  • The purpose of this study was to analyze the effects of aerobic exercise and black garlic intake on blood lipids, lipid peroxidation and BAP in rats. The subjects for the study were 25 male Sprague-Dawley rats composed of the control group (CG, n=6), exercise group (EG, n=6), which trained for 4 weeks (30-60 min/day, speed at 15 m/min), black garlic intake group (BGG, n=7), and exercise with black garlic intake group (EBGG, n=6). Blood lipids, lipid peroxidation, and BAP were measured in all the subjects after the end of the 4 week treadmill exercise period. The findings of this study were as follows; TC and TG were significantly lower in the EBGG compared to other groups, while there were no significant findings for HDL-C and LDL-C levels. TBARS was significantly lower in the EBGG compared to the CG, and the BAP from the EBGG and EG were significantly higher than in the CG and BGG after 4 weeks of treadmill exercise. Therefore, aerobic exercise with black garlic intake was effective in improving TC, TG, TBARS, and BAP. Consequently, aerobic exercise with black garlic intake will be effective for activating antioxidant defenses and decreasing cardiovascular diseases.

  • Anti-hyperlipidemic effect of red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE), which are used frequently in China and Korea as herbal medicines to treat arteriosclerosis, were investigated. Treatments of RG and CF significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. Ginsenoside Rg3 and ursolic acid, the main constituents of RG and CF, respectively, also reduced TG and TC levels in hyperlipidemic mice. RG and CF significantly lowered the high blood TG and TC levels and body and epididymal mass weights induced by long-term feeding of a high-fat diet and increased the high-fat diet-induced decrease in blood HDL cholesterol levels. RG and Rg3 reduced the blood TC levels more than CF and ursolic acid. However, blood TG level were reduced by CF and ursolic acid more than RG and Rg3. RG, CF, and their constituents also inhibited pancreatic lipase and HMG-CoA reductase activities. The most potent inhibitor was Rg3. These findings suggest that RG and CF may be suitable for the therapies of hypercholesterolemia and triglyceridemia, respectively.

  • Abnormal regulation of glucose and impaired lipid metabolism that result from a defective or deficient insulin are the key etiological factor in type 2 diabetes mellitus (T2DM). The our study evaluated the beneficial effect of diet supplementation with Lentinus edodes on hyperglycemia and lipid metabolism in normal and type 2 diabetic rats. The animals were divided into 4 groups: group I(control) rats were fed standard diet (12% of calories as fat); group II (T2DM) rats were fed HFD (40% of calories as fat) for 2 weeks and then injected with STZ (50 mg/kg); group III and group IV rats were continually fed a diet containing 1% and 10% Lentinus edodes for 4 weeks after T2DM induction, respectively. After 4 weeks we determined biochemical parameters such as glucose, insulin concentration, serum total cholesterol (TC), triglycerides (TG), high-density lipoprotein (HDL), and glycosylated hemoglobin (HbA1c) concentration were also measured. There was a significant reduction in serum TC and TG in the Lentinus edodes supplement groups. The Lentinus edodes diet supplementation were found to have a potent lipid metabolism improvement as well as LDL concentration decreased and HDL concentration was increased. Concentrations of blood glucose and HbA1c in the experimental groups II were significantly decreased after 4 weeks compared with the control group. The Lentinus edodes diet supplementation is useful in regulating the glucose level, improves the insulin, HbA1c, serum lipid metabolism in experimental diabetic rats. We suggest that Lentinus edodes supplementation may have the control effects of diabetes mellitus by improving blood glucose control and lipid metabolism.

  • This study aimed to determine the blood lipid profiles, fatty acid composition, and lipogenic enzyme activities in rat adipose tissues as affected by the Angus beef fat (ABF) and Hanwoo beef fat (HBF) containing high oleic acid (OA) content. We assigned 60 Sprague Dawley rats with a mean bodyweight of 249 ± 3.04 g to three groups (n = 20 each) to receive diets containing 7% coconut oil (CON), 7% ABF, or 7% HBF. The OA content was highest in the HBF (45.23%) followed by ABF (39.51%) and CON (6.10%). The final body weight of the HBF-fed group was significantly increased, probably due to increased feed intake, indicating the palatability of the diet. The HBF and ABF significantly increased high-density lipoprotein cholesterol (HDL-C), decreased triglyceride (TG) and total cholesterol (TC) levels, and also tended to attenuate glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) levels in the bloodstream of the rats compared to CON. As compared to CON, lauric, myristic, and palmitic acids were significantly lower, and those of OA and α-linolenic acid (ALA) were significantly higher in the adipose tissues of HBF and ABF-fed groups. The HBF and ABF also reduced lipogenesis as induced by depleted fatty acid synthase (FAS) activity in rat adipose tissues. Nevertheless, between the two fats, HBF showed high feed intake due to its high palatability but reduced lipogenic enzyme activity, specifically that of FAS, and increased HDL-C, decreased TC and TG levels in the bloodstream, reduced saturated fatty acids (SFA), and increased oleic and ALA contents in rat adipose tissues indicating that HBF consumption does not pose significant risks of cardiovascular disease.

  • In this study, we investigated the anti-obese activity of sevennight extract in high fat diet-ob/ob C57BL/6J mice by oral administered for 1 weeks. Mate water extract (MATEWi) was found to lower whole body and epididymal adipose tissue weights and lowered plasma levels of triglyceride (TG) and total cholesterol (TC), HDL and LDL, compared to those in high fat fed ob/ob group. These results suggest that Mate extract ameliorates obesity through activation of lipogenic enzymes and FA oxidation resulting from phosphorylation of AKT and GSK-$3{\beta}$, and could be developed as a potential therapeutic agent for obese mices.

  • This study was performed to investigate the relationships between obese index and major risk factors of cardiovascular disease in CVD patients. Anthropometry, food intake patterns, blood pressure, serum lipids and hematological values were analyzed in 550 patients from June, 1991 to August, 1992. The results are as follows: Total food intakes, especially total vegetable food and alcohol intakes were significantly related with obese index, so that the heavier patients tended to consume more carbohydrate and less fat. The blood pressure were observed to have positive correlation with obese index in man but not in women. However, both in men and women, blood levels of TG, TC, and VLDL tended to be related to obese index positively, while HDL and HDL-C were shown to be oppositive. So, it seems that as the degree of obesity is higher the blood pressure and blood lipids patterns worse. Therefore, these results suggest that obesity is one of the major risk factors of CVD, and for prevention & treatment of the disease, controlling body weight by nutritionally adequate diet along with alcohol restriction is necessary.

  • Objectives : AMP-activated protein kinase (AMPK) is a key metabolic regulator that reduces lipogenesis. AMPK is mainly activated via phosphorylation of liver kinase B (LKB) 1 under energy stress. Here, we highlighted the anti-obesity effect and underlying mechanism of Coix lacryma-jobi var. mayuen Stapf sprout water extract (CSW) sprout extract in connection with the LKB1/AMPK signaling pathway. Methods : C57BL/6 mice (20~25 g) fed HFD to induce obesity and at the same time administered CSW 100 mg/kg (CSWL; (CSWL; CSW low concentration) or CSW 200 mg/kg (CSWH; CSW high concentration) or Garcinia extract (Garcinia) 200 mg/kg orally for 6 weeks. Body weight and food intake were measured at the same time each day. After 6 weeks of CSW administration, liver tissue and serum were obtained through an autopsy. After the end of the experiment, biochemical analysis (triglycerides (TG), total cholesterol (TC), HDL-cholesterol, and LDL-cholesterol) was performed on the serum. And then, protein levels related to TG and TC synthesis were measured through western blot analysis in liver tissue. Results : As a result, serum TG, TC, and LDL-cholesterol levels were significantly increased in the control group and significantly decreased in the CSW administration group. On the other hand, the HDL-cholesterol level was increased in the CSW-administered group. And as a result of Western blot analysis, CSW significantly increased the phosphorylation of LKB1 & AMPK, and remarkably decreased the expression of factors related to TG and TC synthesis. Conclusions : Our findings suggest that CSW influences the TG and TC synthesis to positively affect HFD-induced obesity in C57BL/6 mice.

  • The purpose of this study was to investigate the distribution patterns of serum triglyceride and cholesterol levels and the influence of environmental and dietary factors in 201 healthy male subjects. The subjects were found to be 53.7% for the normal, 15.4% for the TC, 17.4% for TG and 13.5% for TC.TG group, respectively. Among general characteristics, smoking significantly affected serum lipid levels of subjects (p < 0.05). Among anthropometric factors, body weight, BMI and PIBW significantly influenced the blood lipid patterns. Body weight of TG group was significantly higher than that of normal group (p < 0.05). BMI and PIBW of TG group and TC.TG group were significantly higher than those of normal group (p < 0.05) . In addition, diastolic blood pressure of TG group was significantly higher than that of normal group (p < 0.05). For the food intake, average intake of fruit was significantly higher in normal group than that of TG group (p < 0.05). For nutrient intake, calcium intake was significantly higher in TC group than those of normal and TC.TG group (p <0.05). Intakes of vitamin $B_2$ and C were significantly higher in TC group than those of in TG group (p < 0.05). Serum HDL-cholesterol level was negatively related to meat, poultry, and their products (p <0.05), but serum total cholesterol level positively related to animal lipids (p < 0.05). The results of this study suggest that it is crucial for maintaining adequate serum lipid levels to reduce smoking, blood pressure and to lose body weight. As far as food intake concerned, fruit intake was important in maintaining serum triglyceride level normal, while animal food intake affected serum total cholesterol level adversely. • Lipid-lowering effects of the inulin have been demonstrated in animal, yet attempts to reproduce similar effects in humans have generated conflicting results. In this study, the lipid-lowering potential of inulin and especially its effect on bile acid and neutral sterol excretion were investigated in Korean postmenopausal women. Nineteen postmenopausal women were randomly divided into two groups in a double-blind parallel design and consumed one of two supplements for 12 weeks; placebo of 8g maltodextrins/sucrose mixture (placebo group) or 8g inulin (inulin group). There were no significant changes in body weight during the supplementation period in either inulin or placebo group. Dietary consumption of animal fat in both group tended to decrease after 12 weeks of experiment. Intake of cholesterol was lower in placebo group, whereas the decrease of cholesterol intake in inulin group did not reach statistical significance after 12 weeks. The levels of serum total cholesterol (TC) and LDL-cholesterol (LDL-C) were significantly decreased in both placebo (p<0.05) and inulin group (p<0.01) after supplementation for 12 weeks compared with the baseline. The levels of serum triglyceride (TG) and HDL-cholesterol (HDL-C) were not significantly affected by inulin supplements, but atherogenic index (AI) and LDL-C/HDL-C ratio (LHR) as a predictor for coronary heart disease were improved (p<0.01) significantly after inulin supplementation. Therefore, inulin supplement may decrease the risk of cardiovascular disease via improving blood cholesterol level. Fecal weight and pH were not changed after 12 weeks of supplementation. There were no statistically significant changes for the fecal short-chain fatty acids (SCFAs). In inulin group, fecal deoxycholic acid (DCA) was significantly lowered compared with the baseline (p<0.05) whereas other bile acids were not changed. During the 12 weeks of intervention, no differences were found in fecal excretion of neutral sterol in the two groups. In summary, dietary inulin decreases serum TC, LDL-C, AI, LHR and lowers excretion of fecal DCA in the Korean postmenopausal women. These results support the use of inulin for reducing risk factors for hyperlipidemic postmenopausal women. However, the exact mechanism (s) responsible for the blood lipid lowering action of inulin including altered fecal bile acid remain to be elucidated.

  • The purpose of this study is to compare the relative risk of metabolic syndrome (MS) in middle aged adults with different body weights. 155 subjects living in urban Beijing were recruited from 24 neighborhood committees of urban Beijing. They were divided into normal weight, overweight and obese groups according to their BMIs. The general information of the subjects was collected using an interview-administered questionnaire. Standard procedure was followed to measure subject's weight, height and waist. Biochemical parameters (total cholesterol (TC), low- and highdensity lipoprotein cholesterol (LDL-C ; HDL-C), triglyceride (TG), and fasting glucose) and blood pressure were also determined. The results indicated that the systolic and diastolic blood pressure, HDL-C of obese group was lower than that of the normal weight group. Fasting glucose of obese males was significantly higher than that of normal weight males. No significant difference of fasting glucose was found among female groups. No significant difference of TG was found among male groups, while TG of overweight and obese females was both significantly higher than normal weight females. There was no significant difference of TC and LDL-C among normal weight, overweight and obese groups in both males and females. The MS rate of obese males was significantly higher than the normal weight and overweight males, as was the female. The relative risk of MS in obese group was about 11 times higher (OR=11.249, $95\%CI$ = 3.812 - 33.191) than the normal weight group after adjusting for age, gender, smoking, drinking, family economic level and education status. It is concluded that obesity contributed to lower HDL-C, hypertriglyceride, hypertension and MS after controlling the effects of age, gender, socioeconomic status, alcohol drinking and smoking. Obese individuals have a higher risk of having MS than their normal weight counterparts.
