Antioxidant Activity and Chemical Characteristics of Orostachys malacophyllus and Fermented Orostachys malacophyllus (와송과 발효 와송 추출물의 이화학적 특징 및 항산화 활성)

  • Orostachys malacophyllus grow on the old roofing tile or on the rock of mountain and is belong to Crassulaceae family. After air drying for Orostachys malacophyllus (OM), using the mixture of lactic acid bacteria (Lactobacillus bulgaricus, Streptococcus thermophilus, Lactobacillus acidophilus) was fermented (FOM). OM and FOM extracted using water (W), ethanol (E) and methanol (M) and were measured extracts yield, pH and Brix. Extracted OM and FOM were tested by in vitro experimental models of α,α´-diphenyl-β-picrylhydrazyl (DPPH) radical scavenging activity, Fe/Cu reducing power, linoleic acid peroxidation using ferric thiocyanate and thiobarbituric acid (TBA) methods and peroxidation of rat liver homogenate. In addition, the bioactive materials (phenolic compounds, flavonoids and minerals) were measured. The highest phenolic compounds and flavonoids were OME 122.2 mg/100 g and OME 84.0 mg/100 g. OM and FOM′s major minerals were K, Ca and Mg. The highest free radical scavenging activity showed in FOMM (93.9%), OMM (93.4%), FOME (92.1%) and OME (91.9%) at 0.5% additional level. Fe reducing powers were strong in FOME and FOMM and Cu reducing powers were strong in OME and FOMM. Antioxidant activities on lipid peroxidation using rat homogenate as measured by TBARS method showed strong in FOME and on lipid peroxidation of linoleic acid as measured by ferric TBA method showed strong in OME and FOME and measured by ferric thiocyanate showed strong in FOME and FOMM.

An Experimental Study on the Selenium Effects in Lead-exposed Rats (셀레늄이 연폭로 백서에 미치는 영향에 관한 실험적 연구)

  • An experimental study on the selenium effects on toxicity of lead (4ppm intraperitoneal injection) in adult male rats of Spragne-Dawley strain was performed. This study was carried out to reveal the selenium effects concerning treatment and prevention of lead intoxication and differences of the selenium effects according to the administration method between oral and subcutanous. The changes of body weight, liver and kidney weight per body weight, hemoglobin values, hematocrit values, TBA values in serum, and $\delta$-ALAD activity in blood were determined. The results were as follows 1. There were no significant differences among groups about the change of body weight. 2. In the liver weight per body weight, the group of Pb administration after the oral administration of selenium was significantly high, different from the group of Pb administration alone (p < 0.05). 3. There .were no significant differences among groups about the kidney weight per body weight. 4. On the hemoglobin values, the group of Pb administration with selenium was significantly high,different from the group of Pb administration alone (p < 0.05). 5. There were no significant differences among groups about the hematocrit values. 6. On TBA values in serum, all the selenium treated groups were significantly low different from the group of Pb administration alone (p < 0.01). 7. On $\delta$-ALAD activity, all selenium treated groups were significantly high different from the group of Pb adminsitration alone (p < 0.01). Considering from the results of this experiment, selenium seems to reduce Pb toxicity and preventive administration of selenium seems to be also effective. And the difference of selenium effect according to the administration method between oral and subcutaneous could not be found.

Effect of Packaging Method on the Storage Stability of Hair Tail Products (포장방법이 칼치제품의 저장성에 미치는 영향)

  • To improve the individual packaging method and extend the shelf life of hair tail(Trichiurus japonicus), salted an unsalted hair tail chunk (cut in 8-10cm) were packaged in laminated plastic film bag(Nylon/PE: $20{\mu}m,\;12{\times}15cm$) filled with with free-$O_2$ absorber, in vacuum, and stored at 0 and/or $5^{\circ}C$. The other samples were packaged in plastic foam trays, overwraped with oxygen permeable film(control), and stored at same temperature. Volatile basic nitrogen (VBN), trimethylamine (TMA) and viable cell counts (VCC) were progressed with increase of storage time, but thiobarbituric acid (TBA) values decreased gradually after reaching at a maximum peak in 5-15 days. Judging from 4 chemical components, VBN was the most available component in quality judgement of hair tail chunk and its upper limiting content was 29 mg%. Regression equation for shelf life prediction of hair tail chunk with sensory evalution and VBN component was determined.

Quality Characteristics of Dried Squid (Todarodes Pacificus) by Cold Air Drying Process (냉풍건조공정을 이용한 마른오징어의 품질특성)

  • In an attempt to find ways of improving the quality and sanitary state of dried squid, this study compared artificial drying methods i.e. hot air drying and cold air drying with natural drying method. The drying rates of squid were in the order of hot air, cold air and natural drying. However, cold-air drying was slower than natural drying at $7^{\circ}C$. The drying rates increased with increasing drying temperature. When drying temperature was close to the dew point on a dehumidifier surface, which was the case with cold air drying at $7^{\circ}C$, the driving force for dehumidification was lower than under other drying conditions, which resulted in a lower drying rate. There were little color differences between the drying methods and temperature. Squid dried by cold air-drying had better mechanical texture and sensory qualities than with other drying methods. In addition, the cold air drying method maintained superior quality in terms of the contents of amino acids, taurine, EPA, DHA, other fatty acids and a low TBA value than the other drying methods.

Quality and Shelf-life of Vacuum and Aerobic Packed Chevon Patties under Refrigeration

  • Patties were prepared using Barbari male goats meat (age about 4 yrs) and packed in HDPE under vacuum (VP) and aerobically (AP). Packed patties were stored at 4$\pm$1$^{\circ}C$ and evaluated for physico-chemical, microbiological and sensory changes on days 0, 5, 10, 15, 20 and 25. Overall mean water activity ($a_{w}$), moisture (%), fat (%), pH, TBA number and W-B shear force values (kg/$cm^{2}$) of patties were 0.983, 61.93, 18.39, 6.38, 0.150 and 0.86, respectively. Except pH that was significantly higher and TBA number significantly lower in VP patties, treatment had no significant (p>0.05) effect on other physico-chemical traits. However, storage period significantly (p<0.05) affected physico-chemical traits. Moisture (63.79%) and $a_{w}$ (0.985) were significantly (p<0.05) higher on day 25. Patties became firmer on day 20 onwards as indicated by higher W-B shear force. Though packaging method had no significant effect, storage period influenced microbial counts. The standard plate counts (SPC), which were initially log 5.98 CFU/g decreased significantly (p<0.05) on day 10 followed by steady increase and reaching log 4.89 on day 25. Almost similar trend was observed for psychrotrophic bacteria counts. Lactic acid bacteria counts declined as the storage period progressed. Coliforms, and yeast and mould counts were either not detected by the method used or were very low in numbers. All samples of AP patties revealed swollen, greasy and sticky surface with spongy texture on day 20 whereas only some of the VP patties shown such changes on day 20. Results indicated that vacuum packaging had definite advantage in preserving the sensory quality of patties than aerobic packaging but it did not help in extending the shelf-life beyond 15 days.

Antioxidative Effects of Several Compositae Plants (수 종의 국화과 식물의 항산화 쵸과)

  • Aerial parts of Compositae plants were harvested from pastures and crop fields of Sunchon area at vegetative stage to determine their antioxidant activity. Among 13 species, three species Taraxacum officinate, Breea segeta, and Artemisia princeps were selected as potent plant materials. Through measurement by Rancimat and TBARS method, antioxidative effect was the greatest in Taraxacum officinate among plant species. Antioxidative index (AI) investigated by Rancimat showed Siegesbeckia glabrescensi was 1.51 while commonly-used antioxidants, BHT and ascorbic acid were 3.86 and 2.43, respectively. The highest TBARS value was also obtained from Taraxacum officinate(0.18) at 4 weeks after storage, indicating the plant had the strongest antioxidant activity during long time. Even though TBARS value (0.18) in Taraxacum officinate sample showed lower in all plants than two synthetic antioxidants(0.05-0.09), antioxidant activities of the plants were much higher than that of a control (1.01). The results suggest that the plants have antioxidant potential as an alternative natural antioxidant based on natural plant resources.

Fabrication and Characterization of Biphasic Calcium Phosphate Scaffolds with an Unidirectional Macropore Structure Using Tertiary-Butyl Alcohol-Based Freeze-Gel Casting Method (동결-젤 주조 공정 기반 삼차부틸알코올을 이용한 단일방향 기공구조를 가지는 이상인산칼슘 세라믹 지지체의 제조 및 특성평가)

  • Porous biphasic calcium phosphate scaffolds were fabricated by a freeze-gel casting technique using a tertiary-butyl alcohol (TBA)-based slurry. After sintering, unidirectional macropore channels of scaffolds aligned regularly along the TBA ice growth direction were tailored simultaneously with micropores formed in the outer wall of the pore channels. The crystallinity, micro structure, pore configuration, bulk density, and compressive strength for the scaffolds were investigated with X-ray diffractometery, scanning electron microscopy analysis, a water immersion method, and a universal test machine. The results revealed that the sintered porosity and pore size generally resulted in a high solid loading which resulted in low porosity and small pore size, which relatively increased the higher compressive strength. After being sintered at $1100-1300^{\circ}C$, the scaffolds showed an average porosity and compressive strength in the range 35.1-74.9% and 65.1-3.0 MPa, respectively, according to the processing conditions.

A Simple and Accurate Method for Determining Antioxidative Activity

  • Antioxidative activity is an important factor in inhibiting oxidative stress. The usual methods for determining antioxidative activity are time-consuming and cumbersome. They are also indirect processes that use biological material such as brain or liver microsome. This study therefore proposed a new method. Redoxpotential was determined using galvanic cell with or without the addition of various antioxidants or herbal extracts in zinc sulfate solution. The result was compared with the results from the TBA method and the peroxide value from sodium thiosulfate titration. All methods showed significant and dose-dependent enhancement of antioxidative activity by adding ascorbic acid, quercetin, ginseng, or gingko biloba extract. The result of redox potential using galvanic cell showed the smallest standard deviation and took the shortest time among the three methods. Therefore, the antioxidative potential of chemical substances and herbal extracts can be determined simply, directly and accurately in a short period of time using galvanic cell.

Assessment of Allelopathic Potential and Antioxidant Activity of Leaf Extracts from Three Compositae Plants

  • Some Compositae plants are known to contain biologically active substances that are allelopathic to weeds species as well as antioxidant to foods. Aqueous extracts from leaves of 3 plant species, Cirsium japonica, Kalimeris yomena, and Lactuca saliva, were bioassayed against alfalfa (Medicago saliva) to determine their allelopathic effects. The extracts applied on filter paper in bioassay significantly inhibited root growth of alfalfa. Extracts of 20 g dry tissue $\textrm{L}^{-1}$ from Lactuca sativa showed the most inhibitory effect on alfalfa seedling growth and followed by Cirsium japonica and Kalimeris yomena. Oxidative stability by Rancimat method and antioxidant activity by TBA method for the ground samples were the greatest in Lactuca sativa although were less than that of a commonly used antioxidant, 1% ascorbic acid. Antioxidant activity of methanol extracts on storing meat was stably kept for 28days and was excellent compared to control. These results suggest that three Compositae plants have potent allelopathic and antioxidant effects, and that their activities differ, depending on plant species.

Effect of Frying Methods of Chickens on the Physicochemical Properties of Frying Oil and Fried Chickens in the School Foodservice (학교급식에서의 닭튀김 방법에 따른 튀김유와 튀김닭의 이화학적 특성)

  • The purpose of this study was to investigate the effect of frying methods of chickens ; deep-fat frying with raw chickens(A), with pre-cooked chickens(B) on the physicochemical properties of frying oil and fried chickens in the school foodservice. Acid, peroxide and TBA value of frying oil were significantly increased by increasing the number of frying times and those of method A were higher than those of method B. On the contrary, Iodine value was decreased significantly with increasing the number of frying times but there is no significant difference between method A and B. In fatty acids, the content of saturated fatty acid increased while that of unsaturated fatty acid decreased as the frying times increased. Also viscosity, yellowness and redness increased significantly by increasing the number of frying times. The content of vitamin $B_1 $, of fried chicken by the frying methods was not significantly different, but vitamin $B_2 $the method A was higher than by the method B. Ca, P, Na, K and Fe contents of fried chickens by the method A were higher than those by the method B.
