삼원공중합에 있어서 공단량체 및 여러 가지 중합조건이 미치는 영향 (Effects of Comonomer and Various Polymerization Conditions on Terpolymerization)

  • 본 연구에서는 다리 구조의 메탈로센 촉매인 rac-$Et[Ind]_2ZrCl_2$를 이용하여 새로운 구조의 poly(ethylene-ter-1-hexene-ter-divinylbenzene) 삼원공중합체를 제조하였다. 공단량체인 1-hexene이 중합에 미치는 효과에 관한 연구를 수행하였다. 여러 가지 중합 조건의 영향을 살펴보았는데 공촉매/촉매 몰비가 3,000일 때, 촉매활성도는 8,000이 넘는 매우 높은 수준의 활성도를 보여주었다. 또한, 중합 시간에 따라 중량 평균 분자량이 일정 수준까지 증가하는 경향을 나타내었고, 중합시간이 50분일 때는 무정형 상태에 가까워지는 것을 확인하였다. 삼원공중합체의 중량 평균 분자량은 110,000-200,000, 밀도는 $0.85-0.89g/cm^3$ 수준이었다. 또한, 삼원 공중합체의 열적 성질과 구조를 확인하였다.

  • rac-$Et(Ind)_2ZrCl_2$ 메탈로센 촉매와 메틸알루미늄옥산 공촉매를 이용하여 polyethylene, poly(ethylene-co-1-decene), poly(ethylene-co-p-methylstyrene) 및 poly(ethylene-ter-1-decene-ter-p-methystyrene)를 합성하였다. $^{13}C$ NMR과 $^1H$ NMR 및 FT-IR을 이용하여 삼원공중합체의 특성을 분석하였다. 삼원공중합의 최적조건을 확립하기 위해 동일한 중합조건을 유지한 채 촉매량, 공촉매/촉매 몰비, 중합 시간 및 중합 온도를 변경하여 실험하였다. 촉매량이 증가할수록 삼원공중합체의 촉매활성도 및 중량평균분자량은 증가하였으나, 중합시간이 30분을 초과하자 촉매활성도는 감소하였다. 공촉매/촉매 몰비를 증가시킨 결과 중량평균분자량은 감소하였고, 촉매활성도는 어느 정도 증가하였다. 촉매활성도는 중합온도가 증가함에 따라 상승하였으나 중량평균 분자량은 감소하였다.

  • 지르코늄계 메탈로센 촉매/Borate계 공촉매 시스템을 이용하여 poly(ethylene-ter-high ${\alpha}$-olefin-ter-p-methylstyrene)를 합성하였으며 이 때 사용한 high ${\alpha}$-olefin인 1-hexene, 1-octene, 1-decene, 1-dodecene의 사슬길이 변화가 삼원공중합체에 미치는 여러 가지 영향을 분석하였다. $^{13}C$ NMR과 $^1H$ NMR을 이용하여 삼원공중합체의 구조 및 조성을 확인하였고 high ${\alpha}$-olefin 사슬길이가 촉매활성도, 수율, 분자량 및 분자량분포도 등에 미치는 영향을 살펴보았다. 또한, 사슬 길이 변화에 따른 삼원공중합체의 morphology 변화, 결정화도 변화 및 열적 거동을 비교하였다.

Kinetics of Malonyl-CoA Synthetase from Rhizobium trifolii and Evidences for Malonyl-AMP Formation as a Reaction Intermediate

  • The catalytic mechanism of malonyl-CoA synthetase from Rhizobium trifolii was investigated by the steady state kinetics and intermediate identification. Initial velocity studies and the product inhibition studies with AMP and PPi strongly suggested ordered Bi Uni Uni Bi Ping-Pong Ter Ter system as the most probable steady state kinetic mechanism of malonyl-CoA synthetase. Michaelis constants were $0.17{\pm}0.04 {\mu}M,\;0.24{\pm}0.18 {\mu}M\;and\;0.045{\pm}0.26 {\mu}$M for ATP, malonate and CoA, respectively. The TLC analysis of the $^{32}P-labelled$ products in reaction mixture containing $[{\gamma}-^{32}P]$ ATP in the absence of CoA showed that PPi was produced after the sequential addition of ATP and malonate. Formation of malonyl-AMP, suggested as an intermediate in the kinetically deduced mechanism, was confirmed by the analysis of $^{31}P-NMR$ spectra of AMP product isolated from the $^{18}O$ transfer experiment using $[^{18}O]$malonate. Two resonances were observed, corresponding to AMP labelled with zero and one atom of $^{18}O$, indicating that one atom of $^{18}O$ transferred from $[^{18}O]$malonate to AMP through the formation of malonyl-AMP. Formation of malonyl-AMP was also confirmed through the TLC analysis of reaction mixture containing $[{\alpha}-^{32}P]$ATP. These results strongly support the ordered Bi Uni Uni Bi Ping-Pong Ter Ter mechanism deduced from the initial velocity and product inhibition studies.

  • 본 연구에서는 가중주성분분석으로부터 정준대응분석을 유도하는 Legendre와 Legendre (2012)의 알고리즘을 고찰하였다. 그리고, 가중주성분분석에 기반한 Legendre와 Legendre (2012)의 정준대응분석이 가우시안 반응모형에 기초한 Ter Braak (1986)의 정준대응분석과 동일함을 다루었다. 생태학에서 종의 발현 정도를 잘 설명할 수 있는 가우시안 반응곡선에서 도출된 Ter Braak (1986)의 정준대응분석은 종 패킹 모형(species packing model)이라는 기본 가정을 사용한 후 일반화선형모형과 정준상관분석을 결합시키는 방법으로 도출된다. 그런데 Legendre와 Legendre (2012)의 알고리즘은 이러한 가정없이 Benzecri의 대응분석과 상당히 유사한 방법으로 계산되는 특징을 지닌다. 그러므로 가중주성분석에 기초한 정준대응분석을 사용하면, 결과물 활용에 약간의 유연성을 지닐 수 있게 된다. 결론적으로 본 연구에서는 서로 다른 모형에서 출발한 두 방법이 장소점수(site score), 종 점수(species score) 그리고 환경변수와의 상관관계가 서로 동일함을 보인다.

High mRNA expression of GABA receptors in human sperm with oligoasthenoteratozoospermia and teratozoospermia and its association with sperm parameters and intracytoplasmic sperm injection outcomes

  • Objective: This study investigated the mRNA expression of gamma-aminobutyric acid (GABA) receptors in the sperm of oligoasthenoteratozoospermic (OAT) and teratozoospermic (TER) men compared to normozoospermic (NOR) men, as well as the relationships between GABA receptor expression and sperm parameters, fertilization rate, and embryo quality. Methods: The mRNA expression of GABA A-α1 and GABA B-R2 receptors in sperm was examined using reverse transcription-polymerase chain reaction in three groups of patients: NOR (n=32), OAT (n=22), and TER (n=45). The fertilization rate and embryo quality were assessed in 35 patients undergoing intracytoplasmic sperm injection (ICSI; 10 NOR, 10 OAT, and 15 TER men). Results: OAT men had significantly higher mRNA expression of GABA A-α1 and GABA B-R2 receptors in sperm than NOR men; however, the difference between TER and NOR men was not significant. High levels of these receptors were significantly correlated with low sperm concentration, motility, and morphology, as well as the rate of good-quality embryos (GQEs) at the cleavage stage after ICSI. Patients whose female partners had a >50% GQE rate at the cleavage stage had significantly lower levels of GABA A-α1 receptor expression than those whose partners had a ≤50% GQE rate. Conclusion: Our findings indicate that mRNA levels of GABA receptors in human sperm are correlated with poor sperm quality and associated with embryo development after ICSI treatment. The GABA A-α1 receptor in sperm has a stronger relationship with embryo quality at the cleavage stage than the GABA B-R2 receptor.

당뇨병성 망막병증에 대한 보양환오탕(補陽還五湯)의 효과: 체계적 문헌 고찰 및 메타분석 (The Effect of Boyanghwano-tang for Diabetic Retinopathy: A Systematic Review and Meta-Analysis)

  • Objectives : The purpose of this study is to assess the effect of using Boyanghwano-tang(BYHWT) for diabetic retinopathy. Methods : We searched randomized controlled trials(RCTs) that used BYHWT for diabetic retinopathy in 10 databases(RISS, KISS, ScienceON, KCI, OASIS, DBpia, CNKI, Wanfang Database, PubMed, Cochrane Library) on August 12, 2023. Outcome measurements included total effective rate(TER) and visual acuity as the primary outcome measurements, and the secondary outcome measurements included fasting blood glucose(FBG), 2hPG, HbA1c, ET-1, NO, etc. The indicator of stability included adverse reactions. A meta-analysis was conducted according to TER, visual acuity, FBG. Results : A total of 8 randomized control trials were selected. The treatment group which used BYHWT showed significant improvement effects in TER and visual acuity compared to the control group which used western medicine or chinese patent medicine alone. TER: (RR: 1.22, CI 1.14 to 1.31, P=<0.00001), visual acuity: (RR: 2.06, CI 1.40 to 3.03, P=0.0002). Secondary outcome measurements including FBG also showed improvement in the treatment group compared to the control group. FBG: (MD: -0.15, CI -0.43 to 0.31, P=0.28). Adverse reactions were only mentioned in one study, which showed that no adverse reactions occurred in both the treatment and control groups. Conclusions : This study suggests that BYHWT is effective for diabetic retinopathy. However these findings should be interpreted cautiously due to the insufficient number of studies and unknown or high risk of bias in the included trials.

Development of a Zebrafish Larvae Model for Diabetic Heart Failure With Reduced Ejection Fraction

  • Background and Objectives: Diabetes mellitus (DM)-associated heart failure (HF) causes high morbidity and mortality. In this study, we established a zebrafish larvae model for in vivo research on diabetic HF. Methods: DM-like phenotypes were induced by treating zebrafish larvae with a combination of D-glucose (GLU) and streptozotocin (STZ). HF was induced by treatment with terfenadine (TER), a potassium channel blocker. Additionally, myocardial contractility, motility, and viability were evaluated. Results: The zebrafish larvae treated with a combination of GLU and STZ showed significantly higher whole-body glucose concentrations, lower insulin levels, and higher phosphoenolpyruvate carboxykinase levels, which are markers of abnormal glucose homeostasis, than the group treated with only GLU, with no effect on viability. When treated with TER, DM zebrafish showed significantly less myocardial fractional shortening and more irregular contractions than the non-DM zebrafish. Furthermore, in DM-HF with reduced ejection fraction (rEF) zebrafish, a significant increase in the levels of natriuretic peptide B, a HF biomarker, markedly reduced motility, and reduced survival rates were observed. Conclusions: We established a DM-HFrEF zebrafish model by sequentially treating zebrafish larvae with GLU, STZ, and TER. Our findings indicate the potential utility of the developed zebrafish larvae model not only in screening studies of new drug candidates for DM-HFrEF but also in mechanistic studies to understand the pathophysiology of DM-HFrEF.

고속액체크로마토그라피에 의한 백지근 중 Byakangelicin의 정량 (Determination of Byakangelicin in Angelicae dahuricae Radix by High Performance Liquid Chromatography)

  • A new method for quantitative determination of byakangelicin in Angelicae dahuricae Radix by high performance liquid chromatography was established. A reversed-phase system with a ${\mu}Bondapak$ $C_{18}$ column using THF : dioxane : MeOH : HAc : 5% $H_3PO_4$ : $H_2O$=72.5 : 62.5 : 25 : 10 : 1 : 329 as a mobile phase was developed. Byakangelicin together with ter-O-byakangelicin and oxypeucedanin methanolate, and isooxypeucedanin as an internal reference were detected at 350 nm and the analysis was successfully carried out within 30 min.
