나초(糯草) 에탄올추출물이 Human 유래 Jurkat 세포와 THP-1 세포의 알러지 및 염증 사이토카인에 미치는 영향 (Effect of 'DaoCao' on the inflammatory cytokines in Human Jurkat cell and THP-1 cell)

  • Objectives : The aim of this study is to investigate anti-inflammatory activity using various extracts of rice straw (DaoCao) extract (RS).Methods : To investigate the anti-inflammatory effect of RS, we examined the effect of RS on cytokines production on THP-1 cell. Cells were cultured in incubator (37℃, CO25%, 0.5% FBS-RPMI, 1X106cells/ml). One hour after,Dermatophagoides pteronissinus(Dp., 10 ug/ml) was treated into cell and at 6 hour after, each different concentrations(0.1, 1 and 10 ug/ml) of RS were treated. The cells were incubated for 16 hours and harvest the supernatant. The levels of IL-4, IL-5, IL-6, IL-8, MCP-1 and TNF-αwere determined using a commercially available ELISA kit.Results : We investigated whether RS has the inhibition of inflammatory response in Jurkat cells and THP-1 cells. RS suppressed secretion of IL-4, IL-5, and TNF-αinduced by house dust mites in Jurkat cells. It showed significant effects for all concentrations. RS suppressed the increased expression of IL-6, IL-8 and MCP-1 after treatment with mite in THP-1 cells. These results suggest that RS may be used as a valuable agent for treating allergic diseases such as atopy due to its anti-inflammatory property.Conclusions : RS showed significant biological activities with anti-inflammatory in the human T cells. These results suggest that RS may be used as a valuable agent for treating allergic diseases such as atopy due to its anti-inflammatory property. In terms of Korean traditional medicine, we expect the results to contribute to building of EBM (Evidence-Based Medicine).

A Structure-Function Relationship Exists for Ginsenosides in Reducing Cell Proliferation and Inducing Apoptosis in THP-1 Cells

  • Ginsenosides of 20(S)-protopanaxadiol and 20(S)-protopanaxatriol classification including the aglycones, PD, PI and ginsenosides Rh2, Rhl were shown to posses characteristic effects on proliferation of THP-l human leukaemia cells. A similar result was not apparent for ginsenoside Rg3 or dexamathasone. The concentration to inhibit $50\%$ of cells $(LC_{50})$ for PD, Rh2, PI and Rhl were 13 ${\mu}g/mL,\;15{\mu}g/mL,\;19{\mu}g/mL\;and\;210\;{\mu}g/mL$ respectively. Cell cycle analysis showed apoptosis with PD and PI treatment of THP-1 cells resulting in a build up of sub-G1 cells after 24, 48 and 72 hours of treatment. Rh2, and dexamathasone treatments also increased apoptotic cells after 24 hours, where as Rhl did not. After 48 and 72 hours Rh2, Rhl and dexamathasone similarly increased apoptosis, but these effects were significantly (P<0.05) lower than observed for both PD and PI treatments. Furthermore, treatments that produced the largest build up of apoptotic cells were also found to have the largest release of lactate dehydrogenase (LDH). It can be concluded from these studies that the presence of sugars to PD and PI aglycone structure reduces the potency to induce apoptosis, and alternately alter membrane integrity. These cytotoxic effects to THP-l cells were different from dexamethasone.

Chemotactic Effect of the House Dust Mite Allergen, Dermatophagoides pteronyssinus on Human Monocytic THP-1 Cells

  • House dust mites (HDMs) play an important role in the occurrence of allergic diseases such as asthma and atopic dermatitis. Dermatophagoides pteronyssinus (Der p) is one of the most prevalent HDMs. It mediates the activation of T cells and monocytes, and induces the elevation of immunoglobulin E levels in allergic diseases. However, the effects of Der p on human monocytes have not been fully understood. In the present study, we investigated whether or not Der p has a great effect on the chemotactic activity of the human monocytic cell line, THP-1 cells, as induced by CC chemokines. We also show that the Der p extract (DpE) increased the chemotactic activity of THP-1 cells in response to MCP-1, RANTES, MIP-1${\alpha}$, and TARC, but had no effect on the expressions of CC chemokine receptors (CCRs) binding to CC chemokines in THP-1 cells. Protease inhibitors, such as aprotinin and E64, blocked the increased chemotaxis, while cytoplasmic $Ca^{2+}$ influx mediated by these chemokines was inhibited by DpE. These results indicate that DpE increases the chemotactic activity of THP-1 cells in response to CC chemokines by regulating the cells' protease-dependent mechanism. This finding may be useful in identifying the pathogenesis of allergic diseases induced by Der p.

Inhibition of Lipopolysaccharide-stimulated Inflammatory Cytokine Production by LY303511 in Human Macrophagic THP-1 Cells

  • We have previously shown that the specific phosphatidylinositol 3-kinase inhibitor LY294002 (LY29), and its inactive analog LY303511 (LY30), inhibit a monocyte chemoattractant protein-1 (MCP-1) expression in human umbilical vein endothelial cells; these results suggest the potential of LY30 as an anti-inflammatory drug. In this study, we determined the effects of LY30 on the production of various inflammatory cytokines in human macrophagic THP-1 cells which were stimulated with lipopolysaccharide (LPS). LY30 selectively suppressed the mRNA expression of IL-12 p40, $TNF-{\alpha}$, and MCP-1 without affecting the expression of $IL-1{\alpha}$, IL-6, and IL-8. Inhibition of the production of IL-12 and $TNF-{\alpha}$ by LY30 was also demonstrated using ELISA assays. In order to elucidate the mechanisms of the action of LY30, we examined the role played by the mitogen-activated protein kinases and the key transcription factors, AP-1 and $NF-{\kappa}B$ in LPS-stimulated THP-1 cells. The results revealed that LY30 inhibited LPS-induced activation of ERK, but not p38 or JNK. Furthermore, the AP-1 DNA binding activity was suppressed by LY30 based upon the dosage, whereas $NF-{\kappa}B$ DNA binding was not affected. These results suggest that LY30 selectively inhibits cytokine production in the LPS-stimulated macrophagic THP-1 cells by down-regulating the activation of ERK and AP-1.

수질(水蛭)이 THP-1세포의 사이토카인 유전자 발현에 미치는 영향 (Effects of Whitmania pigra on the expressions of cytokine genes in human THP-1 cells)

  • Objective : This study was performed to evaluate the effect of immune reaction inductive substances such as PMA, LPS, DPE, DNCB and WP, the whitman pigra extracting substance at simultaneously on the translocation of $NF{\kappa}B$ towards to the nucleus and the mRNA expression patterns of various cytokine genes in THP-1 cells, monocytes of human. Methods : To analyze the cytokine genes expressions, the RT-PCR method was used, and measuring TNF-${\alpha}$ that had been secreted during cell culture by the ELISA method. The morphological changes were observed during THP-1 cell by a scanning electron microscope and the quantitative distribution of $NF{\kappa}B$ in the cell that was analyzed through immunocytochemistry and a confocal microscopy. Results : WP showed different influences onto the mRNA expression patterns of cytokine genes with PMA, LPS. DPE and DNCB according to the types of immune inductive substances in the THP-1 cells. Upon treating PMA and DPE on the THP-1 cells at the same time or either additionally treating WP thereon, the movement of $NF{\kappa}B$ increase towards the nucleus from cell cytoplasm was able to be observed. The expressions of IL-$1{\alpha}$ and IFN-${\gamma}$ induced by PMA and PMA+DNCB were suppressed by WP while the expression of TGF-${\beta}$ was promoted. Regarding the secretion pattern of TNF-${\alpha}$ according to the treatment of PMA, its secretion amount was incredibly increased by concurrent treatment of WP, however, in case of co-treatment of WP with PMA and DNCB, it was found that the secretion amount of TNF-${\alpha}$ decreased. Conclusions : In this study, the WP extracting substance was confirmed that it had an influence on expression patterns of cytokine genes according to the actions of a variety kinds of immune reaction inductive substances treated on the THP-1 cells. Especially, WP co-treatment with PMA and DNCB was suppressed the expression of inflammatory cytokines, such as IL-$1{\alpha}$, IFN-${\gamma}$ and TNF-${\alpha}$.

상황버섯 균사체 추출물의 면역증진 효능 (Immunostimulating Effect of Mycelium Extract of Phellinus linteus)

  • In order to investigate the immunostimulating effect of mycelia extract of Phellinus linteus (PLM) on human monocyte THP-1 and rat peritoneal macrophage cell, we examined measuring cytokine secretion (IL-6 and TNF-${\alpha}$). The production of IL-6 and TNF-a in human monocyte THP-1 was slight increased dose-dependently when the cells were challenged with PLM for 72 hrs. It was also observed that the treatment of PLM with LPS augmented the production of IL-6 and TNF-a in human monocyte THP-1. It was also observed that the treatment of PLM with LPS augmented the production of IL-6 and TNF-${\alpha}$ in human monocyte THP-1. The production of IL-6 and TNF-${\alpha}$ in rat peritoneal macrophage was significantly enhanced when the cells were treated PLM with LPS for 72 hrs. Moreover, the proliferation rate of rat spleen cells was increased in a dose dependent manner as the cells were treated with PLM and Concanavalin A.

  • Purpose: Interleukin-8 (IL-8) is an important mediator of immune and inflammatory reactions and is produced by a variety of different cell types. This study was undertaken to investigate the effects of lipopolysaccharides (LPSs) from Prevotella intermedia and Prevotella nigrescens, the major causes of inflammatory periodontal disease, on the production of IL-8 and the expression of IL-8 mRNA in differentiated THP-1 cells, a human monocytic cell line. Methods: LPSs from P. intermedia ATCC 25611 and P. nigrescens ATCC 33563 were prepared by the standard hot phenol-water method. THP-1 cells were incubated in the medium supplemented with phorbol myristate acetate to induce differentiation into macrophage-like cells. Results: We found that LPS preparations from P. intermedia and P. nigrescens can induce IL-8 mRNA expression and stimulate the release of IL-8 in differentiated THP-1 cells without additional stimuli. Conclusions: There are no previous reports of the ability of P. intermedia and P. nigrescens LPS to stimulate the release of IL-8, and the present study clearly shows, for the first time, that LPSs from P. intermedia and P. nigrescens fully induced IL-8 mRNA expression and IL-8 production in differentiated human monocytic cell line THP-1. The ability of P. intermedia and P. nigrescens LPS to promote the production of IL-8 may be important in the pathogenesis of inflammatory periodontal disease.

Ursodeoxycholic Acid Inhibits Inflammatory Cytokine Expression in THP-1 Cells Infected with Aggregatibacter actinomycetemcomitans

  • Background: Periodontitis is an inflammatory disease characterized by the breakdown of tooth-supporting tissues, leading to tooth loss. Aggregatibacter actinomycetemcomitans are major etiologic bacterium causing aggressive periodontitis. Ursodeoxycholic acid (UDCA), a hydrophilic gall bladder acid, has been used as an effective drug for various diseases related to immunity. The aim of this study was to investigate the effect of UDCA on the inflammatory response induced by A. actinomycetemcomitans. Methods: A human acute monocytic leukemia cell line (THP-1) was differentiated to macrophage- like cells by treatment with phorbol 12-mystristate 13-acetate (PMA) and used for all experiments. The cytotoxic effect of UDCA was examined by MTT assay. THP-1 cells were pretreated with UDCA for 30 min before A. actinomycetemcomitans infection and the culture supernatant was analyzed for various cytokine production by ELISA. The effect of UDCA on bacterial growth was examined by measuring optical densities using a spectrophotometer. Results: UDCA showed no cytotoxic effect on THP-1 cells, up to $80{\mu}M$ Ed highlight: Please confirm technical meaning. UDCA pretreatment inhibited the A. actinomycetemcomitans-induced $IL-1{\beta}$, $TNF-{\alpha}$, and IL-17A secretion in a dose-dependent manner. UDCA also inhibited IL-21 production at $60{\mu}M$. The production of IL-12 and IL-4 was not influenced by A. actinomycetemcomitans infection. Conclusion: These findings indicate that UDCA inhibits the production of inflammatory cytokines involved in innate and Th17 immune responses in A. actinomycetemcomitans-infected THP-1- derived macrophages, which suggests its possible use for the control of aggressive periodontitis.

Protein Kinase C-delta Stimulates Haptoglobin Secretion

  • Haptoglobin (Hp) is a glycoprotein that is produced by hepatic cells and secreted into the circulation. While studying the physiologic functions of Hp, we found that Hp synthesized in THP-1 monocytic cells was largely retained within cells, although Hp is considered a secretory protein. To investigate the molecular mechanism on Hp secretion in THP-1 cells, in the present study, we examined the effect of protein kinase C (PKC) on Hp secretion. When several inhibitors of PKC isoforms were tested, only Rottlerin, a specific inhibitor of PKC-$\delta$, completely blocked Hp secretion from cells to culture medium. To confirm the role of PKC-$\delta$ in Hp secretion, Hp-overexpressing COS7 cells were transiently transfected with a wild-type or a dominantnegative mutant of the PKC-$\delta$ gene. Mutant PKC-$\delta$ significantly inhibited Hp secretion, whereas the wild-type gene slightly increased Hp secretion. These results demonstrate that the PKC-$\delta$ signal is involved in Hp secretion.

단핵구세포주 THP-1의 분화과정에서 Ref-1의 역할 (The Role of Ref-1 in the Differentiation Process of Monocytic THP-1 Cells)

  • Redox factor (Ref)-1은 세포질과 핵을 오가며 산화환원(redox) 환경에 민감한 transcription factors의 조절과 손상된 DNA의 교정 등 다양한 기능을 수행하는 단백질이다. 하지만, 단핵구(monocyte)의 대식세포(macrophage)로의 분화과정에서 Ref-1의 역할은 잘 알려져 있지 않다. 본 연구에서는 인간 단핵구세포주 THP-1을 이용하여 Ref-1의 단핵구 분화과정에 미치는 영향을 조사하였다. 분화제 phorbol 12-myristate 13-acetate (PMA)는 시간이 지날수록 세포의 부착능을 증가시키고 포식기능의 현저한 증가를 보이지만, Ref-1의 세포 내 양을 현저히 감소시켰다. Ref-1의 억제제인 E3330와 siRNA 기법을 이용한 Ref-1 knock- down은 PMA에 의한 세포 부착능과 막표면 분화인자의 발현을 현저히 감소시켰다. 이는 PMA에 자극을 받은 THP-1 세포의 분화 초기과정에는 Ref-1의 역할이 절대적으로 필요하다는 것을 의미한다. 단핵구 분화과정에서 Ref-1의 작용기전을 조사하기 위하여, PMA로 자극한 THP-1 세포의 세포질과 핵에서 Ref-1의 분포를 조사하였다. 놀랍게도, PMA 자극은 Ref-1을 빠르게 핵으로 이동하는 결과를 나타내었다. Ref-1의 핵으로의 이동이 단핵구 분화에 필요함을 증명하기 위하여, nuclear localization sequence (NLS)가 제거된 Ref-1 vector를 사용하였다. 그 결과, 핵으로의 이동이 제한된 ∆NLS Ref-1의 과발현은 PMA 자극에 의한 막표면 단백질의 발현 억제와 포식기능의 현저한 감소를 보였다. 이를 종합하면, Ref-1은 분화제 자극에 의한 분화 유도 초기과정에 핵으로 이동하여 다양한 분화인자의 발현에 관여하는 것으로 보인다.
