연성 이종 재료 시료의 상온 절편 제작법 (Sample Preparation of Ductile Heterogeneity Materials by Ultramicrotomy)

  • For TEM study of biological samples or polymers that are contained in organic structure, it is often required that the sample is prepared by using ultramicrotome and stained with proper agents to increase the contrast of organic structure. In this study, we investigated an efficient TEM sample preparation method for ductile heterogeneity material by using ultramicrotomy. Cryo-ultramicrotomy is a suitable method that is capable of rendering sample hardness for various ductile materials. However, it has several factors to consider, such as experimental cost, working time and finding the optimal staining conditions. To satisfy these considerations, we prepared TEM sample by using ultramicrotome without cryofunction, and secured the sample hardness by applying the staining process prior to ultrathin sectioning. The cross-linked polyethylene structure in the sample was stained with the 2% $RuO_4$ solution in a sealed test tube for 24 hours at $4^{\circ}C$. After the sample staining, ultrathin sections of sample were prepared using ultramicrotome. As a result, it was revealed that the difficulties associated with staining of ultrathin sections prepared by low-temperature conditions were improved. In addition, appropriate staining depth of sample could be selected for sectioning process. The quality of TEM sample obtained by using this method was better than that of cryo-ultramicroscopy. Finally, it is expected that our method could be effectively applied in TEM sample preparation for a variety of nano-bio convergence materials.

Microstructure and Third Order Optical Nonlinearities of Ion-Implanted and Thermally Annealed $Cu-SiO_2$ Thin Films

  • The crystal structure and optical properties of copper nanoparticles, prepared in fused silica by ion-implantation and subsequent heat-treatment, were characterized by X-ray, TEM, linear absorption, and degenerate four-wave mixing (DFWM) technique. The X-ray data show fcc lattice structure of the nanocrystals and their size was measured as 8-20 nanometer by high resolution TEM. Using DFWM, the third-order nonlinear optical coefficient of the Cu-SiO2 thin films was measured as 0.4-1.1×10-8 esu in the surface plasmon resonance absorption region (540-570 nm).

Bacillus thuringiensis BT-1, BT-2가 생산하는 δ-endotoxin의 특성 규명 (Characterization of a δ-endotoxin produced by Bacillus thuringiensis BT-1, BT-2.)

  • B. thuringiensis는 많은 해로운 곤충들을 박멸시키는데 널리 사용되는 생물학적 살충제로 잘 알려져 있다. 그것은 측포자 형태의 결정체($\delta$-내독소)를 생산하고 내생포자들을 형성한다. 본 논문에서는 B. thuringiensis BT-1과 BT-2에 의해 생산되는 내독소의 특성을 주사전자현미경(SEM), 투과전자현미경(TEM), SDS-PACE, 알칼리 용액에서의 용해 활성을 통하여 규명하였다. BT-1, BT-2 균주는 GBY 배지에서 배양되었고, 두 균주의 내독소는 당밀도구배법을 이용하여 정제되어, 주사전자현미경(SEM), 투과전자현미경(TEM)으로 관찰하였다. 형태학적으로, BT-1의 내독소는 사각형 및 납작형이고 크기는 $1.73{\mu}m{\times}0.7{\mu}m$, 그리고 BT-2 내독소는 구형이며 $1.1{\mu}m$, 폭이 $0.9{\mu}m$인 것으로 밝혀졌다. SDS-PACE 방법으로 분석한 결과, BT-1의 분자량은 28 kDa, 21 kDa, 반면에 BT-2의 분자량은 50 kDa, 35 kDa, 22 kDal으로 밴드가 형성되었다. 이들의 결정체는 알칼리 완충용액 내에서 시간에 지나감에 따라서 점차로 용해되었으며 3시간 후에는 거의 완전하게 용해되었다. 이 결과들을 통하여 BT1과 BT-2 결정체의 내독소가 곤충의 중장 내에서 시간이 흐름에 따라 비활성 상태에서 활성상태로 전환되는 것으로 보여진다.

Synthesis, Self-assembly, and Catalytic Activity of 1H-Imidazole Amphiphiles

  • We prepared polycatenar 1H-imidazole amphiphiles having a structure in which a 1H-imidazole head was connected through a benzene ring to a pheny group having two or three oligo(ethylene glycol) chains and studied their supramolecular assembly by fluorescence spectroscopy, transmission electron microscopy (TEM) and atomic force microscopy (AFM). When the aqueous solutions of the amphiphiles ($5{\times}10^{-5}M{\sim}10^{-3}M$) were deposited onto a carbon-coated copper grid and dried, twisted structures with diameters of ~200-300 nm were imaged by TEM and AFM. We presume that the structures comprised a chain of the amphiphile dimers formed via successive hydrogen bonding between the 1H of the imidazole group and 3N of the neighboring one. In a solution of pH 4, entangled fibers with diameters of several nanometers were observed by TEM. In a pH 10 solution, film-like aggregates formed exclusively. The 1H-imidazole amphiphiles hydrolyzed tetraethoxysilane to induce gelation to form fibrous and spherical silica structures at neutral pH in aqueous solutions. No silica was formed when imidazole was used instead of the amphiphiles, suggesting that the selfassembled aggregates of the amphiphiles were responsible for the gelation.

  • 이상과 같은 실험결과를 다음과 같은 요약할 수 있다. 1) $Hg_{1-x}\;Tl_{x}\;Ba_{2}(Ca_{0.86}\;Sr_{0.14})_{2}\;Cu_{3}\;O_{8+\delta}$의 구조는 Hg-1223과 같은 구조를 이루며 Hg 위치에 T1이 치환되어 있음을 알 수 있다. 2) 격자상수는 탈륨이 증가할수록 감소하는 경향을 나타냈으나 x=0.25 시료에서는 현저히 감소됨을 보였다. 3) x=0.25 시료에서는 적층 결함 1234구조가 1223구조와 intergrowth함을 보였고 불순물 상이 다소 나타났다. 4) 탈륨이 증가할 수록 보다 치밀하고 결함이 없는 안정된 조직을 보였다. 5) 모든 시료가 관찰 도중 전자빔에 대하여 손상되었으나 탈륨의 양이 증가할 수록 손상에 대한 저항이 커져 보다 안정된 상을 보였다.

복행실패로 발생한 CFIT사고의 공통요인 및 외항사 복행게이트 운영 실태에 대한 연구 (한국 대표적 CFIT사고의 TEM 분석을 중심으로) (The Study on Common Factors of Typical CFIT Accident with Go-around Failure and Go-around Gate Operation of Foreign Carriers (An Analysis of Korean CFIT Accidents through TEM))

  • There have been CFIT(Controlled Flight Into Terrain) accidents that can be prevented if the crew executed go-around. This study is to analyse the common factors of three typical CFIT accidents of Korea in TEM(threat and error management) frame, and the examples of go-around gate and the countermeasures of eight airlines through the survey facilitating go-around to prevent CFIT. The common factors found in three typical CFIT accidents occurred in Korea or by Korean carriers turned out to be in mountainous terrain, in bad weather while in non-precision approach or circling approach by captain as PF(Pilot Flying) when crew make monitoring errors and communication errors. It also turned out that the crew in all three typical tragic CFIT accidents did not execute go-around in unstabilized approaches. The captains did not respond immediately when first officers advised them to go-around until it is too late. Seven out of eight Airlines answered that they use stabilized approach height as 1,000 feet to be stabilized earlier to have more safety margin by enhancing go-around gate regardless of the weather to prevent CFIT in the survey.

몬모릴로나이트를 이용한 열방성 액정 폴리에스테르의 나노복합재료 (Nanocomposites Based on Montmorillonite and Thermotropic Liquid Crystalline Polyester)

  • 히드로퀴논과 p-히드록시 벤조산, 그리고 1,6-디브로모헥산을 이용하여 유연한 알킬기를 주사슬에 가지는 열방성 액정 고분자(TLCP)를 얻었다. TLCP의 녹음 전이온도 이상에서 도데실 암모니움-몬모릴로나이트(C$_{12}$-MMT)를 첨가하여 나노복합재료를 만들었다. $C_{12}$-MMT를 TLCP에 대해 1 wt%만 섞어도 액정성은 파괴되었으며, 그 이상의 $C_{12}$-MMT양에 대해서도 마찬가지였다. 첨가된 $C_{12}$-MMT중 일부는 TLCP에 잘 분산되었으나, 일부는 뭉쳐진 형태로 존재하였다. 합성된 나노복합재료의 열적 성질과 몰폴로지는 시차주사 열분석기 (DSC), 열중량 분석기 (TGA), 편광 현미경, 그리고 전자 현미경 (SEM, TEM) 등을 이용하여 분석하였다.다.

Viola속 식물의 원형질체 및 융합세포의 전자현미경 관찰 (Electron Microscopic Observations of Protoplast and Fusion Cell of Viola Species)

  • To obtain a basic information on the development of Genus Viola, ultrastructure and electrofusion process between the two protoplasts from wild Viola callus cells and pansy mesophyll cells were observed with a scanning electron microscopy(SEM) and transmission electron microscopy(TEM). In the ultrastructural observation of wild viola callus protoplasts and pansy mesophyll protoplasts using SEM, their cell walls were removed completely. A knob-like formation was observed on the enlarge surface of viola callus protoplasts. On the surface of pansy mesophyll protoplasts net-like chloroplasts were observed. In SEM observation of pansy mesophyll protoplasts, chloroplasts devoid of membrane were observed on the surface the protoplasts. Pearl chain was formed by applying AC field of 200 V/cm at 1.0 MHz for 43 sec. The lysis of plasma membranes and fusion process occurred by applying a 1,600 V/cm DC pulse twice for 1 sec. After 1-2 hours of a DC pulse application, it was observed that the two protoplasts were fused completely into one cell. In TEM observation of the fused cell, many small vacuoles were located in the fusion area of the two protoplasts. Indeed, two distinct regions were observed during fusing process; in one region, a nucleus was found, while in the other region, both nucleus and nucleous were found.

우리나라 일부 석면광산 지역에서 ABS를 이용한 석면노출 및 위해성 평가 (Asbestos Exposure and Risk Assessment by ABS(Activity Based Sampling) for Former Asbestos Mining Areas in Korea)

  • Objectives: The aim of this study was to investigate the exposure and risk assessment of residents near asbestos mines in Korea. Methods: To assess asbestos types and airborne concentrations, air monitoring was performed in the neighborhoods of Kwangcheon (KC) and Sinsuk (SS) mines, which were leading South Korean mines in the past. In addition, activity-based-sampling (ABS) of residents' particular activities were conducted in order to estimate the Excess Lifetime Cancer Risks (ELCRs) for the residents. Conclusions: The average concentration of airborne asbestos in KC was 0.0014 f/cc and 0.0015 f/cc by PCM and TEM, respectively. In SS it was equal at 0.0012 f/cc by PCM and TEM. No statistically significant difference was found in the average concentration of airborne asbestos between the two mines. The average asbestos concentration of ABS was 0.0048 f/cc (PCM) and 0.0042 f/cc (TEM) in KC, while it was 0.0137 f/cc (PCM) and 0.0125 f/cc (TEM) in SS. It was found that the average asbestos concentration of ABS in SS was statistically significantly higher than that of KC (p<0.01). The results of ELCRs by scenario in KC showed that the scenarios of bicycle, car, weed control, weed whacking, child playing in the dirt, and physical training fell within $1{\times}0^{-6}-1{\times}10^{-4}$, which is the acceptable range of ELCR. The scenarios of motorcycle, walker, digging, and field sweeping, however, exceeded the acceptable range. In SS, only the scenario of car fell within the acceptable range, while all of the other scenarios exceeded the acceptable range.

단일 이온빔 스퍼터링법을 이용한 AIN 박막의 증착 (Deposition of AIN Thin Films by Single Ion Beam Sputtering)

  • Reactive Single Ion Beam Sputtering 방법을 이용하여 AIN박막을 증착하고 물성을 분석하였다. 반응성 가스로 질소 가스 또는 암모니아 가스를 이용하였다. 증착된 AIN박막의 구조적, 화학적, 광학적 물성을 분석하기 위해 XRD, GAXRD, TEM, SEM, XPS, UV/VIS spectrophotometer, FT-IR등을 이용하였다. XRD, GAXRD분석결과에 의하면 증착된 모든 AIN박막은 비정질이었으나 TEM분석결과에서는 비정질 속에 육방정의 AIN미세결정들이 분포해 있었다. 그리고 FT-IR과 XPS분석을 통하여 Al-N결합을 확인하였으며, 화학양론적인 조성이 됨에 따라 UV-VIS spectrophotometery 분석에서 투광성이 증가하며 광학적 밴드갭은 6.2eV까지 증가함을 확인하였다. 또한 단면과 표면 형상관찰에서는, 반응성 가스로 질소 가스나 암모니아 가스에 관계없이, 결정입계가 전혀 관찰되지 않는 아주 평활한 현상이었으며 굴절율은 1.6~1.7의 값을 갖는다.
