Toxicity of Bis(tri-n-butyltin)oxide (TBTO) on the Organ Structure of the Flounder, Paralichthys olivaceus (넙치(Paralichthys olivaceus)의 기관계 구조에 미치는 bis(tri-n-butyltin)oxide (TBTO) 독성)

  • The histopathologieal effects of his(tri-n-butyltin)oxide (TBTO) on the flounder, Paralichthys otivaceus were examined by means of histological methods. The experimental fishes were exposed to 0.17, 0.36, 0.60, 3.20, 6.30, 12.50 ${\mu}g \;L^{-1}$TBTO concentrations for 42 days. Histopathological change of the fish exposed to TBTO is dependent on the exposure duration and concentration. In the lower concentrations early histological changes included activated mucous cells and chloride cells, capillary hyperemia and epithelial hyperplasia in the gill; hepatocyte activation, degeneration of bile duct and pancyeatic zymogen reduction in the hepatopancreas; and capillary hyperemia, appearance of eosinophilic cell and melano-macrophagocytes in the kidney. At the higher concentrations histological changes of dysfunctionality included epithelial lifting and deformation of the lamellae in the gill; pycnosis and cytoplasmic degeneration of hepatocyte; pycnosis of haemopoietic cell and deformation of renal tubules and glomerulus in the kidney. It is indicated that TBTO induced histopathological changes in the fish as other aquatic pollutants.

The Effect of TBT Toxicity on Survival and Growth of Olive Flounder, Paralichthys olivaceus (넙치 생존과 성장에 미치는 TBT의 독성)

  • Toxic effect of TBTO on larva flounder was studied by the use of a food-chain system in which indirect toxicity from seawater or/and plankton can be measured, Under the treatment of 0.5 ng/L TBTO, the combined effect of diets ( Chlorella and rotifer) and seawater was significant by synergism, although the sole effect from TBTO treated diets or seawater was equally not, The values of $LT_{50}$ from results of acute-toxicity experiments for juvenile flounder were estimated to be 230.0, 48.0, 24.0, 14.6, 9.3 5.5 3.0 and 1.7 hr at 1, 10, 25, 50, 100, 250, 500 and 1000 ng/L of TBTO, respectively, and $96hr-LC_{50}$ was 3.5 ng/L. From the above results, the experiments for chronic toxicity of TBTO was executed at the concentration range of $1\sim10 ng/L$. In long-term experiments for four months, the weight and the total length of the juvenile flounder in all TBTO treated experiments slowly increased when compared to control. No significant differences in the growth and survival of the juvenile flounder were found in the treatment of 1 ng/L TBTO(P>0.05), But, $90\%$ of the juvenile flounder died in 20 days under TBTO treated seawater at both concentrations of 5 and 10 ng/L, The TBTO treated on seawater was more effective and significantly different in the growth and survival of the juvenile flounder when compare with that on artificial diets (P<0.05). From the all results, TBTO should be regulated below 5 ng/L in a coast.

Changes of Inorganic Matter and Enzyme Activity in the Hemolymph of Oyster, Crassostrea gigas Exposed to TBTO (TBTO의 노출에 따른 참굴, Crassostrea gigas의 hemolymph내 무기성분 및 효소활성의 변화)

  • The purpose of this study is to find out the effects of various bis(tri-n-butyltin)oxide (TBTO) on changes of inorganic matter and enzyme activity in the hemolymph of Pacific Oyster, Crassostrea gigas. Oyster were exposed to 5, 10, 20, 50, 80and $100{\mu}g$/L of TBTO for 20 days. Survival rate of the oyster wass significantly affected by $\geq80{\mu}g$/L TBTO concentration at 10 days, while the diminution of survival was found at 20 days with a lower concentration of $\geq50{\mu}g$/L TBTO. Calcium concentration in the hemolymph increased significantly after 20 days at the TBTO concentration $20{\mu}g$/L. However no change of magnesium and inorganic phosphate in the hemolymph was showed. A significant increment of GOT activities in the hemolymph was observed after 20 days at more than $20{\mu}g$/L TBTO concentraion, without typical changes of GPT activities. These results indicate that oysters can be affected by TBTO in terms of calcium concentration and GOT activity in the hemolymph when they were exposed to the TBTO concentration $20{\mu}g$/L.

Acute Toxicity of TBT Influencing on the Production of Coastal Olive Flounder (연안 어중의 넙치 생산성에 영향을 미치는 TBT의 급성 독성)

  • The toxic effect of TBTO on Chlorella and Rotifer was observed. The value of 48hr-LC50 for Chlorella (3.3$\mu\textrm{g}$/L) estimated to be almost 500 times as high as that for Rotifer (6.7ng/L). A fertilized egg of olive flounder exposed in an embryo-formation stage was mostly influenced by TBTO toxicity when the fertilized egg at each stage until hatching was exposed to TBTO at the concentrations of 5 to 200ng/L. The values of LT50 were estimated to be 68.0, 41.0, 21.0, 13.0, 7.7 and 4.7 hours at 5, 10, 25, 50, 100 and 200ng/L of TBTO, respectively when the fertilized egg in a morula stage was exposed to TBTO, and the 48hr-LC50 was 8 ng/L. In case of TBTO treatment in an embryo-formation stage, the values of LT50 were 33.0, 12.5, 3.5, 1.3, 0.5 and 0.2 hours at 5, 10, 25, 50, 100 and 200ng/L of TBTO, respectively, and the value of 48hr-LC50 was 4ng/L. The values of LT50 were estimated to be 17.0, 11.0, 6.2, 4.0, 2.6 and 1.7 hours at 5, 10, 25, 50, 100 and 200ng/L of TBTO, respectively when the fertilized egg in a stage just before hatching was exposed to TBTO, and the 48hr-LC50 was below 1ng/L. The percentages of hatching were 46.2, 20.6, 21.9, 20.6 and 13.2% at 25, 50, 100, 250 and 500ng/L of TBTO, respectively when the fertilized egg in the stage just before hatching was exposed to TBTO and the measurement was done at second day after the completion of hatching. However no survival after the completion of hatching was found in all cases. With the treatment of 1, 5 and 10ng/L of TBTO, the percentages of hatching were 80.5, 70.0 and 44.1%, respectively. The percentages of survival until second day after the completion of hatching were 80.0, 63.3 and 9.1%, respectively. The percentages of hatching and survivability after the completion of hatching for the control were 84.5 and 82.5%, respectively.

  • The influence of TBTO (Tributyltin-Oxide) at a series of concentrations (0.52, 1.41 and 3.05 $\mu$g/L) for a period of 3 weeks on estradiol-17$\beta$, testosterone and cortisol levels in male Korean rockfish, Sebastes schlegeli was investigated. Compared to the stable concentration of estradiol-17$\beta$, plasma testosterone level was decreased significantly at the 3rd week after TBTO exposure in plasma of fish (>1.41 $\mu$g/L). The most profound physiological variation in TBTO exposed fish was a dramatic increase in plasma cortisol level at 3rd week. Conclusively, TBTO exposure caused increase level of cortisol as well as alteration of testosterone in the Korean rockfish. These results suggest that TBTO at environmentally relevant (nanomolar) concentrations disrupt endocrine secretions.

  • The purpose of this study finds out the effect of red ginseng extract (1.0g/kg) on TBTO (10, 20, and 40mg/kg) which poisons against some organs like thyroids gland, liver, kidney, testis, ovary, sex hormone activity of rats are examined by gastric tubing for 3 weeks. The weight of each organ in treated group were increased, especially liver in female and those of testis in males were significantly increased at 10, 20 and 40mg/kg (P<0.05, P<0.01). In case of sex hormone activity of each sex, the estradiol activity of female and testosterone activity of males were significantly decreased rather than the control group (P<0.05, P<0.01) According to between the TBTO treated group and 10+ rGe group of the testosterone activity each sex were significantly increased (P<0.01).

  • This study find out the effect of red Ginseng extract(1.0g/kg) against TBTO(10, 20 and 40mg/kg) poisoning on each organs, hematological, serum biochemistry in rat for 3 weeks. 1. Each organ weight per body weight ration in treated group, all rats liver were significantly increased. (P<0.05,P<0.01)2. Iin TBTO treated group, the hematological parameters such as WBC, RBC, Hgb, Hct, PLt, MCV, MCH and MCHC in serum were remarkably decreased in comparison to that of control group. Only added red Ginseng extract group were slightly decreased but not significanted. 3. In TBTO treated group, the biochemical parameters such as AST, ALT in serum were remarkable elevated in comparison to that control group (P>0.05).4. In TBTO treated group, the activity of Triglyceride in serum of male rat were significantly increased in comparison to that of control group (P<0.05, P<0.01).5. In case of B.U.N activity in treated group(10, 20 and 40 mg/kg) were almost increased in comparison to that of control group, but not added red Ginseng extract.

Effects of Trialkyltin in vitro on the Microsomal Monooxygenase System of Digestive Gland in the Clam, Coelomactra antiquata (유기주석화합물이 명주조개 (Coelomactra antiquata)의 약물대사효소계에 미치는 영향)

  • This study was carried out to measure the in vitro interaction of trialkyltin with the microsomal monooxygenase (MFO) system of the clam, Coelomactra antiquata. Cytochrome P450 (CYP) level and 7-ethonvesorufin-O-deethylase (EROD) activity were invetigated in the microsome isolated from digestive gland of the clam (C. antiquata) exposed to tributyltin chloride (TBTC), bis-tributyltinoxide (TBTO) and triphenyltin chloride (TPTC). The specific contents of CYP in clam microsome exposedto 0.4 mM TBTC, TBTO and TPTC for 20 minutes were decreased 52, 72 and $40\%$, respectively, compared to control group. The EROD activities also were inhibited by exposure to TBTO ($92\%$) and TPTC ($85\%$) except for TBTC, The level of CYP and the EROD actintles were decreased according to the OTC exposure concentrations. The toxic effects on the level of CYP and the EROD activities were in order of TPTC>TBTC>TBTO in this study. The measurement of CYP level and EROD activity could be applied as a biomarker for environmental study.

  • The purpose of this study finds out the effect of red ginseng extract(1.0 g/kg) on bis(tributyltin)oxide(TBTO)(10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kidney, testis, ovary. Serum immunity and sex hormone activity of rats are examined by gastric tubing for 3 weeks. The weights of each in treated group were increased, especially liver in females and those of testis in males were signignificantly increased at 10, 20 and 40 mg/kg (P<0.05, P<0.01). In group treated with TBTO(10, 20 and 40 mg/kg) for females, the serum IgG level was significantly reduced in comparison with control group(P<0.05). The group of females which were poisonned by TBTO and rGe was significantly recovered in TBTO 10 mg/kg + rGe 1.0 g/kg and TBTO 20 mg/kg + rGe 1.0 g/kg. In case of sex hormone activity of each sex, the estradiol activity of females and testosterone activity of males were signignificantly decreased rather than the control group. And control group by bis(tributyltin) oxide and red ginseng extract is just increased estiradiol activity.
