correlated.

Properties of Pulsed Photostimulated Luminescence and Thermoluminescence for Detection of Gamma-Irradiated Teas during Storage

  • Green, black and oolong teas were irradiated by $^{60}$ Co-gamma rays (0~10 kGy) and were investigated for detection of irradiation treatment using pulsed photostimulated luminescence (PPSL) and thermoluminescence (TL) during storage. Teas irradiated at 2.5 kGy or more showed a photon count of greater than 5000 counts/60 sec while the non-irradiated yielded only 650~1000 count/60 sec. Correlation coefficients between irradiation dose and photon counts/60 sec were 0.8951, 0.7934 and 0.9007 for green, black and oolong teas, respectively. The TL glow curves for minerals isolated from the non-irradiated teas were situated at about 30$0^{\circ}C$ with a low intensity, but for irradiated samples were approximately 15$0^{\circ}C$ with a high intensity. The TL ratios (TL$_1$/TL$_2$), calculated from values after initial radiation and then after re-irradiation of the teas, were below 0.1 for the non-irradiated samples and higher than 1.44 for all irradiated samples, enhanced the reliability of the identification results for TL. The signal intensity of PPSL and TL for irradiated teas decreased with the lapse of post-irradiation storage time at room temperature but was still distinguishable from that of the non-irradiated samples even after one year.

Observation of Several Detection Factors Derived from Thermoluminescence of Mineral Separated from Irradiated Korean Sesame and Perilla Seeds Stored under Different Storage Conditions

  • This study was carried out to observe changes in several detection factors derived from thermoluminescence (TL) of minerals separated from irradiated Korean perilla and sesame seeds during storage under normal room and darkroom conditions. The TL intensities of the first glow curves increased from 0 to 5 kGy but only slightly increase from 5 to 10 kGy. Maximum TL temperatures of the first glow curves in all irradiated samples were around 20$0^{\circ}C$, ranging from 150 to 25$0^{\circ}C$. Since the control (0 day of storage) glow curve ratios of G3 and G4, calculated from re-irradiated (1 kGy) sample were over 0.5, detection of irradiation was possible. However, because Gl ratios were below 0.1, they were classified as non-irradiated. There was n unique first glow curve shape that could be clearly seen in all irradiated samples, regardless of storage conditions, that was never seen in non-irradiated samples. In all samples, the maximum TL temperatures and shape of the second glow curve was in a lower temperature range than that of the first glow curve. Therefore, detection of irradiated Korean perilla and sesame seeds was possible fur up to 3 months after irradiation, regardless of storage conditions, by examining several TL detection factors; including TL intensity, glow curve ratios maximum TL temperatures, and the shapes of glow curves.

  • An investigation was made of the dependences of the lattice constants and the energy gap on the composition of $TlGa_xIn_{1-x}Se_2$ single cystals grown by Bridgman method. It was found that a discontinuity in $TlGa_xIn_{1-x}Se_2$ solid solutions occurred in the composition range 0.25$0.0{\leq}X{\leq}0.25$) to the monoclinic structure ($0.65{\leq}X{\leq}1.0$) was observed in this composition range. The temperature dependences of the energy gap and the dielectric constant in $TlGaSe_2$ single crystal have shown that the anomalies appeared at 107 K and 120 K corresponding to first-order and second-order phase transitions, respectively.

Global Transcriptional Analysis Reveals Upregulation of NF-${\kappa}B$-responsive and Interferon-stimulated Genes in Monocytes by Treponema lecithinolyticum Major Surface Protein

  • MspTL is the major surface protein of Treponema lecithinolyticum associated with periodontitis and endodontic infections. Our recent investigation revealed that MspTL induces proinflammatory cytokines and intercellular adhesion molecule 1 in THP-1 cells and periodontal ligament cells. In this study we conducted oligonucleotide microarray analysis to investigate the global transcriptional regulation in THP-1 cells stimulated with purified recombinant MspTL. MspTL upregulated the expression of 90 genes in THP-1 cells at least four fold, and the functions of these genes were categorized into adhesion, apoptosis/antiapoptosis, cell cycle/growth/differentiation, chemotaxis, cytoskeleton organization, immune response, molecular metabolism, proteolysis, signaling, and transcription. The majority of the modified genes are known to be NF-${\kappa}B$-responsive and interferon-stimulated genes (ISGs). The expression of 12 selected genes was confirmed by real-time RT-PCR. Because prostaglandin $E_2(PGE_2)$ is an important inflammatory mediator and Cox-2 was found to be induced by MspTL in the microarray analysis, we determined the level of $PGE_2$ in the culture supernatants of MspTL-treated cells and found that MspTL significantly increased $PGE_2$. Our results provide insight into the gene regulation of host cells in response to MspTL, and may contribute to the understanding of the molecular mechanism in periodontitis.

Determination of Si/Al Ratio of Faujasite-type Zeolite by Single-crystal X-ray Diffraction Technique. Single-crystal Structures of Fully Tl+- and Partially K+-exchanged Zeolites Y (FAU), |Tl71|[Si121Al71O384]-FAU and |K53Na18|[Si121Al71O384]-FAU

  • Large colorless single crystals of faujasite-type zeolite with diameters up to 200 μm have been synthesized from gels with the composition of 3.58SiO2:2.08NaAlO2:7.59NaOH:455H2O:5.06TEA:1.23TCl. Two of these, colorless octahedron about 200 μm in cross-section have been treated with aqueous 0.1 M TlC2H3O2 and KNO3 in order to prepare Tl+- and K+-exchanged faujasite-type zeolites, respectively, and then determined the Si/Al ratio of the zeolite framework. The crystal structures of |Tl71|[Si121Al71O384]-FAU and |K53Na18|[Si121Al71O384]-FAU per unit cell, a = 24.9463(2) and 24.9211(16) A, respectively, dehydrated at 673 K and 1 × 10-6 Torr, have been determined by single-crystal X-ray diffraction techniques in the cubic space group Fd m at 294 K. The two single-crystal structures were refined using all intensities to the final error indices (using only the 905 and 429 reflections for which Fo > 4σ(Fo)) R1/R2 = 0.059/0.153 and 0.066/0.290, respectively. In the structure of fully Tl+-exchanged faujasite-type zeolite, 71 Tl+ ions per unit cell are located at four different crystallographic sites. Twenty-nine Tl+ ions fill site I' in the sodalite cavities on 3-fold axes opposite double 6-rings (Tl-O = 2.631(12) A and O-Tl-O = 93.8(4)o). Another 31 Tl+ ions fill site II opposite single 6-rings in the supercage (Tl-O = 2.782(12) A and O-Tl-O = 87.9(4)o). About 3 Tl+ ions are found at site III in the supercage (Tl-O = 2.91(6) and 3.44(3) A), and the remaining 8 occupy another site III (Tl-O = 2.49(5) and 3.06(3) A). In the structure of partially K+-exchanged faujasite-type zeolite, 53 K+ ions per unit cell are found at five different crystallographic sites and 18 Na+ ions per unit cell are found at two different crystallographic sites. The 4 K+ ions are located at site I, the center of the hexagonal prism (K-O = 2.796(8) A and O-K-O = 89.0(3)o). The 10 K+ ions are found at site I' in the sodalite cavity (K-O = 2.570(19) A and O-KO = 99.4(9)o). Twenty-two K+ ions are found at site II in the supercage (K-O = 2.711(9) A and O-K-O = 94.7(3)o). The 5 K+ ions are found at site III deep in the supercage (K-O = 2.90(5) and 3.36(3) A), and 12 K+ ions are found at another site III' (K-O = 2.55(3) and 2.968(18) A). Twelve Na+ ions also lie at site I' (Na-O = 2.292(10) and O-Na-O = 117.5(5)o). The 6 Na+ ions are found at site II in the supercage (Na-O = 2.390(17) A and O-Na-O = 113.1(11)o). The Si/Al ratio of synthetic faujasite-type zeolite is 1.70 determined by the occupations of cations, 71, in two single-crystal structures.

Growth Characteristics of Cultured Dark-Banded Rockfish, Sebastes inermis Cuvier (양식산 볼락, Sebastes inermis Cuvier의 성장 특성)

  • The objective of this study is to elucidate the growth pattern of dark-banded rockfish, Sebastes inermis Cuvier, produced by aquaculture in Korea. Larvae from 250 maternal full-sibs were reared until 1,350 days after parturition. During the rearing period, growth equations on days after parturition (X) to the total length (TL) and body weight (BW) were estimated as TL=0.0217X $(r^2=0.8867)$ and $BW=0.0001X^2+0.0713X\;(r^2=0.9858)$, respectively. The relationship between BW and TL was $BW=0.0072TL^{3.4373}\;(r^2=0.9945)$. An increases in the eye diameter (ED) of dark-banded rockfish larvae on days after parturition (X) could be expressed as $ED=-2E-5X^2+0.0326X\;(r^2=0.9886)$. Relative growth equations on TL to head length (HL), snout length (SL), ED and postorbital head length (PHL) were calculated as $HL=0.2652TL\;(r^2=0.9758),\;SL=0.0613TL\;(r^2=0.9602),\;ED=0.0752TL\;(r^2=0.9715)$ and $PHL=0.2652TL\;(r^2=0.9501)$, respectively. Since there were few reports on the long-term growth experiment of this species due to difficulties in rearing techniques, results of this study would give a useful information to the biology and commercial culture techniques of dark-banded rockfish.
