The penetration of outside material into skin is not easy. It is since the skin, which is a very hard barrier, protects the body against outside chemical and physical stimulation. Microneedle system which can help improve drug penetration into skin is advancing variously in transdermal drug delivery system (TDDS) in the field of skin care. After inserting microneedle into skin by using electrical or artificial forces, it makes microhole and drug penetration easily and induces natural skin rejuvenation. Diffusion and penetration of drug by optical and electrical force of microneedle is better for fast and effective TDDS. This is more developed than the traditional method such as the manual stamp, roller, and meso gun. The drug absorbed into dermal layer by microneedle helps revive and repair damaged skin. In the future, utilization of microneedle for skin care will progress constantly because of its human-friendly biodegradable materials and the development of the no pain microneedle.
점착제는 경피흡수제제(transdermal drug delivery system, TDDS)의 중요한 구성요소 중의 하나이다. TDDS용 점착제는 일반적인 점착제의 역할 외에도 부착되는 피부에 적합해야 하고, 이것에 포함되는 약물 및 첨가제들과 양립하면서 약물의 전달을 효과적으로 지속해야한다. 본 총설에서는 흔히 사용되는 TDDS용 점착제인 polyisobutylenes, polyacrylates, silicones와 최근에 개발된 제품들을 소개한다.
The purpose of this study was to develop transdermal drug delivery system (TDDS) for the combination of physostigmine and procyclidine. The effects of various pressure sensitive adhesives (PSA) on the percutaneous absorption of procyclidine across hairless mouse skin were evaluated to select an appropriate PSA. In addition, the influences of various vehicles on the percutaneous absorption of procyclidine from PSA matrix across hairless mouse skin were evaluated using flow-through diffusion cell system at $37^{\circ}C$. Physostigmine did not have any influence on the permeation rate of procyclidine. The flux of procyclidine was the highest in silicone and PIB and was relatively lower in SIS, Acryl, and SBS adhesive matrices, however, their use was limited by the crystallization of the drug in the matrix. Among acrylic adhesives, the permeability of procyclidine was the highest from poly (ethylene oxide) grafted acrylic adhesive. Some enhancers show different enhancing effect depending on the drug, however, many of the tested enhancers showed enhancing effect for the permeation of both procyclidine and physostigmine to some extent. $Crovol^{\circledR}$ EP 40 showed the highest enhancing effect on the permeation of both compounds. The size of TDDS to provide required permeation rate was estimated to be $35\;cm^2$ based on available information.
최근 산화철 나노입자는 외부의 자기장에 반응하는 자성의 특성과 생체적합성이 뛰어나 약물전달시스템(drug delivery system, DDS)에 관한 많은 연구의 소재로 사용되어져 왔다. 본 연구에서는 마이크로니들(microneedles, MNs)의 매트릭스 물질로 HA (hyaluronic acid)와 CMC (carboxy methyl cellulose)를 이용하여 SIONs (superparamagnetic iron oxide nanoparticles)이 함유된 HA-SMNs (hyaluronic acid-superparamagnetic microneedles)와 CMC-SMNs (carboxy methyl cellulose-superparamagnetic microneedles)를 제조하였으며, SEM (scanning electron microscopic), SQUD-VSM (superconducting quantum interference device-vibrating sample magnetometer), FMMD (frequency mixing magnetic detection), 고분자 및 바이오 멤브레인을 이용하여 SMNs의 다양한 특성을 조사하였다. SQUID-VSM 측정 결과 SIONs이 포함된 HA-SMNs와 CMC-SMNs에서 초상자성의 특성이 나타났으며, FMMD 측정에서는 SIONs 농도가 증가함에 따라 신호 강도에 변화가 확인되었다. 또한 SMNs의 바이오 막을 통한 HA-SMNs와 CMC-SMNs의 투과도 분석에서는 각각 평균 92.5%와 98.5%의 피부 투과율이 조사되었다. 이러한 결과를 통해 SMNs 제형은 경피약물전달시스템(transdermal drug delivery system, TDDS) 및 MR(magnetic mesonance) molecular imaging 분야의 전달소재로 활용될 수 있을 것으로 기대한다.
The purpose of this study was to investigate the feasibility of developing transdermal drug delivery system (TDDS) for fentanyl used for the management of chronic cancer pain. The effect of type of pressure sensitive adhesive on the permeation of fentanyl from polyisobutylene (PIB), silicone and acrylic adhesive was evaluated. Due to the good adhesive force and relatively steady flux for 3 days, both acrylic and PIB adhesives were chosen for further study. The permeation rate of fentanyl was the highest from acrylic adhesive with hydroxyl functional group. Permeation rate increased linearly as the concentration of fentanyl in acrylic adhesive was increased from 2.5% to 10%. In case of PIB adhesive, crystals of fentanyl were developed above 5% drug load. $Crovol^{(R)}$ A40, $Crovol^{(R)}$ PK40 and Plurol $oleique^{(R)}$ provided higher flux of fentanyl.
본 연구의 목적은 클로로겐산이 함유된 피부 패치를 피부에 부착하여 체지방과 체중에 미치는 변화를 규명하여, 유용성을 평가하는 것이다. 시험군 8명, 대조군 8명으로 총 16명의 대상자가 연구에 참여하였고, 연구 기간은 2020년 08월 03일부터 2020년 08월 31일까지 4주간 실시하였다. 군-간 효과검정 결과 체지방의 변화는 전체적으로 유의한 차이가 없었으나(F=4.38, p<.055), 1주 후와 4주 후 검사에서는 의미있는 변화(p<.001)가 있었다. 또한, 체중은 유의미한 차이로 시험군이 대조군보다 체중 감소가 큰 것으로 나타났다(F=9.43, p<.008). 이번 연구의 결과는 비만 관련 연구에 패치를 적용한 예비연구로써 의미가 있다고 할 수 있으며, 연구의 결과가 향후 연구될 피부 패치 연구의 기초 자료로 활용될 수 있을 것으로 판단된다.
본 연구는 allbanggae starch (ABS), polyvinyl alcohol (PVA), alginic acid (SA)를 이용하여 naproxen (NP) 각인 starch 기반 다층 바이오소재를 제조하고, 물리화학적 특성과 약물 방출 제어 효과를 조사하였다. 또한, FE-SEM과 FT-IR 분석에 의해 제조한 다층 바이오소재의 특성을 조사하였다. 약물 방출 제어 효과와 경피 약물 전달 시스템의 적용 가능성을 확인하기 위해 NP 각인 다층 바이오소재로부터 NP 방출 특성을 사람의 체온인 36.5 ℃에서 다양한 pH buffer solution과 인공 피부를 이용하여 확인하였다. NP는 낮은 pH보다 높은 pH에서 1.3배 더 빠른 방출을 나타냈고, single-layer 바이오소재에서보다 multi-layer 바이오소재에서 약 4.0배 느린 방출이 일어남을 확인하였다. 인공 피부 방출에서도 동일하게 single-layer 바이오소재보다 multi-layer 바이오소재에서 약 4.0배 더 느린 약물 방출 결과를 나타내었다. 또한, double-layer와 triple-layer 바이오소재 모두 12시간 동안 지속적으로 NP가 방출되었음을 확인 하였다. NP 방출 mechanism을 규명하기 위해 수학적 모델링에 적용하여 비교했을 때, pH buffer solution에서의 방출은 Fickian diffusion mechanism, 인공 피부 방출은 empirical mechanism에 적합한 것을 확인하였다.
