  • (Z)-12-Tetradecen-1-yl acetate (1), (E)-12-tetradecen-1-yl acetate (2) and tetradecan-1-yl acetate (3), the active components of the sex pheromone of the Asian Corn Borer moth were synthesized by two different methods, one from acetylene and 11-bromo-1-undecanol THP ether by acetylenic route, the other from 12-acetoxy-1-dodecanal and ethylidenetriphenylphosphonium ylide by Wittig route.

The Chemokine SDF-1α Suppresses Fibronectin-mediated In Vitro Lymphocytes Adhesion

  • Stromal cell-derived factor (SDF-1) is a CXC chemokine that selectively activates the CXCR4 chemokine receptor. Fibronectin is an intracellular matrix component that binds integrin and mediates cell-matrix adhesion. Activation of the integrin receptor can occur in two ways: by ligand binding (outside-in signaling), and in response to intracellular events (inside-out signaling). In the current study we showed that SDF-$1{\alpha}$ inhibited adhesion of T lymphocyte Jurkat cells resulting from binding high concentrations of fibronectin as well as that of THP-1 monocytes. The effect of SDF-$1{\alpha}$ on fibronectin-mediated adhesion was partly reversed by the CXCR4 receptor antagonist T140. Our results suggest that an SDF-1/CXCR4 signal pathway modulates fibronectin-mediated lymphocytes adhesion.

Inhibitory Effects of Licochalcone A and Isoliquiritigenin on Monocyte Adhesion to TNF-$\alpha$-activated Endothelium

  • Numerous natural herbal compounds have been reported to inhibit adhesion and migration of leukocytes to the site of inflammation Licorice extracts, which have been widely used in traditional Chinese medicinal preparation, possess various pharmacological effects. Isoliquiritigenin, a biogenetic precursor of flavonoids with various pharmacological effects, is a natural pigment present in licorice. We attempted to explore whether licorice extracts and isoliquiritigenin mitigate monocyte adhesion to tumor necrosis factor-$\alpha$ (TNF-$\alpha$)-activated human umbilical vein endothelial cells (HUVEC). In addition, it was tested whether the inhibition of monocyte adhesion to the activated HUVEC accompanied a reduction in vascular cell adhesion molecule-l expression(VCAM-l). Dry-roasted licorice extracts in methylene chloride but not in ethanol markedly interfered with THP-l monocyte adhesion to INF-$\alpha$-activated endothelial cells. licochalcone A compound isolated from licorice extract in methylene chloride appeared to modestly inhibit the interaction of THP-l monocytes and activated endothelium. In addition, isoliquiritigenin abolished the monocyte adhesion with attenuating VCAM-l protein expression on HUVEC induced by INF-$\alpha$. These results demonstrated that non-polar components from dry-roasted licorice extracts containing licochalcone A as well as isoliquiritigenin were active in blocking monocyte adhesion to cytokine-activated endothelimn, which appeared to be mediated most likely through the inhibition of VCAM-l expression on HUVEC. Therefore, licorice may hamper initial inflammatory events on the vascular endothelium involving induction of endothelial cell adhesion molecules.

폴리벤조옥사졸 전구체의 광투과도 연구 (Optical Transmittance of Polybenzoxazole Precursor)

  • 폴리벤조옥사졸 전구체로써 poly(ο-hydroxyamide)를 2,2'-bis(3-amino-4-hydroxy phenyl)hexafluoropropane과 여러 가지 bis-acid를 사용하여 축중합법에 의해 합성하였으며, 또한 이를 3,4-dihydro-2H-pyran과 부가반응시켜 산민감기인 tetrahydropyran이 부착된 방향족 폴리아미드를 합성하였다. Bis-acid의 구조에 따른 365 nm의 파장에서의 광투과도를 조사한 결과, 4,4'-oxydibenzoic acid로부터 합성된 중합체의 광투과도가 가장 우수하였다. 이러한 현상은 전자받게 성질을 갖고 있는 his-acid에 전자를 공여할 수 있는 구조를 도입하면 전자받게 성질이 감소되어 분자내 전하 이동 착물 (intra-CTC) 형성이 감소됨에 따라 광투과도가 증가된다고 사료된다. 또한 산민감기의 치환율이 높을수록 광투과도가 증가하는 경향을 보였다. 이는 방향족 폴리아미드에 산민감기인 THP의 치환율이 높아질수록 사슬과 사슬간의 조밀함이 떨어지기 때문에 분자간 전하 이동 착물 (inter-CTC) 형성을 줄여주는 효과를 얻게 되어 광투과도가 증가된다고 사료된다.

Oxidized LDL induces phosphorylation of non-muscle myosin IIA heavy chain in macrophages

  • Oxidized LDL (oxLDL) performs critical roles in atherosclerosis by inducing macrophage foam cell formation and promoting inflammation. There have been reports showing that oxLDL modulates macrophage cytoskeletal functions for oxLDL uptake and trapping, however, the precise mechanism has not been clearly elucidated. Our study examined the effect of oxLDL on non-muscle myosin heavy chain IIA (MHC-IIA) in macrophages. We demonstrated that oxLDL induces phosphorylation of MHC-IIA (Ser1917) in peritoneal macrophages from wild-type mice and THP-1, a human monocytic cell line, but not in macrophages deficient for CD36, a scavenger receptor for oxLDL. Protein kinase C (PKC) inhibitor-treated macrophages did not undergo the oxLDL-induced MHC-IIA phosphorylation. Our immunoprecipitation revealed that oxLDL increased physical association between PKC and MHC-IIA, supporting the role of PKC in this process. We conclude that oxLDL via CD36 induces PKC-mediated MHC-IIA (Ser1917) phosphorylation and this may affect oxLDL-induced functions of macrophages involved in atherosclerosis.

Antioxidant and Anti-inflammatory Activities of Allium victorialis subsp. platyphyllum Extracts

  • This study was conducted to investigate antioxidant activity and anti-immunological inflammatory effect of Allium victorialis subsp. platyphyllum extracts (AVPEs). Antioxidant activities of AVPEs were determined by free radical scavenging assay and reducing power test. Leaf-part extract had comparatively better antioxidant activity than other-part extracts. Antioxidant activity of extracts had protective effect for human umbilical vein endothelial cells (HUVECs) against superoxide anions secreted from activated neutrophils. Also, we observed AVPEs had inhibitory effects on the adherence of monocytic THP-1 to HUVEC monolayer to the basal level. Inhibitory effect on cell adhesion was caused by suppression of tumor necrosis factor-${\alpha}\;(TNF-{\alpha})-upregulated$ expression of vascular cellular adhesion molecule-1 (VCAM-1) and E-selectin in HUVECs. From these results, we expect to support the evidence of anti-immunological inflammatory effects of Allium victorialis subsp. platyphyllum (AVP) as a Korean traditional pharmaceutical.

종양괴사인자에 의하여 유도된 혈관내피세포의 Cell Adhesion Molecules 발현을 억제시키는 플라보노이드 선별 (Selection of Flavonoids Inhibiting Expression of Cell Adhesion Molecules Induced by Tumor Necrosis Factor- a in Human Vascular Endothelial Cells)

  • 염증성 cytokines의 분비 또는 혈관손상으로 인한 백혈구의 adhesion과 transmigration을 통하여 죽상경화과정이 시발되는데, 본 연구에서는 이러한 죽상경화의 초기과정에서 플라보노이드가 억제작용을 발휘하는 지를 규명하고자 하였다. 본 연구에서는 화학적인 구조가 서로 다른 플라보노이드를 사용하여 화학적인 구조와 항동맥경화작용과의 상관성을 착인하였다. TNF-$\alpha$는 혈관내피세포를 활성화시켜 THP-1 단핵구의 adhesion을 유의적으로 증가시켰다. 여러형태의 플라보노이드를 전처리하고 TNF-$\alpha$를 가하여 혈관내피세포를 활성화 시 켰을 때, flavonols인 quercetin과 flavones의 luteolin과 apigenin은 THP-1 단핵구의 adhesion억제효과를 보여주었다. 그러나, catechins과 flavanones의 플라보노이드는 이러한 억제효과를 전혀 보여주지 못하였다. 이러한 adhesion 억제작용을 가지는 플라보노이드는 CAMs 단백질의 발현도 차단시킨다는 것을 확인할 수 있었다. Quercetin, luteolin과 apigenin은 TNF-$\alpha$에 의하여 증가된 VCAM-1, ICAM-1 및 E-selectin의 단백질 발현을 일률적으로 감소 또는 차단시켰다. 그 대신, 단핵구의 adhesion을 차단시키지 못한 (-)epigallo-catechin gallate와 (+)catechin은 TNF-$\alpha$에 의한 이러한 CAMs의 발현을 전혀 억제시키지 못하였다. 또한 quercetin, luteolin과 apigenin의 CAMs단백질 발현 억제작용은 유전자 전사단계에서 mRNA의 down-regulation으로 인하여 나타난다는 사실을 알 수 있었다. 결론적으로 quercetin, luteolin, apigenin과 같은 플라보노이드는 TNF-$\alpha$와 같은 염증성 cytokines에 의한 단핵구의 adhesion을 혈관내피세포의 CAMs 단백질 발현을 억제하므로서 차단시킨다는 것이 확인되었다. 여기서 모든 플라보노이드가 이러한 활성을 다 지니고 있지 않아서 화학적인 구조와 초기 항동맥경화작용에는 서로 연관성이 있다는 것이 제시되었다. 또한, 선별된 플라보노이드의 초기 항동맥경화작용은 활성산소를 소거하는 플라보노이드의 항산화능과는 무관한 것 같다고 할 수 있다.

IL-12 Regulates B7-H1 Expression in Ovarian Cancer-associated Macrophages by Effects on NF-κB Signalling

  • Background and Aim: B7-H1, a co-inhibitory molecule of the B7 family, is found aberrantly expressed in ovarian cancer cells and infiltrating macrophage/dendritic-like cells, and plays a critical role in immune evasion by ovarian cancer. IL-12, an inducer of Th1 cell development, exerts immunomodulatory effects on ovarian cancer. However, whether IL-12 regulates B7-H1 expression in human ovarian cancer associated-macrophages has not been clarified. Therefore, we investigated the effects of IL-12 on the expression of B7-H1 in ovarian cancer-associated macrophages and possible mechanisms. Methods: PMA induced THP-1-derived macrophages or human monocyte-derived macrophages were treated with recombinant IL-12 (rIL-12) or infected with adenovirus carrying human IL-12 gene (Ad-IL-12-GFP) for 24 h, then cocultured with the SKOV3 ovarian cancer cell line for another 24 h. Macrophages were collected for real-time PCR and Western blot to detect the expression of B7-H1, and activation of the NF-${\kappa}B$ signaling pathway. Moreover, supernatants were collected to assay for IL-12, IFN-${\gamma}$ and IL-10 by ELISA. In addition, monocyte-derived macrophages treated with IFN-${\gamma}$ were cocultured with SKOV3 and determined for the expression of B7-H1. Furthermore, the expression of B7-H1 in monocyte-derived macrophages was also evaluated after blocking NF-${\kappa}B$ signaling. Results: The expression of B7-H1 was significantly upregulated in monocyte-derived macrophages treated with rIL-12 or Ad-IL-12-GFP compared with the control groups (p<0.05), accompanied by a remarkable upregulation of IFN-${\gamma}$ (p<0.05), a marked downregulation of IL-10 (p<0.05) and activation of NF-${\kappa}B$ signaling. However, the upregulation of B7-H1 was inhibited by blocking the NF-${\kappa}B$ signaling pathway (p<0.05). Expression of B7-H1 was also increased (p<0.05) in monocyte-derived macrophages treated with IFN-${\gamma}$ and cocultured with SKOV3. By contrast, the expression of B7-H1 in THP-1-derived macrophages was significantly decreased when treated in the same way as monocyte-derived macrophages (p<0.05), and IL-10 was also significantly decreased but IFN-${\gamma}$ was almost absent. Conclusions: IL-12 upregulates the expression of B7-H1 in monocyte-derived macrophages, which is possible though inducing the secretion of IFN-${\gamma}$ and further activating the NF-${\kappa}B$ signal pathway. However, IL-12 downregulates the expression of B7-H1 in THP-1-derived macrophages, associated with a lack of IFN-${\gamma}$ and inhibition of expression of IL-10.

별갑이 아토피 피부염에서의 알러지성 염증 반응에 미치는 영향 (Effects of Amyda sinensis on Allergic Inflammation Mechanism related Atopy Dermatitis)

  • Objectives: This study aimed to investigate the effects of Amyda sinensis (AS) on allergic inflammation mechanism related atopy dermatitis. Methods: To investigate the effects of AS, We study inhibitory effect of AS on the levels of pro-inflammatory cytokines released from Raw264.7 cell stimulated with LPS (lipopolysaccaride), and EoL-1, THP-1, Jutkat cell stimulated with DP (Dermatophagoides pteronyssinus), and LPS indused acute inflammatory BALB/c mouse model. Result: AS reduced the levels of IL-$1{\beta}$ released from Raw264.7 cell stimulated with LPS at 20 ug/ml, 5 ug/ml concentration, and reduced the levels of IL-6 in a dose-dependent. AS significantly reduced the levels of MCP-1 released from EoL-1 cell stimulated with DP (Dermatophagoides pteronyssinus) at all the concentration, and significantly reduced the level of IL-8 at 0.1 ug/ml concentration. AS significantly reduced the levels of MCP-1 released from THP-1 cell stimulated with DP (Dermatophagoides pteronyssinus) at 1 ug/ml concentration, and reduced the level of IL-6 in a dose-dependent. AS significantly reduced the levels of IL-4 released from Jutkat cell stimulated with DP at all the concentration, and significantly reduced the level of IL-5 at 0.1 ug/ml. 1 ug/ml concentration,. and reduced the level of TNF-${\alpha}$ in a dose-dependent. AS significantly reduced the levels of TNF-${\alpha}$, IL-6, IL-$1{\beta}$, in LPS indused acute inflammatory BALB/c mouse model, in a dose-dependent. Conclusion: These result suggested that AS has suppressive effect on pro-inflammatory cytokines in various cell lines through the regulation of immune system. AS could be applied on the medicinal sources for treatment of immune abnormal diseases such as atopy dermatitis afterward.
